Abstract

An in-line electron hologram of an individual f1.K phage was recorded with a purpose-built low energy electron point source (LEEPS) microscope. Cryo-microscopic methods were employed to prepare the specimen so that a single phage could be presented to the coherent low energy electrons: An aqueous phage suspension was applied to a thin carbon membrane with micro-machined slits. The membrane was rapidly cooled to freeze the remaining water as an amorphous ice sheet, which was then sublimated at low temperatures and pressures to leave individual free-standing phages suspended across slits. An image of a phage particle, depicted as the amplitude of the object wave, was reconstructed numerically from a digitized record of the hologram, obtained using 88eV coherent electrons. The reconstructed image shows a single phage suspended across a slit in a supporting carbon membrane, magnified by a factor of 100,000. The width and shape in the reconstructed image compared well with a TEM image of the same filament. It is thus possible to record and reconstruct electron holograms of an individual phage. The challenge now is to improve the resolution of reconstructed images obtained by this method and to extend these structural studies to other biological molecules

Abstract

Background: To help adapt cardiovascular disease risk prediction approaches to low-income and middle-income countries, WHO has convened an effort to develop, evaluate, and illustrate revised risk models. Here, we report the derivation, validation, and illustration of the revised WHO cardiovascular disease risk prediction charts that have been adapted to the circumstances of 21 global regions. Methods: In this model revision initiative, we derived 10-year risk prediction models for fatal and non-fatal cardiovascular disease (ie, myocardial infarction and stroke) using individual participant data from the Emerging Risk Factors Collaboration. Models included information on age, smoking status, systolic blood pressure, history of diabetes, and total cholesterol. For derivation, we included participants aged 40–80 years without a known baseline history of cardiovascular disease, who were followed up until the first myocardial infarction, fatal coronary heart disease, or stroke event. We recalibrated models using age-specific and sex-specific incidences and risk factor values available from 21 global regions. For external validation, we analysed individual participant data from studies distinct from those used in model derivation. We illustrated models by analysing data on a further 123 743 individuals from surveys in 79 countries collected with the WHO STEPwise Approach to Surveillance. Findings: Our risk model derivation involved 376 177 individuals from 85 cohorts, and 19 333 incident cardiovascular events recorded during 10 years of follow-up. The derived risk prediction models discriminated well in external validation cohorts (19 cohorts, 1 096 061 individuals, 25 950 cardiovascular disease events), with Harrell's C indices ranging from 0·685 (95% CI 0·629–0·741) to 0·833 (0·783–0·882). For a given risk factor profile, we found substantial variation across global regions in the estimated 10-year predicted risk. For example, estimated cardiovascular disease risk for a 60-year-old male smoker without diabetes and with systolic blood pressure of 140 mm Hg and total cholesterol of 5 mmol/L ranged from 11% in Andean Latin America to 30% in central Asia. When applied to data from 79 countries (mostly low-income and middle-income countries), the proportion of individuals aged 40–64 years estimated to be at greater than 20% risk ranged from less than 1% in Uganda to more than 16% in Egypt. Interpretation: We have derived, calibrated, and validated new WHO risk prediction models to estimate cardiovascular disease risk in 21 Global Burden of Disease regions. The widespread use of these models could enhance the accuracy, practicability, and sustainability of efforts to reduce the burden of cardiovascular disease worldwide. Abstract

In French Polynesia, both currently recognized manta ray species, Mobula alfredi and M. birostris, are observed. Despite being an important cultural asset and generating significant economic benefits through manta ray watching tourism, published data on the ecology and threats to these species in the region are scarce. Based on an 18-year dataset of sighting records collected by citizen scientists and during two scientific expeditions, this study provides the first insights into the population characteristics and regional distribution of the two manta ray species in French Polynesia. A total of 1347 manta ray photographs (1337 for M. alfredi and 10 for M. birostris) were examined for the period January 2001-December 2017, with photo-identification techniques leading to the successful identification of 317 individual M. alfredi and 10 individual M. birostris throughout the Society, Tuamotu and Marquesas Islands. We provide the first confirmation of sympatric distribution of both species in the Society Islands. Our results highlight strong and long-term site fidelity of M. alfredi individuals to certain aggregation sites (> 9 years for 16 individuals) and reveal some degree of connectivity between populations, with 10 individuals recorded moving between islands located up to 50 km apart. Analysis of photographs of individuals bearing sub-lethal injuries (n = 68) suggests that M. alfredi are more likely to be injured at inhabited islands (Maupiti or Bora Bora; 75% of all injured individuals) than at uninhabited islands, with 75% of injuries related to boat propeller strikes and fishing gear entanglements. Our findings emphasize the need for further research to allow for a comprehensive evaluation of population structure, size and threats to manta rays in this region.

Abstract

We have studied the dynamic spall process for copper samples in contact with detonating low-performance explosives. When a triangular shaped shock wave from detonation moves through a sample and reflects from the free surface, tension develops immediately, one or more damaged layers can form, and a spall scab can separate from the sample and move ahead of the remaining target material. For dynamic experiments, we used time-resolved velocimetry and x-ray radiography. Soft-recovered samples were analyzed using optical imaging and microscopy. Computer simulations were used to guide experiment design. We observe that for some target thicknesses the spall scab continues to run ahead of the rest of the sample, but for thinner samples, the detonation product gases accelerate the sample enough for it to impact the spall scab several microseconds or more after the initial damage formation. Our data also show signatures in the form of a late-time reshock in the time-resolved data, which support this computational prediction. A primary goal of this research was to study the wave interactions and damage processes for explosives-loaded copper and to look for evidence of this postulated recompression event. We found both experimentally and computationally that we could tailor the magnitude of the initial and recompression shocks by varying the explosive drive and the copper sample thickness; thin samples had a large recompression after spall, whereas thick samples did not recompress at all. Samples that did not recompress had spall scabs that completely separated from the sample, whereas samples with recompression remained intact. This suggests that the hypothesized recompression process closes voids in the damage layer or otherwise halts the spall formation process. This is a somewhat surprising and, in some ways controversial, result, and the one that warrants further research in the shock compression community. [ABSTRACT FROM AUTHOR]

Abstract

Objectives: Increases in electronic media communication (EMC) and decreases in face-to-face peer contact in the evening (FTF) have been thought to explain the recent decline in adolescent substance use (alcohol, tobacco, cannabis). This study addresses this hypothesis, by examining associations between (time trends in) EMC, FTF, and substance use in more than 25 mainly European countries. Methods: Using 2002–2014 data from the international Health Behaviour in School-aged Children (HBSC) study, we ran multilevel logistic regression analyses to investigate the above associations. Results: National declines in substance use were associated with declines in FTF, but not with increases in EMC. At the individual level, both EMC and FTF related positively to substance use. For alcohol and cannabis use, the positive association with EMC was stronger in more recent years. Associations between EMC and substance use varied across countries, but this variation could not be explained by the proportion of young people using EMC within countries. Conclusions: Our research suggests that the decrease in FTF, but not the increase in EMC, plays a role in the recent decrease in adolescent substance use.

Meanwhile, the natural environment, once imagined as a refuge from this kind of technologised hyperactivity, is under threat, in crisis, and on the verge of collapse. While some recent studies have advanced posthumanist understandings of the interconnectedness of these changing social, technological, economic, and ecological spheres, the road to psychic and ecological rehabilitation remains unclear. This paper examines Berlin-based artist Timur Si-Qin’s project New Peace amid these atmospheric conditions. Si-Qin’s work suggests one avenue to recovery might lie in reimagining spirituality through the cool efficiency of corporate branding.

Abstract

Contrasting other European studies, Dutch research has consistently shown that Dutch immigrant adolescents report more externalizing problems than their non-immigrants peers, while they show equally high amounts of internalizing problems. This cross-sectional study advances our understanding of mechanisms explaining the link between immigration and Dutch adolescent mental health, by investigating to what extend SES, perceived discrimination, and social support mediate this association. Data of the Dutch HBSC study (2017) were used, with a sample of 8,190 adolescents (Mage = 13.38, SD = 1.63). Linear Regression analyses revealed that, when compared to their non-immigrant peers, immigrant adolescents reported more externalizing problems, equally high amounts of internalizing problems, remarkably higher levels of perceived discrimination, lower SES and less social support. Perceived discrimination had a stronger positive contribution to externalizing than to internalizing problems. Family affluence and peer support were negatively related to internalizing problems, while education level was negatively associated with externalizing problems. Comparably strong negative associations of parental support with both types of mental health problems were found. These findings stress the need for interventions targeting immigrant adolescents’ perceived discrimination, to reduce their relatively high amounts of externalizing problems. Upcoming research should investigate which predictors explain perceived discrimination.
