153. Allicin Reversed the Process of Frailty in Aging Male Fischer 344 Rats With Osteoporosis.

Abstract

The research and development of pharmaceutical intervention is insufficient for the frail older adults, especially in preclinical stage for the frail individuals with osteoporosis. Garlic exerts an antiosteoporotic effect and its vital component allicin could protect organisms against aging. The present study aimed to investigate the effect of long-term intragastric administration of allicin (low dose of 4 mg·kg-1·d-1; middle dose of 8 mg·kg-1·d-1; high dose of 16 mg·kg-1·d-1) on frailty with osteoporosis in aging male Fischer 344 rats. Frailty was assessed with a 27-item frailty index based on quantifying health-related deficits in adult male rats varied from 13 to 21 months and in control rats from 6 to 9 months. Osteoporosis was appraised by bone mineral density detected by dual-energy X-ray absorptiometry, biomechanical properties measured by a three-point bending test, and bone metabolic analysis using ELISA. Allicin could attenuate frailty index scores by reducing the accumulation of health deficits in aging male Fischer 344 rats. Meanwhile, allicin could protect against senile osteoporosis, and the underlying mechanism may involve in increasing low bone turnover through elevation of both bone formation and bone resorption, and subsequently lead to increase of bone mineral density, contributing to reversing deleterious bone biomechanical features associated with aging. The present study reveals firstly that long-term oral administration with allicin attenuated frailty with osteoporosis during the process of aging, which provides a preclinical evidence for intervention of frailty. Abstract

OBJECTIVE: To evaluate telomerase activity as a marker for the differentiation of benign and malignant ascites. METHODS: Exfoliated cells were harvested from various kinds of ascitic fluid and telomerase activity was determined by TRAP-PCR-ELISA (tartrate-resistant acid phosphatase–polymerase chain reaction–enzyme-linked immunosorbent assay). RESULTS: Telomerase activity could not be detected in the ascites of hepatic cirrhosis or peritoneal tuberculosis but was detected in 29 of 34 (85.2%) cancerous ascites. CONCLUSIONS: Telomerase activity may be present in cancerous ascites and could be used as a marker for the differential diagnosis of benign and malignant ascites.

Abstract

OBJECTIVE: To investigate the effects of As2O3 on the peritoneal metastasis of colon cancer in nude mice. METHODS: BALB/c-nu/nu mice were injected intraperitoneally with Lovo cells and were allocated to five groups at random: control, epirubicin and varying concentrations of As2O3. The length of their survival period and rates of tumorigenesis and ascitic fluid formation were evaluated. RESULTS: Epirubicin and low doses of As2O3 clearly inhibited ascitic fluid formation and extended the survival period of tumor-bearing nude mice. Moderate and large doses of As2O3 were able to eradicate cancer cells through apoptosis. CONCLUSIONS: As2O3 can induce apoptosis in intraperitoneal colon cancer cells, inhibit ascitic fluid formation and extend the survival periods of nude mice to varying degrees.

In the paper, we studied and designed an optimization algorithm for bilevel programming problems based on Electromagnetism-like mechanism. The experiment results showed that the proposed algorithm is feasible and more efficient that existing algorithms based on evolutionary mechanism.

Abstract

Abstract: There is a great challenge to overcome the limitation of tissue penetration depth, while maximizing the benefit of light‐triggered biochemical cascades in a well‐defined mode simultaneously. Here, a new method of near‐infrared (NIR) light‐triggered release of nitric oxide (NO) by developing upconversion nanoparticles (UCNPs)‐based conjugate chemistry is reported. As the key nanotransducer in the design, core–shell‐structured UCNPs are encapsulated with a layer of SiO2 and then covalently linked with a potent NO‐releasing donor (S‐nitroso‐N‐acetyl‐ dl‐penicillamine, SNAP). It is featured with highly localized breakage of chemical bonds of SNAP molecules by NIR–UV upconversion, enabling simultaneous NO release in a light dosage‐dependent manner. The biological effects of NO releasing are demonstrated by cellular imaging and inhibition of platelet aggregation from blood samples. This work provides a flexible and robust platform to generate cell‐signaling gas molecules trigged by NIR laser with deep tissue penetration. [ABSTRACT FROM AUTHOR]

180. Genetic transformation of the endangered Tibetan medicinal plant Przewalskia tanguticaMaxim and alkaloid production profiling revealed by HPLC

Abstract

To achieve a high yield of tropane alkaloids (TA) and exploit the alpine plant sustainably, an optimized protocol for induction and establishment of hairy roots culture of Prezwalskia tanguticaMaxim was developed through selection of appropriate Agrobacteriumstrain and the explant type. The hypocotyl is more readily facile to induce the HR than the cotyledon is when infected with the three different agrobacterium strains. MUS440 has an efficiency (of up to 20%), whereas the ATCC10060 (A4) can induce HR on both types of explants with the highest frequency (33.33%), root length (21.17 ± 2.84 cm), and root number (10.83 ± 1.43) per explant than the other strains. The highest HR production resulted from using hypocotyl as explants. Independent transformed HR was able to grow vigorously and to propagate on a no-hormone 1/2MS liquid medium. The presence of pRi rolB gene in transformation of HR was confirmed by PCR amplification. In the liquid medium, the HR growth curve appeared to be “S” shaped, and ADB had increased to 4.633 g/l. Moreover, HPLC analysis showed that HR lines have an extraordinary ability to produce atropine (229.88 mg/100 g), anisodine (4.09 mg/100 g), anisodamine (12.85 mg/100 g), and scopolamine (10.69 mg/100 g), which were all more significant than the control roots. In conclusion, our study optimized the culture condition and established a feasible genetics reactor for P. tanguticagreen exploration and biological study in the alpine region.

Abstract

Direct, reactive incorporation of molecular groups into surfaces was demonstrated to be a new surface modification method. Reactive soft-landing of ion beams provided a means of covalently attaching intact molecular moieties to give-well defined products in contrast to earlier experiments using ion treatment. The fragmentation of the reactive projectiles was reduced or prevented by the very low collision energies used in the experiments. A superficial similarity to the classical Kolbe alkane synthesis labels the results an interfacial Kolbe reaction.

190. Turing instability in a network-organized epidemic model with delay.

Abstract

In this paper, we show the impact of both network and time delays on Turing instability and demarcate the role of diffusion in the epidemic. The stability and bifurcation of equilibrium points are analyzed to reveal the epidemic state, which is the precondition of pattern formation. The network could lead to the transition from the endemic to the periodic outbreak via negative wavenumber, which provides a way to prevent significant harm or decrease the damage of the epidemic to humans by the delay, the connection rate, and the infection rate. Also, the threshold value of time delay is proportional to the minimum eigenvalue of the network matrix, which provides a way to control the periodic behavior. Finally, numerical simulations validate these analytical results and the mechanisms of frequent outbreaks. • Turing instability is investigated through wavenumber. • The transition mechanism of the epidemic from the endemic is showed. • The critical value of delay is proportional to the least eigenvalue of network.
