Background: A large amount of epidemiological evidence indicates that persistent HCV infection is the main risk factor for HCC. We aimed to study the effects of persistent HCV infection on the interaction of the virus and host cell to identify cancer gene profiles. Methods: Next-generation sequencing (NGS) was used to identify differentially expressed genes between uninfected Huh7.5.1 control cells, short-term HCV (S-HCV), early long-term HCV (eL-HCV), and long-term HCV (L-HCV) infections, which were analyzed using different dynamic bioinformatics and analytic tools. mRNA expression was validated and quantified using q-PCR. One hundred ninety-six serum samples of HCV patients with IFN/RBV treatment were used to study chemokine levels. Results: S-HCV activates an inflammatory response and drives cell death and apoptosis through cell cycle arrest via MAPK signaling. L-HCV promotes cell growth and alters cell adhesion and chemokine signaling via CXCL8-mediated-SRC regulation. A total of 196 serum samples from the HCV and HCV-HCC cohorts demonstrated significantly upregulated pro-inflammatory CXCL8 in non-SVR (persistent HCV infection) patients in the HCV-HCC group. Conclusions: Persistent infection with HCV induced pro-inflammatory CXCL8 and the oncogene SRC, thereby triggering and promoting hepatocarcinogenesis. CXCL8 may be a potential biomarker for monitoring HCV-related HCC progression.

In present work, the thermal decomposition behaviour and kinetics of proteins, carbohydrates and lipids is studied by use of models derived from mass-loss data obtained from thermogravimetric analysis of Scenedesmus microalgae. The experimental results together with known decomposition temperature range values obtained from various literature were used in a deconvolution technique to model the thermal decomposition of proteins, carbohydrates and lipids. The models fitted well (R2 > 0.99) and revealed that the proteins have the highest reactivity followed by lipids and carbohydrates. Generally, the decomposition kinetics fitted well with the Coats-Redfern first and second order kinetics as evidenced by the high coefficients of determination (R2 > 0.9). For the experimental conditions used in this work (i.e. high heating rates), the thermal decomposition of protein follows second order kinetics with an activation energy in the range of 225.3-255.6 kJ/mol. The thermal decomposition of carbohydrate also follows second order kinetics with an activation energy in the range of 87.2-101.1 kJ/mol. The thermal decomposition of lipid follows first order kinetics with an activation energy in the range of 45-64.8 kJ/ mol. This work shows that the thermal decomposition kinetics of proteins, carbohydrates and lipids can be performed without the need of experimentally isolating the individual components from the bulk material. Furthermore, it was shown that at high heating rates, the decomposition temperatures of the individual components overlap resulting in some interactions that have a synergistic effect on the thermal reactivity of carbohydrates and lipids.

Microbial fuel cell (MFC) technology turns chemical energy into bioelectricity in a clean and efficient manner, lowering carbon emissions and increasing bioenergy production. It is a multifaceted technique that has the potential to be a panacea for clean water scarcity and sustainable, renewable energy. In this review, the approach focuses on scaling-up and application prospects at a commercial scale. An outlook on various, previously tried methodologies was generated in order to establish a viable bioelectricity scaling-up approach that is also cost-effective in its design. Precise themes were followed to evaluate previously produced models and methodologies for MFCs: principle and anatomical mechanisms, basic applications, bioelectricity scaling-up potentials from previous work and limitations, then an outlook on MFC feasibility and its wastewater treatment plant (WWTP) energy supply chain. The goal of this paper is to derive a viable approach from prior research in order to comprehend how MFC technology may be scaled-up for commercial and practical power output. Essentially, this article summarizes the current energy predicaments faced by South Africa and proposes MFCs as a new knowledge-contributing technology with electricity scaling-up potential. Conclusively, more research on MFC technique scaling-up operating factors is recommended.

Hepatitis Delta Virus (HDV) is an RNA virus that requires the presence of hepatitis B surface antigen (HBsAg) to propagate into hepatocytes, with Genotype I being more prevalent globally. However, the prevalence of HDV genotypes in Taiwan is unknown. Accordingly, a cohort including 24 chronic HBV patients who received nucleos(t)ides (NUCs) between January 2002 and July 2018 was used to determine HDV genotypes and genotype specific serological association in chronic HBV carriers. HDV-positive genotypes in 18/24 (75%) males and 6/24 (25%) females were identified among chronic HBV patients. Viremia was lower in HDV-IV patients than in patients affected with other HDV genotypes (1.34 log10 copies/mL vs. 3.30 log10 copies/mL, p = 0.009). A logistics regression analysis revealed that HDV-IV was inversely proportional to HDV RNA (odds ratio [OR]/95% confidence intervals [CI]: 0.370/0.164–0.830, p = 0.017). The serologic association study indicated lower levels of creatinine (p = 0.047) and HDV-RNA (p = 0.009) in the HDV-IV group than the non-HDV-IV group but did not indicate any significant differences in the AST, ALT, bilirubin levels or other laboratory test factors. The three genotypes evident in Taiwan were HDV-I (4/24, 16.7%), HDV-II (6/24, 25.0%), and HDV-IV (14/24, 58.3%), and HDV-IV is the predominant HDV genotype in Taiwan. These results anticipate a clear understanding of HDV genotype serological association in chronic HBV carriers.

Due to the difficulties experienced in identifying the exact location of underground cable faults, it is therefore evident that there is a need to employ optimal methods that will have the capability to swiftly identify the fault location and henceforth reduce the downtime of the system. This paper presents a study that combines both Wavelet Transform and Adaptive Neuro-Fuzzy Inference System (ANFIS) technique for fault location in underground cables. A 9 km, 11 kV, 50 Hz transmission line was developed, and a single line to ground fault was simulated at different line locations. The wavelet transform was used to extract noteworthy features that were subsequently utilised as inputs for training and testing the Adaptive Neuro-Fuzzy Inference System (ANFIS). The results obtained from the ANFIS were compared with the actual simulated test values. From the results obtained, the technique was able to achieve a 99.94% accuracy in fault location.

Standard Western demographic survey protocols fail to capture dynamics, such as circular migration and support networks, that profoundly influence the health of non-Western domestic social groups, typically called households. Enhanced protocols are needed because survey data provide the primary evidence base for health policy and planning globally. We present the participatory development, implementation and analysis of a novel demographic survey protocol, that aimed to better capture domestic social dynamics in rural Eswatini, southern Africa. The multiple-method study incorporated participatory health research about a community affected by HIV/AIDS, of which the survey formed part, and an ethnography of the participatory survey development process. Analysis of the data revealed limitations in the reliability and validity of standardised survey questions for measuring household membership, in contexts where circular migration and polygamy are common. Standard survey protocols potentiate double-counting members and misclassifying 'child-headed' and 'female-headed' households. They neglect social and economic dynamics that are known to influence health. Our novel demographic survey protocol provides a simple alternative method for capturing core data about circular migration and its impact on health. The study illustrates the contributions participatory and ethnographic research can make to enhancing demographic surveys.

Research on transactional sexual relationships has largely focused on women's perspectives. Better understanding the men's views-especially regarding relationships with adolescent girls and young women-can inform HIV prevention efforts. In 2017, 134 in-depth interviews were conducted with the male partners of girls and young women aged 19-47 years, 94 in Uganda and 40 in Eswatini. Respondents were recruited at venues such as bars where men and potential partners meet and through other young women. Most respondents believed that providing money/gifts was the way to establish relationships with women in their communities, a context that some found undesirable. Young women were mainly perceived as actively pursuing transactional sex for material goods, but respondents also described economically impoverished women who were manipulated into relationships. Men described conflict with longer term partners as a driver to seeking younger partners, who were more compliant. Transaction dominates the male partners of adolescent girls and young women's understanding of sexual relationships, and inequitable power dynamics are reinforced by seeking younger partners. However, some respondents' discontent with this dynamic suggests an opportunity for change. HIV prevention programmes should directly address the underlying drivers of transactional relationships (e.g. gender norms) and work with men who question the practice.

Reports in the literature indicate that species of Hepatozoon commonly occur in African wild dog (AWD) or painted wolf (Lycaon pictus) populations. These findings were based on examination of blood smears by microscopy, and specific identity of the Hepatozoon sp. gamonts seen could not be confirmed. We present the first in-depth molecular data on the prevalence of species of Hepatozoon in a free-ranging AWD population. In a general health survey of AWDs in the Kruger National Park, blood specimens (n = 74) collected from 54 individuals were examined for the presence of Hepatozoon spp. At first sampling, specimens from 42 of 54 individuals (77.7%) were positive, based on the primer set HepF300 and HepR900. Twenty individuals were resampled between 51 and 69 days after first sampling; one of these was resampled twice. Samples from six individuals that had tested negative previously now reacted positive. Assuming that all 54 individuals were still alive, the prevalence had therefore increased to 48 individuals infected, or 88.8%. Resultant 18S rDNA sequences isolated from these specimens share high similarity to other Hepatozoon canis genotypes. The quality of freshwater and its supply, particularly for domestic and industrial purposes are waning due to urbanization and inefficient conventional wastewater treatment (WWT) processes. For decades, conventional WWT processes have succeeded to some extent in treating effluents to meet standard discharge requirements. However, improvements in WWT are necessary to render treated wastewater for re-use in the industrial, agricultural, and domestic sectors. Three emerging technologies including membrane technology, microbial fuel cells and microalgae, as well as WWT strategies are discussed in this chapter. These applications are a promising alternative for manifold WWT processes and distribution systems in mitigating contaminants to meet acceptable limitations. The basic principles, types and applications, merits, and demerits of the aforementioned technologies are addressed in relation to their current limitations and future research needs. The development in WWT blueprints will augment the application of these emerging technologies for sustainable management and water conservation, with re-use strategies.

Water may be treated differently in different communities depending on the quality of the water that enters the treatment plant. Typically, surface water requires more treatment and filtration than groundwater because lakes, rivers, and streams contain more sediment and pollutants and are more likely to be contaminated than groundwater. Some water supplies may also contain disinfection by-products, inorganic chemicals, organic chemicals, and radionuclides. Specialized methods for controlling formation or removing them can also be part of water treatment. The study presents an approach to prioritize Water treatment plant maintenance activities based on system reliability and cost-effectiveness. The object of this approach is to minimize the weighted average system reliability index (WASRI) by ranking maintenance tasks based on their marginal benefit to cost ratios, where the benefit is defined as improvement in WASRI. Estimations of WASRI improvement are obtained by utilizing the linear relationship between the benefit obtained from a maintenance task and the change of failure rate of the maintained component.

During the progression from hepatitis to fibrosis, cirrhosis, and liver failure, the accumulation of stressed/damaged hepatocyte elements associated with liver inflammation is critical. The causes of hepatocyte injuries include viral hepatitis infections, alcoholic hepatitis, and non-alcoholic fatty liver disease. Hepatocyte-derived extracellular vesicles (Hep-EVs) released from stressed/damaged hepatocytes are partly responsible for liver disease progression and liver damage because they activate non-parenchymal cells and infiltrate inflammatory cells within the liver, which are in turn are an important source of EVs. This cell-to-cell signaling is prevalent during inflammation in many liver diseases. Accordingly, special emphasis should be placed on liquid biopsy methods for the long-term monitoring of chronic liver diseases. In the present review, we have highlighted various aspects of current liquid biopsy research into chronic liver diseases. We have also reviewed recent progress on liquid biopsies that focus on cell-free DNA (cfDNA), long non-coding RNA (lncRNA), and the proteins in EVs as potential diagnostic tools and novel therapeutic targets in patients with viral hepatitis, fatty liver steatosis, and alcoholic liver diseases.

Comprehensive theories of health justice can supplement rights-based approaches like primary health care, by conceptualizing key terms, and systematizing knowledge about structural factors that influence health. Our aim was to use "health capability" as a theoretical lens for understanding how primary health care approaches might address structural factors impeding health in a rural Swazi community. We conducted abductive, interpretive, analysis of a mixed-method (QUAL+quan) data set about "health capability deprivations," generated through participatory action research. Four themes are discussed: illness and disease, unhealthy daily living environments, inability to move freely, and gendered expectations and norms. The analysis demonstrates that there were complex interrelationships between health capability deprivations, material and ideological deprivation prevented community members from aspiring to or securing their right to health, health capability theory can augment primary health care approaches and vice versa, and qualitatively driven, mixed-method research can generate unique insights about structural factors that influence health.
