Context Between December 2010 and January 2011, 20 cases of mycobacteriosis, consisting of grossly nodular granulomatous and caseous lesions in pig carcases from five different fattening farms, were submitted to the Slaughterhouse Support Service in Barcelona, Spain. Lesions involved lymph nodes, lungs, liver and spleen. Main conclusion A mycobacteriosis outbreak caused by Mycobacterium avium subspecies avium was detected in the five pig farms. Approach Gross pathology, histopathology and bacteriological isolation followed by molecular identification of culture isolates and epidemiological investigation. Results Histopathological examination showed multifocal to coalescent, granulomatous and necrotising lymphadenitis, pneumonia, hepatitis and splenitis. Acid-fast bacilli compatible with mycobacteria were detected in some of the samples. The presence of acid-fast bacilli in some cases revealed mycobacteriosis. Bacteriological culture was performed to confirm diagnoses and identify the aetiological agent (to rule out potential involvement of Mycobacterium tuberculosis complex mycobacteria, which includes those that cause human and animal tuberculosis). PCR from culture isolates confirmed the growth of M avium complex. Further sequencing analysis determined it was M avium subspecies avium. Interpretation The most likely source of the outbreak was considered to be the feed that was shared by the five farms, which might have been contaminated with M avium subspecies avium. The fact that most of the animals presented with a clear involvement of abdominal viscera is consistent with an oral route of infection. Significance of findings The establishment of a meat inspection support network allowed for rapid management of sample submission, diagnosis and subsequent molecular identification of the mycobacteriosis outbreak, leading to an understanding of the associated risks to public health. Abstract: Porcine respiratory disease complex (PRDC) is the name used to describe a clinical presentation characterized by respiratory signs and poor growth in growing-finishing pigs. Porcine circovirus type 2 (PCV2) is one of the pathogens potentially involved in the presentation of this complex, although, it has been mainly linked to PCV2-systemic disease (PCV2-SD). However, some reports have suggested that PCV2 can cause respiratory lesions in the absence of systemic lymphoid damage; this apparently different condition has been referred as PCV2-lung disease (PCV2-LD). The present study analysed retrospectively the relation between PRDC and PCV2 infection from a pathological point of view. Of the 317 selected pigs suffering from PRDC and submitted for necropsy between 1998 and 2011, the most prevalent lesions were subacute interstitial pneumonia (27.7%) and the combination of subacute interstitial pneumonia plus catarrhal-purulent bronchopneumonia (28.4%). Two hundred and twenty-six pigs were positive for PCV2 by in situ hybridization (ISH); using immunohistochemical techniques, 77 were positive for porcine reproductive and respiratory syndrome virus and 3 for swine influenza virus. Among positive pigs for PCV2 ISH, 184 animals suffered from PCV2-SD. The rest of 42 PCV2 ISH positive pigs had no lymphoid lesions and low amount of viral nucleic acid in these tissues and, therefore, did not qualify for PCVD-SD; only 9 had concomitant PCV2 ISH positivity in the lung, also to a low amount. In conclusion, all PRDC-PCV2 ISH positive cases had evidence of viral systemic infection. These results suggest that PCV2-LD is probably a negligible condition and PCV2 mainly contributes to PRDC in relation to PCV2-SD occurrence. [Copyright &y& Elsevier]

A total of 1279 Eurasian wild boar ( Sus scrofa) sera were collected from 2000 to 2011 in the Iberian Peninsula to reveal time changes in serum antibody prevalences against selected infectious agents (porcine circovirus type 2, PCV2; porcine reproductive and respiratory syndrome virus, PRRSV; hepatitis E virus, HEV; and Erysipelothrix rhusiopathiae) and to identify putative individual or population factors driving such changes. Overall seroprevalences were 48%, 26%, 2% and 15% for PCV2, HEV, PRRSV and E. rhusiopathiae, respectively. The global observed prevalence of antibodies against PCV2 and HEV remained stable during the study period, while the global mean antibody seroprevalence against E. rhusiopathiae declined. The mean increment in prevalence was always lower for open than for fenced sites. This study evidenced for the first time that wild boar from the Iberian Peninsula have widespread contact with E. rhusiopathiae, and confirmed high prevalences of antibodies against PCV2 and HEV. Maintained high prevalences of transmissible agents in wild boar suggest that epidemiological drivers such as aggregation and high density are still acting. This will most probably also affect the transmission rates of other disease agents and should be taken into account regarding disease control at the wildlife-livestock interface. Summary: The aim of this study was to assess the effect of simultaneous experimental inoculation of porcine circovirus type 2 (PCV2; intranasal delivery) and Mycoplasma hyopneumoniae (Mhyo; transtracheal delivery) into conventional, seropositive 6-week-old piglets. Thirty-six male piglets were assigned randomly to four groups: control (n =6), PCV2 (n =6), Mhyo (n =12) and PCV2+Mhyo (n =12). Blood samples and faecal and nasal swabs were collected at 0, 7, 14 and 21days post inoculation (dpi). No significant clinical signs attributable to PCV2 infection were observed during the experiment. Coughing was recorded in three pigs from the Mhyo group and six from the PCV2+Mhyo group. No significant differences in mean body weight and rectal temperature were observed between the groups. Mild microscopical lesions similar to those reported for post-weaning multisystemic wasting syndrome were observed in two PCV2 pigs and in one PCV2+Mhyo animal. Mhyo-compatible lung lesions were observed in 21/24 pigs inoculated with Mhyo (10 from the Mhyo group and 11 from the PCV2+Mhyo group). PCV2 was detected by in-situ hybridization in 3/12 PCV2 and in 4/12 PCV2+Mhyo animals. No significant differences in PCV2 load (serum and nasal and faecal swabs), duration of viraemia or antibody titre were detected between PCV2-inoculated groups. No significant differences in Mhyo load in nasal swabs, percentage of Mhyo-seropositive pigs and mean lung score was detected between Mhyo-inoculated groups. Under the conditions of the present study, concurrent inoculation of PCV2 and Mhyo did not result in potentiation of clinical signs and lesions attributed to either infection. Summary The newly established family Anelloviridae includes a number of viruses infecting humans (Torque teno viruses) and other animal species. The ones infecting domestic swine and wild boar are nowadays named Torque teno sus viruses (TTSuV), which are small circular single-stranded DNA viruses highly prevalent in the pig population. So far, two genetically distinct TTSuV species are infecting swine. Both TTSuVs appear to efficiently spread by vertical and horizontal transmission routes; in fact, foetuses may be infected and the prevalence and viral loads increase by age of the animals. Detailed immunological studies on TTSuVs are still lacking, but it seems that there are no efficient immunological responses limiting viraemia. These viruses are currently receiving more attention due to the latest results on disease association. Torque teno sus viruses have been circulating unnoticed in pigs for a long time, and even considered non-pathogenic by themselves; there is increasing evidence that points to influence the development of some diseases or even affect their outcome. Summary: Porcine circovirus type 2 (PCV2) is the essential infectious agent of post-weaning multisystemic wasting syndrome (PMWS), one of the most important diseases of swine. Although several studies have described different biological properties of the virus, some aspects of its replication cycle, including ultrastructural alterations, remain unknown. The aim of the present study was to describe for the first time a complete morphogenesis study of PCV2 in a clone of the lymphoblastoid L35 cell line at the ultrastructural level using electron microscopy techniques. Cells were infected with PCV2 at a multiplicity of infection of 10 and examined at 0, 6, 12, 24, 48, 60 and 72h post-infection. PCV2 was internalized by endocytosis, after which the virus aggregated in intracytoplasmic inclusion bodies (ICIs). Subsequently, PCV2 was closely associated with mitochondria, completing a first cytoplasmic phase. The virus entered the nucleus for replication and virus assembly and encapsidation occurred with the participation of the nuclear membrane. Immature virions left the nucleus and formed ICIs in a second cytoplasmic phase. The results suggest that at the end of the replication cycle (between 24 and 48h), PCV2 was released either by budding of mature virion clusters or by lysis of apoptotic or dead cells. In conclusion, the L35-derived clone represents a suitable in-vitro model for PCV2 morphogenesis studies and characterization of the PCV2 replication cycle. [ABSTRACT FROM AUTHOR]

Summary: Necrotizing lymphadenitis is observed in approximately 2% of pigs affected by post-weaning multisystemic wasting syndrome (PMWS). The pathogenesis of the lesion has been linked to apoptosis induced by porcine circovirus type 2 (PCV2). The aim of the present study was to gain further insights into PCV2-associated lymphoid necrosis in pigs with PMWS. Three groups of animals were studied: (1) PMWS-affected pigs with necrotizing lymphadenitis (n =5), (2) PMWS-affected pigs without necrotizing lymphadenitis (n =5) and (3) healthy pigs with no PMWS-related lesions (n =5). Investigations performed included immunohistochemical evaluation of the expression of cleaved caspase-3 and von Willebrand factor, Mallory’s staining for fibrin and in-situ hybridization for detection of the PCV2 genome. The results of the study suggested that lymphoid necrosis in PMWS-affected pigs may be related to hypertrophy and hyperplasia of high endothelial venules (HEVs). The mechanism underlying these changes in HEVs was not clearly defined, but necrotizing lymphadenitis in pigs with PMWS may develop following vascular damage with thrombosis and subsequent follicular necrosis. Summary: Post-weaning multisystemic wasting syndrome (PMWS) is one of the most significant porcine diseases worldwide. The causative agent is porcine circovirus type 2 (PCV2), the smallest virus known to infect animals. Data related to the structural and ultrastructural aspects of this infectious disease are sparse and there is little knowledge of the subcellular localization of PCV2 and its replication in the tissues of pigs naturally affected by PMWS. The present study describes the cellular localization of PCV2 in the lymph nodes of pigs affected by PMWS by application of immunolabelling techniques for light and transmission electron microscopy (TEM). PCV2 particles were exclusively detected in histiocytes. Ultrastructural alterations including marked dilatation of rough endoplasmic reticulum and swelling of mitochondria were associated with PCV2-labelled intracytoplasmic inclusions (ICIs) with recognizable virions. Within the ICIs icosahedral virus-like particles were specifically labelled with a PCV2 capsid antibody, whereas particles with a granular appearance were not labelled. Colocalization studies with confocal microscopy and double immunolabelling with TEM indicated a close relationship between virus and the mitochondria, suggesting that these organelles may play an important role in the replication of PCV2. Abstract: The aim of the study was to determine the presence of swine hepatitis E virus (HEV) RNA and antibodies in postweaning multisystemic wasting syndrome-affected (n =114) and non-affected (n =46) pigs and the possible association with hepatitis lesions. Forty-four pigs were RT-PCR positive (28.2%); 25 of them were PMWS cases, while 19 were non-PMWS pigs. In both groups, HEV RT-PCR results were associated with hepatitis (OR=5.61 for PMWS-affected pigs and OR=5.17 for non-PMWS affected pigs; p =0.01). No interaction was detected in a logistic regression between PMWS occurrence and HEV infection for the development of hepatitis lesions. Seropositivity to HEV was more likely to occur in pigs with hepatitis (51.9%) compared to pigs without hepatitis (36.1%; p =0.03). Significant differences in optical densities were notices comparing the lesional stage of pigs (p =0.009). While pigs with slight or moderate hepatitis were seropositive, pigs with more severe lesions were seronegative to HEV. These results indicate that swine HEV infection can be a significant contributor to the development of moderate hepatitis in pigs regardless of the PMWS status. [Copyright &y& Elsevier]

We evaluated the effects of a 6% spray-dried porcine plasma (SDPP) and a plant extracts mixture (XT; 5% carvacrol, 3% cinnamaldehyde, and 2% capsicum oleoresin) on the productive performance, intestinal morphology, and leukocyte cell subsets of early-weaned pigs compared with a control group. Morphometry of the jejunum, ileum, and colon, and immune cell analysis of blood, ileocolic lymph node (LN), and ileal Peyer's patches were done in 24 weaned pigs (20 ± 2 d) at 19 or 21 d postweaning. Although SDPP and XT treatments did not increase ADG or ADFI, SDPP improved the G:F ratio (P = 0.024) compared with the control group. Dietary SDPP reduced the percentages of blood monocytes (P = 0.006) and macrophages in ileal Peyer's patches and LN (P = 0.04), of B lymphocytes (P = 0.04) and γδ T cells in LN (P = 0.009), and of intraepithelial lymphocytes (P = 0.026) as well as the density of lamina propria cells in the colon (P < 0.01). Dietary XT reduced intraepithelial lymphocyte numbers in jejunum (P = 0.034) and the percentages of blood cytotoxic cells (P = 0.07) and B lymphocytes in LN (P = 0.03); however, XT increased blood monocytes (P = 0.038) and the density of lamina propria lymphocytes in the colon (P = 0.003). These results indicate that dietary SDPP and plant extracts can affect intestinal morphology and immune cell subsets of gut tissues and blood in weaned pigs. Histopathological and toxicological analyses confirmed a clinical diagnosis of selenium (Se) intoxication in pigs from a farm in Spain. After an initial episode of diarrhoea, animals presented both dermatological and neurological signs; the most obvious sign was a marked hind limb paresis. Cutaneous lesions consisted on diffuse alopecia, multifocal skin necrosis and coronary band necrosis of the hooves. Central nervous system lesions involved the cervical and lumbar intumescences of the spinal cord and consisted of a severe, bilateral symmetrical poliomyelomalacia of the ventral horns; pons and medulla oblongata also presented lesions of polioencephalomalacia. Analyses of feed and sera from clinically affected pigs revealed a marked increase in Se concentration. Abstract: An exploratory case–control study was carried out in Spain in 2002/2003, involving 62 pig farms of different production systems to assess risk factors that, in association with PCV2 infection, induce postweaning multisystemic wasting syndrome (PMWS) expression. To achieve this objective two groups of farms selected according to their PMWS status were compared: “cases” (farms with clinical PMWS, n =32) and “controls” (farms without clinical PMWS, n =30). A filled-in questionnaire and 45 blood samples (15 sows, and two groups of 15 pigs of 12 and 20 weeks of age, respectively) were obtained from each farm. Additionally, two to three diseased pigs were necropsied and relevant tissues to diagnose PMWS collected when PMWS was clinically suspected (“case” farms). A statistical analysis to compare “case” versus “control” farms was performed with the variables obtained from the questionnaire (191 variables) and the serologic test results (20 variables). Data were analysed using conditional logistic regression with a nested n:m matched design taking into account the farm size. Three variables were found significant in the final model: two related to vaccination scheme and one to PCV2 seroprevalence in growing pigs. Vaccination of gilts against PRRSV increased the odds of PMWS expression and vaccination of sows against atrophic rhinitis was related to decreased odds of the disease; however, the possibility that those two factors could be spurious effects (due to the small sample size) or confounding variables cannot be ruled out. On the other hand, a higher prevalence of antibodies to PCV2 at 12 weeks of age was observed in pigs from “case” farms than in pigs from “control” farms. This result suggests that an earlier infection with PCV2 might be a risk factor for PMWS expression. [Copyright &y& Elsevier]

The similarities between the porcine and human cardiovascular systems make the pig a useful animal model for the study of vascular biology. However, a standardized method is needed to describe the normal histological properties of porcine arteries in order to evaluate pathologic lesions in future studies. Descriptive and morphometric analyses were done on 16 porcine femoral arteries. For these purposes, three histological stains (haematoxylin eosin, Masson's trichrome, and orcein), four immunohistochemical methods (using antibodies anti-α-actin, anti-CD3, anti-L1 and anti-lysozyme), and a glycohistochemical method (usingDolichos bifloruslectin) were performed. The porcine femoral arteries evaluated had a mean total area of 6.25 ± 1.99 mm2 and a diameter of 2.79 ± 0.41 mm. The majority of the total area was occupied by the medial layer (42.97 ± 5.38%) and was mainly constituted by smooth muscle cells (94.58 ± 2.65%). All the cell markers used reacted with porcine paraffin-embedded tissue. However, the anti-lysozyme antibody was excluded from this histological analysis because of cytoplasmatic reactivity in smooth muscle cells. In summary, this study proposes histological methods to describe the normal characteristics of the porcine femoral artery and raises the possibility of applying this methodology in future studies on porcine vascular research. Background: Nowadays, the most common presentation of PCV-2 is the subclinical infection in piglets after weaning. The success of PCV-2 vaccination is associated with the control of the clinical disease as well as the improvement of production parameters. In consequence, the objective of the present study was to analyse the effect of PCV-2 maternally derived antibody (MDA) levels on vaccine efficacy in piglets vaccinated at three weeks of age with a commercial PCV-2 subunit vaccine. The study was performed analysing a database with 6112 wean-to-slaughter piglets from 4 different European regions. Results: Results showed that the use of the vaccine was able to decrease the PCV-2 viremia calculated as area under the curve (AUC = 60.29 ± 3.73), increase average daily weight gain (ADWG = 0.65 ± 0.01 kg/day) and reduce mortality (7%) in vaccinated piglets compared to non-vaccinated ones (AUC of 198.27 ± 6.14, 0.62 ± 0.01 kg/day and 11% respectively). The overall difference of ADWG between both groups was close to 30 g per day (p < 0.05), also when they were split for low and high levels of MDA titres. Moreover, the animals with the highest ADWG were observed in the group of piglets vaccinated with high or extremely high antibody titres (0.66 and 0.65 kg/day respectively). Considering only animals with extremely high antibody titres, both study groups performed similar, however there was a numerical difference of 10 g/day in favour of vaccinated piglets. Likewise, lack of correlation between ADWG and MDA was observed suggesting that no maternal antibody interference was present with the tested vaccine because the vaccinated animals grew faster compared to unvaccinated control animals, regardless of the level of maternal antibodies present at the time of vaccination. Conclusions: The results of the present study demonstrated that the MDA against PCV-2 transferred through the colostrum intake has a protective effect against this viral infection. The vaccine used in the present study (Ingelvac CircoFLEX®) was effective when applied at three weeks of age and was not affected by the level of MDA at the time of vaccination. [ABSTRACT FROM AUTHOR]
