The ISWI family of ATP-dependent chromatin remodelers represses transcription by changing nucleosome positions. ISWI regulates nucleosome positioning by requiring a minimal length of extranucleosomal DNA for moving nucleosomes. ISW2 from Saccharomyces cerevisiae, a member of the ISWI family, has a conserved domain called SLIDE (SANT-like ISWI domain) that binds to extranucleosomal DNA ~19 base pairs from the edge of nucleosomes. Loss of SLIDE binding does not perturb binding of the ATPase domain or the initial movement of DNA inside of nucleosomes. Not only is extranucleosomal DNA required to help recruit ISW2, but also the interactions of the SLIDE domain with extranucleosomal DNA are functionally required to move nucleosomes.

The utilization of shock waves plays a pivotal role in the advancement of multiple scientific domains like aerospace, defense, geology, environment, medicine and many more. They serve as essential tools in scientific investigations, enabling the exploration of material behavior under extreme conditions, viz. elevated pressure and temperature. The present study is specifically dedicated to scrutinizing the repercussions of shock waves on an l‐ascorbic acid single crystal, to which they were intentionally applied to assess their influence on structural, optical and third‐order nonlinearity properties. Powder X‐ray diffraction analysis unveiled a discernible overall enhancement in the crystalline quality of the grown crystal following exposure to shock waves. This observation was consistently corroborated by high‐resolution X‐ray diffraction data, particularly on the (200) crystallographic planes. Furthermore, the optical transmittance of the crystal exhibited a notable increase upon the application of shock waves, while the material's band gap remained unaffected. In parallel, the third‐order nonlinearity of the crystal was found to undergo a significant augmentation as a consequence of the shock treatment, as confirmed through Z‐scan measurements. These empirical findings unequivocally demonstrate the substantial enhancement in the structural, optical and nonlinear properties of the grown crystal when subjected to shock waves, rendering it well suited for nonlinear optical applications. Lime is an important flux used in the iron and steel-making processes. Tata Steel Jamshedpur uses a twin-shaft Parallel Flow Regenerative type kiln to calcine limestone. Previously, the kiln was operated based on the temperature measurement at only three locations. This led to suboptimal kiln performance and difficulty in maintaining lime quality. A thermal digital twin was developed using first principles to address these issues. A set of unsteady-state partial-differential equations representing the thermophysical phenomena is solved numerically using the finite volume method. The model was validated with the measured flue gas temperature and lime temperatures. The model is implemented online with the real-time operating parameters to estimate the temperature profile of solid and gas and the degree of conversion of limestone. This real-time visibility enables the operators to optimize the process and achieve lime conversion up to 94%, resulting in energy savings of 31.8 GJ/day and stable lime quality. Abstract Background/aims Crohn’s disease (CD) and intestinal tuberculosis (ITB) are gastrointestinal (GI) inflammatory disorders with overlapping clinical presentations but diverging etiologies. The study aims to decipher CD and ITB-associated gut dysbiosis signatures and identify disease-associated co-occurring modules to evaluate whether this dysbiosis signature is a disease-specific trait or is a shared feature across diseases of diverging etiologies. Methods Disease-associated gut microbial modules were identified using statistical machine learning and co-abundance network analysis in controls, CD and ITB patients recruited as part of this study. Module reproducibility was reinvestigated through meta-network analysis encompassing >5400 bacteriomes and ~900 mycobiomes. Subsequently, >1600 Indian gut microbiomes were analyzed to identify a central-core gut microbiome of 46 taxa, whose abundances aided in the formulation of an India-specific Core Gut Microbiome Score (CGMS) to measure the degree of core retention. Results Both diseases witness similar patterns of alterations in [alpha]-diversity, characterized by a significant reduction in gut bacterial (i.e., bacterial/archaeal) diversity and a concomitant increase in the fungal [alpha]-diversity. Specific bacterial taxa, along with the diverging mycobiome enabled distinction between the diseases. Co-abundance network analysis of these taxa, validated by integrated meta-network analysis, revealed a ‘disease-depleted’ module, consistent across multiple cohorts, with >75% of this module constituting the central-core Indian gut microbiome. CGMS robustly assessed the core-microbiome loss across different stages of gut inflammatory disorders, in Indian and international cohorts. Conclusions While the disease-specific gain of detrimental bacteria forms an important component of gut dysbiosis, loss of the core microbiome is a shared phenomenon contributing to various GI disorders.

Arterial desaturation following surgical repair of tetralogy of Fallot (TOF) is rare. In most instances, it results from residual right ventricular outflow tract obstruction, causing right-to-left shunt across residual interatrial or interventricular communication. In this report, we present an unusual scenario of arterial desaturation due to a recanalized left cardinal vein in a child with repaired TOF. We also discuss stepwise evaluation that led to successful identification and occlusion of the abnormal venous channel by percutaneous device closure.

Abstract Brain cancer is regarded as one of the most life-threatening forms of cancer worldwide. Oxidative stress acts to derange normal brain homeostasis, thus is involved in carcinogenesis in brain. The Nrf2/Keap1/ARE pathway is an important signaling cascade responsible for the maintenance of redox homeostasis, and regulation of anti-inflammatory and anticancer activities by multiple downstream pathways. Interestingly, Nrf2 plays a somewhat, contradictory role in cancers, including brain cancer. Nrf2 has traditionally been regarded as a tumor suppressor since its cytoprotective functions are considered to be the principle cellular defense mechanism against exogenous and endogenous insults, such as xenobiotics and oxidative stress. However, hyperactivation of the Nrf2 pathway supports the survival of normal as well as malignant cells, protecting them against oxidative stress, and therapeutic agents. Plants possess a pool of secondary metabolites with potential chemotherapeutic/chemopreventive actions. Modulation of Nrf2/ARE and downstream activities in a Keap1-dependant manner, with the aid of plant-derived secondary metabolites exhibits promise in the management of brain tumors. Current article highlights the effects of Nrf2/Keap1/ARE cascade on brain tumors, and the potential role of secondary metabolites regarding the management of the same.

Abstract Due to climate and human activity, water supply in Jharkhand, India, fluctuates. This study measured AWS and AWA in Jharkhand, India. MODIS NDVI and Terra Climate data processed by Google Earth Engine (GEE) are used in the analysis. Based on mean annual values for NDWI, run-off, precipitation, and ET, AWS and AWA are categorised. Results show significant regional differences in ET, precipitation, runoff, AWS, and AWA in Jharkhand. Purbi Singhbhum has far higher ET rates than Garhwa, Palamu, and Chatra. High AWS levels in Sahibganj, Godda, Pakur, Garhwa, and Kodarma indicate serious water scarcity. Therefore, these locations need focused water management approaches. Different districts including Giridih, Chatra, Jamtara, Latehar, Simdega, and Hazaribagh have different levels of AWS, highlighting the need for individualised treatments. The AWA index helps resource managers make educated decisions by detecting water-scarce places. Climate variability affects temperatures and precipitation, worsening AWS in high-AWS areas. Water scarcity mitigation requires local infrastructure development and water management measures. To verify remote sensing data accuracy, future study should improve data resolution and include in-situ measurements. These findings can help policymakers and stakeholders in Jharkhand create efficient water resource management strategies to reduce water stress and assure water supply.

Background: Family planning (FP) services are pivotal in assessing a country’s healthcare efficacy. Despite India’s strides in FP promotion, disparities persist in its utilization rates. This study analyzes Karnataka’s FP trends by using National Family Health Survey (NFHS) rounds 4 and 5, comparing its indicators with national averages. Methods: A retrospective record review-based descriptive study design was employed. Data from NFHS-4 (2015–16) and NFHS-5 (2019–21) were analyzed using MS Excel 365. Key variables included age at marriage, total fertility rate (TFR), and various contraceptive methods. Results: Karnataka exhibited a stable rate of early marriages for women and a decline among men aged 25–29 years. TFR in Karnataka slightly decreased, with increased contraceptive usage and significant growth in modern contraceptive methods. However, male sterilization rates remained low. There was an improvement in health workers’ engagement with female non-users and in providing information on contraceptive side effects. Conclusion: While Karnataka has progressed in FP, challenges such as early marriages persist. Enhancing integration, leveraging technology, and empowering women are essential for comprehensive FP services in India.

Abstract Multiple sclerosis (MS) is an inflammatory demyelinating disease with heterogeneous clinical presentations and variable long-term disability accumulation. There are currently no standard criteria to accurately predict disease outcomes. In this study we investigated the cross-sectional relationship between disease phenotype and immune-modulating cytokines and chemokines in cerebrospinal fluid (CSF). We analyzed CSF from 20 DMT-naïve MS patients using Olink Proteomics’ Target 96 Inflammation panel and correlated the resulting analytes with respect to (1) disease subtype, (2) patient age and sex, (3) extent of clinical disability, and (4) MRI segmental brain volumes. We found that intrathecal IL-4 correlated with higher Expanded Disability Status Scale (EDSS) scores and longer 25-foot walk times, and CD8A correlated with decreased thalamic volumes and longer 9-hole peg test times. Male sex was associated with higher FGF-19 expression, and Tumefactive MS with elevated CCL4. Several inflammatory markers were correlated with older age at the time of LP. Finally, higher intrathecal IL-33 correlated with increased MS lesion burden and multi-compartment brain atrophy. This study confirms immune heterogeneity underlying CSF profiles in MS, but also identifies several inflammatory protein biomarkers that may be of use for predicting clinical outcomes in future algorithms.
