Abstract: Bacillus amyloliquefaciens subsp. plantarum strain Fito_F321 is a naturally occurring strain in vineyard, with the ability to colonise grapevine and which unveils a naturally antagonistic potential against phytopathogens of grapevine, including those responsible for the Botryosphaeria dieback, a GTD disease. Herein we report the draft genome sequence of B. amyloliquefaciens subsp. plantarum Fito_F321, isolated from the leaf of Vitis vinifera cv. Merlot at Bairrada appellation (Cantanhede, Portugal). The genome size is 3,856,229 bp, with a GC content of 46.54% that contains 3697 protein-coding genes, 86 tRNA coding genes and 5 rRNA genes. The draft genome of strain Fito_F321 allowed to predict a set of bioactive compounds as bacillaene, difficidin, macrolactin, surfactin and fengycin that due to their antimicrobial activity are hypothesized to be of utmost importance for biocontrol of grapevine diseases. Abstract: International audience; Remote sensing techniques are particularly suitable for geological studies in arid regions such as the Arab world. Here, we present an overview of the main applications of satellite sensors to geology for geological mapping, tectonic and structural geology, hydrogeology, mining geology, geoarchaeology, oil and gas prospection, earthquake and seismicity, landslides and coastal erosion. The main characteristics of the sensors are discussed. In addition, three examples of modern remote sensing imagery show geology in Arab countries: The Dead Sea Fault in the Middle East as seen with the large-scale ALOS PALSAR (ScanSAR mode) image, the Bou Azzer Anticline (Morocco) viewed by multispectral Landsat TM and the evolution of the coastal front in Nouakchott (Mauritania), monitored with multi-temporal Landsat data.

Abstract: Due to the high surface to volume ratio, nanowire based components benefit from new properties typical of the nanoscale. ZnO nanowires have already proved their usefulness in the realization of multiple electronic components, such as FET transistors, gas detectors, photodetectors, LEDs or even solar cells. ZnO nanowires have also shown themselves to be very promising UV detectors thanks to their significant photoconductive gain, as high as G =10^8 [1]. This makes them suitable for single photon detection, or at least detection of very dim light. The main current drawback is the recovery time (minimum time between 2 detections) which we develop last hours. The device we developed is a good candidate for opto-electronic applications. Our device is a photodetector with ohmic contact and it behaves like a transistor. Our experiments stress out the importance of surface effect on the electrical by taking measurements in different atmospheres (oxygen, air, vacuum and argon). These surface states are the reason for the existence of a photoconductive gain, we obtain a maximum gain of G =6.10^6. In counterpart of this great gain, the persistence of the photocurrent (which can last up to several hours) prevents the device from operating at high frequency. Abstract: This article deals with the elaboration and the characterization of an innovative 100% plant-based green composite made solely of beet pulp (BP) and potato starch (S). Using this type of material in insulation applications seems a good solution to reduce the CO₂ gas emissions in building. The influence of the starch amount on composite characteristics was studied. Four mixtures were considered with different S/BP mass ratios (0.1, 0.2, 0.3 and 0.4). The physical properties of these materials were studied in terms of porosity, apparent and absolute densities, thermal conductivity, and hygric properties. The influence of humidity content on acoustical properties was studied as a function of frequency. Test results show a real impact of both starch and humidity contents on the hygrothermal and acoustical properties of the studied material due to the porosity. The composite with the lowest amount of starch (S/BP = 0.1) seems to be the optimal composition in terms of the hygrothermal and acoustical behaviors.

Abstract: International audience; Eight undescribed triterpenoid saponins, scabiostellatosides A-H, together with three known compounds were isolated from the whole plant of Scabiosa stellata L. Their structures were established by spectroscopic analyses (1D, 2D-NMR and HRESIMS) and chemical methods. Scabiostellatosides A-H were evaluated for their cytotoxicity against human fibrosarcoma cell line (HT1080). Scabiostellatoside F, a heptasaccharide derivative of oleanolic acid, exhibited moderate cytotoxicity against HT1080 cell line with IC50 value of 12.0 μM.

Abstract: This article deals with the influence of both morphology and amount of Typha on hygrothermal behavior of a Typha–clay composite for building application. An agromaterial containing the fiber mix of Typha Australis and clay was made in three samples: three fiber mixtures were prepared with different amounts Typha and cut type (transversal or longitudinal). The physical properties of these materials were studied in terms of porosity, apparent and absolute density, thermal conductivity, and hygric properties. Results show a real impact of the Typha fraction type and its volume content on hygrothermal properties of the studied material due to the porosity. The transversal fraction of Typha (80% in volume weight) seems to be the optimal composition for a better hygrothermal behavior.

Abstract: Polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are two families of hydrophobic compounds sharing some similar environmental properties. Previous studies have demonstrated that these compounds could affect fish life-history traits. However, few studies have focused on environmental situations. This thesis is divided into three parts. The first part aimed to evaluate the effects of a realistic mixture of PCBs and PBDEs on the life-history traits of zebrafish. The results showed a slower growth, but to a larger asymptotic length, and delayed spawning probability in exposed fish. In addition, offspring issued from early spawning events of exposed fish exhibited a lower larval survival under starvation condition. The second part aimed to identify, on the basis of a bioenergetic model (DEB), the physiological modes of action (PMoA) of PCBs and PBDEs. Two potential PMoAs have been revealed: the first one was through an increase of the fraction of energy allocated to somatic maintenance and growth and the second one was through an increase of the cost of production of an egg. The third part focused on the population dynamical consequences of the individual life-history effects of PCBs and PBDEs. The results showed a lower abundance, a higher biomass and a higher risk of overexploitation in exposed population compared to control population. The results of this thesis could be a helpful tool for sustainable management of fish stocks that take into account the effects of multiple contaminations that are added to the fishing pressure.; Les polychlorobiphényles (PCB) et les polybromodiphényléthers (PBDE) sont deux familles de composés hydrophobes partageant certaines propriétés physico-chimiques. Des travaux antérieurs ont montré que ces composés pouvaient affecter les traits d’histoire de vie des poissons. Il s’avère cependant que les études approchant des situations environnementales sont rares. Cette thèse se divise en trois parties. La première partie s’est attachée à évaluer les effets d’une exposition à un mélange réaliste de PCB et de PBDE sur les traits d’histoire de vie du poisson-zèbre. Les résultats ont montré une taille asymptotique plus élevée, une probabilité de ponte retardée chez les individus exposés et une faible survie des larves F1 en situation de jeûne issues des pontes précoces des individus exposés. La seconde partie a cherché à identifier, sur la base d’un modèle bioénergétique (DEB), les modes d’action physiologique (PMoA) de PCB et PBDE. Deux PMoAs probables ont été révélés: le premier est lié à l’augmentation de la fraction d’énergie allouée à la maintenance et la croissance somatique et le deuxième est lié à l’augmentation des coûts de production d’un œuf. La troisième partie de la thèse s’est intéressée à évaluer les conséquences de la contamination sur la dynamique de population de poissons. Les résultats ont montré une réduction de l’abondance numérique, une augmentation de la biomasse et un grand risque de surexploitation chez la population exposée, Les résultats de cette thèse pourraient constituer un outil d’aide à une gestion durable des stocks halieutiques qui prennent en compte les effets de multiples contaminations qui se surajoutent à la pression de pêche.

Abstract: International audience; Hemp-concrete is a green material which has become nowadays highly recommended in the construction field. It finds applications as internal or external thermal insulator in wooden frame walls. At wall scale, studies proved that using Hemp-concrete in building envelope can improve indoor hygrothermal comfort. However, at building scale, hemp-concrete is scarcely studied. In that context, a French building in Grand-Est region, Champagne-Ardennes, employing hemp-concrete as external insulator is selected and studied. An apartment is monitored for several months. Indoor temperatures, relative humidities, thermal heat flux as well as external weather conditions are measured using sensors installed inside the apartment and a weather station at the building roof. Measurements underline the hemp-concrete ability to dampen external weather conditions by showing good results for both indoor temperature and relative humidity. Experimental approach is then coupled with a numerical validation at the wall and room scales using SPARK simulation tool. Investigations are conducted on thermal heat flux through the wall, indoor office air temperature, and relative humidity. Results show a good agreement between numerical values and experimental measurements.

Abstract: International audience; The aim of the present study was to investigate the neuro-soothing activity of a water-soluble hydrolysate obtained from the red microalgae Rhodosorus marinus Geitler (Stylonemataceae). Transcriptomic analysis performed on ≈100 genes related to skin biological functions firstly revealed that the crude Rhodosorus marinus extract was able to significantly negatively modulate specific genes involved in pro-inflammation (interleukin 1α encoding gene, IL1A) and pain detection related to tissue inflammation (nerve growth factor NGF and its receptor NGFR). An in vitro model of normal human keratinocytes was then used to evaluate the ability of the Rhodosorus marinus extract to control the release of neuro-inflammation mediators under phorbol myristate acetate (PMA)-induced inflammatory conditions. The extract incorporated at 1% and 3% significantly inhibited the release of IL-1α and NGF secretion. These results were confirmed in a co-culture system of reconstructed human epithelium and normal human epidermal keratinocytes on which a cream formulated with the Rhodosorus marinus extract at 1% and 3% was topically applied after systemic induction of neuro-inflammation. Finally, an in vitro model of normal human astrocytes was developed for the evaluation of transient receptor potential vanilloid 1 (TRPV1) receptor modulation, mimicking pain sensing related to neuro-inflammation as observed in sensitive skins. Treatment with the Rhodosorus marinus extract at 1% and 3% significantly decreased PMA-mediated TRPV1 over-expression. In parallel with these biological experiments, the crude Rhodosorus marinus extract was fractionated by centrifugal partition chromatography (CPC) and chemically profiled by a recently developed 13C NMR-based dereplication method. The CPC-generated fractions as well as pure metabolites were tested again in vitro in an attempt to identify the biologically active constituents involved in the neuro-soothing activity of the Rhodosorus marinus extract. Two active molecules, namely, γ-aminobutyric acid (GABA) and its structural derivative GABA-alanine, demonstrated a strong capacity to positively regulate skin sensitization mechanisms related to the TRPV1 receptors under PMA-induced inflammatory conditions, therefore providing interesting perspectives for the treatment of sensitive skins, atopia, dermatitis, or psoriasis.

Abstract: International audience; Cyclic nucleotide phosphodiesterase type 4 (PDE4), that controls intracellular level of cyclic nucleotide cAMP, has aroused scientific attention as a suitable target for anti-inflammatory therapy in respiratory diseases. Here we describe the development of two families of pyridazinone derivatives as potential PDE4 inhibitors and their evaluation as anti-inflammatory agents. Among these derivatives, 4,5-dihydropyridazinone representatives possess promising activity, selectivity towards PDE4 isoenzymes and are able to reduce IL-8 production by human primary polymorphonuclear cells.

Abstract: Isotopic labeling of plants is useful in tracking the fate of carbon (C) from different plant parts in a soil-plant system when these parts decompose simultaneously. Pulse labeling is a relatively simple technique and is amenable for use in the field. Therefore, we evaluated a 13CO2 pulse-labeling method to label crop plants under subtropical field conditions for simultaneous root and shoot decomposition studies. Wheat (Triticum aestivum L.), pea (Pisum sativum L.), and vetch (Vicia sativa L.) plants were grown inside polyvinyl chloride (PVC) cylinders and pulse labeled once a week for a total of 11 times. After harvest, ‘’paired’’ treatments were designed by combining 13C-labeled shoots with unlabeled roots and unlabeled shoots with 13C-labeled roots, resulting in six treatments (2 combinations × 3 species), plus an unamended control treatment. The 13C enrichment of plant parts, chemical fractions, 13C recovery, and distribution in roots, shoots, and soil were determined. Soil CO2 emissions were measured continuously by the alkaline trap method for 180 days. Plant dry matter production and chemical composition were not modified by 13C labeling. The maximum level of 13C enrichment (δ13C) in plants was +495 ‰ in wheat, +426 ‰ in pea, and +378 ‰ in vetch plants. All three crops showed similar patterns of 13C distribution in the following order: shoots > roots > soil. On average, 81 to 89 % of the recovered 13C was in the shoots, 7 to 14 % was in the roots, and 2.7 to 4.3 % was in the soil. The rate of C mineralization and cumulative C mineralization were not different between ‘’paired’’ treatments of the three crops, showing that the paired treatments were equally degradable. The pulse-labeling technique used under field conditions allowed for production of sufficiently labeled wheat, pea, and vetch plants. Therefore, it is a practical approach with respect to resource demand (tracer and labor costs), and it is suitable for in situ labeling.

Abstract: International audience; Glucosinolates (GLs) constitute a class of plant secondary metabolites that are characteristic of the order Brassicales. They each contain a common hydrophilic moiety connected to a mostly hydrophobic side chain whose constitution is the most frequent structural variant. Their transformations by myrosinases lead to intensively studied and highly reactive compounds of biological relevancy. In other respects, the enzymatic desulfation of GLs produces derivatives (DS-GLs) that are useful for GL analysis. A collection of 31 compounds, GLs and DS-GLs, representing 17 different side chains was established in order to report accurate descriptions of the molecules’ 1H-, 13C-, and 15N-NMR parameters. The descriptions of the 1H-NMR spectra were achieved using the PERCH software, which accurately analyzed the complex coupling patterns that arose from strongly coupled nuclei. The chemical shift assignments were supported by 2D COSY, HSQC, and HMBC spectra. The impact of desulfation and the influence of the nature of the side chains on the chemical shift values are discussed. The results of the spectroscopic analysis and the 3D chemical-structure models of the studied molecules were grouped in structure-and-data-format (SDF) files. The NMR parameters were also collected in a simple text file, a spreadsheet file, and a relational database.

Abstract: International audience; Early detection of malignant tumours and, especially, micrometastases and disseminated tumour cells is still a challenge. In order to implement highly sensitive diagnostic tools we demonstrate the use of nanoprobes engineered from nanobodies (single-domain antibodies, sdAbs) and fluorescent quantum dots (QDs) for single-and two-photon detection and imaging of human micrometastases and disseminated tumour cells in ex vivo biological samples of breast and pancreatic metastatic tumour mouse models expressing human epidermal growth factor receptor 2 (HER2) or carcinoembryonic antigen (CEA). By staining thin (5–10 µm) paraffin and thick (50 µm) agarose tissue sections, we detected HER2-and CEA-positive human tumour cells infiltrating the surrounding tissues or metastasizing to different organs, including the brain, testis, lung, liver, and lymph nodes. Compared to conventional fluorescently labelled antibodies the sdAb-HER2-QD and sdAb-CEA-QD nanoprobes are superior in detecting micrometastases in tissue sections by lower photobleaching and higher brightness of fluorescence signals ensuring much better discrimination of positive signals versus background. Very high two-photon absorption cross-sections of QDs and small size of the nanoprobes ensure efficient imaging of thick tissue sections unattainable with conventional fluorescent probes. The nanobody–QD probes will help to improve early cancer diagnosis and prognosis of progression by assessing metastasis.

Open access article Even though NMR has found countless applications in the field of small molecule characterization, there is no standard file for the NMR data relevant to structure characterization of small molecules. A file format is introduced to associate the NMR parameters extracted from 1D and 2D spectra of organic compounds to the assigned chemical structure. These NMR parameters, which we shall call NMReDATA, include chemical shift values, signal integrals, intensities, multiplicities, scalar coupling constants, lists of 2D correlations, relaxation times and diffusion rates. The file format is an extension of the existing SDF (Structure Data Format), which is compatible with the commonly used MOL format. The association of an NMReDATA file with the raw and spectral data from which it originates constitutes an NMR record. This format is easily readable by humans and computers and provides a simple and efficient way for disseminating results of structural chemistry investigations, automating the verification of published result, and for assisting the constitution of highly needed open-source structural databases.

In Barbarea vulgaris R.Br. (Brassicaceae), the dominant glucosinolate in the investigated “G-type” is glucobarbarin, (S)-2-hydroxy-2-phenylethylglucosinolate. Formation of the nitrile from glucobarbarin was observed in vitro, while a previously suggested thioamide (synonym thionamide) was not confirmed. Resedine (5-phenyl-1,3-oxazolidin-2-one) was detected after glucobarbarin hydrolysis in crushed B. vulgaris leaves and siliques, but not in intact parts. The abundance increased for several hours after completion of hydrolysis. The corresponding 1,3-oxazolidine-2-thione (OAT), with the common name barbarin, was also formed, and appeared to be the precursor of resedine. Addition of each of two non-endogenous OATs, (S)-5-ethyl-5-methylOAT and (R)-5-vinylOAT (R-goitrin), to a leaf homogenate resulted in formation of the corresponding 1,3-oxazolidin-2-ones (OAOs), confirming the metabolic connection of OAT to OAO. Formation of OAOs was inhibited by prior brief heating of the homogenate, suggesting enzyme involvement. We suggest the conversion of OATs to OAOs to be catalyzed by an enzyme (“oxazolidinethionase”) responsible for turnover of OAT formed in intact plants. Resedine had been reported as an alkaloid from another species - Reseda luteola L. (Resedaceae) - naturally containing the glucosinolate glucobarbarin. However, resedine was not detected in intact R. luteola plants, but formed after tissue damage. The formation of resedine in two families suggests a broad distribution of putative OATases in the Brassicales; potentially involved in glucosinolate turnover that needs myrosinase activity as the committed step. In agreement with the proposed function of OATase, several candidate genes for myrosinases in glucosinolate turnover in intact plants were discovered in the B. vulgaris genome. We also suggest that biotechnological conversion of OATs to OAOs might improve the nutritional value of Brassicales protein. HPLC-MS/MS methods for detection of these glucobarbarin products are described.

Abstract: The growing interest in the preservation of our environment stimulates solutions for developing less impacting materials.Thus, the development of composites from renewable resources and recyclable end-of-life resources seems an interesting alternative. This motivated the reason to focus our work on the use of agricultural by-products and waste recycling. Our purpose is the reprocessing of composites based on polypropylene (PP) reinforced with olive husk flour (OHF) at various load rates with adding PP-g-MA as a compatibilizer by six cycles of extrusion.The Young modulus of the composites has slightly increased with the rise of the OHF rate and the number of cycles. An FTIR analysis shows two types of degradation after reprocessing: thermal degradation characterized by a splitting of chains and thermal oxidation justified by the increase in the carbonyl index even when the intensity of the absorption peak has been noticed at 1690-1640 cm(-1) referring to the vinyl groups after the 6th cycle.Moreover, the TGA analysis showed a better thermal stability of the composites after the 6th cycle compared to the virgin PP. The rate of crystallization increases with the number of extrusion cycle.

Abstract: Recycling of carbon (C) and nitrogen (N) from plants into soils is decisive for maintaining soil organic matter and soil fertility. Therefore, we quantified plant biomass and C and N in the shoots and roots from the topsoil layer for a wide range of annual crops grown under subtropical conditions. We grew 26 species, 13 main crops, and 13 cover crops, in the field in standard sowing arrangements. Root biomass was recovered from the 0.00-0.20 m soil layer at flowering, and shoot biomass was measured at flowering for all crops and at maturity only for the main crops. Root dry matter (DM) exhibited an average of 14.9 ± 5.7 % of the total shoot biomass at flowering, and the mean shoot DM to root DM ratio was 6.9 (2.8-15.0) for the 26 crops considered. Leguminous species had less root DM (0.5 to 1.0 Mg ha-1) than grass species (1.1 to 2.3 Mg ha-1). The shoot C to root C ratio varied consistently with DM, while the root N to shoot N ratio varied considerably among species. Proportionally more biomass, C, and N was allocated to the root systems of grasses (Poaceae species) than non-grass species (especially Fabaceae species). The findings of this study contribute to designing rotations to include species that promote cycling of N and have high potential for adding C to the soil through roots. In this sense, the use of intercropped grasses and legumes is a promising strategy, especially for cover crops.

Abstract: International audience; In a previous study, the Comet assay was optimized for Palaemon serratus prawns in order to propose a biomarker for sperm quality in this species. However, better knowledge of its basal level and its natural variability, related to intrinsic biotic and environmental abiotic factors, is required before any relevant use of this biomarker in the field. To fulfill this goal, the present study proceeded in three steps: (i) the temporal variability of DNA integrity was followed monthly in a reference population over a 2-year period, (ii) the correlation between the main intrinsic biotic (i.e. size, weight and molting stage) and abiotic factors (i.e. water temperature) were recorded in the field, and the basal DNA integrity was assessed in order to scrutinize any confounding influence of factors unrelated to toxic response, (iii) the baseline level was used to discriminate biomarker response among different stations displaying contrasting contamination levels. The results of the two-year monitoring in the reference population revealed no correlation between the levels of spermatozoa DNA damage and temperature, body size, weight or molting stage. Only a slight variability between monthly samplings was detected. On the basis of these field-collected data, we defined a reference distribution (i.e. 52.6 ± 5.6 A.U) with a threshold value (i.e. 61.7 A.U). Finally, this threshold value proved its relevance to discriminate among stations with contrasting pollution levels around the Seine Bay. Indeed, the results suggest significant DNA damage in populations nearest the Seine estuary, a major source of contaminants in the Bay, and a lower effect in populations further away from the estuary. The overall conclusion was that the Comet assay on P. serratus spermatozoa could be a useful tool for the monitoring of the toxicological print within sperm and main globally the contamination exposure of crustaceans in marine waters

Abstract: Rev.1 is a stable live attenuated vaccine, which has significantly contributed to reduce the incidence of brucellosis due to Brucella melitensis in many countries, provided a sufficient coverage of sheep and goat populations was achieved. As regards the control of the brucellosis outbreak in Alpine ibex, an experiment to compare the safety of conjunctival Rev.1 vaccination between domestic goats and Alpine ibex was undertaken in spring 2017. Twenty-four adult animals were included in the study, divided into two batches of 6 males and two batches of 6 non-pregnant females of each species, for a period of 45 or 90 days. The Rev.1 vaccine resulted in a humoral response in all vaccinated animals, with a higher intensity in ibex compared to goats, regardless of age and sex. The risk resulted in the Rev.1 transmission from vaccinated ibex to one control animal. Despite what the genetic proximity between goat and ibex might suggest, the distribution of Rev.1 in organs and the risk of urogenital excretion were much more marked in ibex than in goats, all ages and sexes combined. These results highlight a reduced ability of Alpine ibex to contain the spread, multiplication and excretion of Rev.1 vaccine, suggesting a risk of spread of the vaccine strain in the ibex population. In nature, the uncontrolled diffusion of the vaccine strain may impair surveillance and management of this outbreak, inducing seroconversions of animals, without difference between animals infected from wild and vaccine strains., Rev.1 est un vaccin vivant atténué stable, ayant permis dans de nombreux pays, une réduction significative de l’incidence de la brucellose à Brucella melitensis, à la condition d’une couverture suffisante des populations ovines et caprines. Dans le cadre de la gestion du foyer de brucellose chez les bouquetins du Bargy, une expérimentation visant à comparer l’innocuité du vaccin Rev.1 conjonctival entre la chèvre domestique et le bouquetin des Alpes a été entreprise au printemps 2017. Au total 24 animaux adultes ont été inclus dans l’étude, répartis en deux lots de 6 mâles et deux lots de 6 femelles non gestantes de chaque espèce, puis suivis pendant une période de 45 ou 90 jours. Le vaccin Rev.1 a entraîné une réponse humorale chez tous les animaux vaccinés, avec une intensité supérieure chez les bouquetins. Contrairement à ce que la proximité génétique entre chèvre et bouquetin pouvait laisser penser, la distribution de la souche Rev.1 au sein des organes ainsi que le risque d’excrétion urogénitale ont été beaucoup plus élevés chez le bouquetin que chez la chèvre, tous âges et sexes confondus. Ce risque s’est concrétisé par la transmission de la souche vaccinale de bouquetins vaccinés à un animal témoin non vacciné. Ces travaux mettent en évidence une capacité moindre du bouquetin à contenir la diffusion, la multiplication et l’excrétion du vaccin Rev.1, suggérant un risque plus important de diffusion de cette souche vaccinale chez cette espèce sauvage. En nature, une diffusion non maitrisée de la souche vaccinale pourrait compromettre la surveillance et la gestion du foyer, en induisant des séroconversions impossibles à distinguer entre animaux infectés par les souches sauvage et vaccinale.

Abstract: Background and aims The use of summer cover crops (SCCs) as residue inputs in agricultural systems can lead to a potential environmental trade-off between the amount C and N retained and the emission of N as the potent greenhouse gas, nitrous oxide (N2O). Our objectives were to relate N2O fluxes to the quality of SCCs residues and to select SCCs species with high C and N addition and low N2O emissions.Methods We measured SCC biomass production and N2O fluxes after SCCs termination - velvet bean, pearl millet, dwarf pigeonpea, sunn hemp, showy rattlebox and jack bean - under subtropical no-till conditions.Results The N2O fluxes in the first 30 days after SCCs termination corresponded to 65% of cumulative N2O emissions measured during 165 days in 2010 and 150 days in 2011. The cumulative N2O emissions varied from 0.46 to 1.38 kg N ha(-1) and were not proportional to aboveground biomass N addition. Cumulative N2O emissions were positively correlated to water-soluble C and N and the N content of crop residues and negatively correlated to cellulose and hemicellulose content. The N2O emission factor values varied from 0.24 to 0.64% and did not differ among SCCs.Conclusions Sunn hemp and dwarf pigeonpea are the most suitable species for inclusion in crop systems because they combined high C and N input and low N2O emissions.

Abstract: International audience; Innate immunomarkers reflect both environmental contamination and fish health status, providing useful information in environmental risk assessment studies. Nevertheless, the lack of knowledge about the effect of confounding factors can lead to data misinterpretation and false diagnoses. The aim of this study was to evaluate the impact of three confounding factors (season, sex and body size) on three-spined stickleback innate immunomarkers in laboratory conditions. Results shown strong seasonal variations in stickleback innate immunomarkers, with higher immune capacities in late winter-early spring and a disturbance during the spawning period in late spring-summer. Sex and body size had a season dependant effect on almost all tested immunomarkers. Reference ranges were established in laboratory-controlled conditions (i.e. laboratory reference ranges) and compared with data obtained from in vivo chemical expositions. The predictive power of the statistical model depended on the immunomarker, but the control data of the in vivo experiments, realized in same laboratory conditions, were globally well include in the laboratory reference ranges. Moreover, some statistical effects of the in vivo exposures were correlated with an augmentation of values outside the reference ranges, indicating a possible harmful effect for the organisms. As confounding factors influence is a major limit to integrate immunomarkers in biomonitoring programs, modelling their influence on studied parameter may help to better evaluated environmental contaminations.

Abstract: The use of natural fibers such as hemp (Cannabis sativa L.) as substitutes for nonrenewable fibers increases the life cycle performance of composite materials. The management of retting in fields as a natural pretreatment prior to fiber extraction remains challenging due to a lack of knowledge about the relative importance of environmental and biotic factors, which continually interact under field conditions. Here, we studied the dynamics of hemp retting under controlled air temperature and humidity conditions and with simulated rain. We tracked the color and infrared spectral absorbance of the stem surface, the chemical composition and microbial enzyme activities of the bast tissues, and the stem architecture over 42 days at 15 °C. Color changes on the stem surfaces were the first indicators of retting progress, with a significant decrease in the L*, a* and b* values (CIELAB) from day 14 onward. These findings were closely correlated with the surface colonization progress as revealed by scanning electron microscopy, the changes in enzymatic activities and the decohesion of the bast tissues. Additional investigations are needed to study other environmental scenarios to provide an accurate assessment of the retting process over time.

Abstract: A previously undescribed triterpenoid saponin, 3-O-[α-l-rhamnopyranosyl-(1→2)-{β-d-glucopyranosyl-(1→6)-}β-d-galactopyranosyl-(1→2)-β-d-glucuronopyranosyl]-sophoradiol (1), in addition to twenty-nine known constituents (2–30) were isolated from the aerial parts of Genista numidica Spach. Structures elucidation was performed by comprehensive 1D- and 2D-NMR analyses and HRESIMS. The extracts, fractions and isolated compounds were evaluated for their antibacterial, antioxidant and tyrosinase inhibitory activities. The experimental findings indicated that genistin (16), isosalipurpol (27), and koaburaside (29) have moderate to low antibacterial activity against E. faecalis, S. aureus, S. epidermidis and P. aeruginosa bacteria with MICs ranging from 31.2 to 125 μg/mL. Compounds 19 and 27 exhibited a good antiradical activity potential (IC50 11.8 and 11.1 μg/mL, respectively). Only compounds 23, 27 and 28 exhibited low inhibitory effect against mushroom tyrosinase (IC50 from 90.2 to 225.6 μg/mL).

Abstract: International audience; This study presents the chemical profile investigation of a 70% ethanol extract obtained from Scabiosa stellata, a medicinal herbaceous traditionally used to treat heel cracks. A 13C NMR-based dereplication methodology was firstly applied on centrifugal partition chromatography-generated fractions in order to quickly identify the major compounds of the extract. The dereplication process was then completed by semi-preparative high-performance liquid chromatography in order to identify unknown or minor compounds. Two new bis-iridoids, namely 7-O-caffeoyl-sylvestroside I (1) and 7-O-(p-coumaroyl)-sylvestroside I (2), together with ten known compounds (3 − 12) were isolated. Their structures were elucidated by spectroscopic methods including NMR and HR-ESI-MS. The antibacterial, anti-tyrosinase and DPPH radical scavenging activities of the crude extract, fractions, and isolated compounds were evaluated. A significant antibacterial activity was observed for nine isolated compounds, particularly 1 and 2 which yielded MIC values of 31.2 μg/mL against Enterococcus faecalis and 62.5 μg/mL against Staphylococcus epidermidis. The cytotoxic activity of these new bis-iridoids was evaluated on a fibrosarcoma cell line (HT1080) and only compound 1 exhibited a moderate cytotoxic activity (IC50 35.9 μg/mL).

Abstract: Atomic force microscopy (AFM) experiments with functionalized tips are currently one of the most powerful tools to locally measure adhesion forces via single-molecule force spectroscopy (SMFS) measurements.The main difficulty is to precisely control the attachment of biomolecules to the cantilever. Different chemistry procedures have been developed including the use of spacer molecules. Even if a process works well for small biomolecules such as antibodies, issues remain regarding nanoparticles or larger objects such as cellulose nanocrystals because it is difficult to precisely control their coverage and homogeneity. In this work, an original procedure based on the Langmuir-Blodgett (LB) technique was implemented for lever functionalization with cellulose nanocrystals and compared with classical chemical strategies. LB shows to be almost 6.0-fold more efficient than chemical procedure in terms of cellulose nanocrystals coverage attachment.Moreover, the LB technology provides advantage of not requiring linker molecules, which could have detrimental effects such as overestimation of the interaction force. The structural characterization and SMFS measurements of lignocellulosic polymers show that this strategy enables the precise control of the lever coverage, which improves the accuracy of the adhesion measurements. Such methodology is expected to strongly impact the AFM tip/tipless functionalization and SMFS measurements in different fields.

Abstract: Increasing the accumulation of organic carbon (C) in soils is a crucial challenge both for soil fertility and for climate change mitigation. Heterotrophic microbial communities are key drivers of C cycling in the soil and are influenced by cultural practices, among other factors. However, whether changes in microbial communities in turn affect their C degradation functions is not well understood. Here, we studied the effects of prior soil management on the microbial taxonomic composition and activity of soils amended with wheat litter. Prior soil management was either conventional (CONV) (i.e., full inversion ploughing) or reduced tillage (RT) during a 5 year period in the same loamy soil in northern France. Soil samples taken from the top 5 cm of field plots were incubated with C-13-labelled litter of either flowering wheat or mature wheat for 29 days at 15 degrees C. We measured the C-CO2 and C-13-CO2, microbial biomass C (MBC) and C-13, and hydrolytic enzyme activities during decomposition. The initial bacterial and fungal community diversity was studied via high-throughput sequencing of ribosomal genes. The results showed that the MBC in the RT soil was initially 1.5-fold greater than that in the CONV soil; contrasting taxonomic compositions were also recorded. The soil biotic legacy impacted the degradation functions when the soils were amended with wheat litter. Compared with that in the CONV soil, the enzymatic efficiency of microorganisms in the RT soil increased by 49% and 61% in the presence of mature and flowering wheat litter, respectively. Enzyme efficiency was positively correlated with microbial litter C use efficiency (CUE) (r = 0.92, P-Value < 0.001) but negatively associated with the priming effect (PE) (r = -0.85, P value < 0.001) across all soils and litter treatments. These findings demonstrated that the RT soil benefited both from an increase in litter C incorporated in the microbial biomass and from a reduction in soil C loss due to the PE, regardless of the quality of the decomposed litter. Our study indicated that agricultural practices such as RT, which enriches the amount of soil organic C (SOC) in the topsoil layer, can lead to positive feedback against C stabilization functions.

Abstract: Biogeochemical cycles of carbon (C), nitrogen (N), and phosphorus (P) are naturally coupled in terrestrial ecosystems by elemental stoichiometry of plant autotrophy and of soil biologic heterotrophy. The shift from natural to cropped intensive systems led to major changes in crop land use, nutrient availability, and soil management. This chapter presents the fundamental processes of C, N, and P coupling and how the intensification of agriculture has “opened” and “decoupled” the terrestrial nutrient cycles, leading to major environmental impacts, particularly with nitrogen. Intensification of agriculture has reduced the diversity of plant species and lowered organic carbon inputs to soils, increased the N saturation of agrosystems by fertilization, and to a lesser extent P, the territorial specialization of agriculture has increased nutrient fluxes and imbalances both at regional and world scales. These major changes have modified in turn the nature and amount of plant biomass recycled to soils, the diversity and activities of soil communities, and affected the balance of nutrient mineralization to immobilization by heterotrophs. Lack of synchrony between nutrients supply and plant demand, enhanced by the simplification of crop rotations, is also a major cause of decoupling between cycles. Therefore, new farming strategies should aim, at plot scale, at diversifying crops, increasing the presence of legumes in rotations, and the annual soil occupancy by crops, and at increasing, at farm and regional scales, the complexity of cropping systems (particularly by mixing arable and livestock farming) to enhance local recycling of nutrients and decrease the use of synthetic fertilizers.

Abstract: International audience; A new in silico method is introduced for the dereplication of natural metabolite mixtures based on HMBC and HSQC spectra that inform about short-range and long-range H–C correlations occurring in the carbon skeleton of individual chemical entities. Starting from the HMBC spectrum of a metabolite mixture, an algorithm was developed in order to recover individualized HMBC footprints of the mixture constituents. The collected H–C correlations are represented by a network of NMR peaks connected to each other when sharing either a 1H or 13C chemical shift value. The network obtained is then divided into clusters using a community detection algorithm, and finally each cluster is tentatively assigned to a molecular structure by means of a NMR chemical shift database containing the theoretical HMBC and HSQC correlation data of a range of natural metabolites. The proof of principle of this method is demonstrated on a model mixture of 3 known natural compounds and then on a real-life bark extract obtained from the common spruce (Picea abies L.).

Abstract: Two novel magnesium(II) tetraphenylporphyrin-based six-coordinate complexes; bis(hexamethylenetetramine)(5,10,15,2O tetrakis[4(benzoyloxy)phenyl]porphinato) magnesuim(II) (1) and bis(1,4-diazabicyclo(2.2.2)octane) (5,10,15,2O-tetrakis[4- (benzoyloxy)phenyl]porphinato)magnesium(II) (2) have been synthesised and confirmed by proton nuclear magnetic resonance, mass spectrometry, elemental analysis and IR spectroscopy. Both crystal structures were determined and described by single crystal X-ray diffraction analysis and Hirshfeld surfaces computational method. All Mg(II) atoms are surrounded by four porphyrin nitrogen atoms and two axial ligands coordinated to the metal ion through one nitrogen atom, forming a regular octahedron. In both complexes, molecular structures and three-dimensional framework are stabilised by inter-and intramolecular C–H ⋯ O and C–H ⋯ N hydrogen bonds, and by weak C–H ⋯ Cg π interactions. UV–visible and Fluorescence investigations, respectively, show that studied complexes have a strong absorption in red part and exhibit an emission in the blue region. The HOMO-LUMO energy gap values, modelled using the DFT approach, indicates that both studied compounds can be classified as semiconductors. The role of these complexes as novel antibacterial agents was also performed.

Abstract: Crop residues left as mulch on the soil surface generally increase the risk of ammonia (NH3) volatilization from fertilizer-N, but the roles of mulch as a physical barrier and as a biological support of N transformation are not well known, particularly in response to straw removal. Thus, our goal was to assess how NH3 volatilization from urea-N was affected by different amounts of residues left on the soil surface after harvest in the sugarcane crop by mimicking different rates of straw removal. We ran a replicated field experiment during two consecutive years in the first ratoon sugarcane with 4 amounts of straw (0, 4, 8 and 12 Mg dry matter ha−1) and with 0 or 100 kg urea-N ha−1. Over approximately 2 weeks, we measured the NH3 emissions, levels of inorganic N in soil and water-soluble total N and NH4+ of straw. The dynamics of N in the soil and in straw supported the hypothesis that mulch acts as a physical barrier to the incorporation of urea in soil but also that NH3 emissions occurred directly from the mulch. The NH3 losses were different according to rain pattern of each year, but we showed that the emissions of NH3 were higher with higher amounts of straw (12 > 8 > 4 > 0) for both years (9.2–27.6% in 2016 and 30.7–60.8% in 2017). These results suggest that straw removal contributes to a reduction in NH3-N losses, but since total straw removal is not a recommended practice, N fertilization strategies should be implemented in systems with residue mulches on the soil surface to prevent N loss and to improve the environmental footprint of these cropping systems.

Abstract: The distribution of lignin, 8-5′ and 8-8′ linked lignin substructure, and noncellulosic polysaccharides in hemp (Cannabis sativa L.) phloem fibers were explored based on histochemical and immunological methods. Ultraviolet absorption and potassium permanganate staining were observed mainly in the compound middle lamella (CML) and S1 layers, and rarely in the G-layer of phloem fibers, suggesting that lignin concentration is high at the CML and S1 layers, and very low at the G-layer of hemp fibers. Acriflavine staining, uniform KM1 labeling (8-5′ linked lignin substructure), and no KM2 labeling (8-8′ linked structure) were observed in the G-layer, suggesting that there is a small amount of lignin-like compound with 8-5′ linked structure in the G-layer. In addition, some fiber cells showed a multilayered structure. Uniform arabinogalactan protein (AGP) labeling was observed on the S1 layers and G-layers using JIM14, but little appeared in the CML of hemp fibers, indicating that these layers of the phloem fibers contain AGP. Immunogold labeling of xylan (LM11) and glucomannan (LM21) showed that xylan and glucomannan were mainly present in the S1 layers and the G-layers, respectively. In some phloem fibers, LM21 immunofluorescence labeling showed multilayered structure, suggesting the heterogeneous distribution of glucomannan.

Abstract: International audience; Automatic structure elucidation of small molecules by means of the “logic for structure elucidation” (LSD) software is introduced in the context of the automatic exploitation of chemical shift correlation data and with minimal input from chemical shift values. The first step in solving a structural problem by means of LSD is the extraction of pertinent data from the 1D and 2D spectra. This operation requires the labeling of the resonances and of their correlations; its reliability highly depends on the quality of the spectra. The combination of COSY, HSQC, and HMBC spectra results in proximity relationships between nonhydrogen atoms that are associated in order to build the possible solutions of a problem. A simple molecule, camphor, serves as an example for the writing of an LSD input file and to show how solution structures are obtained. An input file for LSD must contain a nonambiguous description of each atom, or atom status, which includes the chemical element symbol, the hybridization state, the number of bound hydrogen atoms and the formal electric charge. In case of atom status ambiguity, the pyLSD program performs clarification by systematically generating the status of the atoms. PyLSD also proposes the use of the nmrshiftdb algorithm in order to rank the solutions of a problem according to the quality of the fit between the experimental carbon‐13 chemical shifts, and the ones predicted from the proposed structures. To conclude, some hints toward future uses and developments of computer‐assisted structure elucidation by LSD are proposed.

Abstract: International audience; A water-based polyurethane (PUR) acrylate water emulsion was selected as a radiation curable matrix for preparing nanocomposites including cellulose nanocrystals (CNC) prepared by controlled hydrolysis of Ramie fibers. Cross-linking polymerization of samples prepared in the form of films or of 1 mm-thick bars was either initiated by exposure to the 395 nm light of a high intensity LED lamp or by treatment with low energy electron beam (EB). The conversion level of acrylate functions in samples submitted to increasing radiation doses was monitored by Fourier Transform Infrared Spectroscopy (FTIR). Differential Scanning Calorimetry (DSC) and Dynamic Mechanical Analysis (DMA) were used to characterize changes in the glass transition temperature of the PUR-CNC nanocomposites as a function of acrylate conversion and of CNC content. Micromechanical testing indicates the positive effect of 1 wt% CNC on Young's modulus and on the tensile strength at break (σ) of cured nanocomposites. The presence of CNC in the PUR acrylate matrix was shown to double the σ value of the nanocomposite cured to an acrylate conversion level of 85% by treatment with a 25 kGy dose under EB, whereas no increase of σ was observed in UV-cured samples exhibiting the same acrylate conversion level. The occurrence of grafting reactions inducing covalent linkages between the polysaccharide nanofiller and the PUR acrylate matrix during the EB treatment is advanced as an explanation to account for the improvement observed in samples cured under ionizing radiation.

Abstract: From the aerial parts of Salvia barrelieri Etl. (Lamiaceae), 12 compounds including a new triterpene, 3β-acetoxy-olean-18-ene-2α-ol, were isolated. Their structures were established by the combination analyses of spectroscopy including 1D-, 2D-NMR and HRESIMS and in comparison with the reported data in the literature. The antibacterial activity of these compounds was evaluated by bioautography on Staphylococcus aureus followed by the determination of MIC values by serial dilution technique against five bacteria. Three compounds were active against Enterococcus faecalis, S. aureus, Staphylococcus epidermidis, Escherichia coli and Pseudomonas aeruginosa (MIC 15.1 to 125 μg/mL). Two compounds showed moderate DPPH radical scavenging activity (IC50 79.1 and 21.2 μg/mL). These compounds did not show significant tyrosinase inhibitory activities (IC50> 1.5 mg/mL). Their cytotoxic activity was evaluated against promyelocytic leukaemia (HL60), human erythromyeloblastoid leukaemia (K562) and fibrosarcoma (HT1080) cell lines and four compounds exhibited a moderate cytotoxic activity (IC50 28.75 to 85.0 μM).

Abstract: Crop residues left on the soil surface as mulch greatly influence the fate of pesticides in conservation agricultural practices because most of the applied pesticide is intercepted by mulch before passing to the soil. Modelling of pesticide losses from wash-off and leaching will greatly improve our understanding of the environmental consequences of pesticides in these systems. The PASTIS model, which simulates water transfer, mulch decomposition, and pesticide dynamics, was adapted in this new version to model the interactions between pesticides and mulch in order to simulate the impact of mulch on pesticide dynamic. Parameters of mulch dynamics and pesticide degradation and retention processes were estimated using independent incubation experiments. The PASTIS model was tested with experimental laboratory data that were obtained from two pesticides (Glyphosate and s-metolachlor) applied to soil columns where mulch composed of maize and dolichos was placed at the soil surface impacted by two rain intensities (a high and infrequent intensity and a light and frequent intensity). Simulations indicated good agreement between simulated and experimental values. After 1 day, 45–46% of the pesticides leached from the mulch and 54–55% remained in the mulch for both pesticides and both rain intensities. During the experiment, pesticide wash-off was greater for the high and infrequent rain (56–57%) compare to light and frequent rain (39–45%) for both pesticides. A smaller amount of S-metolachlor washed off with the light and frequent rain intensity (39%) than glyphosate (45%) because of its lower desorption rate from mulch residues. Glyphosate was more degraded (37–45%) than s-metolachlor (17–37%), which agrees with preliminary incubation experiments that were used for parameter estimation. A sensitivity analysis indicated that the saturation index of mulch at which pesticides started their diffusion in the rainwater and the time of the first rainfall were the two parameters that influenced the most output variables of our model. This study suggests that the PASTIS model developed for pesticide dissipation in mulch is a useful tool to evaluate the potential risk of pesticide leaching to the groundwater in conservation agriculture systems.

Abstract: International audience; The development of new drugs will certainly benefit from an ever improving knowledge of the living beings chemistry. However, identification of drugable molecules within the immense biodiversity of forests, soils or oceans still requires considerable investments in technical equipments, time and human resources. An important part of this process is the quick identification of known substances in order to concentrate the efforts on the discovery of new ones. A range of “dereplication” procedures are currently emerging to meet this challenge as key strategies to improve the performance of natural product screening programs. Initially defined in 1990 as “a process of quickly identifying known chemotypes”, dereplication is today a not so univocal concept and has evolved over the last years in different ways. The present review covers all dereplication-related sudies in natural product research from 1990 to 2014. Its writing brought to light five distinct dereplication workflows that can be characterized by the nature of starting materials, by the selected analytical technique, and above all by the final objective. Dereplication can be used as an untargeted workflow for the rapid identification of the major compounds whatever their chemical class in a single sample or for the acceleration of bioactivity-guided fractionation procedures. In other cases dereplication is fully integrated in metabolomic studies for the untargeted chemical profiling of natural extract collections or for the targeted identification of a predetermined class of metabolites. Finally a quite distinct dereplication approach mainly based on gene-sequence analyses is frequently used for the taxonomic identification of microbial strains.

Abstract: The aim of this study was to evaluate the effects of caging constraints on multiple fish biomarkers used during ecotoxicological studies (biometric data, immune and antioxidant systems, and energetic status). Two of these constraints were linked to caging: starvation and fish density in cages, and one in relation to the post-caging handling: a short transport. Three in situ experiments were conducted with three-spined sticklebacks (Gasterosteus aculeatus). The first experiment compared the effects of three densities (low, medium, and high). The second experiment compared effects of starvation in fish fed every two days with fish that were not fed. Finally comparisons between sticklebacks which have suffered a short car transport after caging and sticklebacks killed without preliminary transport were made. The lack of food had no effect on fish energetic reserves but negatively affected their condition index and their immune system. Transport and high density induced oxidative stress, defined as an overproduction of reactive oxygen species and a stimulation of the antioxidant system. These two constraints also harmed the leucocyte viability. In order not to have any impact on ecotoxicity biomarkers during in situ experiments, it is preferable to decrease fish density in cages, prevent transport before dissections, and feed fish when the caging lasts more than two weeks.

Abstract: International audience; Securigera varia (Fabaceae) is a common herbaceous perennial plant widely growing in Europe and Asia and purposely established for erosion control, roadside planting, and soil rehabilitation. The aim of this study was to determine the radical scavenging activity of a crude methanol extract of S. varia aerial parts by using the free radical DPPH (1,1-diphenyl-2-picrylhydrazyl) and to rapidly identify the compounds involved in this activity. The crude extract was initially separated in five fractions on Diaion HP20 resin and the most active part was fractionated by Centrifugal Partition Extraction (CPE). Known compounds were directly identified by a 13C-NMR-based dereplication method. Semi-preparative high performance liquid chromatography purification experiments were further performed to identify unknown or minor active compounds. As a result, one new (13) and twelve known flavonoid glycosides together with three nitropropanoylglucopyranoses were isolated, including astragalin (1), kaempferol-3-O-(6-O-acetyl)-β-D-glucopyranoside (2), kaempferol-3,4′-di-O-β-D-glucopyranoside (3), trifolin (4), isoquercitrin (5), hyperoside (6), isovitexin (7), isoorientin (8), isovitexin 4′-O-β-D-glucopyranoside (9), apigenin 7-O-β-D-glucuronopyranoside (10), luteolin 7-O-β-D-glucuronopyranoside (11), apigenin 7-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucuronopyranoside (12), apigenin 7-O-β-D-glucopyranosyl-(1→2)-β-D-glucuronopyranoside (13), 6-O-(3-nitropropanoyl)-β-D-glucopyranoside (14), coronillin (16) and coronarian (15). 120 mg of the most active compound isoorientin against the free radical DPPH was recovered by CPE with an HPLC purity of 99%

Abstract: International audience; One new 5-alkylresorcinol glucoside (1) was isolated from leaves of Cybianthus magnus, along with 12 known compounds (2-13), isolated from four plants belonging to Myrsinaceae family. Their structures were determined on the basis of spectroscopic analysis and by comparison of their spectral data with those reported in the literature. Among the tested molecules, only compound 2 displayed a strong cytotoxic activity with IC50 values ranging between 22 and 100 μM for all cell lines tested. One new 5-alkylresorcinol glucoside (1) was isolated from leaves of Cybianthus magnus, along with 12 known compounds, isolated from four plants belonging to Myrsinaceae family (2, 3 isolated from C. magnus; 4-7, 10 and 11 isolated from Myrsine latifolia; 4, 8 and 9 isolated from Myrsine sessiflora; 6, 7, 10, 12 and 13 isolated from Myrsine congesta). Their structures were determined on the basis of spectroscopic analysis and by comparison of their spectral data with those reported in the literature. So far, only nine 5-alkylresorcinol glucosides were isolated from leaves of Grevillea robusta. Since resorcinols are known to exhibit strong cytotoxic activity, compounds 1 and 2 were tested against cell lines 3T3, H460, DU145 and MCF-7 for cytotoxicity in vitro and compounds 3-13 were tested for their antileishmanial activity. Compound 2 displayed a strong cytotoxic activity with IC50 values ranging between 22 and 100 μM for all tested cell lines. Compounds 3-13 were not active against Leishmania amazonensis amastigotes.

Abstract: The Bureau Commun de Référence (BCR) sequential extraction scheme and micro-synchrotron-based X-ray fluorescence (μ-SXRF) analysis were used to determine the Cu fractionation in a calcareous vineyard soil and a synthetic soil (mixture of seven constituents: calcite, birnessite, ferrihydrite, goethite, lignocellulosic residue, kaolinite, and quartz) at different Cu contamination rates (190, 1270, and 6350 mg kg(-1) of Cu) and aging times (1, 30, 92, and 181 days). The Cu distribution in the spiked vineyard and synthetic soils was different from the original vineyard one and was influenced by the loading level. The newly added Cu was preferentially present in the acid soluble fraction. Aging of the contaminated vineyard and synthetic soils during 6 months led to the redistribution of Cu from the weakly bound acid soluble fraction to the strongly bound reducible one. The evolution with time could satisfactorily be simulated by the Elovich diffusion model for the synthetic soils. It was less significant as less marked in the contaminated vineyard soil than in the synthetic one, even though the trends observed in both were similar. This study supported the hypothesis that "simple" synthetic models could be used to approach the Cu fractionation and its evolution with time in vineyard soils.

Abstract: The Blue Mussel (Mytilus edulis, L. 1758) is an ecologically important and commercially relevant bivalve. Because of its ability to bioconcentrate xenobiotics, it is also a widespread sentinel species for environmental pollution, which has been used in ecotoxicological studies for biomarker assessment. Consequently, numerous proteomics studies have been carried out in various research contexts using mussels of the genus Mytilus, which intended to improve our understanding of complex physiological processes related to reproduction, adaptation to physical stressors or shell formation and for biomarker discovery. Differential-display 2-DE proteomics relies on an extensive knowledge of the proteome with as many proteoforms identified as possible. To this end, extensive characterization of proteins was performed in order to increase our knowledge of the Mytilus gill proteome. On average, 700 spots were detected on 2-DE gels by colloidal blue staining, of which 122 different, non-redundant proteins comprising 203 proteoforms could be identified by tandem mass spectrometry. These proteins could be attributed to four major categories: (i) “metabolism”, including antioxidant defence and degradation of xenobiotics, (ii) “genetic information processing”, comprising transcription and translation as well as folding, sorting, repair and degradation, (iii) “cellular processes”, such as cell motility, transport and catabolism, (iv) “environmental information processing”, including signal transduction and signalling molecules and interaction. The role of cytoskeleton proteins, energetic metabolism, chaperones/stress proteins, protein trafficking and the proteasome are discussed in the light of the exigencies of the intertidal environment, leading to an enhanced stress response, as well as the structural and physiological particularities of the bivalve gill tissue.

Abstract: Hydrophobic ionic liquids (ILs) were generated by association between a cationic ester derivative of betaine and bis(trifluoromethylsulfonyl)imide (Tf2N−) or dicyanamide (Dca−) anions. Extraction of Cu(II), Ni(II), Pb(II), and Cd(II) from water was performed with these ILs at room temperature. The use of Dca− anion greatly enhances the extraction efficiency of IL phase towards metal ions extraction. The metal could be back-extracted from the ionic liquid phase with aqueous EDTA solutions. The metal extractability of the ionic liquid after the back-extraction is equivalent to that of the fresh mixture showing that ionic liquid can be reused for several extraction and back-extraction cycles.

Abstract: Much of the uncertainty in crop and grassland model predictions of how arable and grassland systems respond to changes in management and environmental drivers can be attributed to differences in the structure of these models. This has created an urgent need for international bench- marking of models, in which uncertainties are estimated by running several models that simulate the same physical and management conditions (ensemble modelling) to generate expanded envelopes of uncertainty in model predictions (Asseng et al. , 2013). Simulations of C and N fluxes, in particular, are inherently uncertain because they are driven by complex interactions (Sandor et al. , 2016) and complicated by considerable spatial and temporal variability in the measurements. In this context, the Integrative Research Group of the Global Research Alliance (GRA) on Agricultural Greenhouse Gases promotes a coordinated activity across multiple international projects (e.g. C and N Models Inter-comparison and Improvement to assess management options for GHG mitigation in agrosystems worldwide (C-N MIP) and Models4Pastures of the FACCE-JPI, https://www.faccejpi.com) to benchmark and compare simulation models that estimate C – N related outputs (including greenhouse gas emissions) from arable crop and grassland systems (http://globalresearchalliance.org/e/model- intercomparison-on-agricultural-ghg-emissions). This study presents some preliminary results on the uncertainty of outputs from 12 grassland models, whereas exploring differences in model response when increasing data resources are used for model calibration.

Abstract: Lignocellulosic biomass (LB) represent abundant and renewable resources and a sustainable alternative to fossil carbon. LB is composed of various polymers such as cellulose, hemicellulose and lignin. These polymers are interconnected through a variety of covalent and noncovalent interactions in plant cell walls thus forming a highly complex organized network that vary according to plant species and development, and growing conditions. Such an organization plays a key role in plants growth, mechanical properties, and biological recalcitrance but can be a bottleneck to the development of sustainable and cost-efficient biorefineries.Given the increasing interest in LB it has become critical to characterize LB architecture at multiscale levels and to understand structural parameters that control the LB properties and its susceptibility to transformation into chemicals, materials and biofuels. Notably, understanding the effect of the interactions between individual polymers on the cell wall structure and subsequent impact on the their properties is still challenging considering the hierarchical organization of the lignified cell walls. In this context, bioinspired systems based on lignocellulosic polymers can be a valuable tool to adress the role of interactions between cell wall polymers in plant cell walls properties and to provide rationale for designing lignin-based nanomaterials.
