The development of a responsive fluorescent probe for the detection of a particular biomolecule in a specific site at the desired moment is important in the fields of bioanalysis and imaging, molecular biology and biomedical research. In this work, we report the development of a remote-light activatable nanoprobe for the fluorescence detection of sulphite in pure aqueous solution and its imaging applications in living cells. The nanoprobe, Poly-Cm-SP, is fabricated simply by wrapping photochromic molecules (Cm-SP) into a polymer nanoparticle. BackgroundCerebrospinal fluid (CSF) leak is typically diagnosed by detecting a protein marker β2-transferrin (β2-Tf) in secretion samples. β2-Tf and β1-transferrin (β1-Tf) are glycoforms of human transferrin (Tf). A novel affinity capture technique for sample preparation, called microprobe-capture in-emitter elution (MPIE), was incorporated with high-resolution mass spectrometry (HR-MS) to analyze the Tf glycoforms and elucidate the structures of β1-Tf and β2-Tf.MethodsTo implement MPIE, an analyte is first captured on the surface of a microprobe, and subsequently eluted from the microprobe inside an electrospray emitter. The capture process is monitored in real-time via next-generation biolayer interferometry (BLI). When electrospray is established from the emitter to a mass spectrometer, the analyte is immediately ionized via electrospray ionization (ESI) for HR-MS analysis. Serum, CSF, and secretion samples were analyzed using MPIE-ESI-MS.ResultsBased on the MPIE-ESI-MS results, the structures of β1-Tf and β2-Tf were solved. As Tf glycoforms, β1-Tf and β2-Tf share the amino acid sequence but have varying N-glycans. β1-Tf, the major serum-type Tf, has two G2S2 N-glycans on Asn413 and Asn611. β2-Tf, the major brain-type Tf, has an M5 N-glycan on Asn413 and a G0FB N-glycan on Asn611.ConclusionsThe structures of β1-Tf and β2-Tf were successfully elucidated by MPIE-ESI-MS analysis. The resolving power of the novel MPIE-ESI-MS method was demonstrated in this study. On the other hand, knowing the N-glycan structures on β2-Tf allows for the design of other novel test methods for β2-Tf in the future.

The warning label is identified as a simple front-of-package labeling format that assists consumers in making healthier food choices at the point of purchase. The color and/or shape of the design elements of the packaging labels can influence the understanding and purchase intention outcomes. This study aimed to explore the perceptions of consumers regarding differently designed warning labels (WLs) to identify a possibly suitable warning label design in the Chinese context. Using a qualitative approach, we collected data through 12 focus group discussions consisting of 116 participants residing in 6 provinces. Participants were selected by purposive sampling. Data were coded following inductive thematic analysis and summarized into three themes: (1) perceived preference for the appearance of the WLs, (2) perceived benefits of the WLs, and (3) suggestions for promoting the WLs. Participants preferred the black shield label showing a high content of the nutrients of concern (e.g., sodium, added sugar, and saturated fat) in both Chinese and English. Consumers generally agreed that the WLs were easy to understand and offered a simple method for choosing healthier foods. They anticipated that WLs could encourage the food industry to reformulate their products. Our study results will support the design and facilitate the use of WLs in China.

Climate change impacts ocean nutrient availability and will likely alter the marine food web. While climate models predict decreased average ocean productivity, the extent of these changes, especially in the marginal seas upon which large human populations depend, is not well understood. Here, we reconstructed changes in seawater phosphate concentration and nitrate source over the past 400 years, which reveals a more than 50% decline in residence time of seawater phosphate, and 8%–48% decline in subsurface nitrogen supply following the coldest period of Little Ice Age. Our data indicates a link between surface ocean nutrient supply and the East Asian Summer Monsoon strength in an economically important marginal sea. As climate models predict that the East Asian Summer monsoon will strengthen in the future, our study implies that surface ocean primary productivity may increase in the South China Sea, contrary to the predicted decrease in global average ocean productivity. Ultra-high molecular weight polyethylene (UHMWPE)/high-density polyethylene (HDPE) blend with lower viscosity is more suitable for melt spinning compared to pure UHMWPE; however, the mechanical property of the blend fiber is hard to dramatically improve (the maximum tensile strength of 998.27 MPa). Herein, different content modified-nano-SiO2 is incorporated to UHMWPE/HDPE blend fiber. After adding 0.5 wt% nano-SiO2, the tensile strength and initial modulus of UHMWPE/HDPE/nano-SiO2 fiber are increased to 1211 MPa and 12.81 GPa, respectively, 21.57% and 43.32% higher than that of UHMWPE/HDPE fiber. Meanwhile, the influence of the nano-SiO2 content on the performance for as-spun filament and fiber are emphatically analyzed. The crystallinity and molecular chain orientation of as-spun filament reduces with the addition of nano-SiO2. On the contrary, for fiber, the addition of nano-SiO2 promoted the crystallinity, molecular chain orientation and grain refinement more obvious at a lower content. Furthermore, the possible action mechanism of nano-SiO2 in the as-spun filament extrusion and fiber hot drawing stage is explained.

In this study, a rubber-composite-nanoparticle-modified epoxy powder composite coating with low curing temperature and high toughness was successfully fabricated. The effects of N,N-dimethylhexadecylamine (DMA) carboxy-terminated nitrile rubber (CNBR) composite nanoparticles on the microstructure, curing behavior, and mechanical properties of epoxy-powder coating were systematically investigated. SEM and TEM analysis revealed a uniform dispersion of DMA-CNBR in the epoxy-powder coating, with average diameter of 100 nm. The curing temperature of the epoxy-composite coatings had reduced almost 19.1% with the addition of 1phr DMA-4CNBR into the coating. Impact strength tests confirmed that DMA-CNBR-modified epoxy-composite coatings showed significant improvements compared with the neat EP coating, which was potentially attributed to the nanoscale dispersion of DMA-CNBR particles in epoxy coatings and their role in triggering microcracks. Other mechanical properties, including adhesion and cupping values, were improved in the same manner. In addition, thermal and surface properties were also studied. The prepared epoxy composite powder coating with the combination of low curing temperature and high toughness broadened the application range of the epoxy coatings.

The detection of changes in the reactive oxygen species (ROS)/reactive sulfur species (RSS) couple is important for studying the cellular redox state. Herein, we developed a 1,8-naphthalimide-based fluorescence probe (NI) for the reversible detection of bisulfite (HSO3- ) and hydrogen peroxide (H2 O2 ) in vitro and in vivo. NI has been designed with a reactive ethylene unit which specifically reacts with HSO3- by a Michael addition reaction mechanism, resulting in the quenching of yellow fluorescence at 580 nm and the appearing of green fluorescence at 510 nm upon excitation at 500 nm and 430 nm, respectively. The addition product (NI-HSO3 ) could be specifically oxidized to form the original C=C bond of NI, recovering the fluorescence emission and color. The detection limits of NI for HSO3- and NI-HSO3 for H2 O2 were calculated to be 2.05 μM and 4.23 μM, respectively. The reversible fluorescence response of NI towards HSO3- /H2 O2 couple can be repeated for at least five times. NI is reliable at a broad pH range (pH 3.0-11.5) and features outstanding selectivity, which enabled its practical applications in biological and food samples. Monitoring the reversible and dynamic inter-conversion between HSO3- and H2 O2 in vitro and in vivo has been verified by fluorescence imaging in live HeLa cells, adult zebrafish and nude mice. Moreover, NI has been successfully applied to detect of HSO3- levels in food samples.

Monogeneans of the genus Paradiplozoon were found on the gills of specimens of five species of schizothoracid caught using fyke nets in the upper stream of the Yarkand River, Xinjiang, China in May–August 2019. The preserved parasite were stained with boric acid magenta and hematoxylin, respectively. Morphological observations, line drawings, photomicrographs and measurements were made in Nikon ECLIPSE E200 imaging optical microscope and digitally edited. The molecular analysis included the study of the sequence of the second internal transcribed spacer (ITS 2) of the ribosomal DNA region, calculation and analysis of genetic distance, with phylogenetic reconstructions based on the Bayesian inference and Maximum Likelihood analysis. The natural infection rate of host fish was 10–88%. Morphological analysis indicated that the average length of the new species was 2.125 mm while the width was 0.69 mm. The anterior part was 1.387 mm in length and the average length of the posterior part was 0.545 mm. The vitellaria was well-developed and located in the front of the body. A single ovary (oval shaped) was located at the back end of the reproductive binding area. A testis (irregular mass) was located behind or parallelled to the ovary. The new species can be distinguished from all the recorded Paradiplozoon species in terms of morphological characteristics such as haptor, clamp and central hook morphology, intestine shape and body size. In addition, the second internal transcribed spacer (ITS 2) of the ribosomal DNA region of the diplozoid was compared with that of known diplozoids previously published. It indicated that there were significant differences between the new species and the published diplozoids. Both morphological and molecular analysis support that the diplozoid is a new species. Based on the sampling location, the new species was named Paradiplozoon yarkandense n. sp.

Objective Severe hypertriglyceridemia (SHTG; plasma triglycerides >1000 mg/dL) is a rare but serious complication in children who develop diabetic ketoacidosis (DKA) from uncontrolled or new-onset type 1 diabetes. Methods We present the case of a severely malnourished 16-year-old with a 10-month history of presumed type 2 diabetes managed with lifestyle modifications and metformin, who presented with SHTG, acute pancreatitis, and DKA. On examination, there was no evidence of lipemia retinalis, cutaneous xanthomas, or xanthelasma. He was initially treated with an insulin infusion and intravenous fluids. Despite this treatment, his pancreatitis symptoms worseneed and lipase level increased, necessitating 2 courses of plasmapheresis that immediately resolved his symptoms and dramatically improved his clinical status. He was discharged on hospital day 5. During his hospital admission, islet cell antigen 512, insulin, glutamic acid decarboxylase 65, and zinc transporter 8 autoantibodies were positive in the presence of insulinopenia, consistent with type 1 diabetes. Results Hypertriglyceridemia and hypercholesterolemia did not recur during follow-up, suggesting that the underlying mechanism for SHTG was insulin deficiency. Conclusion This report of SHTG, DKA, and pancreatitis in an adolescent highlights the safe, early initiation of plasmapheresis as an effective treatment. To our knowledge, plasmapheresis has rarely been used so early in the course of treatment for an adolescent with SHTG, DKA, and acute pancreatitis.

We previously reported that a multifunctional opioid/neuropeptide FF receptor agonist, DN-9, achieved peripherally restricted analgesia with reduced side effects. To develop stable and orally bioavailable analogues of DN-9, eight lactam-bridged cyclic analogues of DN-9 between positions 2 and 5 were designed, synthesized, and biologically evaluated. In vitro cAMP assays revealed that these analogues, except 7, were multifunctional ligands that activated opioid and neuropeptide FF receptors. Analogue 1 exhibited improved potency for κ-opioid and NPFF2 receptors. All analogues exhibited potent, long-lasting, and peripherally restricted antinociception in the tail-flick test without tolerance development after subcutaneous administration and produced oral analgesia. Oral administration of the optimized compound analogue 1 exhibited powerful, peripherally restricted antinociceptive effects in mouse models of acute, inflammatory, and neuropathic pain. Remarkably, orally administered analogue 1 had no significant side effects, such as tolerance, dependence, constipation, or respiratory depression, at effective analgesic doses.

NLRP3 inflammasome-mediated pyroptosis contributes to the mechanism of neuroinflammation. It has been reported that dendritic cell-associated C-type lectin-1 (Dectin-1) activation triggers inflammation in neurological diseases. However, the role of Dectin-1 on NLRP3 inflammasome-mediated pyroptosis after ICH remains unclear. Here, we aimed to explore the effect of Dectin-1 on NLRP3 inflammasome-mediated pyroptosis and neuroinflammation after ICH.Methods: Adult male C57BL/6 mice were used to establish the ICH model. Laminarin, an inhibitor of Dectin-1, was administered for intervention. Expression of Dectin-1 was evaluated by Western blot and immunofluorescence. Brain water content and neurobehavioral function were tested to assess brain edema and neurological performance. Western blot was conducted to evaluate the level of GSDMD-N. ELISA kits were used to measure the levels of IL-1β and IL-18. qRT–PCR and Western blot were performed to evaluate the expressions of NLRP3 inflammasome, IL-1β, and IL-18.Results: The expression of Dectin-1 increased following ICH, and Dectin-1 was expressed on microglia. In addition, inhibition of Dectin-1 by laminarin decreased brain edema and neurological impairment after ICH. Moreover, inhibition of Dectin-1 decreased the expression of pyroptosis-related protein, GSDMD-N, and inflammatory cytokines (IL-1β and IL-18). Mechanistically, Dectin-1 blockade inhibits NLRP3 inflammasome activation, thereby alleviating neuroinflammatory injury by attenuating NLRP3 inflammasome-mediated pyroptosis both in vivo and in vitro.Conclusion: Our study indicates that the inhibition of Dectin-1 alleviates neuroinflammation by attenuating NLRP3 inflammasome-mediated pyroptosis after ICH.Keywords: intracerebral hemorrhage, Dectin-1, NLRP3 inflammasome, pyroptosis, microglia

The study aimed to observe the therapeutic effect of static progressive stretching (SPS) combined with extracorporeal shock wave therapy (ESWT) on extension knee joint contracture in rats and the effect on the MAPK/ERK pathway in the development of joint capsule fibrosis. Thirty-six Sprague Dawley rats were randomly divided into blank control group, immobilization model group, natural recovery group, ESWT intervention group, SPS intervention group, and SPS combined with ESWT intervention group. The left knee joints of the rats, except for the control group, were fixed with an external fixation brace for four weeks at full extension to form joint contractures. The therapeutic effect of each intervention was assessed by evaluating total and arthrogenic contracture, the number of total cells and collagen deposition in the anterior joint capsule, the protein levels of TGF-β1, FGF-2, and ERK2 in the anterior joint capsule, the mean optical density of upstream RAS and downstream ERK2 positive expression in the MAPK/ERK pathway. SPS in combination with ESWT was more effective in relieving joint contracture, improving the histopathological changes in the anterior joint capsule, and suppressing the high expression of target proteins and the overactivated MAPK/ERK pathway. The overactivated MAPK/ERK pathway was involved in the formation of extension knee joint contracture in rats. SPS in combination with ESWT was effective in relieving joint contracture and fibrosis of joint capsule. Moreover, the inhibition of the overactivated MAPK/ERK pathway may be the potential molecular mechanism for its therapeutic effect.

The iron and steel industry is a major CO2 emitter and an important subject for the implementation of carbon emission reduction goals and tasks. Due to the complex ore composition and low iron grade, vanadium–bearing titanomagnetite smelting in a blast furnace consumes more coke and emits more carbon than in an ordinary blast furnace. Injecting hydrogen–rich gas into blast furnace can not only partially replace coke, but also reduce the carbon emission. Based on the whole furnace and zonal energy and mass balance of blast furnace, the operation window of the blast furnace smelting vanadium–bearing titanomagnetite is established in this study on the premise that the thermal state of the blast furnace is basically unchanged (raceway adiabatic flame temperature and top gas temperature). The effects of different injection amounts of hydrogen–rich gases (shale gas, coke oven gas, and hydrogen) on raceway adiabatic flame temperature and top gas temperature, and the influence of blast temperature and preheating temperature of hydrogen–rich gases on operation window are calculated and analyzed. This study provides a certain theoretical reference for the follow–up practice of hydrogen–rich smelting of vanadium–bearing titanomagnetite in blast furnace.

Topical application of double-stranded RNA (dsRNA) can induce RNA interference (RNAi) and modify traits in plants without genetic modification. However, delivering dsRNA into plant cells remains challenging. Using developing tomato (Solanum lycopersicum) pollen as a model plant cell system, we demonstrate that layered double hydroxide (LDH) nanoparticles up to 50 nm in diameter are readily internalized, particularly by early bicellular pollen, in both energy-dependent and energy-independent manners and without physical or chemical aids. More importantly, these LDH nanoparticles efficiently deliver dsRNA into tomato pollen within 2–4 h of incubation, resulting in an 89% decrease in transgene reporter mRNA levels in early bicellular pollen 3-d post-treatment, compared with a 37% decrease induced by the same dose of naked dsRNA. The target gene silencing is dependent on the LDH particle size, the dsRNA dose, the LDH–dsRNA complexing ratio, and the treatment time. Our findings indicate that LDH nanoparticles are an effective nonviral vector for the effective delivery of dsRNA and other biomolecules into plant cells.

High-performance multifunctional materials for water splitting driven by low voltage are crucial for hydrogen evolution reaction (HER), but developing such materials is challenging. Herein, a simple strategy was designed to build a MoS2/Co9S8/MoC@CNT-N (MCM@CNT-N) heterostructure with a large number of interfaces. Regarding the HER, the synthesized MCM@CNT-N heterostructure catalyst showed high efficiency and stable electrocatalytic performance, with a low overpotential of 174.2 mV and a small Tafel slope of 84.7 mV dec−1 at a current density of 10 mA cm−2 in 0.5 M H2SO4. In addition to the function of heterojunctions, the excellent activity is also attributed to the introduction of Co and N atoms and the formation of carbon nanotubes. This work provides a new approach to build efficient and low-cost electrocatalysts for electrochemical reactions.

In this paper, relaxor ferroelectric Ba0.8Sr0.2Zr0.9Ti0.1O3–Na0.5K0·5NbO3 (BSZT-KNN) ceramics were achieved by designing microstructures and adding nano-additives. The fine grains (192 nm) and relaxor behaviour of the BSZT-KNN ceramics were enhanced by utilizing the BSZT@KNN powders. The relative density (97%) and temperature stability of the ceramics were increased by adding nano-additives. Finally, BSZT-KNN ceramics with a large polarization difference (16.85 μC/cm2) and high breakdown field strength (352 kV/cm) were obtained. Accordingly, a recoverable energy storage density of 2.96 J/cm3 and an energy storage efficiency of 98.0% were achieved. These results suggest that BSZT-KNN ceramics with high comprehensive energy storage performance can be applied to electrical energy storage.

Ascorbate (Vitamin C) has been proposed as a promising therapeutic agent against sepsis in clinical trials, but there is little experimental evidence on its anti-septic efficacy. We report that Toll-like receptor 4 (TLR4) activation by LPS stimuli augments ascorbate uptake in murine and human tubular cells through upregulation of two ascorbate transporters SVCT-1 and -2 mediated by Fn14/SCFFbxw7α cascade. Ascorbate restriction, or knockout of SVCT-1 and -2, the circumstance reminiscent to blockade of ascorbate uptake, endows tubular cells more vulnerable to the LPS-inducible apoptosis, whereas exogenous administration of ascorbate overrides the ruin execution, for which the PINK1-PARK2, rather than BNIP3-NIX axis is required. Ascorbate increases, while SVCT-1 and -2 knockout or ascorbate restriction dampens tubular mitophagy upon LPS stimuli. Treatment of endotoxemic mice with high-dose ascorbate confers mitophagy and substantial protection against mortality and septic acute kidney injury (AKI). Our work provides a rationale for clinical management of septic AKI with high doses of ascorbate.

Sepsis-induced acute lung injury (ALI) is associated with dysregulated inflammatory reactions. MiR-19b-3p level was reported to be downregulated in patients with sepsis. In order to evaluate the role of miR-19b-3p in sepsis, cecum ligation and puncture (CLP)-induced mouse sepsis model and lpopolysaccharide (LPS)-treated pulmonary microvascular endothelial cells (PMVECs) were used. For in vivo study, lung tissue was harvested for hematoxylin and eosin (H&E) staining, TNF-α, IL-6, IL-1β and p-p65, p-IκB measuring. Cell apoptosis was assessed by TUNEL assay. For in vitro study, cell proliferation and apoptosis were detected by CCK-8 and flow cytometry, respectively. Methylation of miR-19b-3p promoter was measured by methylation-specific PCR (MSP) assay. The target of miR-19b-3p was determined by dual-luciferase reporter gene assay. The level of miR-19b-3p was determined to be downregulated in vitro and in vivo. In addition, miR-19b-3p protected mice from inflammation injury through inhibiting NF-κB signaling pathway. Overexpression of miR-19b-3p increased cell viability, decreased apoptosis and pro-inflammatory cytokines secretion in LPS-treated PMVECs. Reactive oxygen species (ROS) play an essential role in the progression of many chronic diseases like atherosclerosis and rheumatoid arthritis. For decades, antioxidant compounds have always been considered as potential treatments for these ROS-related diseases. Concomitantly, noninvasive imaging systems such as magnetic resonance imaging (MRI) have also been widely used in the diagnosis of diseases, especially atherosclerosis. In this study, we investigated the feasibility to develop chitosan nanococktails containing both nanoceria and superparamagnetic iron oxide nanoparticles for ROS-related theranostics. Nanoceria utilized as therapeutic modules capable of ROS scavenging and iron-oxide nanoparticles utilized as imaging agents for MRI have been synthesized separately. Subsequently, two versions of theranostic chitosan nanococktails containing both nanoceria and iron oxide nanoparticles (Chit-IOCO and Chit-TPP-IOCO) were successfully synthesized via two different mechanisms, electrostatic self-assembly, and ionic gelation. In vitro studies such as cytotoxicity, MRI, and ROS scavenging were performed. These theranostic nanococktails demonstrated effective ROS scavenging and MRI contrast as a potential platform for treatment and diagnosis of ROS-related diseases. Results indicated that both Chit-IOCO and Chit-TPP-IOCO can reduce the ROS level of the lipopolysaccharide-stimulated macrophage J774A.1 to the baseline level. Chit-IOCO was less toxic to the cells than Chit-TPP-IOCO. In addition, Chit-IOCO exhibited higher MRI relaxivity than Chi-TPP-IOCO (308 and 150 mM-1 s-1, respectively), indicating that Chi-IOCO was more effective than Chit-TPP-IOCO as an MRI contrast agent in macrophages. Taken together, Chit-IOCO nanococktail demonstrates outstanding potential for treatment and diagnosis of ROS-related diseases. Potentially, this nanococktail can be easily modified to include new modules, allowing future application of personalized medicine.
