This study develops an original co-infection model in mice using Brucella melitensis, the most frequent cause of human brucellosis, and Trypanosoma brucei, the agent of African trypanosomiasis. Although the immunosuppressive effects of T. brucei in natural hosts and mice models are well established, we observed that the injection of T. brucei in mice chronically infected with B. melitensis induces a drastic reduction in the number of B. melitensis in the spleen, the main reservoir of the infection. Similar results are obtained with Brucella abortus-and Brucella suis-infected mice and B. melitensis-infected mice co-infected with Trypanosoma cruzi, demonstrating that this phenomenon is not due to antigenic cross-reactivity. Comparison of co-infected wild-type and genetically deficient mice showed that Brucella elimination required functional IL-12p35/IFNγ signaling pathways and the presence of CD4+ T cells. However, the impact of wild type and an attenuated mutant of T. brucei on B. melitensis were similar, suggesting that a chronic intense inflammatory reaction is not required to eliminate B. melitensis. Finally, we also tested the impact of T. brucei infection on the course of Mycobacterium tuberculosis infection. Although T. brucei strongly increases the frequency of IFNγ+CD4+ T cells, it does not ameliorate the control of M. tuberculosis infection, suggesting that it is not controlled by the same effector mechanisms as Brucella. Thus, whereas T. brucei infections are commonly viewed as immunosuppressive and pathogenic, our data suggest that these parasites can specifically affect the immune control of Brucella infection, with benefits for the host. Tuberculosis (TB) caused by Mycobacterium tuberculosis mainly affects the lungs, but can spread to other organs. TB chronically activates the immune and endocrine systems producing remarkable functional changes.So far, it is unknown whether pulmonary non-disseminated TB cause changes in the female reproductive system and lung endocrinology. To investigate whether pulmonary TB produces immunoendocrine alterations of the female mice reproductive organs, and lung estradiol synthesis. BALB/c mice were infected intratracheally with Mycobacterium tuberculosis (Mtb) strain H37Rv. Groups of six non-infected and infected animals were euthanized on different days. Bacillary loads were determined in the lungs, ovaries and uterus. Immunohistochemistry and morphometry studies were performed in histological sections. Serum estradiol wasassayed, and supernatantfrom cultured lung cells was analyzed by Thin Layer Chromatography (TLC). Mtb only grew in lung tissue. Histopathology revealed abnormal folliculogenesis and decreased corpora lutea. Altered ovarian expression of IL-6, IL-1β was found. The infection increased serum estradiol. Estradiol synthesis by infected lung cells triplicate after 30 pi days.Aromatase immunostaining was found in the alveolar and bronchial epithelium, being stronger in the infected lungs, mainly in macrophages. Objectives To compare the effects of regional anaesthesia and opioid administration on adrenocortical and glycaemic responses, postoperative pain and recovery quality in dogs undergoing stifle surgery. Study design Prospective, blinded clinical study. Animals Forty-five dogs anaesthetized for tibial tuberosity advancement or tibial plateau levelling osteotomy, and 15 healthy dogs undergoing noninvasive orthopaedic diagnostic procedures. Methods The baseline behaviour of each dog was classified on a descriptive scale before anaesthesia. Dogs were anaesthetized using a standard protocol. Those undergoing surgery were randomly assigned to one of three intraoperative analgesia protocols: 1) peripheral (femoral and sciatic) nerve block (bupivacaine 0.5%; 0.15 mL kg -1 in each nerve; group PNB); 2) spinal anaesthesia (isobaric bupivacaine 0.5%; 0.05 mL kg -1; group SPI); and 3) intravenous fentanyl variable rate infusion and postoperative methadone (0.2 mg kg -1; group FEN). Blood samples were collected for measurement of cortisol and glucose concentrations on arrival (TAR), at induction (TIND), at tracheal extubation (TEXT), and 1 hour post-extubation (TPEX). After extubation, a researcher unaware of the dog's group assignment evaluated pain and recovery quality using the Glasgow Composite Measure Pain Scale (Short- Form) and a descriptive scale, respectively. Results Median recovery quality was significantly worse in group FEN (p < 0.0001) compared with all other groups. Postoperative pain scores were lower in group PNB compared with groups SPI and FEN. Cortisol and glucose concentrations increased significantly from TAR at TEXT and TPEX (p < 0.0001) in group FEN, and were unchanged from TAR in the other groups. Conclusions and clinical relevance Analgesia with a peripheral nerve block or spinal anaesthesia prevented the glycaemic and cortisol responses to surgery, promoted better recovery quality, and decreased postoperative pain scores compared with FEN. Persistent infection with high-risk human papillomaviruses is the main etiological factor in cervical cancer (CC). The human papillomavirus type 16 (HPV16) E7 oncoprotein alters several cellular processes, regulating the expression of many genes in order to avoid cell cycle control. Retinoic acid receptor beta (RARB) blocks cell growth, inducing differentiation and apoptosis. This tumor suppressor gene is gradually silenced in late passages of foreskin keratinocytes immortalized with HPV16 and in various tumors, including CC, mainly by epigenetic modifications. We investigated the effect of E7 oncoprotein on RARB gene expression. We found that HPV16 E7 increases RARB mRNA and RAR-beta protein expression both in vitro and in the cervix of young K14E7 transgenic mice. In E7-expressing cells, RARB overexpression is further increased in the presence of the tumor suppressor p53 (TP53) R273C mutant. This effect does not change when either C33-A or E7-expressing C33-A cell line is treated with Trichostatin A, suggesting that E7 enhances RARB expression independently of histone deacetylases inhibition. In many cases parasites display highly complex life cycles that include the penetration and permanence of the larva or adults within host organs, but even in those that only have one host, reciprocal, intricate interactions occur. Evidence indicates that steroid hormones have an influence on the development and course of parasitic infections. The host gender's susceptibility to infection, and the related differences in the immune response are good examples of the host-parasite interplay. However, the capacity of these organisms to synthesize their own steroidogenic hormones still has more questions than answers. It is now well-known that many parasites synthesize ecdysteroids, but limited information is available on sex steroid and corticosteroid synthesis. This review intends to summarize some of the existing information in the field. In most, but not all parasitosis the host's hormonal environment determines the susceptibility, the course, and severity of parasite infections. In most cases the infection disturbs the host environment, and activates immune responses that end up affecting the endocrine system. Furthermore, sex steroids and corticosteroids may also directly modify the parasite reproduction and molting. Available information indicates that parasites synthesize some steroid hormones, such as ecdysteroids and sex steroids, and the presence and activity of related enzymes have been demonstrated. More recently, the synthesis of corticosteroid-like compounds has been shown in Taenia solium cysticerci and tapeworms, and in Taenia crassiceps WFU cysticerci. In-depth knowledge of the parasite's endocrine properties will contribute to understand their reproduction and reciprocal interactions with the host, and may also help designing tools to combat the infection in some clinical situations. Mycobacterium tuberculosis H37Rv, constitutively expressing a second copy of the transcriptional regulator DosR ( Rv3133c) under control of the hsp60 promoter, was compared to wild-type M. tuberculosis for in vitro expression of the target genes of the DosR dormancy regulon, for its in vitro growth characteristics in liquid 7H9 culture medium, and for its capacity to replicate in murine macrophages. Under aerobic conditions, hsp60-driven DosR significantly induced the expression of 39 out of 44 DosR regulon genes, as assessed by real time qPCR. Increased DosR regulon gene transcription in vitro did not modify the capacity of the strain to grow under axenic conditions nor to infect murine macrophages as compared to unmodified wild-type bacteria. Summary Background HIV-1 infection is associated with increased risk of tuberculosis and a safe and effective vaccine would assist control measures. We assessed the safety, immunogenicity, and efficacy of a candidate tuberculosis vaccine, modified vaccinia virus Ankara expressing antigen 85A (MVA85A), in adults infected with HIV-1. Methods We did a randomised, double-blind, placebo-controlled, phase 2 trial of MVA85A in adults infected with HIV-1, at two clinical sites, in Cape Town, South Africa and Dakar, Senegal. Eligible participants were aged 18–50 years, had no evidence of active tuberculosis, and had baseline CD4 counts greater than 350 cells per μL if they had never received antiretroviral therapy or greater than 300 cells per μL (and with undetectable viral load before randomisation) if they were receiving antiretroviral therapy; participants with latent tuberculosis infection were eligible if they had completed at least 5 months of isoniazid preventive therapy, unless they had completed treatment for tuberculosis disease within 3 years before randomisation. Participants were randomly assigned (1:1) in blocks of four by randomly generated sequence to receive two intradermal injections of either MVA85A or placebo. Randomisation was stratified by antiretroviral therapy status and study site. Participants, nurses, investigators, and laboratory staff were masked to group allocation. The second (booster) injection of MVA85A or placebo was given 6–12 months after the first vaccination. The primary study outcome was safety in all vaccinated participants (the safety analysis population). Safety was assessed throughout the trial as defined in the protocol. Secondary outcomes were immunogenicity and vaccine efficacy against Mycobacterium tuberculosis infection and disease, assessed in the per-protocol population. Immunogenicity was assessed in a subset of participants at day 7 and day 28 after the first and second vaccination, and M tuberculosis infection and disease were assessed at the end of the study. The trial is registered with ClinicalTrials.gov , number NCT01151189 . Findings Between Aug 4, 2011, and April 24, 2013, 650 participants were enrolled and randomly assigned; 649 were included in the safety analysis (324 in the MVA85A group and 325 in the placebo group) and 645 in the per-protocol analysis (320 and 325). 513 (71%) participants had CD4 counts greater than 300 cells per μL and were receiving antiretroviral therapy; 136 (21%) had CD4 counts above 350 cells per μL and had never received antiretroviral therapy. 277 (43%) had received isoniazid prophylaxis before enrolment. Solicited adverse events were more frequent in participants who received MVA85A (288 [89%]) than in those given placebo (235 [72%]). 34 serious adverse events were reported, 17 (5%) in each group. MVA85A induced a significant increase in antigen 85A-specific T-cell response, which peaked 7 days after both vaccinations and was primarily monofunctional. The number of participants with negative QuantiFERON-TB Gold In-Tube findings at baseline who converted to positive by the end of the study was 38 (20%) of 186 in the MVA85A group and 40 (23%) of 173 in the placebo group, for a vaccine efficacy of 11·7% (95% CI −41·3 to 44·9). In the per-protocol population, six (2%) cases of tuberculosis disease occurred in the MVA85A group and nine (3%) occurred in the placebo group, for a vaccine efficacy of 32·8% (95% CI −111·5 to 80·3). Interpretation MVA85A was well tolerated and immunogenic in adults infected with HIV-1. However, we detected no efficacy against M tuberculosis infection or disease, although the study was underpowered to detect an effect against disease. Potential reasons for the absence of detectable efficacy in this trial include insufficient induction of a vaccine-induced immune response or the wrong type of vaccine-induced immune response, or both. Interleukin-17A ( IL-17A), a pro-inflammatory cytokine acting on neutrophil recruitment, is known to play an important role during Mycobacterium tuberculosis infection, but the role of IL-17 A receptor signalling in immune defence against this intracellular pathogen remains poorly documented. Here we have analysed this signalling using C57 BL/6 mice genetically inactivated in the IL-17 receptor A subunit ( IL-17 RA−/−). Although early after infection bacterial growth was controlled to the same extent as in wild-type mice, IL-17 RA−/− mice were defective in exerting long-term control of M. tuberculosis infection, as demonstrated by a progressively increasing pulmonary bacterial burden and shortened survival time. Compared with infected wild-type mice, IL-17 RA−/− mice showed impaired recruitment of neutrophils to the lungs at the early but not the late stage of infection. Pulmonary tumour necrosis factor-α, IL-6 and particularly IL-10 levels were decreased in the absence of IL-17 RA signalling, whereas IL-1β was increased. CD4+-mediated and γδ-mediated IL-17 A production was dramatically increased in IL-17 RA−/− mice (confirming part of their phenotype), whereas production of interferon-γ and expression of the bactericidal enzyme inducible nitric oxide synthase were not affected. Collectively, our data suggest that early but not late neutrophil recruitment is essential for IL-17 A-mediated long-term control of M. tuberculosis infection and that a functional interferon-γ response is not sufficient to control M. tuberculosis growth when the IL-17 RA pathway is deficient. As treatment of auto-immune diseases with anti- IL-17 A antibodies is actually being tested in clinical studies, our data suggest that caution should be taken with respect to possible reactivation of tuberculosis. Abstract: Few studies have been carried out on the welfare of captive bottlenose dolphins (Tursiops truncatus). Of these, most include information on animals kept in closed facilities or pools. The aim of this study was to assess the welfare of captive bottlenose dolphins in open and closed facilities by measuring states of individual behavior and salivary cortisol concentrations. A total of 23 bottlenose dolphins were studied in 4 different dolphinaria. Dolphinaria A and B have closed facilities, whereas dolphinaria C and D have open facilities. A total of 152 hours of behavioral observations were analyzed using a combination of behavior and scan sampling to obtain information on individual time budgets. Salivary cortisol concentrations were measured using radioimmunoassay in 96 and 180 saliva samples of dolphins kept in closed and open facilities, respectively. In general, the results found that dolphins kept in open facilities spent less time floating (P < 0.05) and swimming in circular patterns than linear ones (P < 0.05) compared with dolphins in closed facilities. Dolphins kept in open facilities also had lower salivary cortisol concentrations than dolphins kept in closed facilities (P < 0.05). For this reason, we suggest that further research should include other welfare indicators such as reproductive function and health measurements to know more about the relationships between the design of pools and dolphin welfare. Summary: Objectives: A fundamental to global tuberculosis (TB) control is timely and accurate diagnosis of infectious cases of the disease. Among various methods, techniques based on nucleic acid amplification are the ones with promising prospects. The present study evaluates the diagnostic value of the recently developed IS6110-based loop-mediated isothermal amplification (LAMP) for detection of Mycobacterium tuberculosis complex (MTBC) in sputum specimens. Methods: In this cross-sectional study (2008–2009), IS6110-LAMP was evaluated on 101 sputum specimens from 93 highly suspected TB patients and compared to Amplicor MTB test and in-house IS6110-PCR and -nested PCR assays. Culture results or clinical recovery following anti-TB therapy was considered as a reference to prove the TB cases. Results: The overall sensitivity of IS6110-LAMP, Amplicor, nPCR, and PCR were respectively 89.6% (69/77 specimens; 95% confidence interval [CI], 80.5–95.4%), 76.6% (59/77 specimens; CI, 65.6–85.5%), 79.2% (61/77 specimens; CI, 68.5–87.6%) and 59.7% (46/77 specimens; CI, 47.9–70.8%). The specificity and positive predictive value (PPV) were 100% for all the tests, and the negative predictive value (NPV) of IS6110-LAMP, Amplicor, nPCR, and PCR were respectively 75%, 57.1%, 60%, and 43.6%. There was an excellent overall agreement between LAMP and nPCR (k 0.828), and between LAMP and Amplicor (k 0.746), in addition to a better tolerance of IS6110-LAMP to inhibitors present in clinical specimens. Conclusion: The better diagnostic performance of IS6110-LAMP compared to Amplicor (p = 0.009), nPCR (p = 0.013) and PCR (p < 0.0001) besides its rapidity, simplicity, and cost-effectiveness makes it a valuable method for the detection of MTBC in clinical samples, particularly in resource-limited settings. The article presents a study which investigates the stress of birds in an urbanizing landscape. The study has measure the physiological traits of Canyon Towhee and Inca Dove birds, such as the levels of immunoglobulin and corticosterone, that occupy three habitat alteration levels. Results show that 30% of the capture towhees in urban areas are suffering chronic stress. Study also reveals the appropriate physiological response of Inca Dove in urban areas.

The article presents a study on the effects of adrenocotropic hormone (ACTH) challenge and age on hair cortisol concentrations in dairy cattle. The researchers compared cortisol concentrations in the hair of cows and calves to test whether cortisol accumulates in bovine hair after ACTH challenges and to determine any link between cortisol levels and hair color in Holstein cows. Assessing cortisol concentration in hair is said to be a useful non-invasive method for assessing stress in cattle.

Facial expressions are essential in animal communication, and facial expression-based pain scales have been developed for different species. Automated pain recognition offers a valid alternative to manual annotation with growing evidence across species. This study applied machine learning (ML) methods, using a pre-trained VGG-16 base and a Support Vector Machine classifier to automate pain recognition in caprine patients in hospital settings, evaluating different frame extraction rates and validation techniques. The study included goats of different breed, age, sex, and varying medical conditions presented to the University of Florida's Large Animal Hospital. Painful status was determined using the UNESP-Botucatu Goat Acute Pain Scale. The final dataset comprised images from 40 goats (20 painful, 20 non-painful), with 2,253 'non-painful' and 3,154 'painful' images at 1 frame per second (FPS) extraction rate and 7,630 'non-painful' and 9,071 'painful' images at 3 FPS. Images were used to train deep learning-based models with different approaches. The model input was raw images, and pain presence was the target attribute (model output). For the single train-test split and 5-fold cross-validation, the models achieved approximately 80% accuracy, while the subject-wise 10-fold cross-validation showed mean accuracies above 60%.
