Abstract The current study evaluated the effects of dietary seaweed supplementation in European seabass juveniles (Dicentrarchus labrax) subjected to rearing temperature and salinity oscillations, simulating natural variations in pond aquaculture conditions. Two experimental diets where formulated: a control diet (CTRL) with no supplementation and one supplemented with 7.5% seaweed mix (SW 2.5% Fucus sp., 2.5% Gracilaria sp. and 2.5% Ulva sp.). Seabass from both dietary groups (40.7 g initial body weight) was subjected to either combined salinity and temperature oscillations, or to fixed conditions. Growth performance, innate immune, and oxidative stress responses were evaluated. Results showed that seaweed supplementation had no significant effect on the analyzed parameters. However, environmental oscillations revealed significant effects on growth performance and oxidative stress response. Fish subjected to salinity and temperature oscillations had a significantly lower weight gain and daily growth index than those subjected to fixed conditions, regardless of dietary treatment. Total glutathione, oxidized glutathione, and catalase increased in fish subjected to oscillatory conditions. Lysozyme and peroxidase were not influenced by either diet or environmental conditions. In conclusion, this particular dietary seaweed mix supplementation did not mitigate the negative effects of environmental oscillations on growth performance and innate immune responses in European seabass.

Abstract Background To study pancreatic fat deposition and beta-cell function in familial partial lipodystrophy (FPLD) patients. Methods In a cross-sectional study, eleven patients with FPLD, and eight healthy volunteers were matched for age and body mass index and studied at a referral center. Body composition was assessed using dual-energy X-ray absorptiometry and the Dixon method of magnetic resonance imaging was used to quantify pancreatic and liver fat. Fasting plasma glucose, insulin, leptin, lipids and homeostasis model assessment of insulin resistance values were measured, and an oral glucose tolerance test was performed. The insulinogenic index, Matsuda insulin sensitivity index and beta-cell disposition index were calculated. Results The FPLD group presented a higher waist-to-hip ratio and fat mass ratio and lower total, truncal and lower-limb fat masses. Pancreatic and liver fat contents (log transformed) were significantly higher in the FPLD group (5.26 ± 1.5 vs. 4.08 ± 0.64, p = 0.034 and 0.77 ± 0.50 vs. 0.41 ± 0.18, p = 0.056, respectively). Pancreatic fat was inversely related to the DI (r = − 0.53, p = 0.027) and HDL-cholesterol (r = − 0.63, p = 0.003) and directly related to WHR (r = 0.60; p = 0.009), HbA1c (r = 0.58; p = 0.01) and serum triglyceride (r = 0.48, p = 0.034). Higher triglyceride and lower HDL-cholesterol levels were observed in the FPLD group. Conclusions This study demonstrated for the first time that pancreatic fat deposition is increased in FPLD. Moreover, an inverse relationship was demonstrated between pancreatic fat and beta-cell function. The findings of this study may be consistent with the expandability hypothesis and the twin cycle hypothesis.

Abstract Background The clinical utility of serum procalcitonin levels in guiding antibiotic treatment decisions in patients with sepsis remains unclear. This patient-level meta-analysis based on 11 randomized trials investigates the impact of procalcitonin-guided antibiotic therapy on mortality in intensive care unit (ICU) patients with infection, both overall and stratified according to sepsis definition, severity, and type of infection. Methods For this meta-analysis focusing on procalcitonin-guided antibiotic management in critically ill patients with sepsis of any type, in February 2018 we updated the database of a previous individual patient data meta-analysis which was limited to patients with respiratory infections only. We used individual patient data from 11 trials that randomly assigned patients to receive antibiotics based on procalcitonin levels (the “procalcitonin-guided” group) or the current standard of care (the “controls”). The primary endpoint was mortality within 30 days. Secondary endpoints were duration of antibiotic treatment and length of stay. Results Mortality in the 2252 procalcitonin-guided patients was significantly lower compared with the 2230 control group patients (21.1% vs 23.7%; adjusted odds ratio 0.89, 95% confidence interval (CI) 0.8 to 0.99; p = 0.03). These effects on mortality persisted in a subgroup of patients meeting the sepsis 3 definition and based on the severity of sepsis (assessed on the basis of the Sequential Organ Failure Assessment (SOFA) score, occurrence of septic shock or renal failure, and need for vasopressor or ventilatory support) and on the type of infection (respiratory, urinary tract, abdominal, skin, or central nervous system), with interaction for each analysis being > 0.05. Abstract l-Carnitine plays a key role in the regulation of energy metabolism and growth in fish. The aim of this study was to determine the effects of dietary l-carnitine levels on carnitine homeostasis and energy metabolism in diploid and triploid trout (Oncorhynchus mykiss). Diploid and triploid trout (29.5 ± 0.6 and 31.8 ± 0.5 g, respectively) were fed with three diets supplemented with 15, 200 or 530 mg l-carnitine/kg, respectively, for 56 days. Compared to 15 mg l-carnitine, total carnitine content increased in the liver by 182% in diploid and by 154% in triploid trout fed 530 mg l-carnitine. In muscle, total carnitine content increased by 60 and 73% in diploid and by 34 and 75% in triploid with the increase in dietary l-carnitine levels. l-Carnitine caused no significant change in the plasma concentrations of metabolites like proteins (ammonia, urea and protein), lipid (triglycerides), carbohydrate (glucose, lactate) and enzyme activities (lipase, lactate, alanine transaminase, lactic acid dehydrogenase). Triploid trout had significantly lower plasma ammonia (P = 0.003), lipase (P = 0.005) and triiodothyronine (T3; P = 0.003) levels than diploid trout. In conclusion, ploidy significantly affected the energy metabolism in rainbow trout, dietary l-carnitine levels altered the l-carnitine homeostasis, but not influence nutritional metabolism.

Abstract In the last decade, several studies have shown that fear memories can be attenuated by interfering with reconsolidation. However, most of the pharmacological agents used in preclinical studies cannot be administered to humans. Caffeine is one of the world’s most popular psychoactive drugs and its effects on cognitive and mood states are well documented. Nevertheless, the influence of caffeine administration on fear memory processing is not as clear. We employed contextual fear conditioning in rats and acute caffeine administration under a standard memory reconsolidation protocol or periodical memory reactivation. Additionally, potential rewarding/aversion and anxiety effects induced by caffeine were evaluated by conditioning place preference or open field, respectively. Caffeine administration was able to attenuate weak fear memories in a standard memory reconsolidation protocol; however, periodical memory reactivation under caffeine effect was necessary to attenuate strong and remote memories. Moreover, caffeine promoted conditioned place preference and anxiolytic-like behavior, suggesting that caffeine weakens the initial learning during reactivation through counterconditioning mechanisms. Thus, our study shows that rewarding and anxiolytic effects of caffeine during fear reactivation can change the emotional valence of fear memory. It brings a new promising pharmacological approach based on drugs widely used such as caffeine to treat fear-related disorders.

The best decision-making is a difficult task in forest management because it involves a large number of market and production variables. We simulated the growth and yield of loblolly pine in high-quality sites, aiming to indicate thinning regimes and planting densities that maximize the production of large diameter logs (minor diameter >35 cm), as well as economically evaluating such regimes. We used the software OpTimber-LP for forest optimization problems. Simulations involved four planting densities (1,111; 1,600; 2,000; and 2,500 plants/ha); three thinnings with different weights, frequencies and ages of application; and clear cut at 25 years. Such combinations totalized in 2,916 simulated regimes. We computed ANPV assuming variations in interest rates, besides costs of production and returns resulting from thinnings and from clear cut. The findings indicated great productions of large diameter logs in regimes with low planting densities. Heavy thinnings (60% removal of trees) highlighted by being ideal to almost all regimes, mainly those ones with densities of 2,000 and 2,500 plants/ha. Heavy intensity also was ideal to the first thinning, regardless the planting density. The most productive regimes presented large diameter log volumes ranging from 480 to 260 m3/ha, for regimes with 1,111 and 2,500 plants/ha, respectively. ANPV ranged from ~1.300 to ~2.100 R$/ha. As conclusion, less dense regimes provide largest productions in large diameter log, but the lowest productions in total volume; as consequence, ANPV was larger in denser regimes, once the revenue obtained from high-value logs did not overcome that one obtained from finer (and consequently cheaper) logs. High-density regimes may produce less timber volume due to only one improper thinning, whereas low-density stands may have production really affected when are subjected to consecutive improper thinnings.

Bees have a diversified natural history, thus the methods applied to study such diversity are varied. When it comes to studies of nesting biology, bees which nest in pre-existing cavities have been reasonably well studied since researchers started using trap-nests. However, bees whose nests are built underground are poorly studied due to the difficulty of finding natural nesting areas and the absence of a method that facilitates bee nest excavation. The latter is evidenced by the lack of accurate descriptions in literature of how nests are excavated. In this study we tested cylindrical rubber refills of eraser pen as a new material to be used as a tracer of underground nest galleries in a natural nesting area of two species of Epicharis Klug, 1807 (Apidae). We compared this technique directly with plaster in powder form mixed with water and our results with other methodological studies describing alternative methods and materials. The rubber refill technique overcame the main issues presented by materials such as plaster, molten metal alloys and bioplastic, namely: death of the organisms by high temperatures and/or formation of plugs and materials unduly following the roots inside the galleries. This study aims to evaluate the thorium (Th) and uranium (U) quantification in soil employing a commercial routine in a portable X‐ray fluorescence (pXRF) spectrometer. Certified reference materials (CRMs), as well as a soil sample set, were measured by pXRF and gamma‐ray spectrometry. The pXRF results showed that the thorium average detection and quantification limits (in mg kg−1) were 3.3 ± 0.5 and 11.0 ± 1.7, respectively. The uranium values were 3.0 ± 0.4 and 9.9 ± 1.3. Quantitative pXRF results for Th were in accordance with the reference reported for most CRMs analyzed, and the same accordance was obtained in the comparison with gamma‐ray spectrometry results for the soil sample set. However, U results were overestimated by pXRF when compared both with the CRM‐reported values and the gamma‐ray spectrometry results. In conclusion, the tested pXRF commercial routine may be a viable technique to identify variations in natural Th levels in soil but needs to be used with caution to identify and quantify U. The global use of pesticides, exceeding 4 million tons annually, poses substantial threats to public health and the environment. Numerous studies emphasize the correlation between pesticide exposure, genotoxicity, and neurobehavioral effects, which particularly impact children and agricultural workers. Recent research underscores the health risks associated with moderately toxic pesticides like chlorpyrifos. Chlorpyrifos contamination in crops, a result of extensive pesticide use since the 1960s, raises significant concerns regarding human health and the environment. Given its potential atmospheric volatilization from crops, alternative detection methods are imperative. This study employs gas chromatography coupled with mass spectrometry (GC/MS) in MS/MS mode, focusing on detecting chlorpyrifos in Chilean lettuce, using insects as indicators. Two extraction methods, micro-QuEChERS and HEADSPACE-SPME, were compared, with HEADSPACE-SPME demonstrating superior sensitivity (6.77 ng/mg per sample vs. 3.99 ng/mg per sample) and offering time- and solvent-saving advantages. Additionally, HEADSPACE-SPME preserves samples for future research, enhancing its utility. The study confirms the presence of pesticide residues in insects from lettuce crops, with 52.3% displaying chlorpyrifos when the SPME method was used. Predatory and parasitoid insect families like Braconidae and Ichneumonidae show high pesticide loads. The findings of this study emphasize the widespread contamination of chlorpyrifos in lettuce crop insects and highlight the efficacy of SPME for detection, suggesting its broader applicability for evaluating pesticide residues in horticultural systems. This optimized method holds promise for advancing chlorpyrifos contamination detection in agroecosystems, contributing to environmental monitoring and food security. [ABSTRACT FROM AUTHOR]

Intensive aquaculture practices involve rearing fish at high densities. In these conditions, fish may be exposed to suboptimal dissolved O2 levels with an increased formation of reactive O2 species (ROS) in tissues. Seaweeds (SW) contain biologically active substances with efficient antioxidant capacities. This study evaluated the effects of dietary supplementation of heat-treated SW (5% Gracilaria vermiculophylla or 5% Ulva lactuca) on stress bioindicators in sea bream subjected to a hypoxic challenge. 168 fish (104.5 g average weight) were distributed in 24 tanks, in which eight tanks were fed one of three experimental diets for 34 days: (i) a control diet without SW supplementation, (ii) a control diet supplemented with Ulva, or (iii) a control diet with Gracilaria. Thereafter, fish from 12 tanks (n=4 tanks/dietary treatment) were subjected to 24 h hypoxia (1.3 mg O2 l−1) and subsequent recovery normoxia (8.6 mg O2 l−1). Hypoxic fish showed an increase in hematocrit values regardless of dietary treatment. Dietary modulation of the O2-carrying capacity was conspicuous during recovery, as fish fed SW supplemented diets displayed significantly higher haemoglobin concentration than fish fed the control diet. After the challenge, survival rates in both groups of fish fed SW were higher, which was consistent with a decrease in hepatic lipid peroxidation in these groups. Furthermore, the hepatic antioxidant enzyme activities were modulated differently by changes in environmental O2 condition, particularly in sea bream fed the Gracilaria diet. After being subjected to hypoxia, the gene expression of antioxidant enzymes and molecular chaperones in liver and heart were down regulated in sea bream fed SW diets. This study suggests that the antioxidant properties of heat-treated SW may have a protective role against oxidative stress. The nature of these compounds and possible mechanisms implied are currently being investigated.
