Abstract

Mass cytometry is a variation of conventional flow cytometry using metal tagged antibodies for cell staining instead of fluorochromes and detection in a mass cytometer, a modified mass spectrometer that allows for separation of discrete masses of these metal tags by time of flight (TOF). Currently, up to 50 different metal tags are available for cell analysis. The lack of any significant mass spectral overlap and autofluorescence background makes mass cytometry uniquely suited for complex high-dimensional phenotypic and functional analysis at the single cell level, thus accelerating biomarker discovery and drug screening. Here we describe a workflow for phenotyping of human peripheral blood mononuclear cells (PBMCs) covering cell staining, instrument setup of a Fluidigm Helios™ mass cytometer, and sample acquisition, and summarize a basic workflow of data analysis.

Abstract

El objetivo de esta investigación fue Determinar la relación que existe entre la gestión organizacional y la calidad de servicio docente en la unidad educativa “Barreiro”-Ecuador- 2018. El estudio se procesó bajo el método cuantitativo. La investigación fue de tipo correlacional, se utilizó un diseño descriptivo - correlacional, la muestra que se seleccionó estuvo conformada por 50 docentes. La recolección de información se realizó aplicando un cuestionario de opinión sobre gestión organizacional y otro cuestionario sobre calidad de servicio docente, los mismos que fueron validados mediante la matriz de validación respectiva. Para el análisis de la información se utilizaron las pruebas estadísticas r de Pearson y la t de student, las que permitieron determinar la relación entre las variables y comprobar las hipótesis. Los resultados mostraron también un predominio del nivel de gestión organizacional regular con un 60% y una prevalencia del nivel de calidad de servicio docente regular con el 66%.También determinaron que existe relación significativa entre ambas variables. El grado de relación según el coeficiente r de Pearson fue de 0,710** (Sig.= 0.000 < 0,01) lo que indica una correlación alta, directa y significativa a nivel 0.01. 158. HUBUNGAN TINGKAT PENGETAHUAN DAN TINGKAT KECEMASAN LANJUT USIA TERHADAP COVID-19

Abstract

Coronavirus (COVID-19) adalah penyakit menular yang dapat ditularkan dari seseorang yang positif Covid-19 melalui percikan cairan dari mulut atau hidung saat orang tersebut batuk dan bersin. Covid-19 ini sudah menimbulkan kecemasan di kalangan masyarakat global khususnya lanjut usia yang sangat rentan kepada penularan penyakit ini. Asumsi dasar penelitian ini adalah bahwa lansia yang memiliki pengetahuan yang baik terhadap Covid-19 dapat menurunkan kecemasan mereka terhadap penularan penyakit ini. Oleh Karena itu penelitian ini bertujuan untuk Menggambarkan tingkat pengetahuan dan tingkat kecemasan lansia terhadap covid-19, dan untuk mengetahui apakah ada hubungan yang signifikan antara kedua variabel tersebut. Metode yang digunakan dalam penelitian ini adalah deskriptif dengan jenis data diperoleh berdasarkan survey menggunakan kuesioner tingkat pengetahuan dan tingkat kecemasan. Populasinya adalah lansia dengan jumlah sampel 32 orang. Data dikumpulkan dengan menggunakan kuesioner yang disebarkan melalui melalui google form. Analisis data menggunakan SPSS. Didapati bahwa tingkat pengetahuan lansia termasuk dalam kategori cukup dengan nilai rata-rata 79.92%. tingkat kecemasan lansia termasuk dalam kecemasan ringan/tanpa gejala sebanyak (63%). Hubungan tingkat pengetahuan dan kecemasan berada pada kategori hubungan lemah dengan nilai r hitung adalah -0,091 dan nilai p-value 0,619>0,05 yang mengindikasikan tidak ada hubungan yang signifikan antara tingkat pengetahuan dan tingkat kecemasan lansia terhadap Covid-19. Kata Kunci: Covid-19, Kecemasan, Pengetahuan Abstract Coronavirus (COVID-19) is an infectious disease that can be transmitted from someone who is positive for Covid-19 through droplets of liquid from the mouth or nose when the person coughs and sneezes. Covid-19 has caused anxiety among the global community, especially the elderly who are very vulnerable to the transmission of this disease. The basic assumption of this study is that the elderly who have good knowledge of Covid-19 can reduce their anxiety about the transmission of this disease. Therefore, this study aims to describe the level of knowledge and anxiety levels of the elderly about Covid-19, and to find out whether there is a significant relationship between the two variables. The method used in this research is descriptive with the type of data obtained based on a survey using a questionnaire level of knowledge and level of anxiety. The population is the elderly with a sample size of 32 people. Data collection is done by distributing questionnaires via google form. Data analysis using SPSS. It was found that the knowledge level of the elderly was in the moderate category with an average score of 79.92%. The level of anxiety in the elderly was classified as mild / asymptomatic as much (63%). The relationship between the level of knowledge and anxiety is in the weak relationship category with the calculated r value of -0.091 and a p-value of 0.619> 0.05, which indicates that there is no significant relationship between the level of knowledge and the level of anxiety in the elderly against Covid-19. Abstract

Verticillium dahliae is a highly detrimental pathogen of soil cultivated strawberry (Fragaria x ananassa). Breeding of Verticillium wilt resistance into commercially viable strawberry cultivars can help mitigate the impact of the disease. In this study we describe novel sources of resistance identified in multiple strawberry populations, creating a wealth of data for breeders to exploit. Pathogen-informed experiments have allowed the differentiation of subclade-specific resistance responses, through studying V. dahliae subclade II-1 specific resistance in the cultivar “Redgauntlet” and subclade II-2 specific resistance in “Fenella” and “Chandler.” A large-scale low-cost phenotyping platform was developed utilizing automated unmanned vehicles and near infrared imaging cameras to assess field-based disease trials. The images were used to calculate disease susceptibility for infected plants through the normalized difference vegetation index score. The automated disease scores showed a strong correlation with the manual scores. A co-dominant resistant QTL; FaRVd3D, present in both “Redgauntlet” and “Hapil” cultivars exhibited a major effect of 18.3% when the two resistance alleles were combined. Another allele, FaRVd5D, identified in the “Emily” cultivar was associated with an increase in Verticillium wilt susceptibility of 17.2%, though whether this allele truly represents a susceptibility factor requires further research, due to the nature of the F1 mapping population. Markers identified in populations were validated across a set of 92 accessions to determine whether they remained closely linked to resistance genes in the wider germplasm. The resistant markers FaRVd2B from “Redgauntlet” and FaRVd6D from “Chandler” were associated with resistance across the wider germplasm. 170. Surface acoustic wave actuated rotations for microfluidics

Abstract

Microfluidics and the lab-on-a-chip concept promises to open up new possibilities in miniaturised portable fluid systems and point-of-care diagnostics. Centrifugal microfluidics, most commonly recognised in the lab-on-a-CD concept, has already shown many applications in chemical and biological systems by exploiting the radial forcing associated with high rotational velocities. Bulk rotation of a 100 mm diameter plastic disc is however a cumbersome approach to driving nano-litres of fluid, and furthermore the rotations require large external motor systems for actuation. Surface Acoustic Wave (SAW) driven microfluidics can address these issues by exploiting a very efficient mechanism of energy transfer to the fluids themselves. SAWs have been shown to actuate a large range of microfluidic processes on a truly miniaturised scale, owing to the very efficient coupling that occurs between the surface waves and fluids that are in contact with them. By clever use of wave blocking and substrate geometries it has been shown that SAWs can generate rotational flows with applications in micromixing and centrifugation [Biomed. Microdev. 9, 647-656 (2007)]. In this thesis we report on the use of focussed SAWs to generate very high rotational velocities in microfluids, and an associated reduction in process time for micromixing and particle concentrations. We then further explore the process by which SAWs promote fluid mixing and show for the first time that the mixing process is in fact dominated by chaotic advection. The last part of the thesis explores these ultra-fast fluid rotations for use as a coupling layer for a high speed miniaturised motor system that could be used as a truly portable centrifugal microfluidics platform, akin to the larger lab-on-a-CD concept. At even further reduced scales, we show that by removing the fluid coupling layer we can drive 1 mm diameter rotors at ∼ 10, 000 rpm by direct contact with focussed SAWs. In this thesis we aim to demonstrate some of the vast potential that SAW driven ultra-fast rotational flows have in actuating centrifugal microfluidics on truly portable, solid state devices.

Abstract

Fingerprints are distinct physical characteristics that remain unchanged throughout an individual's lifetime. This study derived Filipino-specific probability formulae from fingerprints to be used for sex discrimination in human identification cases. Ridge density from three different areas - distal radial area, distal ulnar area, and proximal area - as well as white line counts from fingerprints of 200 male and 200 female Filipinos were collected and analyzed statistically. Ridge densities of radial and ulnar areas emerged as displaying significant differences between the sexes, with 16ridges/25mm(2) or more in radial area and 15ridges/25mm(2) or more in ulnar area being more likely to be female, whereas 13ridges/25mm(2) or less in radial area and 12ridges/25mm(2) or less in ulnar area were more likely to be male. A white line count of 0 was more likely to be male while a white line count of 2 or more was more likely to be female. The results of this study show sex differences in Filipino fingerprints and support the observation of previous studies that females have finer ridges than males.
