151. XUE. Molecular inventory in the inner region of an extremely irradiated Protoplanetary Disk

Abstract: We present the first results of the eXtreme UV Environments (XUE) James Webb Space Telescope (JWST) program, that focuses on the characterization of planet forming disks in massive star forming regions. These regions are likely representative of the environment in which most planetary systems formed. Understanding the impact of environment on planet formation is critical in order to gain insights into the diversity of the observed exoplanet populations. XUE targets 15 disks in three areas of NGC 6357, which hosts numerous massive OB stars, among which some of the most massive stars in our Galaxy. Thanks to JWST we can, for the first time, study the effect of external irradiation on the inner ($< 10$ au), terrestrial-planet forming regions of proto-planetary disks. In this study, we report on the detection of abundant water, CO, CO$_2$, HCN and C$_2$H$_2$ in the inner few au of XUE 1, a highly irradiated disk in NGC 6357. In addition, small, partially crystalline silicate dust is present at the disk surface. The derived column densities, the oxygen-dominated gas-phase chemistry, and the presence of silicate dust are surprisingly similar to those found in inner disks located in nearby, relatively isolated low-mass star-forming regions. Our findings imply that the inner regions of highly irradiated disks can retain similar physical and chemical conditions as disks in low-mass star-forming regions, thus broadening the range of environments with similar conditions for inner disk rocky planet formation to the most extreme star-forming regions in our Galaxy., Comment: Accepted for publication in ApJ Letters. 20 pages, 7 figures

152. ALMA Observations of the Extraordinary Carina Pillars: A Complementary Sample

Abstract: We present a study of six dusty and gaseous pillars (containing the HH 1004 and HH 1010 objects) and globules (that contain the HH 666, HH 900, HH 1006, and HH 1066 objects) localized in the Carina nebula using sensitive and high angular resolution ($\sim$0.3$''$) Atacama Large Millimeter/Sub-millimeter Array (ALMA) observations. This is a more extensive study that the one presented in \citet{Cortes}. As in this former study, we also analyzed the 1.3 mm continuum emission and C$^{18}$O(2$-$1), N$_2$D$^+$(3$-$2) and $^{12}$CO(2$-$1) spectral lines. These new observations revealed the molecular outflows emanating from the pillars, the dusty envelopes$+$disks that are exciting them, and the extended HH objects far from their respective pillars. We reveal that the masses of the disks$+$envelopes are in a range of 0.02 to 0.38 M$_\odot$, and those for the molecular outflows are of the order of 10$^{-3}$ M$_\odot$, which suggests that their exciting sources might be low- or intermediate-mass protostars as already revealed in recent studies at infrared and submillimeter bands. In the regions associated with the objects HH 900 and HH 1004, we report multiple millimeter continuum sources, from where several molecular outflows emanate.

Abstract: The use of formal privacy to protect the confidentiality of responses in the 2020 Decennial Census of Population and Housing has triggered renewed interest and debate over how to measure the disclosure risks and societal benefits of the published data products. Following long-established precedent in economics and statistics, we argue that any proposal for quantifying disclosure risk should be based on pre-specified, objective criteria. Such criteria should be used to compare methodologies to identify those with the most desirable properties. We illustrate this approach, using simple desiderata, to evaluate the absolute disclosure risk framework, the counterfactual framework underlying differential privacy, and prior-to-posterior comparisons. We conclude that satisfying all the desiderata is impossible, but counterfactual comparisons satisfy the most while absolute disclosure risk satisfies the fewest. Furthermore, we explain that many of the criticisms levied against differential privacy would be levied against any technology that is not equivalent to direct, unrestricted access to confidential data. Thus, more research is needed, but in the near-term, the counterfactual approach appears best-suited for privacy-utility analysis., Comment: 47 pages, 1 table

Abstract: We report measurements of the branching fractions and direct $\it{CP}$ asymmetries of the decays $B^0 \to K^+ \pi^-$, $B^+ \to K^+ \pi^0$, $B^+ \to K^0 \pi^+$, and $B^0 \to K^0 \pi^0$, and use these for testing the standard model through an isospin-based sum rule. In addition, we measure the branching fraction and direct $\it{CP}$ asymmetry of the decay $B^+ \to \pi^+\pi^0$ and the branching fraction of the decay $B^0 \to \pi^+\pi^-$. The data are collected with the Belle II detector from $e^+e^-$ collisions at the $\Upsilon(4S)$ resonance produced by the SuperKEKB asymmetric-energy collider and contain $387\times 10^6$ bottom-antibottom meson pairs. Signal yields are determined in two-dimensional fits to background-discriminating variables, and range from 500 to 3900 decays, depending on the channel. We obtain $-0.03 \pm 0.13 \pm 0.04$ for the sum rule, in agreement with the standard model expectation of zero and with a precision comparable to the best existing determinations.

Abstract: We determine the CKM matrix-element magnitude $|V_{cb}|$ using $\overline{B}^0\to D^{*+}\ell^-\bar\nu_\ell$ decays reconstructed in $189 \, \mathrm{fb}^{-1}$ of collision data collected by the Belle II experiment, located at the SuperKEKB $e^+e^-$ collider. Partial decay rates are reported as functions of the recoil parameter $w$ and three decay angles separately for electron and muon final states. We obtain $|V_{cb}|$ using the Boyd-Grinstein-Lebed and Caprini-Lellouch-Neubert parametrizations, and find $|V_{cb}|_\mathrm{BGL}=(40.57\pm 0.31 \pm 0.95\pm 0.58)\times 10^{-3}$ and $|V_{cb}|_\mathrm{CLN}=(40.13 \pm 0.27 \pm 0.93\pm 0.58 )\times 10^{-3}$ with the uncertainties denoting statistical components, systematic components, and components from the lattice QCD input, respectively. The branching fraction is measured to be ${\cal B}(\overline{B}^0\to D^{*+}\ell^-\bar\nu_\ell)=(4.922 \pm 0.023 \pm 0.220)\%$. The ratio of branching fractions for electron and muon final states is found to be $0.998 \pm 0.009 \pm 0.020$. Abstract: There is an ongoing effort to find quantum speedups for learning problems. Recently, [Y. Liu et al., Nat. Phys. $\textbf{17}$, 1013--1017 (2021)] have proven an exponential speedup for quantum support vector machines by leveraging the speedup of Shor's algorithm. We expand upon this result and identify a speedup utilizing Grover's algorithm in the kernel of a support vector machine. To show the practicality of the kernel structure we apply it to a problem related to pattern matching, providing a practical yet provable advantage. Abstract: Massive star-forming regions are thought to be the most common birth environments in the Galaxy and the only birth places of very massive stars. Their presence in the stellar cluster alters the conditions within the cluster impacting at the same time the evolution of other cluster members. In principle, copious amounts of ultraviolet radiation produced by massive stars can remove material from outer parts of the protoplanetary disks around low- and intermediate-mass stars in the process of external photoevaporation, effectively reducing the planet-formation capabilities of those disks. Here, we present deep VLT/MUSE observations of low-mass stars in Trumpler 14, one of the most massive, young, and compact clusters in the Carina Nebula Complex. We provide spectral and stellar properties of 717 sources and based on the distribution of stellar ages derive the cluster age of $\sim$1~Myr. The majority of the stars in our sample have masses $\leqslant$1~$M_\odot$, what makes our spectroscopic catalogue the most deep to date in term of masses, and proves that detailed investigations of low-mass stars are possible in the massive but distant regions. Spectroscopic studies of low-mass members of the whole Carina Nebula Complex are missing. 159. The Impact of Exposed Passwords on Honeyword Efficacy

Abstract: Honeywords are decoy passwords that can be added to a credential database; if a login attempt uses a honeyword, this indicates that the site's credential database has been leaked. In this paper we explore the basic requirements for honeywords to be effective, in a threat model where the attacker knows passwords for the same users at other sites. First, we show that for user-chosen (vs. algorithmically generated, i.e., by a password manager) passwords, existing honeyword-generation algorithms do not simultaneously achieve false-positive and false-negative rates near their ideals of $\approx 0$ and $\approx \frac{1}{1+n}$, respectively, in this threat model, where $n$ is the number of honeywords per account. Second, we show that for users leveraging algorithmically generated passwords, state-of-the-art methods for honeyword generation will produce honeywords that are not sufficiently deceptive, yielding many false negatives. Instead, we find that only a honeyword-generation algorithm that uses the \textit{same} password generator as the user can provide deceptive honeywords in this case. However, when the defender's ability to infer the generator from the (one) account password is less accurate than the attacker's ability to infer the generator from potentially many, this deception can again wane. Abstract: In explainable artificial intelligence (XAI) research, the predominant focus has been on interpreting models for experts and practitioners. Model agnostic and local explanation approaches are deemed interpretable and sufficient in many applications. However, in domains like healthcare, where end users are patients without AI or domain expertise, there is an urgent need for model explanations that are more comprehensible and instil trust in the model's operations. We hypothesise that generating model explanations that are narrative, patient-specific and global(holistic of the model) would enable better understandability and enable decision-making. We test this using a decision tree model to generate both local and global explanations for patients identified as having a high risk of coronary heart disease. These explanations are presented to non-expert users. We find a strong individual preference for a specific type of explanation. The majority of participants prefer global explanations, while a smaller group prefers local explanations. A task based evaluation of mental models of these participants provide valuable feedback to enhance narrative global explanations. This, in turn, guides the design of health informatics systems that are both trustworthy and actionable.

161. Fully Synthetic Data for Complex Surveys

Abstract: When seeking to release public use files for confidential data, statistical agencies can generate fully synthetic data. We propose an approach for making fully synthetic data from surveys collected with complex sampling designs. Our approach adheres to the general strategy proposed by Rubin (1993). Specifically, we generate pseudo-populations by applying the weighted finite population Bayesian bootstrap to account for survey weights, take simple random samples from those pseudo-populations, estimate synthesis models using these simple random samples, and release simulated data drawn from the models as public use files. To facilitate variance estimation, we use the framework of multiple imputation with two data generation strategies. In the first, we generate multiple data sets from each simple random sample. In the second, we generate a single synthetic data set from each simple random sample. We present multiple imputation combining rules for each setting. We illustrate the repeated sampling properties of the combining rules via simulation studies, including comparisons with synthetic data generation based on pseudo-likelihood methods. We apply the proposed methods to a subset of data from the American Community Survey.

162. Into the Mystic: ALMA ACA observations of the Mystic Mountains in Carina

Abstract: We present new observations of the Mystic Mountains cloud complex in the Carina Nebula using the ALMA Atacama Compact Array (ACA) to quantify the impact of strong UV radiation on the structure and kinematics of the gas. Our Band~6 observations target CO, $^{13}$CO, and C$^{18}$O; we also detect DCN J=3-2 and $^{13}$CS J=5-4. A dendrogram analysis reveals that the Mystic Mountains are a coherent structure, with continuous emission over $-$10.5 km s$^{-1}$ $<$ v < $-$2 km s$^{-1}$. We perform multiple analyses to isolate non-thermal motions in the Mystic Mountains including computing the turbulent driving parameter, $b$, which indicates whether compressive or solenoidal modes dominate. Each analysis yields values similar to other pillars in Carina that have been observed in a similar way but are subject to an order of magnitude less intense ionizing radiation. We find no clear correlation between the velocity or turbulent structure of the gas and the incident radiation, in contrast to other studies targeting different regions of Carina. This may reflect differences in the initial densities of regions that go on to collapse into pillars and those that still look like clouds or walls in the present day. Pre-existing over-densities that enable pillar formation may also explain why star formation in the pillars appears more evolved (from the presence of jets) than in other heavily-irradiated but non-pillar-like regions. 163. Fast rotating Blue Straggler Stars in the Globular Cluster NGC3201

Abstract: We used high resolution spectra acquired at the Magellan Telescope to measure radial and rotational velocities of approximately 200 stars in the Galactic globular cluster NGC 3201. The surveyed sample includes Blue Stragglers Stars (BSSs) and reference stars in different evolutionary stages (main sequence turn-off, sub-giant, red giant and asymptotic giant branches). The average radial velocity value ($\langle V_r\rangle = 494.5 \pm 0.5$ km s$^{-1}$) confirms a large systemic velocity for this cluster and was used to distinguish 33 residual field interlopers. The final sample of member stars counts 67 BSSs and 114 reference stars. Similarly to what is found in other clusters, the totality of the reference stars has negligible rotation ($<20$ km s$^{-1}$), while the BSS rotational velocity distribution shows a long tail extending up to $\sim 200$ km s$^{-1}$, with 19 BSSs (out of 67) spinning faster than 40 km s$^{-1}$. This sets the percentage of fast rotating BSSs to $\sim 28\%$. Such a percentage is roughly comparable to that measured in other loose systems ($\omega$ Centauri, M4 and M55) and significantly larger than that measured in high-density clusters (as 47 Tucanae, NGC 6397, NGC 6752 and M30). This evidence supports a scenario where recent BSS formation (mainly from the evolution of binary systems) is occurring in low-density environments. We also find that the BSS rotational velocity tends to decrease for decreasing luminosity and surface temperature, similarly to what is observed in main sequence stars. The relationship between tumor location and its detection/missing by radiologists on mpMRI compared to WMHP as a reference standard was investigated.Statistical testsA weighted chi-square test was performed to examine the statistical differences for rCP, DR, and PPV of the aggregated sectors within the zone, anterior/posterior, left/right prostate, and different levels of the prostate with a statistically significant level of 0.05.ResultsA total of 1665 PCa lesions were identified in 1143 patients, and from those 1060 lesions were clinically significant (cs)PCa tumors (any Gleason score [GS] ≥7). Our sector-based analysis utilizing weighted chi-square tests suggested that the left posterior part of PZ had a high likelihood of missing csPCa lesions at a DR of 67.0%. Aggregated sector analysis indicated that the anterior or apex locations in PZ had the significantly lowest csPCa detection at 67.3% and 71.5%, respectively.Data conclusionSpatial characteristics of the per-lesion-based mpMRI performance for diagnosis of PCa were studied. Research suggests improved accuracy over conventional imaging and other PET radiotracers in many contexts. With multiple approved PSMA-targeting radiotracers, PSMA PET will become even more available in clinical practice. Its increased use requires an understanding of the prospective data available and caution when extrapolating from prior trial data that utilized other imaging modalities. Future trials leveraging PSMA PET for treatment optimization and management decision-making will ultimately drive its clinical utility.

Abstract: 284 Background: Multiparametric MRI (mpMRI) and PSMA-PET are complementary imaging modalities used in the pre-surgical evaluation of patients with prostate cancer (PCa). The aim of this study was to evaluate the imaging and pathology parameters associated with PSMA-PET and mpMRI disagreement with each other and with gold standard histopathology. Methods: Patients undergoing radical prostatectomy (RP) with pre-surgical PSMA-PET and mpMRI were screened for inclusion in this retrospective analysis. Patients had the imaging scans done 3, n=153). mpMRI and PSMA PET identified 138 and 170 lesions, respectively. Sensitivity for ISUP>3 lesions was 79% and 87% for mpMRI and PSMA-PET, respectively. 115/153 (76%) ISUP> 3 lesions were correctly identified, and 14/153 (9%) were missed by both imaging modalities, respectively. 5/153 (3%) were correctly identified by mpMRI and missed by PSMA-PET, 17/153 (11%) were correctly identified by PSMA-PET and missed by mpMRI. Lesion’s ISUP grade group and size on pathology were significantly lower in mpMRI- compared to mpMRI+ and in PET- compared to PET+ lesions. SUVmax and SUVmean were significantly higher in PET+/MRI+ than PET+/MRI- lesions. Lesion’s ISUP grade group and size were significantly correlated to PSMA-PET and mpMRI agreement, whereas PSMA-PET metrics were not. Logistic regression model showed that lesions with lower ISUP, smaller size, and lack of aggressive prostate cancer features on pathology (cribriform pattern and intraductal carcinoma) have higher likelihood of being missed by PET and mpMRI and of PET/MRI disagreement. Conclusions: Higher SUVmax, SUVmean, and tumor volume on PSMA-PET were associated with a TP finding on PSMA-PET, higher ISUP grade group and size on pathology were associated with a TP finding on both PSMA-PET and mpMRI, and with agreement between PSMA-PET and mpMRI.

Abstract: Micro-ultrasound has recently been introduced as a low-cost alternative to multi-parametric MRI for imaging prostate cancer. Early clinical studies have demonstrated promising results; however, robust validation via comparison with whole-mount pathology has yet to be achieved. Due to micro-ultrasound probe design and tissue deformation during scanning, it is difficult to accurately correlate micro-ultrasound imaging planes with ground truth whole-mount pathology slides. In this study, we developed a multi-step methodology to co-register micro-ultrasound and MRI to whole-mount pathology. The three-step process had a registration error of 3.90 ± 0.11 mm and consists of: (1) micro-ultrasound image reconstruction, (2) 3D landmark registration of micro-ultrasound to MRI, and (3) 2D capsule registration of MRI to whole-mount pathology. This process was then used in a preliminary reader study to compare the diagnostic accuracy of micro-ultrasound and MRI in 15 patients who underwent radical prostatectomy for prostate cancer. Micro-ultrasound was found to have equivalent performance to retrospective MRI review for index lesion detection (91.7% vs. 80%), while demonstrating an increased detection of tumor extent (52.5% vs. 36.7%) with similar false positive regions-of-interest (38.3% vs. 40.8%). Prospective MRI review had reduced detection of index lesions (73.3%) and tumor extent (18.9%) but improved false positive regions-of-interest (22.7%) relative to micro-ultrasound and retrospective MRI. Further evaluation is needed with a larger sample size.

Abstract: U.S. prisons were especially susceptible to COVID-19 infection and death; however, data limitations have precluded a national accounting of prison mortality (including but not limited to COVID-19 mortality) during the pandemic. Our analysis of mortality data collected from public records requests (supplemented with publicly available data) from 48 Departments of Corrections provides the most comprehensive understanding to date of in-custody mortality during 2020. We find that total mortality increased by 77% in 2020 relative to 2019, corresponding to 3.4 times the mortality increase in the general population, and that mortality in prisons increased across all age groups (49 and under, 50 to 64, and 65 and older). COVID-19 was the primary driver for increases in mortality due to natural causes; some states also experienced substantial increases due to unnatural causes. These findings provide critical information about the pandemic's toll on some of the country's most vulnerable individuals while underscoring the need for data transparency and standardized reporting in carceral settings.

Abstract: Abstract To examine the morphological impact of deep learning (DL)-quantified biomarkers on point-wise sensitivity (PWS) using microperimetry (MP) and optical coherence tomography (OCT) in intermediate AMD (iAMD). Patients with iAMD were examined by OCT (Spectralis). DL-based algorithms quantified ellipsoid zone (EZ)-thickness, hyperreflective foci (HRF) and drusen volume. Outer nuclear layer (ONL)-thickness and subretinal drusenoid deposits (SDD) were quantified by human experts. All patients completed four MP examinations using an identical custom 45 stimuli grid on MP-3 (NIDEK) and MAIA (CenterVue). MP stimuli were co-registered with corresponding OCT using image registration algorithms. Multivariable mixed-effect models were calculated. 3.600 PWS from 20 eyes of 20 patients were analyzed. Abstract: Abstract Lipoprotein (a) [Lp(a)] is suspected to have antifibrinolytic effects, however, its relevance for the severity of venous thromboembolic events remains unclear. We studied the association of Lp(a) levels with thrombus load in pulmonary embolism (PE). 90 patients (40% female, median age 70 [56–79] years) at our tertiary care hospital with a diagnosis of acute PE, available Lp(a) levels and CT pulmonary angiography (CT-PA) performed between April 2017 and December 2019 were included. All CT-PA scans were reanalyzed and thrombus load was determined via Qanadli CT obstruction index (CTOI) and most proximal thrombus location. Median Lp(a) levels were 11.4 [9.3–29.1] mg/dL, median D-dimer levels were 4.6 [2.1–9.8] mg/L, median CTOI was 23 [8–50], central PE was present in 27 (30%) patients. Abstract: Abstract The Epigravettian human remains from Riparo Tagliente in northern Italy represent some of the earliest evidence of human occupation in the southern Alpine slopes after the Last Glacial Maximum. Genomic analyses of the 17,000-year-old Tagliente 2 mandible revealed the oldest presence of a genetic profile with affinities to the Near East in the Italian peninsula, which later became the most widespread hunter-gatherer ancestry across Europe. However, a comparable biomolecular characterization of the Tagliente 1 burial remains unavailable, preventing us from defining its biological relationships with Tagliente 2. Here, we apply paleogenomic, isotopic, and radiocarbon dating analyses on a femur fragment of Tagliente 1 and compare the reconstructed data with previously reported results from Tagliente 2. Despite their different isotopic signatures and non-overlapping radiocarbon dates, we reveal that the two human remains belong to the same male individual. We determine that the distinct isotopic values can be explained by different dietary practices during lifetime, whereas the non-overlapping radiocarbon dates can be caused by minimal radiocarbon contamination, possibly deriving from chemical treatments for conservation purposes. These findings highlight the importance of interdisciplinary biomolecular studies in offering new perspectives on the Palaeolithic fossil record and addressing long-standing bioarchaeological questions.

Abstract: Abstract We report a determination of the CKM angle ϕ 3, also known as γ, from a combination of measurements using samples of up to 711 fb −1 from the Belle experiment and up to 362 fb −1 from the Belle II experiment. We combine results from analyses of B + → DK +, B + → Dπ +, and B + → D * K + decays, where D is an admixture of D 0 and D ¯ 0 $$ {\overline{D}}^0 $$ mesons, in a likelihood fit to obtain ϕ 3 = (75.2 ± 7.6) ° . We also briefly discuss the interpretation of this result.

Abstract: Abstract Objective The piriform cortex is considered to be highly epileptogenic. Its resection during epilepsy surgery is a predictor for postoperative seizure freedom in temporal lobe epilepsy. Epilepsy is associated with a dysfunction of the blood–brain barrier. We investigated blood–brain barrier dysfunction in the piriform cortex of people with temporal lobe epilepsy using quantitative T1‐relaxometry. Methods Gadolinium‐based contrast agent was administered ictally and interictally in 37 individuals before undergoing quantitative T1‐relaxometry. Postictal and interictal images were co‐registered, and subtraction maps were created as biomarkers for peri‐ictal (∆qT1interictal‐postictal) and interictal (∆qT1noncontrast‐interictal) blood–brain barrier dysfunction. Values were extracted for the piriform cortex, hippocampus, amygdala, and the whole cortex. Results In temporal lobe epilepsy (n = 14), ∆qT1noncontrast‐interictal was significantly higher in the piriform cortex than in the whole cortex (p = 0.02). In extratemporal lobe epilepsy (n = 23), ∆qT1noncontrast‐interictal was higher in the hippocampus than in the whole cortex (p = 0.05). Abstract: Abstract We report measurements of the e + e − → B B ¯ $$ B\overline{B} $$ , B B ¯ ∗ $$ B{\overline{B}}^{\ast } $$ , and B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ cross sections at four energies, 10653, 10701, 10746 and 10805 MeV, using data collected by the Belle II experiment. We reconstruct one B meson in a large number of hadronic final states and use its momentum to identify the production process. In the first 2 – 5 MeV above B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ threshold, the e + e − → B ∗ B ¯ ∗ $$ {B}^{\ast }{\overline{B}}^{\ast } $$ cross section increases rapidly. This may indicate the presence of a pole close to the threshold.

181. Dynamic chromatin accessibility and transcriptome changes following PDGF-BB treatment of bone-marrow derived mesenchymal stem cells

Abstract: Abstract Background Mesenchymal stem cells (MSCs) are multipotent stem cells that are under investigation for use in clinical trials because they are capable of self-renewal and differentiating into different cell types under defined conditions. Nonetheless, the therapeutic effects of MSCs have been constrained by low engraftment rates, cell fusion, and cell survival. Various strategies have been explored to improve the therapeutic efficacy of MSCs, with platelet-derived growth factor (PDGF)-BB emerging as a promising candidate. To enhance our comprehension of the impact of PDGF-BB on the gene expression profile and chromosomal accessibility of MSCs, RNA-sequencing and analysis of chromatin accessibility profiles were conducted on three human primary MSCs in culture, both with and without stimulation by PDGF-BB. Results Integrative analysis of gene expression and chromatin accessibility demonstrated that PDGF-BB treatment modified the chromatin accessibility landscape, marking regions for activation or repression through the AP-1 family transcription factors TEAD, CEBP, and RUNX2. These changes in AP-1 transcription factor expression, in turn, led to cell proliferation and differentiation potential towards osteoblasts, adipocytes, or chondrocytes. The degree of MSC differentiation varies among cells isolated from different donors. The presence of an enrichment of exosome-related genes is also noted among all the differentially expressed genes. Conclusions In conclusion, the observed changes in AP-1 transcription factor expression not only induced cellular proliferation and differentiation, but also revealed variations in the degree of MSC differentiation based on donor-specific differences. Moreover, the enrichment of exosome-related genes among differentially expressed genes suggests a potential significant role for PDGF-BB in facilitating intercellular communication.

Abstract: Abstract Increasing aridity in the Mediterranean region will result in longer and recurrent drought. These changes could strongly modify plant defenses, endangering tree survival. We investigate the response of chemical defenses from central and specialized metabolism in Quercus pubescens Willd. to future Mediterranean drought using a long-term drought experiment in natura where trees have been submitted to amplified drought (~ –30% annual precipitation) since April 2012. We focused on leaf metabolites including chlorophylls and carotenoids (central metabolism) and flavonols (specialized metabolism). Measurements were performed in summer from 2016 to 2022. Amplified drought led to higher concentrations of total photosynthetic pigments over the 2016–2022 period. However, it also led to lower AZ/VAZ and flavonol concentrations. Additionally, chemical defenses of Q. pubescens responded to previous precipitation where low precipitation 1 year and/or 2 years preceding sampling was associated to low concentrations of VAZ, flavonol and high neoxanthin concentrations. Our study indicates that the decline of flavonol concentration under long-term drought is counterbalanced by a higher production of several central metabolites. Such results are potentially due to an adjustment in tree metabolism, highlighting the importance of performing long-term experimental studies in natura for assessing drought legacy effects and thus forest adaptation to climate change.

Abstract: Abstract The ongoing quest to discover new phenomena at the LHC necessitates the continuous development of algorithms and technologies. Established approaches like machine learning, along with emerging technologies such as quantum computing show promise in the enhancement of experimental capabilities. In this work, we propose a strategy for anomaly detection tasks at the LHC based on unsupervised quantum machine learning, and demonstrate its effectiveness in identifying new phenomena. The designed quantum models-an unsupervised kernel machine and two clustering algorithms-are trained to detect new-physics events using a latent representation of LHC data, generated by an autoencoder designed to accommodate current quantum hardware limitations on problem size. For kernel-based anomaly detection, we implement an instance of the model on a quantum computer, and we identify a regime where it significantly outperforms its classical counterparts. We show that the observed performance enhancement is related to the quantum resources utilised by the model.

The current study aimed to determine the reliability of the modified three-stage cervical vertebrae maturation method for estimating the skeletal growth of males and females. Methods Pretreatment lateral cephalometric radiographs of 137 orthodontic patients were utilized to evaluate the intra- and inter-observer reliability of a modified cervical vertebrae maturation method. Four independent examiners categorized each radiograph into one of the three skeletal growth stages according to the cervical vertebrae morphology: (1) a pre-peak group (combining the CS1 and CS2 stages), (2) a peak group (combining the CS3 and CS4 stages), and (3) a post-peak group (combining the CS5 and CS6 stages). The Kappa index and the Fleiss multirater Kappa test were carried out to determine the reproducibility of this method. Results The studied sample included cephalometric radiographs of 68 males (49.6%) and 69 females (50.4%) aged between 7 and 58 years; no significant age difference was found between the sexes (p = 0.189). Abstract: Abstract To evaluate the intra- and interdevice repeatability of microperimetry (MP) assessments in patients with diabetic macular edema (DME) two consecutive MP testings (45 fovea-centered stimuli, 4–2 staircase strategy) were performed using MP3 (NIDEK, Aichi, Japan) and MAIA (CenterVue, Padova, Italy), respectively. Intraretinal fluid (IRF) and ellipsoid zone (EZ) thickness were automatically segmented by published deep learning algorithms. Hard exudates (HEs) were annotated semi-automatically and disorganization of retinal inner layers (DRIL) was segmented manually. Point-to-point registration of MP stimuli to corresponding spectral-domain OCT (Spectralis, Heidelberg Engineering, Germany) locations was performed for both devices. Repeatability was assessed overall and in areas of disease-specific OCT biomarkers using Bland-Altmann coefficients of repeatability (CoR). A total of 3600 microperimetry stimuli were tested in 20 eyes with DME. Global CoR was high using both devices (MP3: ± 6.55 dB, MAIA: ± 7.69 dB). Abstract: This article presents a series of recommendations for the publication of archaeological data, to improve their usability. These 12 recommendations were formulated by archaeological data experts who mined thousands of publications for different data types (including funerary practices, accelerator mass spectrometry dating, stable isotopes, zooarchaeology, archaeobotany and pathologies) during the initial construction of the Big Interdisciplinary Archaeological Database (BIAD). We also include data harmonisation vocabularies utilised for the integration of data from different recording systems. The case studies we cite to illustrate the recommendations are grounded in examples from the published literature and are presented in a problem/solution format. Though practically oriented towards the facilitation of efficient databasing, these recommendations – which we refer to as the BIAD Standards – are broadly applicable by those who want to extract scientific data from archaeological information, those who work with a specific region or theoretical focus and journal editors and manuscript authors. We anticipate that the use of the BIAD Standards will increase the usability, visibility, interoperability and longevity of published data and also increase the citations of those publications from which data were mined. The Standards will also help frame a unified foundation to support the continued integration of the natural sciences with archaeological research in the future.

Abstract: Abstract We present a study of Ξ c 0 → Ξ 0 π 0 $$ {\Xi}_c^0\to {\Xi}^0{\pi}^0 $$ , Ξ c 0 → Ξ 0 η $$ {\Xi}_c^0\to {\Xi}^0\eta $$ , and Ξ c 0 → Ξ 0 η ′ $$ {\Xi}_c^0\to {\Xi}^0{\eta}^{\prime } $$ decays using the Belle and Belle II data samples, which have integrated luminosities of 980 fb −1 and 426 fb −1, respectively. Abstract: Abstract Background Caring for patients with anorexia nervosa (AN) is associated with high levels of moral distress among healthcare professionals. The main moral conflict has been posited to be between applying coercion to prevent serious complications such as premature death and accepting treatment refusals. However, empirical evidence on this topic is scarce. Methods We identified all 19 documentations of ethics consultations (ECs) in the context of AN from one clinical ethics support service in Switzerland. These documentations were coded with a sequential deductive-inductive approach and the code system was interpreted in a case-based manner. Here, we present findings on patient characteristics and ethical concerns. Findings The ECs typically concerned an intensely pretreated, extremely underweight AN patient endangering herself by refusing the proposed treatment. In addition to the justifiability of coercion, frequent ethical concerns were whether further coerced treatment aimed at weight gain would be ineffective or even harmful, evidencing uncertainty about beneficence and non-maleficence and a conflict between these principles. Discussed options included harm reduction (e.g. psychotherapy without weight gain requirements) and palliation (e.g. initiating end-of-life care), the appropriateness of which were ethical concerns in themselves. Overall, nine different types of conflicts between or uncertainties regarding ethical principles were identified with a median of eight per case. Conclusions Ethical concerns in caring for persons with AN are diverse and complex. To deal with uncertainty about and conflict between respect for autonomy, beneficence and non-maleficence, healthcare professionals consider non-curative approaches. However, currently, uncertainty around general justifiability, eligibility criteria, and concrete protocols hinders their adoption.

Patents for many existing biological products are expiring, and generic versions, which are referred to as biosimilars, are produced to serve as an alternative to the reference medicinal product (RMP) cutting down the costs and expanding access. The present paper assesses the analytical similarity between Formycon’s FYB206 pembrolizumab biosimilar candidate and Keytruda®, an RMP that is approved to treat various types of cancer, with the intention of determining FYB206’s suitability to enter clinical biosimilar trials. Methods Monoclonal antibodies (mAbs) are biological medicinal products that are characterized by a high overall heterogeneity. Due to the complex nature of these molecules, a comprehensive comparative analytical assessment was designed to demonstrate analytical similarity in all clinically relevant quality attributes between RMP and the corresponding biosimilar candidate. This exercise addresses physicochemical, biophysical as well as functional characteristics. Results The comparative analytical evaluation results demonstrate that the proposed biosimilar is structurally and functionally highly similar to the RMP, showing only minor differences for some quality attributes that are justified to be noncritical for clinical efficacy and safety. Conclusion Based on physicochemical and biological characteristics, FYB206 is suitable to enter the clinical phase.

Patient characteristics may contribute to postoperative difficulties thereby increasing risk of hospital readmission. The objective of this study was to determine the frequency of and reasons for 30-day readmission and postoperative complications following ORIF for ankle fractures. Methods A retrospective review of the National Surgical Quality Improvement Program (NSQIP) database from 2015 to 2021 identified patients undergoing ORIF for ankle fractures. Patient demographics, complication incidence, and reasons for unplanned hospital readmission were collected. Multivariable analyses identified patient risk factors for any adverse event (AAE) and readmission within 30-days of surgery. Results The 29,905 patients queried who underwent ORIF procedures for ankle fractures between 2015 and 2021 were 49.6 ± 18.40 years of age, 30.9 ± 7.10 kg/m2, and 40.81% male. Of this cohort, 981 (3.30%) experienced 30-day postoperative adverse events, with surgical site infections (SSI; 1.25%) the most common. Unplanned readmission was observed in 2.08% of patients after a mean of 14.64 days. Surgical site related readmissions were 20.55% (n = 128) of reported readmissions with the most common being superficial incisional SSI. Abstract: Abstract We present the result of a search for the charged-lepton-flavor violating decay τ − → μ − μ + μ − using a 424 fb −1 sample of data recorded by the Belle II experiment at the SuperKEKB e + e − collider. The selection of e + e − → τ + τ − events is based on an inclusive reconstruction of the non-signal tau decay, and on a boosted decision tree to suppress background. We observe one signal candidate, which is compatible with the expectation from background processes. We set a 90% confidence level upper limit of 1.9 × 10 −8 on the branching fraction of the τ − → μ − μ + μ − decay, which is the most stringent bound to date.

Abstract: Abstract With the approval of the first two substances for the treatment of geographic atrophy (GA) secondary to age-related macular degeneration (AMD), a standardized monitoring of patients treated with complement inhibitors in clinical practice is needed. Optical coherence tomography (OCT) provides high-resolution access to the retinal pigment epithelium (RPE) and neurosensory layers, such as the ellipsoid zone (EZ), which further enhances the understanding of disease progression and therapeutic effects in GA compared to conventional fundus autofluorescence (FAF). In addition, artificial intelligence-based methodology allows the identification and quantification of GA-related pathology on OCT in an objective and standardized manner. The purpose of this study was to comprehensively evaluate automated OCT monitoring for GA compared to reading center-based manual FAF measurements in the largest successful phase 3 clinical trial data of complement inhibitor therapy to date. Automated OCT analysis of RPE loss showed a high and consistent correlation to manual GA measurements on conventional FAF. EZ loss on OCT was generally larger than areas of RPE loss, supporting the hypothesis that EZ loss exceeds underlying RPE loss as a fundamental pathophysiology in GA progression. Automated OCT analysis is well suited to monitor disease progression in GA patients treated in clinical practice and clinical trials.

Abstract: Abstract Processes ranging from fracture of crystals to peeling of tape have been known for many decades to emit light through a mechanism believed to be associated with electrical charging of separating surfaces. This topic, broadly termed fractoluminescence, has been proposed to be involved in several remarkable phenomena, including medical diagnostics and the generation of X-rays in the lab and earthquake lightning in nature. Here we add the paradoxical finding that two separating surfaces produce entirely different charge patterns, despite originating from the same interface. Further, we report the discovery of a rich variety of new and unexplained patterns, and we examine the hypothesis that the patterns are produced by migration of either polar or non-polar discharge ions onto contact-charged surfaces. This hypothesis may first explain prior findings that charge patterns can extend far beyond points of contact, and second suggests that the ultimate charge imparted on surfaces depends both on well-characterized mechanisms of surface potential and on highly variable discharge ions in the surrounding environment.

Abstract: Abstract Background Large language models (LLMs) such as ChatGPT-4 and Google Gemini show potential for patient health education, but concerns about their accuracy require careful evaluation. This study evaluates the readability and accuracy of ChatGPT-4 and Google Gemini in answering questions about retinal detachment. Methods Comparative study analyzing responses from ChatGPT-4 and Google Gemini to 13 retinal detachment questions, categorized by difficulty levels (D1, D2, D3). Masked responses were reviewed by ten vitreoretinal specialists and rated on correctness, errors, thematic accuracy, coherence, and overall quality grading. Analysis included Flesch Readability Ease Score, word and sentence counts. Results Both Artificial Intelligence tools required college-level understanding for all difficulty levels. Google Gemini was easier to understand (p = 0.03), while ChatGPT-4 provided more correct answers for the more difficult questions (p = 0.0005) with fewer serious errors. ChatGPT-4 scored highest on most challenging questions, showing superior thematic accuracy (p = 0.003). ChatGPT-4 outperformed Google Gemini in 8 of 13 questions, with higher overall quality grades in the easiest (p = 0.03) and hardest levels (p = 0.0002), showing a lower grade as question difficulty increased. Conclusions ChatGPT-4 and Google Gemini effectively address queries about retinal detachment, offering mostly accurate answers with few critical errors, though patients require higher education for comprehension. The implementation of AI tools may contribute to improving medical care by providing accurate and relevant healthcare information quickly.

195. Conflicting Problem Representations and Creativity: Effects on Problem Construction

Abstract: Contradictions and competing demands are common in everyday problems. To address these problems, individuals need to both recognize and integrate these contradictions during problem-solving to find creative solutions. We propose that recognition and integration take place in the problem construction phase of the creative problem-solving process. During problem construction, individuals work to develop their own understanding of the problem at hand. To do so, they sort through and combine problem representations activated through cues in the problem. We argue that the integration of conflicting and paradoxical elements in their understanding of the problem during problem construction is related to the originality and quality of proposed solutions beyond their initial ability to identify these elements. In a laboratory study, participants solved two different problems. We found that individuals identified more conflicting elements in the problem construction phase, but retained fewer of them in the problem-solving phase. Larger conflict between elements retained for problem solving resulted in more original and higher quality solutions to the problems.

Abstract: This study explores how preschoolers develop understandings of the symbolic nature of print in the context of their own writing. Using qualitative methods and a cross-sectional design, this study documents the learning trajectory that begins with children's earliest experiences linking speech and print in writing events and continues as they learn that English print is glottographic and alphabetic. Children's changing approaches to speech-print linking provide evidence of their developing understanding of how print functions as a representational system. Participants were 134 English-speaking 2- to 5-year-olds attending childcare classrooms where preschoolers' writing was frequent and valued. Children completed an open-ended writing task where they wrote a photo caption and read it to an adult. Open and axial coding identified six core approaches to speech-print linking during writing: marks not read, conversational speech without pointing, conversational speech with pointing to the graphic array, segmented speech with pointing to the graphic array, segmented speech matched to specific marks, and phoneme-grapheme matching. A growth curve model provided statistical support for this ordering of the core approaches. Findings show that early writing experiences can be an important context for building foundational literacy skills such as the alphabetic principle. Adult practices that physically materialize speech-print relations in writing may be especially supportive of this learning. We conclude that young children should be offered frequent opportunities to compose their own texts, and to interact with adults around their writing.

197. Examination of Grading Patterns during One Institution's Transition to a Responsibility Center Management Budget Model

Abstract: This study examines grading patterns during one intuition's transition to a responsibility center management (RCM) budget model. RCM is intended to focus an institution on resource growth and cost control, and the model incentivizes and rewards these behaviors. The adoption of RCM is becoming more widespread across the United States, especially as institutions are experiencing declining enrollment and shrinking state support (Barr & McClellan, 2018; State Higher Education Executive Officers Association [SHEEO], 2021; The Pew Charitable Trusts, 2019). This has led to budget management being cited as a top priority for institutional leaders (Bass et al., 2021; Rylee, 2011). This decline in enrollment and shrinking state support makes institutions more reliant on existing and prospective students as their primary source of revenue. Through the lens of Resource Dependency Theory (RDT) it is reasonable to assume that institutions may engage in behaviors and practices to maximize their relationship with students. Moreover, as students continue to see themselves as consumers who deserve high grades and a degree, institutions have changed their grading practices to attract and retain these students, and research has shown that students take advantage of these practices to protect their GPA (Bromley et al., 1978; Goldman, 1985). Additionally, research has shown that students, in part, make course choices based on difficulty and expected grade (e.g., Barr et al., 2009; Butcher et al., 2014; Fournier & Sass, 2000; Sabot & Wakemin-Linn, 1991). Therefore, it is reasonable to hypothesize that institutions may transition to RCM to focus on resource growth, and given their reliance on student enrollment, may engage in practices and behaviors to maximize the relationship between the institution and the student. These actions may have unintended consequences, such as grade inflation. My study begins to examine if grade inflation is an unintended consequence of a transition to RCM by exploring what happened to grades during one institution's transition to RCM. The results of my study show that at Public Research University (PRU), a pseudonym, grades continued to increase during and after the implementation of RCM, with a statistically significant increase occurring in the year immediately following PRU's "hold harmless" year. My results also show that grades continued to rise during and after PRU's transition to RCM across all departments when categorized based on historical GPAs, selectivity levels, and enrollment levels. This study is intended to raise awareness of the far-reaching effects that central university decisions, such as budget model choice, may have across the entire institution -- both intended and unintended. 198. The VLT MUSE NFM view of outflows and externally photoevaporating discs near the Orion Bar

Abstract: We present VLT/MUSE Narrow Field Mode (NFM) observations of a pair of disc-bearing young stellar objects towards the Orion Bar: 203-504 and 203-506. Both of these discs are subject to external photoevaporation, where winds are launched from their outer regions due to environmental irradiation. Intriguingly, despite having projected separation from one another of only 1.65{\arcsec} (660au at 400pc), 203-504 has a classic teardrop shaped ``proplyd'' morphology pointing towards $\theta^2$Ori A (indicating irradiation by the EUV of that star, rather than $\theta^1$ Ori C) but 203-506 has no ionisation front, indicating it is not irradiated by stellar EUV at all. However, 203-506 does show [CI] 8727{\AA} and [OI] 6300{\AA} in emission, indicating irradiation by stellar FUV. This explicitly demonstrates the importance of FUV irradiation in driving mass loss from discs. We conclude that shielding of 203-506 from EUV is most likely due to its position on the observers side of an ionized layer lying in the foreground of the Huygens Region. We demonstrate that the outflow HH 519, previously thought to be emanating from 203-504 is actually an irradiated cloud edge and identify a new compact outflow from that object approximately along our line of sight with a velocity $\sim130$\,km\,s$^{-1}$., Comment: 14 pages, 8 figures. Abstract: We report results from a study of $B^\pm \rightarrow DK^\pm$ decays followed by $D$ decaying to $CP$~eigenstates, where $D$ indicates a $D^0$ or $\bar{D}^{0}$ meson. These decays are sensitive to the Cabibbo-Kobayashi-Maskawa unitarity-triangle angle $\phi_{3}$. The results are based on a combined analysis of the final data set of $772 \times 10^6~B\bar{B}$ pairs collected by the Belle experiment and a data set of $198 \times 10^6~B\bar{B}$ pairs collected by the Belle~II experiment, both in electron-positron collisions at the $\Upsilon(4S)$ resonance. We measure the $CP$ asymmetries to be $\mathcal{ A}_{CP +} =~(+12.5 \pm 5.8 \pm 1.4)\% $ and $\mathcal{ A}_{CP -} =~(-16.7 \pm 5.7 \pm 0.6)\%$, and the ratios of branching fractions to be $\mathcal{ R}_{CP+}=~1.164 \pm 0.081 \pm 0.036 $ and $\mathcal{ R}_{CP-} =~1.151 \pm 0.074 \pm 0.019$. The first contribution to the uncertainties is statistical, and the second is systematic. The asymmetries $\mathcal{A}_{CP +}$ and $\mathcal{A}_{CP -}$ have similar magnitudes and opposite signs; their difference corresponds to 3.5~standard deviations. From these values we calculate 68.3\% confidence intervals of ($8.5^{\circ}<\phi_{3}<16.5^{\circ}$) or ($84.5^{\circ}<\phi_{3}<95.5^{\circ}$) or ($163.3^{\circ}<\phi_{3}<171.5^{\circ}$) and $0.321

Abstract: At the FRS Ion Catcher (FRS-IC), projectile and fission fragments are produced at relativistic energies, separated in-flight, energy-bunched, slowed down, and thermalized in the ultra-pure helium gas-filled cryogenic stopping cell (CSC). Thermalized nuclei are extracted from the CSC using a combination of DC and RF electric fields and gas flow. This CSC also serves as the prototype CSC for the Super-FRS, where exotic nuclei will be produced at unprecedented rates making it possible to go towards the extremes of the nuclear chart. Therefore, it is essential to efficiently extract thermalized exotic nuclei from the CSC under high beam rate conditions, in order to use the rare exotic nuclei which come as cocktail beams. The extraction efficiency dependence on the intensity of the impinging beam into the CSC was studied with a primary beam of 238U and its fragments. Tests were done with two different versions of the DC electrode structure inside the cryogenic chamber, the standard 1 m long and a short 0.5 m long DC electrode. In contrast to the rate capability of 10^4 ions/s with the long DC electrode, results show no extraction efficiency loss up to the rate of 2x10^5 ions/s with the new short DC electrode. This order of magnitude increase of the rate capability paves the way for new experiments at the FRS-IC, including exotic nuclei studies with in-cell multi-nucleon transfer reactions. The results further validate the design concept of the CSC for the Super-FRS, which was developed to effectively manage beams of even higher intensities.
