The giant, single-celled organism Stentor coeruleus has a long history as a model system for studying pattern formation and regeneration in single cells. Stentor [1, 2] is a heterotrichous ciliate distantly related to familiar ciliate models, such as Tetrahymena or Paramecium. The primary distinguishing feature of Stentor is its incredible size: a single cell is 1 mm long. Early developmental biologists, including T.H. Morgan [3], were attracted to the system because of its regenerative abilities-if large portions of a cell are surgically removed, the remnant reorganizes into a normal-looking but smaller cell with correct proportionality [2, 3]. These biologists were also drawn to Stentor because it exhibits a rich repertoire of behaviors, including light avoidance, mechanosensitive contraction, food selection, and even the ability to habituate to touch, a simple form of learning usually seen in higher organisms [4]. While early microsurgical approaches demonstrated a startling array of regenerative and morphogenetic processes in this single-celled organism, Stentor was never developed as a molecular model system. We report the sequencing of the Stentor coeruleus macronuclear genome and reveal key features of the genome. First, we find that Stentor uses the standard genetic code, suggesting that ciliate-specific genetic codes arose after Stentor branched from other ciliates. We also discover that ploidy correlates with Stentor's cell size. Finally, in the Stentor genome, we discover the smallest spliceosomal introns reported for any species. The sequenced genome opens the door to molecular analysis of single-cell regeneration in Stentor.

- Abstract
Urbane Mobilität insbesondere der innerstädtischen Personentransport wird auch zukünftig ein besonderes Augenmerk unter dem Eindruck nachhaltiger Nutzung von Energiesystemen haben. Hierbei besitzt der öffentliche Personennahverkehr(ÖPNV) ein großes Potential zur großskaligen Minderung des Kohlendioxid ausstoßes. Ein sehr wichtiges Konzept hierbei ist der Oberleitungsbus der ganz analog zu einer Straßenbahn über eine Oberleitung fahrbahnbegleitend mit Strom versorgt wird. Da der Bus naturgemäß nicht schienengebunden fährt und somit ein gewisses Maß an Positionsungenauigkeit besitzt, ist das Eindrahten an die Oberleitung allerdings erschwert. In dieser Arbeit präsentieren wir einen Regelungsansatz basierend auf einer neuartigen Sensorkonfiguration, der das automatische Eindrahten eines Stromabnehmers auf einem Bus ermöglichen kann.

Die Industrie 5.0 fordert neue Lernansätze und zeitgleich auch passende Lernumgebungen. Parallel müssen diese neben den didaktischen Herausforderungen auch den Transfer- und Übertragungsgedanken auf die industriellen Anwendungen gerecht werden. Durch die täglich steigende Anzahl vielfältiger KI-Tools insbesondere textgenerierenden Tools, braucht es Systeme mit einem breiten Anwendungsbereich. Im Rahmen des vorliegenden Beitrags geben die Autoren einen Einblick in die Wildauer Smart Production, welche den transdisziplinären Gedanken von Lern- und Transferumgebungen Rechnung trägt, Möglichkeiten der Gestaltung komplexer Produktionssysteme widerspiegelt, die Integration menschzentrierter Ansätze ermöglicht und als Forschungsumgebung eingesetzt wird.

- Abstract
The modern marine megafauna is known to play important ecological roles and includes many charismatic species that have drawn the attention of both the scientific community and the public. However, the extinct marine megafauna has never been assessed as a whole, nor has it been defined in deep time. Here, we review the literature to define and list the species that constitute the extinct marine megafauna, and to explore biological and ecological patterns throughout the Phanerozoic. We propose a size cut-off of 1 m of length to define the extinct marine megafauna. Based on this definition, we list 706 taxa belonging to eight main groups. We found that the extinct marine megafauna was conspicuous over the Phanerozoic and ubiquitous across all geological eras and periods, with the Mesozoic, especially the Cretaceous, having the greatest number of taxa. Marine reptiles include the largest size recorded (21 m; Shonisaurus sikanniensis) and contain the highest number of extinct marine megafaunal taxa. This contrasts with today’s assemblage, where marine animals achieve sizes of >30 m. The extinct marine megafaunal taxa were found to be well-represented in the Paleobiology Database, but not better sampled than their smaller counterparts. Among the extinct marine megafauna, there appears to be an overall increase in body size through time. Most extinct megafaunal taxa were inferred to be macropredators preferentially living in coastal environments. Across the Phanerozoic, megafaunal species had similar extinction risks as smaller species, in stark contrast to modern oceans where the large species are most affected by human perturbations. Our work represents a first step towards a better understanding of the marine megafauna that lived in the geological past. However, more work is required to expand our list of taxa and their traits so that we can obtain a more complete picture of their ecology and evolution.

Hybrid project management is an approach that combines traditional and agile project management techniques. The goal is to benefit from the strengths of each approach, and, at the same time avoid the weaknesses. However, due to the variety of hybrid methodologies that have been presented in the meantime, it is not easy to understand the differences or similarities of the methodologies, as well as, the advantages or disadvantages of the hybrid approach in general. Additionally, there is only fragmented knowledge about prerequisites and success factors for successfully implementing hybrid project management in organizations. Hence, the aim of this study is to provide a structured overview of the current state of research regarding the topic. To address this aim, we have conducted a systematic literature review focusing on a set of specific research questions. As a result, four different hybrid methodologies are discussed, as well as, the definition, benefits, challenges, suitability and prerequisites of hybrid project management. Our study contributes to knowledge by synthesizing and structuring prior work in this growing area of research, which serves as a basis for purposeful and targeted research in the future.

- Abstract
Addressing the need for suicide prevention on campus, I CARE training, developed and facilitated by counseling center clinicians, trains students, staff, and faculty to provide support to students experiencing distress or mental health problems. We assessed its impact using a mixed-methods approach. Quantitative analyses demonstrated significant increases in knowledge of intervention skills and readiness to intervene from pretraining to posttraining. Knowledge and readiness remained significantly higher than preworkshop for the entire follow-up evaluation period, extending 15 months posttraining. Qualitative analyses revealed the value of experiential activities and emotional processing in increasing participants' comfort and preparedness to intervene in challenging situations.

Introduction: Brazil is a developing and an Upper Middle Income, categorized by the World Bank. Therefore, it is a country that needs a special vision for children with oncological diseases who require Pediatric Palliative Care. This study aimed to understand the specificities of services that provide oncology services in comparison to those that do not provide oncological care. Methods: This is a descriptive, cross-sectional, and online survey study. A questionnaire was created by a multidisciplinary group of leaders from the Brazilian Pediatric Palliative Care Network and then the survey was distributed using a snowball strategy. Results: Of the 90 services that answered the questionnaire, 40 (44.4%) attended oncologic patients. The Southeast represented most of the services (57.57%), followed by the Northeast, with 18.89% (17 services), the South with 12.22% (11 services), and the Center West with 8.89% (8 services). No differences were observed in access to opioid prescriptions between the services. It was observed that those services that attended oncologic patients had a tendency to dedicate more time to Pediatric Palliative Care. Discussion: The distribution of services that cover oncology and those that do not, are similar in the different regions of Brazil. In Brazil, there are difficulties in accessing opioids in pediatrics: access to opioid prescriptions without differences revealed that even pediatric oncologists might have difficulty with this prescription, and this should improve. Unfortunately, many people today see the regulation of lies, disinformation, hate speech, and fake news as an infringement of free speech, at least when such speech is ‘political,’ despite the damage that such speech can do. But this very protective attitude toward speech rests on a mistaken understanding of the role of free speech in a liberal society. The right to free speech is based on the liberal value of freedom, and as such can be no broader than freedom itself. And freedom has always been subject to reasonable limits in a liberal society. Indeed, while the principles of toleration and neutrality are often cited as supporting a broad interpretation of the right to free speech, they also tell us that certain limits apply to that right. We need not tolerate speech that encourages intolerance, and while government should be neutral between reasonable conceptions of the good, it need not be neutral between reasonable and unreasonable conceptions. These ideas form the framework of liberal society, and as I shall show, also provide a guide for understanding what kind of speech is protected in a liberal society and what it is not. Abstract Background Prompt diagnosis of juvenile idiopathic arthritis (JIA) is important to avoid long term complications. Elevated serum 14–3-3η levels improve the diagnostic sensitivity of rheumatoid factor (RF) and cyclic citrullinated peptide (CCP) antibody in adult rheumatoid arthritis (RA), and have been associated with more severe phenotype. We investigated the prevalence and clinical significance of serum 14–3-3η in different types of JIA. Methods JIA patients (n = 151) followed by the Pediatric Rheumatology Core at Children’s Hospital of Los Angeles were categorized into 5 groups: polyarticular JIA RF+ (PJIA RF+; n = 39), PJIA RF- (n = 39), psoriatic arthritis (PsA; n = 19), enthesitis-related arthritis (ERA; n = 18), and oligoarticular JIA (OJIA [control group]; n = 36). RF, CCP antibody, and 14–3-3η were measured for all patients. 14–3-3η serum levels > 0.2 ng/mL were considered positive. Disease activity was assessed by the Juvenile Arthritis Disease Activity Score-71 (JADAS-71). Results Elevated 14–3-3η levels were detected in 34/151 (23%) patients, and across all groups tested. Most patients with 14–3-3η had titers ≥4 times above the cutoff value. The majority (22, 65%) of 14–3-3η-positive patients were also positive for RF or CCP antibodies, 16 (47%) were positive for all 3, and 12 (35%) were single-positive for 14–3-3η. The highest prevalence of 14–3-3η was in PJIA RF+ patients (49%), followed by OJIA (22%). Positivity for 14–3-3η was not significantly associated with disease activity or age at diagnosis. Conclusion Serum 14–3-3η can be detected in all forms of JIA tested but appears to be most common in PJIA RF+. 14–3-3η does not appear to correlate with disease activity in JIA.

Abstract Importance Active pediatric COVID-19 pneumonia and MIS-C are two disease processes requiring rapid diagnosis and different treatment protocols. Objective To distinguish active pediatric COVID-19 pneumonia and MIS-C using presenting signs and symptoms, patient characteristics, and laboratory values. Design Patients diagnosed and hospitalized with active COVID-19 pneumonia or MIS-C at Children’s of Alabama Hospital in Birmingham, AL from April 1 through September 1, 2020 were identified retrospectively. Active COVID-19 and MIS-C cases were defined using diagnostic codes and verified for accuracy using current US Centers for Disease Control case definitions. All clinical notes were reviewed for documentation of COVID-19 pneumonia or MIS-C, and clinical notes and electronic medical records were reviewed for patient demographics, presenting signs and symptoms, prior exposure to or testing for the SARS-CoV-2 virus, laboratory data, imaging, treatment modalities and response to treatment. Findings 111 patients were identified, with 74 classified as mild COVID-19, 8 patients as moderate COVID-19, 8 patients as severe COVID-19, 10 as mild MIS-C and 11 as severe MIS-C. All groups had a male predominance, with Black and Hispanic patients overrepresented as compared to the demographics of Alabama. Most MIS-C patients were healthy at baseline, with most COVID-19 patients having at least one underlying illness. Fever, rash, conjunctivitis, and gastrointestinal symptoms were predominant in the MIS-C population whereas COVID-19 patients presented with predominantly respiratory symptoms. The two groups were similar in duration of symptomatic prodrome and exposure history to the SARS-CoV-2 virus, but MIS-C patients had a longer duration between presentation and exposure history. COVID-19 patients were more likely to have a positive SAR-CoV-2 PCR and to require respiratory support on admission. MIS-C patients had lower sodium levels, higher levels of C-reactive protein, erythrocyte sedimentation rate, d-dimer and procalcitonin. COVID-19 patients had higher lactate dehydrogenase levels on admission. MIS-C patients had coronary artery changes on echocardiography more often than COVID-19 patients. Conclusions and relevance This study is one of the first to directly compare COVID-19 and MIS-C in the pediatric population. The significant differences found between symptoms at presentation, demographics, and laboratory findings will aide health-care providers in distinguishing the two disease entities.

ABSTRACT Objective: To compare the cervical sagittal balance in groups of patients submitted to lateral radiographs of the cervical segment, with the upper limbs in two different positions. Methods: This is a cross-sectional, quantitative, prospective, descriptive study using radiographic analysis of 100 adult individuals, aged between 20 and 70 years. Two lateral radiographs were taken of each patient, one image with the upper limbs crossed over the trunk (flexed elbows) and the other with the upper limbs elevated in front (extended elbows). All patients answered a questionnaire about age, sex, presence of pain in the cervical spine, as well as degree of pain based on the Likert scale. The radiographic parameters evaluated were the COBB angle (Cobb), thoracic inlet angle (TIA), T1 slope, neck tilt and distance from the center of gravity (COG) of the skull to C7 (COG-C7) with the elbows in extension (Ext) and flexion (Flex). A p-value of less than 0.05 was considered a statistically significant difference. Results: Sixty patients were female (60%) and forty were male (40%). The data analysis obtained showed statistical significance in Cobb Flex vs. Cobb Ext (p = 0.000), TIA Flex vs. TIA Ext (p = 0.000), T1 Flex vs. T1 Ext (p = 0.000), Neck tilt Flex vs. Neck tilt Ext (p = 0.000), and COG-C7 Flex vs. COG-C7 Ext (p = 0.000). Conclusion: There was statistical significance in relation to the positioning of the upper limbs in the lateral radiographs, yielding higher values in the angulations with the limbs raised towards the front (extended elbows). Syphilis findet in Erasmus’ von Rotterdam Colloquia familiaria mehrfach Erwähnung. Insbesondere ein 1529 in die Sammlung aufgenommener Dialog mit dem Titel „Ἄγαμος γάμος sive Coniugum impar“ („Die ungleiche Ehe“) setzt sich mit dem Ende des 15. Jahrhunderts als neu wahrgenommenen Krankheit auseinander. In diesem Text werden Teile des semantischen Spektrums der Lepra durch Vergleich, Abgrenzung und Überbietung auf die Syphilis übertragen, was eine Lesbarkeit des kranken Körpers nicht nur als infektiös, sondern auch als sünden- und schuldbehaftet ermöglicht. In einer Lesart als Schlüsseltext wird der Dialog im Kontext einer schriftlich geführten Kontroverse zwischen Erasmus und Heinrich Eppendorf verortet, die auf ein früheres Zerwürfnis zwischen Erasmus und Ulrich von Hutten in den Jahren 1522/23 zurückgeht. Es wird herausgestellt, dass der syphilitische Bräutigam des „Coniugum impar“ auf Hutten verweist und mit ihm das Vorbild Eppendorfs diskreditiert. In allegorischer Lesart wird damit auch die Verdorbenheit des Standes der Reichsritter insgesamt herausgestellt.

The fast development of the Internet of Things (IoT) changes our life in many areas, especially in the health domain. For example, remote disease diagnosis can be achieved more efficiently with advanced IoT-technologies which not only include hardware but also smart IoT data processing and learning algorithms, e.g. image-based disease classification. In this paper, we work in a specific area of skin condition classification. This research work aims to provide an implementable solution for IoT-led remote skin disease diagnosis applications. The research output can be concluded into three folders. The first folder is about dynamic AI model configuration supported IoT-Fog-Cloud remote diagnosis architecture with hardware examples. The second folder is the evaluation survey regarding the performances of machine learning models for skin disease detection. The evaluation contains a variety of data processing methods and their aggregations. The evaluation takes account of both training-testing and cross-testing validations on all seven conditions and individual condition. In addition, the HAM10000 dataset is picked for the evaluation process according to the suitability comparisons to other relevant datasets. In the evaluation, we discuss the earlier work of ANN, SVM and KNN models, but the evaluation process mainly focuses on six widely applied Deep Learning models of VGG16, Inception, Xception, MobileNet, ResNet50 and DenseNet161. The result shows that each of the top four models for the major seven skin conditions has better performance for the specific condition than others. Based on the evaluation discovery, the last folder proposes a novel classification approach of the Targeted Ensemble Machine Classify Model (TEMCM) to enable dynamically combining a suitable model in a two-phase detection process. The final evaluation result shows the proposed model can archive better performance.

The cloud ecosystem provides transformative advantages that allow elastically offering on-demand services. However, it is not always possible to provide adequate services to all customers and thus to fulfill service level agreements (SLA). To enable compliance with these agreements, service providers leave the customer responsible for determining the service settings and expect that the client knows what to do. Some studies address SLA compliance, but the existing works do not adequately address the problem of resource allocation according to clients' needs since they consider a limited set of objectives to be analyzed and fulfilled. In previous work, we have already addressed the problem considering a single-objective approach. In that work, we identified that the problem has a multi-objective characteristic since several attributes simultaneously influence the SLA agreement, which can lead to conflicts. This paper proposes a multi-objective combinatorial optimization approach for computational resources provisioning, seeking to optimize the efficient use of the infrastructure and provide the client with greater flexibility in contract closure.
