The analysis of isotope ratios of lead in the mushrooms and soil, where they were grown, assisted with a principal component analysis, offered a new perspective for understanding possible chemical environment in a real setup of those compartments. The content of lead and its isotope compositions were determined in soil samples and mushroom Macrolepiota procera from unpolluted area of Mountain Goč, Serbia. Sequential extraction procedure based on the Commission of the European Community Bureau of Reference (BCR) was applied on soil samples in order to determine the distribution of lead in the labile and un-labile fractions of the soil. Caps and stipes of mushrooms were subjected to microwave acid-assisted digestion prior to measurements by inductively coupled plasma quadrupole mass spectrometer for determination of lead content and lead isotope ratios. Information about the chemical fractionation of Pb in soil, Pb isotopic data from soil fractions and fruiting bodies allowed a more detailed insight on the uptake mechanisms. Lead was predominantly associated with reducible fraction (~ 60%). Only its small portion (∼ 1%) was present in the exchangeable and acid-extractable fractions suggesting the low mobility of Pb. Lead isotope analysis revealed the presence of anthropogenic lead in the surface soil. Significant lower 206 Pb/ 207 Pb compared with other fractions was found in exchangeable and acid-soluble fraction (1.331 ± 0.010), which corresponds to the isotope ratio of European gasoline. The highest 206 Pb/ 207 Pb ratio was observed in reducible fraction (1.162 ± 0.007), while in oxidizable and residual fraction, those values were similar (1.159 ± 0.006 and 1.159 ± 0.004, respectively). Distinction of exchangeable and acid-extractable fractions from others was also confirmed, for the first time, by principal component analysis. The analysis of four isotope ratios ( 206 Pb/ 207 Pb, 208 Pb/ 206 Pb, 206 Pb/ 204 Pb, and 207 Pb/ 204 Pb) indicated that the analyzed M. procera accumulates lead from the first two fractions of topsoil layers., (© 2020. Recent studies show that lanthanides (Ln) are becoming emerging pollutants due to their wide application in new technologies, but their environmental fate, transport, and possible accumulation are still relatively unknown. This study aims to determine major and trace elements including Ln in the Danube River sediment which either belong or close to the Iron Gate Reservoir. The Iron Gate Reservoir is characterized by accumulation of sediments as an effect of building hydropower dam Iron Gate I. The surface sediments were collected on the Danube River-1141 to 864 km and three tributaries along this waterway. Two samples of deep sediments were used for comparison. The results indicate the significant upward enrichment of Zn, Sb, Cr, Nd, and Dy in sediments belongs to the Iron Gate Reservoir. The sample 4-Smed is labelled as a hot spot of contamination with Zn, Cr, As, Sb, Nd, and Dy. Also, a trend of increasing concentration in the time period from 1995 to 2016 was found for elements Zn, Cr, and Ni in sediment samples in the Iron Gate Reservoir. Chemometric analysis shows the grouping of sample sites into clusters characterized by the following properties: (i) increased concentration of all measured elements (samples within the Iron Gate Reservoir); (ii) increased Cu concentration (11-Pek); and (iii) lower concentrations of the measured elements (deep sediments). Preparation of functional products as well as natural-based products requires non-toxic but effective extraction techniques. In this study, subcritical water extraction was used for the extraction of different aronia parts in order to explore their potential. Stems, leaves and berries of Aronia melanocarpa were extracted under the following conditions: temperature 130 °C; pressure 35 bar; time 20 min. The total phenols and flavonoid contents of the produced extracts were evaluated by conventional spectrophotometric methods. Additionally, the main phenolic compounds were also identified and quantified by high performance liquid chromatography with diode array detection (HPLC-DAD). The biological potential of the extracts was evaluated by determining their antioxidant (DPPH, ABTS and lipid peroxidation assays), antimicrobial, enzyme inhibitory (cholinesterase and elastase) and cytotoxic effects (HeLa, A-549, LS-174T, MRC-5 cell lines). The results indicate that leaves and berries extracts exhibited stronger antioxidant action when compared with stems. The strongest cholinesterase and elastase inhibitory activity was also found in berries extract. Similarly, the extracts obtained from leaves and berries showed considerable cytotoxic effects against tested cell lines. A moderate antimicrobial effects was observed too. To close the gap in our knowledge of sex influence on age-related changes in inflammation-oxidation state in spinal cord (SC) relevant to inflammation/oxidative-stress associated neuropathologies, 2-3 month-old (young) and 18-20 month-old (old) rats, exhibiting increased level of IL-6, a commonly used marker of inflamm-aging, were examined for inflammatory/redox status, and the underlying regulatory networks' molecules expression. With age, rat SC microglia became sensitized ("primed"), while SC tissue shifted towards mild inflammatory state, with increased levels of proinflammatory IL-1β (key marker of microglial systemic inflammation-induced neurotoxicity), which was more prominent in males. This, most likely, reflected age- and sex-related impairment in the expression of CX3CR1, the receptor for fractalkine (CX3CL1), the soluble factor which regulates microglial activation and diminishes production of IL-1β (central for fractalkine neuroprotection). Considering that (i) age-related changes in SC IL-1β expression were not followed by complementary changes in SC IL-6 expression, and (ii) the reversal in the direction of the sex bias in circulating IL-6 level and SC IL-1β expression, it seems obvious that there are tissue-specific differences in the proinflammatory cytokine profile. Additionally, old male rat SC exhibited greater oxidative damage than female, reflecting, most likely, their lower capacity to maintain the pro-oxidant-antioxidant balance. In conclusion, these findings, apart from highlighting the significance of sex for age-associated changes in SC inflammation-oxidation, may be relevant for understating sex differences in human inflammation/oxidative-stress related SC diseases, and consequently, for optimizing their prevention/therapy.
