Vascular mimicry has been thoroughly investigated in tumor angiogenesis. In this study, we demonstrate for the first time that a process closely resembling tumor vascular mimicry is present during physiological blood vessel formation in tissue regeneration using the zebrafish fin regeneration assay. At the fin-regenerating front, vasculature is formed by mosaic blood vessels with endothelial-like cells possessing the morphological phenotype of a macrophage and co-expressing both endothelial and macrophage markers within single cells. Our data demonstrate that the vascular segments of the regenerating tissue expand, in part, through the transformation of adjacent macrophages into endothelial-like cells, forming functional, perfused channels and contributing to the de novo formation of microvasculature. Inhibiting the formation of tubular vascular-like structures by CVM-1118 prevents vascular mimicry and network formation resulting in a 70% shorter regeneration area with 60% reduced vessel growth and a complete absence of any signs of regeneration in half of the fin area. Additionally, this is associated with a significant reduction in macrophages. Furthermore, depleting macrophages using macrophage inhibitor PLX-3397, results in impaired tissue regeneration and blood vessel formation, namely a reduction in the regeneration area and vessel network by 75% in comparison to controls. m-Cresol and p-cresol are widely used in medicine and pesticides as important chemical intermediates. They are generally produced as a mixture in industry and are difficult to separate due to the similarities in both chemical structures and physical properties. The adsorption behaviours of m-cresol and p-cresol on zeolites (NaZSM-5 and HZSM-5) with different Si/Al ratios have been compared by static experiments. Selectivity on NaZSM-5(Si/Al = 80) could be greater than 6.0. Adsorption kinetics and isotherms were investigated in detail. The kinetic data was correlated by PFO, PSO, and ID models, the NRMSE of which were 14.03%, 9.41%, and 21.11%, respectively. In the meanwhile, according to the NRMSE of Langmuir (6.01%), Freundlich (57.80%), D-R (1.1%), and Temkin (0.56%) isotherms, adsorption on NaZSM-5(Si/Al = 80) was mainly a monolayer and chemical adsorption process. It was endothermic for m-cresol and exothermic for p-cresol. The Gibbs free energy, entropy, and enthalpy were calculated accordingly. The adsorption of cresol isomers on NaZSM-5(Si/Al = 80) were both spontaneous, and it was exothermic (ΔH = −37.11kJ/mol) for p-cresol while endothermic (ΔH = 52.30kJ/mol) for m-cresol. Additionally, ΔS were respectively −0.05 and 0.20 kJ·mol−1·K−1for p-cresol and m-cresol, which were both close to zero. The adsorption was mainly driven by enthalpy. The result of breakthrough further demonstrated m-cresol and p-cresol could be separated effectively by NaZSM-5(Si/Al = 80). Additionally, the selectivity increased from 7.53 to 14.72 after four cycles regeneration with 9.95% and 53.96% decreases in the adsorption amounts for m-cresol and p-cresol, respectively. Icing interventions on the injured skeletal muscle affect the macrophage-related regenerative events and muscle repair. However, despite its importance for the practice in sport medicine, the influence of different icing protocols on muscle regeneration remains unclear. Here, using a rodent model of mild muscle injury with necrosis in a small fraction of myofibers, the injured animals were allocated to four groups: non-icing control (Con) and a single treatment (Ice-1), three treatments (Ice-3), or nine treatments (Ice-9) with a 30-min icing each time within two days following injury. Muscle regeneration was compared between the groups on post-injury days 1, 3, 5, and 7. The results showed that compared with the Con group, muscle regeneration was faster in the Ice-9 group (but not in the Ice-1 and Ice-3 groups), as indicated by more rapid accumulation of satellite cells within the regenerating area and enlarged size of regenerating myofibers (p <0.05, respectively). There was also less macrophage accumulation (p <0.05) and a trend toward early removal of necrotic myofibers in the damaged/regenerating area in the Ice-9 group (p =0.0535). Divalent cations have captured the interest of researchers in biomedical and dental fields due to their beneficial effects on bone formation. These metallic elements are similar to trace elements found in human bone. Strontium is a divalent cation commonly found in various biomaterials. Since strontium has a radius similar to calcium, it has been used to replace calcium in many calcium-containing biomaterials. Strontium has the ability to inhibit bone resorption and increase bone deposition, making it useful in the treatment of osteoporosis. Strontium has also been used as a radiopacifier in dentistry and has been incorporated into a variety of dental materials to improve their radiopacity. Furthermore, strontium has been shown to improve the antimicrobial and mechanical properties of dental materials, promote enamel remineralization, alleviate dentin hypersensitivity, and enhance dentin regeneration. In recent decades, ungulates have expanded in number and range in Europe. This review aims to analyze the impact of ungulate browsing in different forest ecosystems and identify the main driving factors and trends. In total, 155 studies were analyzed in preparing this review, across 19 European countries. In Europe, the main browsers are represented by roe deer (Capreolus capreolus L.), red deer (Cervus elaphus L.), moose (Alces alces L.), chamois (Rupicapra rupicapra L.), and fallow deer (Dama dama L.). Regarding browsing severity, they frequently exceeded 50%, meaning that over half of the saplings were browsed. Ungulate density was the main driving factor of browsing severity, with areas exhibiting high browsing pressure often having more than ten individuals per square kilometer. The type of silvicultural system used played a vital role in the severity of browsing, and trends in foraging for preferred tree species were identified. Fencing was the most common non-harmful protection method used, while hunting management was the most efficient method for controlling deer numbers and browsing intensity. Large carnivores were missing in most study areas, but in the areas where they were present, they played a significant role in creating a chain reaction of ecological impacts. Considering the significant impact of ungulate browsing on forest ecosystems, there is a pressing need for more research to comprehend and effectively mitigate the effects of deer presence comprehensively. The term Delayed Onset Muscle Soreness (DOMS) refers to a complex injury entity which is based on ultrastructural injury patterns. The injury is usually harmless and heals within a few days without sequelae. However, at the time of the manifestation there are (sometimes considerable) performance limitations and the risk of possible secondary injuries is increased. Thus, prevention and treatment of DOMS, especially in high-performance sports, are of particular value. Insights into muscle biomechanics are necessary to understand the background and mechanisms of DOMS. The definition and classification of overload-related muscle injuries has not yet been uniformly determined. Fluid transitions between physiological, metabolic and neuromuscular fatigue as well as the actual occurrence of injuries with clinical symptoms make clear delineation difficult. The “Munich Classification” terms Delayed Onset Muscle Soreness (DOMS) as Type 1b muscle injury, “delayed onset muscle pain”. The central target of prevention of DOMS is to prevent or alleviate the onset of the initial damage, known as “Exercise Induced Muscle Damage” (EIMD). In case of manifest DOMS, the intention is both to relieve the attendant symptoms and to promote rapid restoration of muscle function. In clinical practice as in scientific research, diverse procedures and interventions have proven valuable, whereby especially physical therapy procedures like cryotherapy, heat therapy or compression therapy show promising study results. The purpose of this study is to remove more Cd2+ ions from the aqueous solutions and increase biosorption capacity of Abies bornmuelleriana cones treated with 0.01 M NaOH. After biosorption, the capacities of normal Abies bornmuelleriana cones (N biosorbent) and modified Abies bornmuelleriana cones (M biosorbent) were compared. Additionally, the selectivity study was carried out in the presence of Hg2+ and Cd2+ ions. The removal mechanism of Cd2+ ions was explained using the zeta potentials, BET areas, total pore volumes, average particle diameters, FTIR, and SEM images-EDX analyses of N and M biosorbent after biosorption. The Langmuir isotherm for N and M biosorbents was selected as the ideal model since it had the greatest coefficient of determination (R2) (0.99, 0.98) values and the lowest mean absolute errors (MAEs) (0.0070, 0.0018) and root mean squared errors (RMSEs) (0.0011, 0.0043). For the removal of Cd2+ ions using N and M biosorbents, the pseudo-second-order (P-S–O) model presented the highest R2 (0.99, 0.99) values as well as the lowest MAE (0.015, 0.007) and RMSE (0.021, 0.01) values. Positive ΔH° for both biosorbents implies that the biosorption process is endothermic, whereas negative ΔG° shows that the biosorption process happens quickly at high temperatures. The augmentation in ΔS° with the temperature displayed the randomness of the biosorption. The Langmuir biosorbent capacities of N and M biosorbents were determined to be 18.4 and 22.1 mg g−1, respectively. When profit-driven renewal is difficult to implement, many historic districts in China become "frozen" under protection. In the recent social context of "building cities for the people", public value-oriented renewal of urban historic areas has become crucial in city transformation. This study investigates strategies for historic district regeneration in China by integrating spatial form, urban governance, and public value. We propose a novel framework to analyze the regeneration of historic districts, using Taiyuan's Bell Tower Street as a case study. The framework distinguishes between two dimensions: spatial and economic–social. In the spatial dimension, we focus on the transformation of spatial form to regenerate public value, emphasizing the attribute of "publicness" as central to urban regeneration efforts. This involves a tiered approach to excavating and upgrading historic districts at macro, meso, and micro levels. The economic–social dimension explores urban governance to enhance public value through collaboration between government, market, and societal actors. This study employs a mixed-methods approach, including document reviews, interviews, field observations, and statistical data analysis. The analysis highlights that a public value-oriented approach to heritage regeneration can balance historical preservation with contemporary urban needs, offering a sustainable model for other cities. Callus formation induced by auxin accumulation is considered the first step of in vitro plant regeneration. In Arabidopsis, degradation of the Aux/IAA protein, IAA14, in response to auxin signaling, which activates the AUXIN RESPONSE FACTOR 7 (ARF7) and ARF19 along with a series of downstream transcription factors, also plays a critical role in this process. However, the specific mechanism by which auxin regulates callus formation remains unclear. By screening mutant library in the solitary root 1 (iaa14/slr) Arabidopsis background we obtained the callus formation related 2 (cfr2) mutant. The cfr2 mutant exhibited a stronger capacity for callus formation, as well as lateral root and adventitious root regeneration from leaf explants than wild type (WT) seedlings, but did not recover gravitropism capability. The auxin signal in cfr2 was significantly enhanced, and the expression of some downstream transcription factors was increased. Map-based cloning, whole genome resequencing, and phenotypic complementation experiments showed that the phenotypes observed in the cfr2 mutant were caused by a point mutation in the IAA14 promoter region. This mutation, which is predicted to disrupt the binding of LBD16, LBD19, and LBD30 to the IAA14 promoter, changed the expression pattern of IAA14 in cfr2. Taken together, our results identified a new mutation in the IAA14 promoter region, which affects the expression pattern of IAA14 and in turn its ability to control plant regeneration. Formation of granulation tissue is a fundamental phase in periodontal wound healing with subsequent maturation leading to regeneration or repair. However, persistently inflamed granulation tissue presents in osseous defects as a result of periodontitis and is routinely disrupted and discarded with non‐surgical and surgical therapy to facilitate wound healing or improve chances of regeneration. Histological assessment suggests that granulation tissue from periodontitis‐affected sites is effectively a chronic inflammatory tissue resulting from impaired wound healing due to persistence of bacterial dysbiotic bioflim. Nevertheless, the immunomodulatory potential and stem cell characteristics in granulation tissue have also raised speculation about the tissue's regenerative potential. This has led to the conception and recent implementation of surgical techniques which preserve granulation tissue with the intention of enhancing innate regenerative potential and improve clinical outcomes. As knowledge of fundamental cellular and molecular functions regulating periodontitis‐affected granulation tissue is still scarce, this review aimed to provide a summary of current understanding of granulation tissue in the context of periodontal wound healing. Hydrogels with high mechanical properties and excellent adsorption capacity are expected in wastewater treatment. Herein, 3D porous hydrogels were prepared by free radical copolymerization using carboxymethyl cellulose nanofibers (CNFs) grafted with N, S atom doped carbon quantum dots (N, S-CDs) and 1-Allyl-2-thiourea. N, S-CDs introduced in the hydrogel can uniformly disperse and increase the adsorption site of Hg(II) by grafting method. Meanwhile, the good mechanical strength of N, S-CDs is beneficial to improve the hydrogel mechanical properties. The experimental results show that the 3D porous hydrogel has great swelling properties (SR = 875.22 g/g) and mechanical properties (elastic modulus = 86.77 MPa). Moreover, compared with the hydrogel without N, S-CDs, the 3D porous hydrogel has an excellent Hg(II) adsorption capacity (943.77 mg/g). In addition, the 3D porous hydrogel has outstanding regeneration performance. The adsorption capacity accounts for 94.6% of the first adsorption capacity after 5 cycles. Skeletal muscle stem and progenitor cells including those derived from human pluripotent stem cells (hPSCs) offer an avenue towards personalized therapies and readily fuse to form human-mouse myofibres in vivo. However, skeletal muscle progenitor cells (SMPCs) inefficiently colonize chimeric stem cell niches and instead associate with human myofibres resembling foetal niches. We hypothesized competition with mouse satellite cells (SCs) prevented SMPC engraftment into the SC niche and thus generated an SC ablation mouse compatible with human engraftment. Single-nucleus RNA sequencing of SC-ablated mice identified the absence of a transient myofibre subtype during regeneration expressing Actc1. Similarly, ACTC1+ human myofibres supporting PAX7+ SMPCs increased in SC-ablated mice, and after re-injury we found SMPCs could now repopulate into chimeric niches. To demonstrate ACTC1+ myofibres are essential to supporting PAX7 SMPCs, we generated caspase-inducible ACTC1 depletion human pluripotent stem cells, and upon SMPC engraftment we found a 90% reduction in ACTC1+ myofibres and a 100-fold decrease in PAX7 cell numbers compared with non-induced controls. We used spatial RNA sequencing to identify key factors driving emerging human niche formation between ACTC1+ myofibres and PAX7+ SMPCs in vivo. This revealed that transient regenerating human myofibres are essential for emerging niche formation in vivo to support PAX7 SMPCs.

Mammals have limited capacity for heart regeneration, whereas zebrafish have extraordinary regeneration abilities. During zebrafish heart regeneration, endothelial cells promote cardiomyocyte cell cycle reentry and myocardial repair, but the mechanisms responsible for promoting an injury microenvironment conducive to regeneration remain incompletely defined. Here, we identify the matrix metalloproteinase Mmp14b as an essential regulator of heart regeneration. We identify a TEAD-dependent mmp14b endothelial enhancer induced by heart injury in zebrafish and mice, and we show that the enhancer is required for regeneration, supporting a role for Hippo signaling upstream of mmp14b. Last, we show that MMP-14 function in mice is important for the accumulation of Agrin, an essential regulator of neonatal mouse heart regeneration. These findings reveal mechanisms for extracellular matrix remodeling that promote heart regeneration.

Abstract Dental diseases such as caries and periodontitis have been common public health problems. Dental disease treatment can be achieved through stem cell-based dental regeneration. Biophysical cues determine the fate of stem cells and govern the success of dental regeneration. Some studies have manifested exosomes derived from stem cells could not only inherit biophysical signals in microenvironment but also evade some issues in the treatment with stem cells. Nowadays, biophysical cue-regulated exosomes become another promising therapy in dental regenerative medicine. However, methods to improve the efficacy of exosome therapy and the underlying mechanisms are still unresolved. In this review, the association between biophysical cues and dental diseases was summarized. We retrospected the role of exosomes regulated by biophysical cues in curing dental diseases and promoting dental regeneration. Our research also delved into the mechanisms by which biophysical cues control the biogenesis, release, and uptake of exosomes, as well as potential methods to enhance the effectiveness of exosomes. The aim of this review was to underscore the important place biophysical cue-regulated exosomes occupy in the realm of dentistry, and to explore novel targets for dental diseases.

Abstract Background Plant breeding played a very important role in transforming strawberries from being a niche crop with a small geographical footprint into an economically important crop grown across the planet. But even modern marker assisted breeding takes a considerable amount of time, over multiple plant generations, to produce a plant with desirable traits. As a quicker alternative, plants with desirable traits can be raised through tissue culture by doing precise genetic manipulations. Overexpression of morphogenic regulators previously known for meristem development, the transcription factors Growth-Regulating Factors (GRFs) and the GRF-Interacting Factors (GIFs), provided an efficient strategy for easier regeneration and transformation in multiple crops. Results We present here a comprehensive protocol for the diploid strawberry Fragaria vesca Hawaii 4 (strawberry) regeneration and transformation under control condition as compared to ectopic expression of different GRF4-GIF1 chimeras from different plant species. We report that ectopic expression of Vitis vinifera VvGRF4-GIF1 provides significantly higher regeneration efficiency during re-transformation over wild-type plants. On the other hand, deregulated expression of miRNA resistant version of VvGRF4-GIF1 or Triticum aestivum (wheat) TaGRF4-GIF1 resulted in abnormalities. Transcriptomic analysis between the different chimeric GRF4-GIF1 lines indicate that differential expression of FvExpansin might be responsible for the observed pleiotropic effects. Similarly, cytokinin dehydrogenase/oxygenase and cytokinin responsive response regulators also showed differential expression indicating GRF4-GIF1 pathway playing important role in controlling cytokinin homeostasis. Conclusion Our data indicate that ectopic expression of Vitis vinifera VvGRF4-GIF1 chimera can provide significant advantage over wild-type plants during strawberry regeneration without producing any pleiotropic effects seen for the miRNA resistant VvGRF4-GIF1 or TaGRF4-GIF1.

Abstract Background Lack of insight into factors that determine purity and quality of human iPSC (hiPSC)-derived neo-cartilage precludes applications of this powerful technology toward regenerative solutions in the clinical setting. Here, we set out to generate methylome-wide landscapes of hiPSC-derived neo-cartilages from different tissues-of-origin and integrated transcriptome-wide data to identify dissimilarities in set points of methylation with associated transcription and the respective pathways in which these genes act. Methods We applied in vitro chondrogenesis using hiPSCs generated from two different tissue sources: skin fibroblasts and articular cartilage. Upon differentiation toward chondrocytes, these are referred to as hFiCs and hCiC, respectively. Genome-wide DNA methylation and RNA sequencing datasets were generated of the hiPSC-derived neo-cartilages, and the epigenetically regulated transcriptome was compared to that of neo-cartilage deposited by human primary articular cartilage (hPAC). Results Methylome-wide landscapes of neo-cartilages of hiPSCs reprogrammed from two different somatic tissues were 85% similar to that of hPACs. By integration of transcriptome-wide data, differences in transcriptionally active CpGs between hCiC relative to hPAC were prioritized. Among the CpG-gene pairs lower expressed in hCiCs relative to hPACs, we identified genes such as MGP, GDF5, and CHAD enriched in closely related pathways and involved in cartilage development that likely mark phenotypic differences in chondrocyte states. Vice versa, among the CpG-gene pairs higher expressed, we identified genes such as KIF1A or NKX2-2 enriched in neurogenic pathways and likely reflecting off target differentiation. Conclusions We did not find significant variation between the neo-cartilages derived from hiPSCs of different tissue sources, suggesting that application of a robust differentiation protocol such as we applied here is more important as compared to the epigenetic memory of the cells of origin. Results of our study could be further exploited to improve quality, purity, and maturity of hiPSC-derived neo-cartilage matrix, ultimately to realize introduction of sustainable, hiPSC-derived neo-cartilage implantation into clinical practice.

The death of photoreceptors is the principal event which is lack of curative treatment. Xaliproden, a highly selective synthetic 5-OH-tryptamine (5HT) 1A receptor agonist, has the neuroprotective potential. However, its application has been limited by the insoluble formulation, low utilization efficiency and side effects caused by systemic administration.Methods: Nanoscale zirconium-porphyrin metal-organic framework (NPMOF) was used as a skeleton and loaded with xaliproden (XAL) to prepare a novel kind of nanoparticle, namely, XAL-NPMOF. The human umbilical vein endothelial cells, zebrafish embryos and larvae were used to test the biotoxicity and fluorescence imaging capability of XAL-NPMOF both in vitro and in vivo. A photoreceptor degeneration model was generated by intense light injury in adult zebrafish and XAL-NPMOF was delivered to the injured retina by intraocular injection. The photoreceptor regeneration, inflammatory response and visual function were explored by immunohistochemistry, quantitative real-time polymerase chain reaction and optomotor response analysis.Results: Following a single XAL-NPMOF intraocular injection, the injured retina underwent the faster photoreceptor regeneration with a recovery of visual function via promoting cell proliferation, suppressing the inflammatory responses and increasing the expression of antioxidases.Conclusion: As an amplifier, NPMOF can enhance the anti-inflammatory efficacy and neuroprotective effect of xaliproden. Abstract Background Vasoactive intestinal peptide (VIP) is a neuronal peptide with prominent distribution along the enteric nervous system. While effects of VIP on intestinal motility, mucosal vasodilation, secretion, and mucosal immune cell function are well-studied, the direct impact of VIP on intestinal epithelial cell turnover and differentiation remains less understood. Intestinal stem and progenitor cells are essential for the maintenance of intestinal homeostasis and regeneration, and their functions can be modulated by factors of the stem cell niche, including neuronal mediators. Here, we investigated the role of VIP in regulating intestinal epithelial homeostasis and regeneration following irradiation-induced injury. Methods Jejunal organoids were derived from male and female C57Bl6/J, Lgr5-EGFP-IRES-CreERT2 or Lgr5-EGFP-IRES-CreERT2/R26R-LSL-TdTomato mice and treated with VIP prior to analysis. Injury conditions were induced by exposing organoids to 6 Gy of irradiation (IR). To investigate protective effects of VIP in vivo, mice received 12 Gy of abdominal IR followed by intraperitoneal injections of VIP. Results We observed that VIP promotes epithelial differentiation towards a secretory phenotype predominantly via the p38 MAPK pathway. Moreover, VIP prominently modulated epithelial proliferation as well as the number and proliferative activity of Lgr5-EGFP+ progenitor cells under homeostatic conditions. In the context of acute irradiation injury in vitro, we observed that IR injury renders Lgr5-EGFP+ progenitor cells more susceptible to VIP-induced modulations, which coincided with the strong promotion of epithelial regeneration by VIP. Finally, the observed effects translate into an in vivo model of abdominal irradiation, where VIP showed to prominently mitigate radiation-induced injury. Conclusions VIP prominently governs intestinal homeostasis by regulating epithelial progenitor cell proliferation and differentiation and promotes intestinal regeneration following acute irradiation injury.

Abstract Background Embryos are regeneration and wound healing masters. They rapidly close wounds and scarlessly remodel and regenerate injured tissue. Regeneration has been extensively studied in many animal models using new tools such as single-cell analysis. However, until now, they have been based primarily on experiments assessing from 1 day post injury. Results In this paper, we reveal that critical steps initiating regeneration occur within hours after injury. We discovered the regeneration initiating cells (RICs) using single-cell and spatial transcriptomics of the regenerating Xenopus laevis tail. RICs are formed transiently from the basal epidermal cells, and their expression signature suggests they are important for modifying the surrounding extracellular matrix thus regulating development. The absence or deregulation of RICs leads to excessive extracellular matrix deposition and defective regeneration. Conclusion RICs represent a newly discovered transient cell state involved in the initiation of the regeneration process.

Abstract Objectives Oral mucosal lesions are prevalent and often cause pain, thus impacting patients' quality of life. Platelet-rich fibrin (PRF) has emerged as a promising autologous biomaterial for wound healing, yet comprehensive evidence regarding its efficacy in treating oral mucosal lesions is limited. This study aims to update the current evidence on the effectiveness of PRF in treating various types of oral mucosal lesions. Materials and methods We conducted a literature search in PubMed, Scopus, Embase, and Web of Science databases until April 2024. The search included studies that investigated the use of PRF in treating oral mucosal lesions. Twelve studies met the inclusion criteria, comprising three case reports, three randomized controlled trials, two animal studies, three split-mouth trials, and one retrospective study. We performed data extraction according to a predefined form. Results PRF was applied in two forms—membranes and injectable gels—to treat a range of oral mucosal lesions, including ulcerative, red and white, pigmented, and potentially malignant or malignant lesions. Compared to control groups or conventional treatments, PRF generally demonstrated superior outcomes regarding faster healing, lesion size reduction, symptom relief, and lower recurrence rates. Histological and molecular analyses from some studies also indicated PRF's regenerative and anti-inflammatory effects. Conclusion PRF shows promise as an effective and safe alternative to current treatments for oral mucosal lesions due to its autologous nature, ease of preparation, and wound-healing capabilities. However, further research is needed to standardize PRF preparation protocols and confirm its long-term efficacy across different lesion types.

Objective(s): Mammals have limited limb regeneration compared to amphibians. The role of Wnt signaling pathways in limb regeneration has rarely been studied. So, this study aimed to investigate the effect of Wnt-signaling using chemicals CHIR99021 and IWP-2 on amputated mice digit tips regeneration in an in vitro organ culture system.Materials and Methods: The distal phalanx of paws from C57BL/6J mouse fetuses at E14.5, E16.5, and E18.5 was amputated. Then, the hands were cultured for 7 days. Subsequently, paws were treated with 1–50 µg/ml concentration of CHIR99021 and 5–10 µg/ml concentration of IWP-2. Finally, the new tissue regrowth was assessed by histological analysis, immunohistochemistry for BC, TCF1, CAN, K14, and P63 genes, and beta-catenin and Tcf1 genes were evaluated with RT-qPCR. Results: The paws of E14.5 and E16.5 days were shrinkaged and compressed after 7 days, so the paws of 18.5E that were alive were selected. Objective: This study aimed to evaluate the calcium ion (Ca2+) release from apical plugs formed by three different bioceramic cements in simulated immature teeth with open apices.Methods: In this in-vitro study, 66 single-rooted lower premolar teeth were divided into three groups, each containing 22 samples, as follows: Group A: ProRoot MTA, Group B: Bio-C Repair, and Group C: Dia-Root Bio MTA. Each sample was prepared to simulate immature teeth with open apices. A 4 mm apical plug was inserted at the open apex area. Calcium ion release was measured on the 7th, 15th, and 30th days using an atomic absorption spectrophotometer. Plant growth regulators (PGRs) control signaling networks and developmental processes involved in plant responses to various biotic and abiotic stresses, making it crucial to study PGRs in vitro . The protocol for micropropagation of Solanum lycopersicum L., following callus induction and regeneration through explants such as internodal segments, leaves, and nodal segments, was established during the present study. Explants were inoculated on Murashige and Skoog (MS) medium supplemented with different plant growth regulators like BA (6-benzylaminopurine), 2,4-D (2,4-dichlorophenoxyacetic acid), BA + 2,4-D, IAA (Indoleacetic acid), IBA (Indolebutyric acid), and NAA (Naphthaleneacetic acid). It was observed that among all explants, the nodal segment showed maximum callus induction (83.33%) and multiplication (86.67%) at 0.25 mg/l of 2,4-D; the highest shoot number (3.33) at 0.5 mg/l of IAA + 0.5 mg/l of BA; the greatest shoot length (7.57 cm) at 0.75 mg/l of BA; root induction (80.95%), root number (21.88), and root length (10.62 cm) at 1 mg/l of IAA. Additionally, the maximum fresh weight (2.448 g) was observed at 0.5 mg/l of BA, while the highest dry weight (0.172 g) and dry matter content (14.25%) were noted at 1 mg/l of BA + 1 mg/l of 2,4-D for the internodal segment. Results of the present study revealed that among different explants, the best response was given by nodal segments, followed by internodal segments. Among the different PGRs, 2,4-D resulted in the highest callus induction and multiplication percentage.

Abstract The mechanism of convective cell initiation and regeneration in the Bandung Basin ( $$107.35^\circ \text{E}-107.95^\circ \text{E};6.75^\circ \text{S}-7.25^\circ \text{S}$$ 107 . 35 ∘ E - 107 . 95 ∘ E ; 6 . 75 ∘ S - 7 . 25 ∘ S ), Indonesia, is investigated using a Weather Research and Forecasting model. Based on the Cumulative Distribution Function and Regeneration Index, model simulations were conducted using case studies in February–March 2019. Upslope wind flows to the mountain peak in north and south of the basin due to solar heating, carrying water vapor from the bottom of the basin. Therefore, low-level convergence is formed due to the convergence of winds from the bottom of the basin and from outside the basin. The low-level convergence causes the developing updraft, makes atmosphere unstable, and generates convective initiation. The convective system will be developed in the mountains region, produced precipitation, and formed cold pools on the surface. The cold pool will fall down the slopes and convective activity from the mountains will continue to the bottom of the basin. The convective system’s outflow causes the cold pool to move in the west–east direction. The cold pool will collide with warmer air from the opposite direction, resulting in an updraft at the cold pool leading edge, and convection will be re-initiated (convective regeneration) in the Bandung Basin, with the new convective system tending to move eastward.

Abstract Key message Multi-temporal airborne laser scanning (ALS) data were used to estimate regeneration stem height growth within gaps in uneven-aged deciduous forests. The height and height growth measured in the field were used to calibrate and validate ALS estimates. This method provided highly precise estimates of height and unbiased height increment estimates of regeneration at stem level. Context Assessing regeneration height growth is essential for evaluating forest dynamics and optimizing silvicultural operations. However, regeneration description at high spatiotemporal resolution has remained limited to restricted areas by the limiting cost constraints of field measurements. Highly precise airborne laser scanning (ALS) data are currently acquired over wide areas. Such datasets are promising for characterizing regeneration dynamics. Aims We aimed to estimate height and height growth within regenerating areas at the stem level using multi-temporal ALS data. Methods ALS data were acquired from 56,150 ha of uneven-aged deciduous forest in Belgium in 2014 and 2021. Stem tops were detected using local maxima (LM) within regenerating areas in both ALS datasets and matched. Field data were collected in 2021 and used to calibrate the ALS-estimated heights using linear and non-linear models at stem level. Height growth estimation was then validated using field-measured increments. Results Without height calibration, the 2021 ALS-estimated height had a − 1.06 m bias and 1.39 m root-mean-squared error (RMSE). Cementum plays a crucial role in linking periodontal and dental tissues and maintaining tooth root stability. Regenerating cementum is essential for functional periodontal tissue regeneration. It is a significant focus in tissue engineering. Periodontal disease, prevalent among the middle-aged and elderly, can severely impact overall health. Effective periodontal tissue regeneration is vital for enhancing the elderly's health and quality of life. Despite the progress, the lack of ideal scaffold materials and methods makes cementum regeneration a persistent challenge. This review discusses the current state of cementum tissue engineering materials and addresses existing challenges, providing a foundation for future scaffold material development.

Abstract Background Foxn1−/− deficient mice are a rare model of regenerative skin wound healing among mammals. In wounded skin, the transcription factor Foxn1 interacting with hypoxia-regulated factors affects re-epithelialization, epithelial-mesenchymal transition (EMT) and dermal white adipose tissue (dWAT) reestablishment and is thus a factor regulating scar-forming/reparative healing. Here, we hypothesized that transcriptional crosstalk between Foxn1 and Hif-1α controls the switch from scarless (regenerative) to scar-present (reparative) skin wound healing. To verify this hypothesis, we examined (i) the effect of hypoxia/normoxia and Foxn1 signalling on the proteomic signature of Foxn1−/− (regenerative) dermal fibroblasts (DFs) and then (ii) explored the effect of Hif-1α or Foxn1/Hif-1α introduced by a lentiviral (LV) delivery vector to injured skin of regenerative Foxn1−/− mice with particular attention to the remodelling phase of healing. Results We showed that hypoxic conditions and Foxn1 stimulation modified the proteome of Foxn1−/− DFs. Hypoxic conditions upregulated DF protein profiles, particularly those related to extracellular matrix (ECM) composition: plasminogen activator inhibitor-1 (Pai-1), Sdc4, Plod2, Plod1, Lox, Loxl2, Itga2, Vldlr, Ftl1, Vegfa, Hmox1, Fth1, and F3. We found that Pai-1 was stimulated by hypoxic conditions in regenerative Foxn1−/− DFs but was released by DFs to the culture media exclusively upon hypoxia and Foxn1 stimulation. We also found higher levels of Pai-1 protein in DFs isolated from Foxn1+/+ mice (reparative/scar-forming) than in DFs isolated from Foxn1−/− (regenerative/scarless) mice and triggered by injury increase in Foxn1 and Pai-1 protein in the skin of mice with active Foxn1 (Foxn1+/+ mice). Then, we demonstrated that the introduction of Foxn1 and Hif-1α via lentiviral injection into the wounded skin of regenerative Foxn1−/− mice activates reparative/scar-forming healing by increasing the wounded skin area and decreasing hyaluronic acid deposition and the collagen type III to I ratio. We also identified a stimulatory effect of LV-Foxn1 + LV-Hif-1α injection in the wounded skin of Foxn1−/− mice on Pai-1 protein levels. Conclusions The present data highlight the effect of hypoxia and Foxn1 on the protein profile and functionality of regenerative Foxn1−/− DFs and demonstrate that the introduction of Foxn1 and Hif-1α into the wounded skin of regenerative Foxn1−/− mice activates reparative/scar-forming healing.

Endometrial injury is the main fact leading to the damage of endometrial basal layer. It is usually caused by traumatic treatment, inflammatory injury, radiation, chemical drug-related injury, and delivery-related diverticulum. The use of hyaluronic acid, decellularized matrix, amniotic membrane, platelet-rich plasma, mesenchymal stem cells (especially scaffold-based therapeutic delivery) has been proved to reduce post-injury scar formation and promote endometrial growth and repair, which provides a new therapeutic strategy for repairing the damaged endometrium with improved tolerability and fertility outcomes.

Abstract The present study aims to investigate the low-energy consumption and high-efficiency removal of arsenic from aqueous solutions. The designed adsorbent Fe/TBC was synthesized by impregnating iron on torrefaction henequen fibers. Isothermal adsorption experiments indicated maximum adsorption capacities of 7.30 mg/g and 8.98 mg/g for arsenic(V) at 25.0 °C and 40.0 °C, respectively. The interference testing showed that elevated levels of pH, HCO3 − concentration, and humic acid content in the solution could inhibit the adsorption of arsenic by Fe/TBC. Characterization of the adsorbent before and after adsorption using FTIR and SEM–EDS techniques confirmed arsenic adsorption mechanisms, including pore filling, electrostatic interaction, surface complexation, and H-bond adhesion. Column experiments were conducted to treat arsenic-spiked water and natural groundwater, with effective treatment volumes of 550 mL and 8792 mL, respectively. Lastly, the life cycle assessment (LCA) using OpenLCA 2.0.3 software was performed to treat 1 m3 of natural groundwater as the functional unit. The results indicated relatively significant environmental impacts during the Fe/TBC synthesis stage. The global warming potential resulting from the entire life cycle process was determined to be 0.8 kg CO2-eq. The results from batch and column experiments, regeneration studies, and LCA analysis indicate that Fe/TBC could be a promising adsorbent for arsenic(V).

Abstract We investigated the effect of repeated sessions of anodal transcranial direct current stimulation (a-tDCS) on subjective and objective measures of recovery, cognitive and sport-specific performance in professional soccer players following a soccer match simulation (SMS). Sixteen soccer players participated in this randomized, crossover, and sham-controlled study. They completed baseline assessments of well-being, total quality recovery (TQR), electromyographic activity (EMG) of the thigh muscles, countermovement jump (CMJ), and cognitive and Loughborough soccer passing test (LSPT) skills. Then, the participants engaged in an SMS routine (2 × 45 min, 15-min intervals). There was no significant difference in rating of perceived exertion (RPE) during the SMS in the anodal (17.25 ± 0.85) and sham (16.93 ± 0.92) conditions (p = 0.19). Following the SMS, the participants were randomized to receive three sessions of a-tDCS (2 mA, 20 min, +F3/−F4) targeting the left dorsolateral prefrontal cortex (DLPFC) or sham immediately after, 24 h, and 48 h after the SMS. Finally, the same outcome measures were evaluated 24 and 48 h following the SMS. Relict subtropical coniferous forests in China face severe fragmentation, resulting in declining populations, and some are under significant threat from invasive alien species. Despite the crucial importance of understanding forest dynamics, knowledge gaps persist, particularly regarding the impact of invasive plants on vulnerable natives like Keteleeria evelyniana. In this study, we investigated the impact of invasive plants on the regeneration of forests dominated by K. evelyniana, a subtropical relict species in southwestern China. For this purpose, we characterized forest dynamics of 160 forest plots featuring K. evelyniana as the primary dominant species and determined whether the presence of invasive plants was correlated with regeneration of K. evelyniana. We identified four distinct forest types in which K. evelyniana was dominant. We found that radial growth of K. evelyniana trees is faster in younger age-classes today than it was for older trees at the same age. The population structure of K. evelyniana in each forest type exhibited a multimodal age-class distribution. However, three forest types lacked established saplings younger than 10 years old, a situation attributed to the dense coverage of the invasive alien Ageratina adenophora. This invasive species resulted in a reduction of understory species diversity. Additionally, our analysis uncovered a significant negative correlation in phylogenetic relatedness (net relatedness index) between native and invasive alien plant species in eastern Yunnan. This suggests closely related invasive species face heightened competition, hindering successful invasion. Taken together, our findings indicate that successful establishment and habitat restoration of K. evelyniana seedling/saplings require effective measures to control invasive plants.

Taking into account the imperfect results of treatment of peripheral nerve injuries and despite the large number of already performed clinical and experimental studies on the regeneration of peripheral nerves, the search for new effective methods of treatment of this pathology remains relevant until now. Cell therapy is one of the innovative and promising therapeutic approaches in the field of nerve regeneration. Mesenchymal stem cells (MSCs) were first introduced in 1966 by Friedenstein and colleagues. This is a class of multipotent stem cells of mesodermal origin, which have low immunogenicity, but high capacity for self-renewal and multidirectional differentiation potential. They were found in almost all tissues. According to the International Society for Cell Therapy (ISCT, Vancouver, Canada), human MSCs express CD73, CD105, CD90 and lack hematopoietic stem cell markers (CD34, CD14, CD45, CD11b, CD79a, and HLA-DR). There is no consensus regarding the main mechanism of action of MSCs in stimulating nerve regeneration, but in general, most likely, they perform a trophic function, inhibit proapoptotic pathways and enhance the intrinsic regenerative capacity of the nerve. The ability of MSCs to produce various immunoregulatory factors is also known, which leads to the reduction of an inadequate inflammatory response after nerve damage. MSCs can transdifferentiate into Schwann cells, acquire a Schwann-like phenotype, or stimulate the activity of endogenous Schwann cells, which are one of the key regulators of reparative nerve regeneration. MSCs also take part in the remyelination of damaged nerves due to the synthesis and release of myelin proteins MBP or P0. MSCs create an enhanced neuroprotective microenvironment that prevents nerve degeneration, supports axon growth, neurogenesis, and cellular metabolism. The property of stem cells to produce neurotrophic factors is also well known. It is interesting to note that endogenous stem cells, including those circulating in peripheral blood, are much less studied. A detailed study of various pharmacological, gene-modifying, or biomodifying methods of endogenous stem cell mobilization, their properties, and identification may be promising in regenerative medicine in general and peripheral nerve regeneration in particular.

Abstract Microglia are resident immune cells in the central nervous system, including the retina that surveil the environment for damage and infection. Following retinal damage, microglia undergo morphological changes, migrate to the site of damage, and express and secrete pro-inflammatory signals. In the zebrafish retina, inflammation induces the reprogramming and proliferation of Müller glia and the regeneration of neurons following damage or injury. Immunosuppression or pharmacological ablation of microglia reduce or abolish Müller glia proliferation. We evaluated the retinal architecture and retinal regeneration in adult zebrafish irf8 mutants, which have significantly depleted numbers of microglia. We show that irf8 mutants have normal retinal structure at 3 months post fertilization (mpf) and 6 mpf but fewer cone photoreceptors by 10 mpf. Surprisingly, light-induced photoreceptor ablation induced Müller glia proliferation in irf8 mutants and cone and rod photoreceptor regeneration. Light-damaged retinas from both wild-type and irf8 mutants show upregulated expression of mmp-9, il8, and tnfβ pro-inflammatory cytokines. Our data demonstrate that adult zebrafish irf8 mutants can regenerate normally following acute retinal injury. These findings suggest that microglia may not be essential for retinal regeneration in zebrafish and that other mechanisms can compensate for the reduction in microglia numbers.

Abstract The skin epidermis is continually influenced by a myriad of internal and external elements. At its basal layer reside epidermal stem cells, which fuels epidermal renovation and hair regeneration with powerful self-renewal ability, as well as keeping diverse signals that direct their activity under surveillance with quick response. The importance of epidermal stem cells in wound healing and immune-related skin conditions has been increasingly recognized, and their potential for clinical applications is attracting attention. In this review, we delve into recent advancements and the various physiological and psychological factors that govern distinct epidermal stem cell populations, including psychological stress, mechanical forces, chronic aging, and circadian rhythm, as well as providing an overview of current methodological approaches. Furthermore, we discuss the pathogenic role of epidermal stem cells in immune-related skin disorders and their potential clinical applications.

In this review, we will summarize various nanomedicine, biomaterials, and innovative technologies that contribute to endometrial regeneration, including nanoscale exosomes, nanomaterials, stem cell-based materials, naturally sourced biomaterials, chemically synthesized biomaterials, approaches and methods for functionalizing biomaterials, as well as the application of revolutionary new technologies such as organoids, organ-on-chips, artificial intelligence, etc. The diverse design and modification of new biomaterials endow them with new functionalities, such as microstructure or nanostructure, mechanical properties, biological functions, and cellular microenvironment regulation. It will provide new options for the regeneration of endometrium, bring new hope for the reconstruction and recovery of patients’ reproductive abilities. Abstract Aim This study aims to compare the clinical and radiographic outcomes after complete versus incomplete removal of granulation tissue (GT) during modified minimally invasive surgical technique (M-MIST) for management of periodontitis patients with deep pockets associated with infra-bony defects. Methodology Ten patients with a total of 14 deep non-resolving pockets (≥ 5 mm) associated with a vertical infra-bony defect were recruited for this study. They were randomized into 2 groups; a test group with incomplete removal of GT and a control group with complete removal of GT. Clinical parameters of clinical attachment level (CAL), residual probing depth (rPD) and buccal recession (Rec.) were recorded every 3 months. Radiographic periapicals were taken at baseline, 6 and 9 months. The significance level was set to 0.05. Results None of the results showed statistical significance between the 2 groups (p > 0.05). The test group showed less CAL gain (2 ± 0.87 mm, p = 0.062), more reduction in rPD (3.1 ± 0.96 mm, p = 0.017) and more recession (0.857 ± 0.26 mm, p = 0.017) than control group CAL gain (2.4 ± 0.58 mm, p = 0.009), rPD reduction (2.9 ± 0.3 mm, p = 0.001) and recession (0.5 ± 0.34 mm, p = 0.203) respectively. Control group had linear reduction in depth defect (DD) (0.68 ± 0.287, p = 0.064) compared to an increase in DD in test group (-0.59 ± 0.5, p = 0.914). Conclusions No statistical significance were observed in healing parameters between complete removal of GT in M-MIST and incomplete (partial) removal of GT of deep pockets with infra-bony defects both clinically and radiographically. Further studies with larger samples are needed to confirm the results.

Phytic acid (inositol hexaphosphate/IP6) is a versatile chemical that is abundant in nature and is required for a variety of biological processes. It is harnessed in a wide range of fields, including drug discovery, daily supplies, chemical industries, medicine, and dentistry. IP6 is becoming increasingly popular in dentistry, with promising results. Several properties, such as cariostatic ability, beneficial impact on enamel disintegration, and anti-plaque, anti-tartar, and dental adhesive-forming properties, have been investigated thus far. Due to many constraints in the literature, there was a point in time when IP6 received less attention, which impacted knowledge in this field. Nevertheless, the positive outcomes of the flourishing of IP6 have recently been reconsidered from a number of papers that have improved our understanding of its modes of action in the aforementioned applications. The role of phytic acid in refining the properties and manoeuvring of dental resources is being investigated in novel endeavors in treating diseases of pulp and tissues supporting tooth structure, but to show its novel therapeutic potential, more precisely calibrated clinical trials are needed. This review examines and discusses the various uses proposed in the literature, as well as the applications of IP6 in dentistry.

Abstract Background The starlet sea anemone, Nematostella vectensis, is an emerging model organism with a high regenerative capacity, which was recently found to possess an orthologue to the human Leucine Rich Repeat Kinase 2 (LRRK2) gene. Mutations in this gene are the most common cause of inherited Parkinson’s Disease (PD), highlighting the importance of understanding its function. Despite two decades of research, however, the function of LRRK2 is not well established. Methods To investigate the function of LRRKs in Nematostella vectensis, we applied small molecule inhibitors targeting the kinase activity of LRRK2 to examine its function in development, homeostasis and regeneration in Nematostella vectensis. Results In vivo analyses inhibiting the kinase function of this enzyme demonstrated a role of nvLRRK2 in development and regeneration of N. vectensis. These findings implicate a developmental role of LRRK2 in Nematostella, adding to the expanding knowledge of its physiological function. Conclusions Our work introduces a new model organism with which to study LRRK biology. We report that LRRK kinase activity is necessary for the development and regeneration of Nematostella. Given the short generation time, genetic trackability and in vivo imaging capabilities, this work introduces Nematostella vectensis as a new model in which to study genes linked to neurodegenerative diseases such as Parkinson’s.

Abstract Background The remarkable regenerative abilities observed in planarians and cnidarians are closely linked to the active proliferation of adult stem cells and the precise differentiation of their progeny, both of which typically deteriorate during aging in low regenerative animals. While regeneration-specific genes conserved in highly regenerative organisms may confer regenerative abilities and long-term maintenance of tissue homeostasis, it remains unclear whether introducing these regenerative genes into low regenerative animals can improve their regeneration and aging processes. Results Here, we ectopically express highly regenerative species-specific JmjC domain-encoding genes (HRJDs) in Drosophila, a widely used low regenerative model organism. Surprisingly, HRJD expression impedes tissue regeneration in the developing wing disc but extends organismal lifespan when expressed in the intestinal stem cell lineages of the adult midgut under non-regenerative conditions. Notably, HRJDs enhance the proliferative activity of intestinal stem cells while maintaining their differentiation fidelity, ameliorating age-related decline in gut barrier functions. Conclusions These findings together suggest that the introduction of highly regenerative species-specific genes can improve stem cell functions and promote a healthy lifespan when expressed in aging animals.

Salinity stress is a major abiotic stress factor that affects plant growth, physiological activities and developmental processes. This study involves establishing efficient somatic embryogenesis and plantlet regeneration system using immature inflorescence of kodo millet (Paspalum scrobiculatum L. cv. TNAU86) under NaCl-salinity stress conditions. To begin with, the immature inflorescence (0.5cm) of kodo millet was excised from the 40-45 days old field growing plants followed by surface sterilization and inoculation in Murashige and Skoog (MS) medium supplemented with the various concentrations of NaCl (10mM, 25mM, 50mM, 75mM, 100mM, 150mM, and 200mM) salt along with 2,4-Dichlorophenoxyacetic acid (2,4-D) (1.5 mg/L). The results reveal that the maximum mean frequency (89.3±0.3%) of somatic embryogenesis was obtained from embryogenic callus that was growing with medium added with NaCl (50mM) while it was found to be the least mean frequency (12±4.1%) with 150mM of NaCl-salt treatments. Moreover, the highest concentration of NaCl (200mM) salinity was found to be lethal and explants were observed to get gradually necrosed. Later, embryogenic calli showing differentiation of somatic embryos were sub-cultured on basal medium supplemented with 0.5mg/L of 6-Benzylaminopurine (BAP) along with respective concentrations of NaCl-salt for the germination of somatic embryos into plantlets. Significantly, 100mM of NaCl-treatment was proved to show strong inhibitions and thus minimum salt tolerant plantlets regeneration (4±2.21%) was recorded. Further, in vitro grown salinity stress tolerant plantlets were transferred to plastic cups and gradually acclimatized under greenhouse conditions.

Clinical use of small-diameter vascular grafts remains a challenging issue in neovessel regeneration in view of thrombosis and intimal hyperplasia. Developing a vascular graft with structure and function similar to those of the native vessels necessitates a major direction of vascular tissue regeneration. Thus, this study sought to design and fabricate a range of tri-phasic scaffolds (0, 2, and 5 wt% gastrodin-polyurethane (PU)) with spatiotemporally defined structure and gastrodin-release for regulating the highly coordinated processes in growth of the intima and media. While the small pores of inner layer guided infiltration of human umbilical vein endothelial cells (HUVECs), the bigger pores of medial layer could offer smooth muscle cell (SMC)-friendly habitat, and external fibers conferred adequate mechanical properties. Correspondingly, spatial distribution and differential regulation of key proteins in HUVECs and SMCs were mediated by hierarchical release of gastrodin, of which rapid release in inner layer elicited enhanced HUVEC proliferation and migration against those of the SMC via activated endothelial nitric oxide synthase (eNOS) and heat shock protein 70 (HSP70) signal. Of note, superior anti-coagulation was reflected in 2 wt% gastrodin-PU ex vivo extracorporeal blood circulation experiment. After in vivo implantation for 12 weeks, there was no formation of obvious thrombosis and intimal hyperplasia in 2 wt% gastrodin-PU. The scaffold maintained high patency and improved vascular remodeling, including the formation of thin endothelialization in lumen and dense extracellular matrix deposition in medial layer. Taken together, the results demonstrate the positive function of hierarchical releasing system that responded to tri-phasic structure, which not only suppressed intimal thickening but also tightly controlled tissue regeneration.

An outstanding mystery in biology is why some species, such as the axolotl, can regenerate tissues whereas mammals cannot1. Here, we demonstrate that rapid activation of protein synthesis is a unique feature of the injury response critical for limb regeneration in the axolotl (Ambystoma mexicanum). By applying polysome sequencing, we identify hundreds of transcripts, including antioxidants and ribosome components that are selectively activated at the level of translation from pre-existing messenger RNAs in response to injury. By contrast, protein synthesis is not activated in response to non-regenerative digit amputation in the mouse. We identify the mTORC1 pathway as a key upstream signal that mediates tissue regeneration and translational control in the axolotl. We discover unique expansions in mTOR protein sequence among urodele amphibians. By engineering an axolotl mTOR (axmTOR) in human cells, we show that these changes create a hypersensitive kinase that allows axolotls to maintain this pathway in a highly labile state primed for rapid activation. This change renders axolotl mTOR more sensitive to nutrient sensing, and inhibition of amino acid transport is sufficient to inhibit tissue regeneration. Together, these findings highlight the unanticipated impact of the translatome on orchestrating the early steps of wound healing in a highly regenerative species and provide a missing link in our understanding of vertebrate regenerative potential.

High-mobility group box 1 was first discovered in the calf thymus as a DNA-binding nuclear protein and has been widely studied in diverse fields, including neurology and neuroscience. High-mobility group box 1 in the extracellular space functions as a pro-inflammatory damage-associated molecular pattern, which has been proven to play an important role in a wide variety of central nervous system disorders such as ischemic stroke, Alzheimer’s disease, frontotemporal dementia, Parkinson’s disease, multiple sclerosis, epilepsy, and traumatic brain injury. Several drugs that inhibit high-mobility group box 1 as a damage-associated molecular pattern, such as glycyrrhizin, ethyl pyruvate, and neutralizing anti-high-mobility group box 1 antibodies, are commonly used to target high-mobility group box 1 activity in central nervous system disorders. Although it is commonly known for its detrimental inflammatory effect, high-mobility group box 1 has also been shown to have beneficial pro-regenerative roles in central nervous system disorders. In this narrative review, we provide a brief summary of the history of high-mobility group box 1 research and its characterization as a damage-associated molecular pattern, its downstream receptors, and intracellular signaling pathways, how high-mobility group box 1 exerts the repair-favoring roles in general and in the central nervous system, and clues on how to differentiate the pro-regenerative from the pro-inflammatory role. Research targeting high-mobility group box 1 in the central nervous system may benefit from differentiating between the two functions rather than overall suppression of high-mobility group box 1.

Axolotls are known for their remarkable regeneration ability. Exploring their transcriptome provides insight into regenerative mechanisms. However, the current annotation of the axolotl transcriptome is limited, leaving the role of unannotated transcripts in regeneration unknown. To discourse this challenge, we exploited long-read sequencing technology, which enables direct observation of full-length RNA transcripts, greatly enhancing the coverage and accuracy of axolotl transcriptome annotation. By utilizing this method, we identified 222 novel gene loci and 4775 novel transcripts, which were quantified using short-read sequencing data. Through the inclusive analysis, we discovered novel homologs, potential functional proteins, noncoding RNAs, and alternative splicing events in key regeneration pathways. In particular, we identified novel transcripts with high protein-coding potential implicated in cell cycle regulation and musculoskeletal development, and regeneration were identified. Interestingly, alternative splice variants were also detected across diverse pathways critical to regeneration. This specifies that these novel transcripts potentially play vital roles underpinning the robust regenerative capacities of axolotls. Single-cell transcriptomic analysis further revealed these isoforms to predominantly exist in axolotl limb chondrocytes and mature tissue cell populations. Overall, the findings significantly advanced consideration of the axolotl transcriptome and provided a new perspective for understanding the mechanisms of regenerative abilities of axolotls.

Lasers have numerous advantageous tissue interactions such as ablation or vaporization, hemostasis, bacterial killing, as well as biological effects, which induce various beneficial therapeutic effects and biological responses in the tissues. Thus, lasers are considered an effective and suitable device for treating a variety of inflammatory and infectious conditions of periodontal disease. Among various laser systems, the Er:YAG laser, which can be effectively and safely used in both soft and hard tissues with minimal thermal side effects, has been attracting much attention in periodontal therapy. This laser can effectively and precisely debride the diseased root surface including calculus removal, ablate diseased connective tissues within the bone defects, and stimulate the irradiated surrounding periodontal tissues during surgery, resulting in favorable wound healing as well as regeneration of periodontal tissues. The safe and effective performance of Er:YAG laser-assisted periodontal surgery has been reported with comparable and occasionally superior clinical outcomes compared to conventional surgery. This article explains the characteristics of the Er:YAG laser and introduces its applications in periodontal surgery including conventional flap surgery, regenerative surgery, and flapless surgery, based on scientific evidence from currently available basic and clinical studies as well as cases reports.

The present study aims to determine the ability of algae in sequestering Lambda‐cyhalothrin (L-C) from aqueous solution. A series of experiments were carried out in batch mode to find equilibrium data for sorption of Lambda‐cyhalothrin (L-C), FTIR analysis was utilized to investigate the impacts of functional groups of algae in the biosorption process. Pseudo second-order kinetics model (R2=0.991) well describe the kinetics of adsorption of L-C onto algae sites while the adsorption mechanisms was controlled by external mass transfer as well as intraparticle diffusion. Langmuir isotherm model better fit the experimental data than Freundlich isotherm model. The higher adsorption capacity was found to be 6.954mg/g. Thermodynamic parameters indicating that sorption of L-C onto algae was endothermic in nature. Only 40% reduction in the sequestration efficiency was noticed after five sequential regeneration cycles. The maximum sorption efficiency was found to be (95.6%) under the best conditions adsorbent dosage=1g/100ml, pH=7, initial L-C concentration=10 mg/l with a contact time of 60 minutes at 25°C. This work demonstrated that algae are a promising adsorbent for L-C removal from aqueous solution.

Decellularized grafts have shown promising results in tissue engineering. The aim of decellularization is to create a scaffold that is devoid of immunogenic components, has natural tissue architecture, and can provide cellularity again. The aim of our study is to obtain a living/regenerative valve by coculture of HUVEC and human dermal fibroblast cells on the pulmonary heart valves of decellularized young sheep and subsequently perform their in vitro characterization to determine its suitability for clinical studies. Various characterization tests were applied to sheep pulmonary heart valves decellularized by the detergent-based method. HUVEC and dermal fibroblast cells were seeded on the scaffold, and their viability was determined by MTT analysis, adhesion by SEM, and cell infiltration by histological staining. Finally, to determine the regenerative ability of the resulting live cap, collagen type I (Col1), collagen type III (Col3), and elastin (Eln) gene expressions were analyzed by PCR. The results showed that cell proliferation increased day by day on the scaffold. In histological findings, it was observed that cellular regeneration was almost completely achieved, especially in arterial wall samples. According to PCR findings, a significant increase in Col1, Col3, and Eln gene expressions was observed in arterial wall samples. In this study, for the first time in the literature, the regeneration potential of differentiated human cells on decellularized sheep heart valves was observed, and markers related to new ECM production were analyzed. In conclusion, we suggest that the decellularized heart valves of young sheep can be used as a starting matrix in tissue engineering studies, and clinical studies are needed. [ABSTRACT FROM AUTHOR]

The dye removal effect of the anisotropic polysaccharide-ZIF-8 aerogel via adsorption and catalysis was observed, highlighting the great potential of the hybrid aerogel as a highly effective, sustainable, and low energy-consuming environmental remediation material. [Display omitted] Assembling microscopic metal–organic frameworks into macroscopic polymeric scaffolds to develop highly renewable materials has been a promising yet challenging area of research. Herein, chitosan (CS) blended with nano-cellulose (NC) was unidirectionally transformed into an aerogel with oriented macropores and then biomineralized with zeolite imidazolate frameworks-8 (ZIF-8) to form a hierarchical structured chitosan-nanocellulose/zeolite imidazolate frameworks-8 (CS-NC-ZIF-8) hybrid aerogel. Incorporating ZIF-8 significantly increases the versatility and mechanical strength with a Young's modulus of 14.18 MPa of the CS-NC aerogel. The incorporation of ZIF-8 into the aerogel not only enhances its adsorption capacity for methylene blue, rhodamine B, acid fuchsin, and methyl orange, but also facilitates the generation of electrons from water that can be transferred to degrade > 90 % of malachite green within 90 min in each catalytic cycle, and this capability was maintained for at least 10 consecutive cycles. Remarkably, the hybrid aerogel was highly renewable after the adsorption of cationic dyes and catalytic removal of malachite green. With its facile production process, high removal efficiency, affordable and green nature, and excellent regeneration feasibility, the CS-NC-ZIF-8 aerogel stands as a promising solution for addressing challenges associated with dye-contaminated water treatment. The efficient recycling of waste graphite anode from used lithium-ion batteries (LIBs) has attracted considerable concerns mainly owing to the environment protection and reutilization of resources. Herein, we reported a rational and facile strategy for the synthesis of holey graphite coated by carbon (hG 0.01 @C 0.10) through the separation, purification and creation of holey structures of waste graphite by using NaOH and carbon-coating by using phenolic resin. The holey structures facilitate the hG 0.01 @C 0.10 with the quick penetration of electrolytes and rapid diffusion of Li+. The carbon coating is more favorable for hG 0.01 @C 0.10 with improved electronic conductivity and less alleviated volume during the cycles. Benefiting from the synergistic effect of holey structures and carbon coating, the hG 0.01 @C 0.10 as anode for LIBs displays a high reversible capacity of 377.6 mAh g-1 at 0.5 C and superior rate capabilities (e.g., 348.0 and 274.7 mAh g-1 at 1 and 2 C, respectively) and maintains a high reversible capacity of 278.7 mAh g-1 at 1 C after 300 cycles with an initial capacity retention of 80.0 %.
