152. Molecular Identification of Rickettsia Species in Haemaphysalis Ticks Collected from Southwest China.

Abstract

Rickettsia species are obligate intracellular Gram-negative bacteria that can infect a wide range of vertebrate hosts, including humans, through arthropod vectors such as Ixodid ticks. These ticks are a threat to humans and animals because they are the primary vectors or reservoirs for rickettsiae, which is of public health importance. In this study, we report the identification and percent of positive of Rickettsia spp. in ticks collected from Cangxi County, Southwest China. Haemaphysalis longicornis comprised 48.4% of the 188 ticks collected followed by Haemaphysalis flava (29.3%), H. doenitzi (12.2%), and Haemaphysalis hystricis (10.1%). A total of 63 (33.5%) ticks were positive with Rickettsia spp., with 48 (57%) of those being H. longicornis and 15 (27.3%) being H. flava. The other two tick species, however, did not have any ticks positive for rickettsial DNA. In addition, two different Rickettsia spp. were identified using gltA and ompA as molecular markers. The sequence of Rickettsia sp. infecting H. longicornis ticks was found to be identical to the Rickettsia sequences from Northeastern China and Japan (KF728367, AB516964). Phylogenetic analyses using these molecular markers support the notion that Rickettsia species from H. flava is the most close to a member of the Candidatus Rickettsia gannanii subgroup. The high percentage of Rickettsia positive in this Southwest China region suggests potential public health threat in the future and warrants to be monitored. [ABSTRACT FROM AUTHOR]

Abstract

In this report, we present a breast imaging technique combining high-resolution near-infrared (NIR) light induced photoacoustic tomography (PAT) with NIR dye-labeled amino-terminal fragments of urokinase plasminogen activator receptor (uPAR) targeted magnetic iron oxide nanoparticles (NIR830-ATF-IONP) for breast cancer imaging using an orthotopic mouse mammary tumor model. We show that accumulation of the targeted nanoparticles in the tumor led to photoacoustic contrast enhancement due to the high absorption of iron oxide nanoparticles (IONP). NIR fluorescence images were used to validate specific delivery of NIR830-ATF-IONP to mouse mammary tumors. We found that systemic delivery of the targeted IONP produced 4- and 10-fold enhancement in photoacoustic signals in the tumor, compared to the tumor of the mice that received non-targeted IONP or control mice. The use of targeted nanoparticles allowed imaging of tumors located as deep as 3.1 cm beneath the normal tissues. Our study indicates the potential of the combination of photoacoustic tomography and receptor-targeted NIR830-ATF-IONP as a clinical tool that can provide improved specificity and sensitivity for breast cancer detection.

Abstract

This paper presents a strategy for immobilizing biomolecules onto a solid surface. We used the free thiol group directing method to immobilize Fab’ fragments to the MPTMS derived quartz substrate and results of X-ray photoelectron spectroscopic studies on the binding of MTPMS and Fab’ fragments on the quartz surfaces are reported. We also prepared a new type of immuno-labeled protein which was the rare earth element labeled antigen (human IgG) to recognize Fab’ fragment. Four characteristic peaks has been determined on the surface specifically bound human IgG labeled Tb 3+ .

Abstract

We present a simple method for rapid preparation of crystalline colloidal arrays (CCAs) by a strong electric field dialysis (SEFD). This method is based on rapid removing of ionic impurities in colloidal suspensions by applying a strong electric field. In a SEFD process, the negatively charged ions in colloidal suspensions are rapidly driven to the anode, the positively charged ones are rapidly driven to the cathode, and the colloidal particles are withheld in the dialysis tube. It was shown that the colloidal particles aggregated on the wall of the dialysis tube could block the SEFD process, which could be overcome by reversing the direction of the electric field. The purified colloidal particles can self-assemble into a crystalline colloidal array, which has an electrostatically stabilized three-dimensional periodic array of colloidal particles with a characteristic lattice spacing that can be varied by dilution. The reflection spectra show distinct peaks due to diffraction from CCAs. Atomic force microscopy (AFM) image illustrates the non-contacted ordering of the colloidal particles in the CCAs embedded in gels. This indicates the formation of high-quality single CCAs. Using a SEFD method, the preparation time of CCAs can be reduced. This new technique will greatly speed up the process of preparing polymerized crystalline colloidal arrays (PCCAs) into real-world application in the analytical field.

Abstract

Surface-enhanced Raman scattering (SERS) based on the doubleantibody sandwich format is reported for the determination of carcinoembryonic antigen. Ordered gold nanobowl arrays were fabricated and conjugated with anticarcinoembryonic as capturing substrates, and gold nanoshells, adsorbed with 4-mercaptobenzonic acid, were modified with anticarcinoembryonic antigen as labeling tags. After the carcinoembryonic antigen was captured on ordered gold nanobowl arrays, the labeling tags were bonded to the captured carcinoembryonic antigen. The interaction of SERS substrates (ordered gold nanobowl arrays) and SERS labels (gold nanoshells) showed high sensitivity and a low detection limit for carcinoembryonic antigen. The linear dynamic range of SERS for carcinoembryonic antigen was from 5 pg/mL to 100 ng/mL with a linear relationship between carcinoembryonic antigen concentration and SERS intensity. The detection limit was 1.73 pg/mL. SERS detection may be used for other cancer biomarkers and provides potential for the clinical diagnosis of cancer biomarkers. [ABSTRACT FROM AUTHOR]

Abstract

Using a biomedical sensor setup RIfS we have investigated the kinetic behavior of bovine serum albumin (BSA), human fibrinogen (FIb) and human IgG adsorbed onto surfaces of hydroxyapatite (HA) and polyurethane (PU) H50-50. According to the operation principle of RIfS and the molecular dimensions of three kinds of proteins, a formula to calculate the adsorbed layers of proteins onto the surface of HA and PU H50-50 has been introduced. The results show that the adsorbed layers of three kinds of proteins on the surface of HA are 1.0751 for IgG 0.9684 for BSA, 0.7464 for FIb and that of PU H50-50 are: 0.8199 for IgG 0.7964 for BSA, 0.6120 for FIb. It is shown that RIfS can perform a kinetic, real time and in situ analysis of plasma proteins adsorbed on a surface of biomaterials. From this study the potential application of RIfS as a new analytical tool in the evaluation of the biocompatibility of biomaterials was confirmed and the experiences of preparing suitable nanograde film from inorganic and organic biomaterials for a RIfS experiment were accumulated.

Conjugation of a HER2 antibody, Herceptin®, or a single chain fragment (ScFv) of antibody against epidermal growth factor receptor (ScFvEGFR) to PEO-b-PγMPS-coated IONPs resulted in HER2-targeted or EGFR-targeted IONPs (anti-HER2-IONPs or ScFvEGFR-IONPs). The anti-HER2-IONPs bound specifically to SK-BR-3, a HER2-overexpressing breast cancer cell line, but not to MDA-MB-231, a HER2-underexpressing cell line. On the other hand, the ScFvEGFR-IONPs showed strong reactivity with MDA-MB-231, an EGFR-positive human breast cancer cell line, but not with MDA-MB-453, an EGFR-negative human breast cancer cell line. Transmission electron microscopy revealed internalization of the receptor-targeted nanoparticles by the targeted cancer cells. In addition, both antibody-conjugated and non-antibody-conjugated IONPs showed reduced nonspecific uptake by RAW264.7 mouse macrophages in vitro. The developed IONPs showed a long blood circulation time (serum half-life 11.6 hours) in mice and low accumulation in both the liver and spleen.
