Understanding the factors that cause population divergence has long been of interest to marine biologists in their attempts to interpret the effect of humanmediated vectors. Broadcast-spawning species with limited dispersal capability are excellent candidates to measure the present-day patterns of genetic diversity. The tunicate Ciona intestinalis (Ascidiacea) is comprised of a complex of morphologically cryptic species that form vigorous aggregates in eutrophic habitats (harbors, gulfs and lagoons) where they can compete with the epibenthic community and cause biofouling problems. This study investigated biogeographic variability and migration patterns of C. intestinalis sp. A along Northeast Atlantic and Mediterranean coasts using microsatellite markers. Data presented here on 371 specimens collected from 17 populations reveal high genetic polymorphism, but with a deficit of heterozygote deficiency. Absence of evidence for isolation by distance suggests that the genetic patterns do not reflect the geographic distribution of sampled populations. Substantial gene flow and artificial potential for dispersal boost high levels of within-population genetic variability and prevent genetic differentiation within and between seas. A predominant eastward migration pattern was revealed by the data set, with very limited opportunity for C. intestinalis sp. A to travel westward. This directional movement indicates that other properties (e.g. habitat quality, genetic traits, mating system, life cycle) may cause adaptive divergence at a large biogeographic scale. Spatial and temporal heterogeneity in flowering occur in many plant species with abiotic pollination and may confer fitness advantages through mechanisms such as predator satiation or pollination efficiency. Environmental factors such as light quality or quantity and temperature play an important role in inducing synchronization on wide geographic scales. On a smaller geographic scale, external factors such as resource availability and herbivory are theorized to trigger flowering, while genetic factors may also play an important role., In this study, we assessed the importance of ecological and genetic factors in shaping seascape-level spatial heterogeneity in flowering of the seagrass Posidonia oceanica. By investigating spatially close sites (<20 km) with similar seascape configurations and depth, we assume that major environmental drivers (temperature and light) were equivalent., We assessed four ecological factors (productivity, leaf nitrogen and carbon content and herbivory) and three genetic factors (heterozygosity, relatedness and clonality) to assess three hypotheses for synchronized flowering in P. oceanica: (i) clone synchronization (internal clock hypothesis), (ii) variation in nutrient availability, potentially caused by spatial heterogeneity in herbivory rates or nutrient translocation via clonal integration (resource budget hypothesis) or (iii) kin selection and sibling synchronization., Internal relatedness and heterozygosity had a significant positive effect on the abundance of flowers. Moreover, productivity and genotypic richness (clonality) were negatively associated with flower density, although at a lower level of significance. In addition, we found that clones were almost exclusively shared among mass-flowering patches and patches without mass-flowering, respectively., Synthesis. The results shed new light on seagrass flowering patterns and on the mechanisms of flower synchronization at the patch level within a wider spatial scale. We found support for the kin selection hypothesis and indirect evidence for the resource budget hypothesis. Thus, a combination of mainly genetic but also ecological factors causes the observed heterogeneous flowering patterns in Posidonia oceanica seascapes. In addition, we found a strong positive relationship between the number of flowers and heterozygosity, adding evidence to the controversial association between heterozygosity and fitness when a limited number of loci are used. The intensification of anomalous events of seawater warming and the co-occurrence with local anthropogenic stressors are threatening coastal marine habitats, including seagrasses, which form extensive underwater meadows. Eutrophication highly affects coastal environments, potentially summing up to the widespread effects of global climate changes. In the present study, we investigated for the first time in seagrasses, the transcriptional response of different plant organs (i.e., leaf and shoot apical meristem, SAM) of the Mediterranean seagrass Posidonia oceanica growing in environments with a different history of nutrient enrichment. To this end, a mesocosm experiment exposing plants to single (nutrient enrichment or temperature increase) and multiple stressors (nutrient enrichment plus temperature increase), was performed. Results revealed a differential transcriptome regulation of plants under single and multiple stressors, showing an organ-specific sensitivity depending on plants' origin. While leaf tissues were more responsive to nutrient stress, SAM revealed a higher sensitivity to temperature treatments, especially in plants already impacted in their native environment. The exposure to stress conditions induced the modulation of different biological processes. Plants living in an oligotrophic environment were more responsive to nutrients compared to plants from a eutrophic environment. Evidences that epigenetic mechanisms were involved in the regulation of transcriptional reprogramming were also observed in both plants' organs. These results represent a further step in the comprehension of seagrass response to abiotic stressors pointing out the importance of local pressures in a global warming scenario. On the basis of morphological and molecular studies, we identified the Australian endemic green alga known as Caulerpa distichophylla along the coasts of Sicily (Italy, Mediterranean Sea). The slender Caulerpa previously reported as C. taxifolia from southeastern Turkey (Gulf of Iskenderun) also belongs to C. distichophylla. Morphologically, C. distichophylla clearly differs from C. taxifolia in its slender thallus and the lack of large rhizoidal pillars. However, genetic data do not provide undisputed evidence that the species are distinct. Sequences of the tufA cpDNA gene and of the cp 16S rDNA intron-2 sequences separated the two taxa by only one single nucleotide mutation, whereas ITS rDNA sequences did not clearly distinguish them. The new combination Caulerpa taxifolia var. distichophylla is therefore proposed. Western and eastern Mediterranean populations of C. taxifolia var. distichophylla are probably the result of introduction events from southwestern Australia. Although the vector of primary introductions remains unknown (aquarium trade or shipping), maritime traffic appears to be the most likely vector of secondary dispersal. C. taxifolia var. distichophylla is closely related to C. taxifolia, hence interbreeding with the other C. taxifolia strains in the Mediterranean Sea might be expected to occur. Abstract: Aquatic organisms are constantly exposed to both physical (e.g. temperature and salinity variations) and chemical (e.g. endocrine disruptor chemicals, heavy metals, hydrocarbons, diatom toxins, and other toxicants) stressors which they react to by activating a series of defense mechanisms. This paper reviews the literature on the defense systems, including detoxification enzymes and proteins (e.g. glutathione S-transferases, heat shock proteins, superoxide dismutase and catalase), studied in copepods at the molecular level. The data indicate high inter- and intra-species variability in copepod response, depending on the type of stressor tested, the concentration and exposure time, and the enzyme isoform studied. Ongoing -omics approaches will allow the identification of new genes which will give a more comprehensive overview of how copepods respond to specific stressors in laboratory and/or field conditions and the effects of these responses on higher trophic levels. Atoll-like structures of the endemic Mediterranean seagrass Posidonia oceanica were encountered in the innermost area of the Stagnone di Marsala, a semi-enclosed coastal lagoon along the western coasts of Sicily. The area is characterized by limited water exchange with the open sea and by a marked seasonal variation of water salinity and temperature, reaching beyond the theoretical tolerance limits of the species. In the present study we determined the genetic composition of the atoll-like structures, as well as the growth performance and flowering rate of these stands. We also assessed whether and to what extent the atoll-like structures are genetically isolated from plants growing in meadows outside the lagoon. For this purpose we utilized 13 microsatellite markers to genotype single shoots sampled inside and outside the lagoon. Lepidochronological analyses were performed on the same shoots to determine the annual rhizome growth rate, the number of leaves and the inflorescences formed as an estimate of growth- and reproductive performance over the years. The innermost area of the lagoon showed a lower number of alleles, a lower percentage of polymorphic loci, a lower clonal diversity, but higher heterozygosis excess with respect to the other areas analysed. Spatial autocorrelation was here significant, up to slightly below 300 m. Shoots collected in the atolls exhibited a 25% lower vertical growth rate and 16% lower leaf formation in comparison to those in open-sea meadows. No flowering events were recorded during 24 years of investigation, whereas inflorescences were observed frequently in meadows outside the lagoon. Results from Fst and factorial correspondence analysis confirmed the expected genetic isolation of the confined atolls with respect to the meadows outside the lagoon and revealed limited gene flow within the lagoon itself. Apparently, the enclosed system of the Stagnone lagoon is genetically isolated, with a possible selection of genotypes adapted to persistent stressful conditions, consistent with reduced growth and lack of flowering events. Seawater warming and increased incidence of marine heatwaves (MHW) are threatening the integrity of coastal marine habitats including seagrasses, which are particularly vulnerable to climate changes. Novel stress tolerance-enhancing strategies, including thermo-priming, have been extensively applied in terrestrial plants for enhancing resilience capacity under the re-occurrence of a stress event. We applied, for the first time in seedlings of the Mediterranean seagrass Posidonia oceanica , a thermo-priming treatment through the exposure to a simulated warming event. We analyzed the photo-physiological and growth performance of primed and non-primed seedlings, and the gene expression responses of selected genes (i.e. stress-, photosynthesis- and epigenetic-related genes). Results revealed that during the re-occurring stress event, primed seedlings performed better than unprimed showing unaltered photo-physiology supported by high expression levels of genes related to stress response, photosynthesis, and epigenetic modifications. The red alga, Asparagopsis taxiformis, has recently expanded its distribution range into the Western Mediterranean Sea, and populations have now even been found on the Portuguese South coast. All Western Mediterranean populations belong to a single mitochondrial cryptic lineage (referred to as lineage 2 in earlier studies) and probably result from a recent invasion of Indo-Pacific origin. Here we investigate fine-scale population genetic diversity and structure within and among Mediterranean populations of lineage 2 using eight microsatellite loci and compare the obtained patterns with those observed in a Californian population of the same lineage. To generate an appropriate analytical method suitable to the polyploid status of this species, we score microsatellite loci as in a dominant marker system. Thereafter, we produce robust descriptors inferred from frequencies of both microsatellite alleles and multilocus genotypes. Populations from California and the Mediterranean Sea differ considerably in their levels of genetic diversity. In the Mediterranean, populations reproduce predominantly sexually and exhibit high levels of genotypic variation, suggestive either of multiple introductions or of a single introduction by a genetically diverse and large group of individuals. Bayesian clustering revealed one or possibly two weakly supported panmictic subpopulations, indicative of extensive admixture. The expansion of this lineage is rapid, possibly due to the absence of eco–physiological barriers to gene flow throughout its invasive trajectory. [ABSTRACT FROM AUTHOR]

The tropical-subtropical red seaweed Asparagopsis Montagne (Bonnemaisoniales) constitutes the haploid, gametophytic phase in a heteromorphic diplo-haplontic life cycle. The diploid tetrasporophyte is known as the ' Falkenbergia ' stage. The genus contains two species, A. armata and A. taxiformis , both present in the Mediterranean Sea where they are regarded as introduced. A. armata is morphologically distinct from A. taxiformis in that it possesses long stolons bearing harpoon-like hooks. The seemingly morphologically identical ' Falkenbergia ' stages of the two Asparagopsis species and phenotypic variation within these species have caused taxonomic confusion. We defined species boundaries in the Mediterranean Sea by inferring phylogenies from sequence data from a variable region in the nuclear LSU rDNA gene, the plastid RuBisCo spacer, and the mitochondrial cox 2 - 3 spacer of specimens from the Mediterranean, western Europe and the Canary Islands. Results indicate that A. armata and its ' Falkenbergia ' tetrasporophyte are genetically distinct from A. taxiformis and its ' Falkenbergia ' phase. No phylogeographic structure was detected within A. armata, whereas A. taxiformis seems to consist of at least two genetically distinct but morphologically cryptic species, an Atlantic one (from the Canary Islands) and a Mediterranean one. Hypothetical distribution patterns of the two species as reconstructed from critical temperature limits to growth, survival and reproduction and from the summer and winter isotherms in the Mediterranean Sea agree with the actual Mediterranean distribution patterns as gleaned from our data. A new marine microalga from the Mediterranean Sea, Crustomastix stigmatica Zingone, is investigated by means of LM, SEM, TEM, and pigment and molecular analyses (nuclear-encoded small subunit [SSU] rDNA and plastid-encoded rbcL). Pigment and molecular information is also provided for the related species Dolichomastix tenuilepis Throndsen et Zingone. Crustomastix stigmatica has a bean-shaped cell body 3-5 µm long and 1.5-2.8 µm wide, with two flagella four to five times the body length. The single chloroplast is pale yellow-green, cup-shaped, and lacks a pyrenoid. A small bright yellow stigma is located in the mid-dorsal part of the cell under the chloroplast membrane. An additional accumulation of osmiophilic globules is at times seen in a chloroplast lobe. Cells lack flat scales, whereas three different types of hair-like scales are present on the flagella. The main pigments of C. stigmatica are those typical of Mamiellales, though siphonein/siphonaxanthin replaces prasinoxanthin and uriolide is absent. The pigment pool of D. tenuilepis is more similar to that of Micromonas pusilla (Butcher) Manton et Parke and of other Mamiellales. The nuclear SSU rDNA phylogeny shows that the inclusion of C. stigmatica and D. tenuilepis in the Mamiellales retains monophyly for the order. The two species form a distinct clade, which is sister to a clade including all the other Mamiellales. Results of rbcL analyses failed to provide phylogenetic information at both the order and species level. No unique morphological or pigment characteristics circumscribe the mamiellalean clade as a whole nor its two daughter clades. [ABSTRACT FROM AUTHOR]

Marine Heat Waves (MHWs) occurrence has been increasing in the Mediterranean Sea. The effects of field simulated MHWs of different intensity (medium and high temperature) on the transcriptome expression of the endemic seagrass Posidonia oceanica were evaluated considering different origins of the plant. The aim of the study was reached through a common garden transplant experiment in the North-west of Sardinia (Italy), where two P. oceanica meadows characterized by different thermal regimes (cold and warm) were chosen. MHWs were simulated in front of a power plant, that creates a natural laboratory by releasing warm water in the sea. Differential gene expression and GO enrichment analyses highlighted differences in the transcriptomic profiles of plants from cold and warm environments suggesting that the MHWs induced different levels of stress due to different tolerance to the heat event. Plants from both origins activated processes to achieve protein homeostasis, but only cold plants activated an antioxidant defense and altered sugar metabolism, both indicators of heat stress. Within plants of the same origin, a different response to MHW intensity was also detected: while warm plants showed the most complex response at high temperature rather than at medium temperature, cold plants seemed to better cope with the medium temperature intensity rather than with high temperature.
