Abstract

Invasive fungal disease (IFD) can be caused by a range of pathogens. Conventional diagnosis has the capacity to detect most causes of IFD, but poor performance limits impact. The introduction of non-culture diagnostics, including the detection of (1-3)-β-D-Glucan (BDG), has shown promising performance for the detection of IFD in variety of clinical settings. Recently, the Dynamiker® Fungus (1-3)-β-D-Glucan assay (D-BDG) was released as an IFD diagnostic test. This article describes an evaluation of the D-BDG assay for the diagnosis of invasive aspergillosis (IA), invasive candidiasis (IC) and Pneumocystis pneumonia (PCP) across several high-risk patient cohorts and provides comparative data with the Associates of Cape Cod Fungitell® and BioRad Platelia™ Aspergillus Ag (GM) assays. Abstract

The plethora of alternative food labels emerging in the marketplace reflects consumer interest in informed and sustainable purchasing. However, consumers' preferences vary with respect to the sustainability metrics represented by labeling programs. The goal of this project was to characterize public university students' perceptions of product parameters commonly represented by food labels for produce (e.g., cost, taste, certified organic, locally grown). A consumer survey (n = 338) was conducted at two university dining halls, located in close proximity to major residence hall communities which house approximately 2600 students. Culinary, health and cost attributes (taste, nutritional value, price and appearance) were ranked as more influential in determining purchasing decisions than sustainability attributes related to production and sourcing. While sustainability values were not as influential in driving purchasing behaviors as compared with product attributes, they were important to approximately 50% of the survey respondents. By identifying the sustainability values of students and their willingness to pay more for specific types of sustainable food, results from this study can inform efforts to align priorities of campus dining services with the values of their student patrons, as well as identify educational opportunities around agriculture and food production. Abstract

The purpose of this study was to explore professional therapists’ experiences of personal therapy in an attempt to understand its purpose. The study applied a qualitative approach combining phenomenological and hermeneutic perspectives through the model of IPA (Interpretative Phenomenological Analysis). Six psychodynamically oriented therapists were interviewed, using one broad and open question. The transcribed interviews were closely read and analyzed, and in the end four main themes about the use of personal therapy stood out from the material: the meaning of the therapeutic room, the importance of the therapist, personal growth and professional development. Our study showed that the persons asked about their personal therapy had a positive experience from it. They acknowledged that they had learned a great deal from it and that some of the things learned were hard to achieve in any other way. They described an “inside experience” of, for example, what it feels like to sit in the patient´s chair. Our conclusion from this study was that personal therapy is an important part of the education to become a trained therapist.

162. Notch3 Signaling Promotes Adhesion and Tumor Progression in a Murine Epithelial Ovarian Cancer Model

Abstract

Ovarian cancer is the 5th leading cause of cancer death in women in the United States and is the most fatal gynecological malignancy. High grade serous ovarian cancer (HGSC) is the most common and deadly type of ovarian cancer largely due to the rapid metastasis throughout the peritoneum (abdominal cavity wall and organ lining). Metastatic spread of ovarian cancer usually occurs before diagnosis and can lead to bowel obstruction, organ failure, ascites, cachexia, infection and sepsis, and pulmonary embolism all causing death. Current methods to detect early stage ovarian cancer do not increase overall survival. A better understanding of the metastatic ability of ovarian cancers and the mechanism of cancer cell dissemination are critical to the development of new treatments for this devastating disease. In particular, investigation of pathways that affect early metastasis may indicate treatments that will lower disease burden and may suggest biomarkers of recurrent and/or chemotherapy resistant disease. Notch3 expression correlates with worse prognosis, chemotherapy resistance, and increased tumorigenic cell behaviors in HGSC. Here, we demonstrate that Notch3 acts to promote early stages of metastasis in a model of HGSC using the murine ID8 IP2 ovarian surface epithelial cell line. ID8 IP2 cells have little to no endogenous Notch3 expression and model metastatic disease when introduced intraperitoneally. We investigated the role of Notch3 by ectopically expressing the intracellular domain of murine Notch3 to induce constitutive Notch3 signaling in ID8 IP2 cells and verified Notch signal activation by target gene assessment. Induction of Notch3 signaling in ID8 IP2 reduced survival and accelerated disease burden, as measured by ascites accumulation, after intraperitoneal introduction of cells into nude mice. We interrogated downstream targets in Notch3 activated cells by RNA-Seq and found that Notch3 induced a significant enrichment of adhesion and extracellular matrix pathways. Notch3 active cells showed increased ITGA1 expression and increased adhesion on collagens I and IV in vitro, suggesting that increased adhesion to collagen-rich peritoneal surfaces drives the observed increase in tumor burden. Notch3 active cells showed reduced migration on surfaces coated with multiple types of extracellular matrix and no detectable increase in invasion through extracellular matrix, indicating that Notch3 effects may be specific to the initial adhesion of tumor cells and not the later stages of metastasis. These results demonstrate that Notch3 upregulates the expression of specific adhesion genes in ovarian cancer cells and this promotes increased attachment to the collagen-rich extracellular matrix. The implications of this study are that oncogenic Notch signal activation, as documented in human disease, may promote dissemination and metastasis of primary and/or recurrent HGSC by increasing attachment to the peritoneal lining.

Abstract

Recent research has demonstrated that the emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), uses visual and chemical cues when locating potential mates and host trees. Here, we present the first evidence of the production of possible acoustic cues generated by adult males and females. We demonstrate for the first time that simultaneous wing opening and head 'pumping' in both males and females produce specific and distinct sounds. Acoustic recordings demonstrate that these sounds consist of a phrase comprising a 'click' generated by wing opening, followed by two separate chirps corresponding to the outward and subsequent inward movement of the head. Mean primary and secondary chirp durations were 0.332 ± 0.038 and 0.414 ± 0.046 s for males and 0.414 ± 0.046 and 0.389 ± 0.032 s for females, respectively. Overall mean phrase durations were 0.905 ± 0.114 s in males and 1.133 ± 0.133 s in females. Mean maximum phrase frequencies were 8 706.1 ± 141.1 and 7 885.0 ± 499.2 Hz in males and females, respectively. There were no significant differences in sound parameters between males and females. To determine whether these events were related to mating pair formation, we observed the occurrence of these events in adults paired with either another male or female and lone adults, and observed the number of mating pairs in copula formed during or shortly after sound production by either of the mating party. The mean number of sound‐generating events per min was significantly greater in males paired with females (0.75 ± 0.15) and/or other males (0.55 ± 0.23) vs. unpaired males (0.14 ± 0.06). Conversely, the mean number of sound‐producing events for lone females (1.4 ± 0.2) was significantly greater than for females paired with either males (0.49 ± 0.18) or other females (0.38 ± 0.10). No mating pairs in copula were formed during any of our observations. Scanning electron microscopy images showed the presence of possible pars stridens and plectrum structures on the ventral surfaces of the gula and pronotum in both males and females. Furthermore, we observed that the proportion of males and females exhibiting these specific behaviors were significantly greater under both halogen and incandescent light than under darkness, signifying the importance of light source or type on the production of acoustic signals. Although we did not find evidence of these behaviors being related to the courtship sequence, our results indicate the presence of a previously undescribed sound‐generation behavior in A. planipennis that is significantly influenced by light conditions. A trap that utilizes attractive plant and pheromone volatiles to detect emerald ash borer (EAB) (Coleoptera: Buprestidae) adults is the principal method for monitoring EAB populations. This trap exploits visual and chemical cues used by EAB adults to locate host trees and one another. An additional, yet undescribed, sensory modality in EAB is the role of acoustic cues produced by adults. The Dynamiker® Fungus (1-3)-β-D-Glucan Assay (D-BDG) has recently become available in the Western Hemisphere for the diagnosis of invasive fungal disease (IFD). Evaluations of its performance for Pneumocystis pneumonia (PcP) are limited. A retrospective evaluation of D-BDG diagnosis of PcP was performed (23 PcP cases and 23 controls). Sensitivity and specificity were 87% and 70%, respectively, reducing the positivity threshold to 45 pg/ml increased sensitivity (96%), whereas a threshold of 300 pg/ml increased specificity (96%). The performance of D-BDG for the detection of PcP is comparable to other IFD, but sensitivity is below that required to confidently exclude PcP. [ABSTRACT FROM AUTHOR]

Abstract

Now is not the time to increase the strength of copyright law. Copyright law is facing a crisis of legitimacy: consumers increasingly appear to doubt its moral weight. To a large extent, this can be traced to the fact that Australian consumers do not believe they are being treated fairly by (predominantly US-based) copyright producers and distributors. Compared to their overseas peers, Australian consumers pay much more for access to books, films, television, and computer games, and are often subjected to long delays before material is available in Australia. Our research shows that this perceived unfairness increases the willingness of Australian consumers to seek out alternative distribution channels. Put simply, the failure of content distributors to meet consumer demand in Australia is a leading factor in copyright infringement. This submission argues that the best strategy to reduce copyright infringement in Australia, at the present time, is for distributors to focus on providing timely, affordable, convenient and fair access to copyright goods. Until this is done, the prevalence of copyright infringement in Australia should be seen as essentially a market problem, rather than a legal one. The Australian Government, meanwhile, should address the recommendations of the IT Pricing Report as a matter a priority. As a first step, the Government should urgently consider repealing the IP exception to competition law in s 51(3), as recommended by the Ergas committee, the IT Pricing report, and the ALRC. This change alone may go a long way to enhancing the efficiency of the copyright market in Australia.

Abstract

Most endometrial cancers can be classified histologically as endometrioid, serous, or clear cell. Non-endometrioid endometrial cancers (NEECs; serous and clear cell) are the most clinically aggressive of the three major histotypes and are characterized by aneuploidy, a feature of chromosome instability. The genetic alterations that underlie chromosome instability in endometrial cancer are poorly understood. In the present study, we used Sanger sequencing to search for nucleotide variants in the coding exons and splice junctions of 21 candidate chromosome instability genes, including 19 genes implicated in sister chromatid cohesion, from 24 primary, microsatellite-stable NEECs. Somatic mutations were verified by sequencing matched normal DNAs. We subsequently resequenced mutated genes from 41 additional NEECs as well as 42 endometrioid ECs (EECs). We uncovered nonsynonymous somatic mutations in ESCO1, CHTF18, and MRE11A in, respectively, 3.7% (4 of 107), 1.9% (2 of 107), and 1.9% (2 of 107) of endometrial tumors. Overall, 7.7% (5 of 65) of NEECs and 2.4% (1 of 42) of EECs had somatically mutated one or more of the three genes. A subset of mutations are predicted to impact protein function. The co-occurrence of somatic mutations in ESCO1 and CHTF18 was statistically significant (P = 0.0011, two-tailed Fisher's exact test). This is the first report of somatic mutations within ESCO1 and CHTF18 in endometrial tumors and of MRE11A mutations in microsatellite-stable endometrial tumors. Our findings warrant future studies to determine whether these mutations are driver events that contribute to the pathogenesis of endometrial cancer.
