Abstract

International audience; Propagation-based imaging or inline holography in combination with computed tomography (holotomography) is a versatile tool to access a sample's three-dimensional (3D) micro or nano structure. However, the phase retrieval step needed prior to tomographic reconstruction can be challenging especially for strongly absorbing and refracting samples. Near-field ptychography is a recently developed phase imaging method that has been proven to overcome this hurdle in projection data. In this work we extend near-field ptychography to three dimensions and we show that, in combination with tomography, it can access the nano structure of a solid oxide fuel cell (SOFC). The quality of the resulting tomographic data and the structural properties of the anode extracted from this volume were compared to previous results obtained with holotomography. This work highlights the potential of 3D near-field ptychography for reliable and detailed investigations of samples at the nanometer scale, with important applications in materials and life sciences among others. Byline: J. Srnik, F. Pfeiffer The dynamics of continuously variable transmissions (CVT) is characterised by the discrete structure of the chain, which causes the polygonal action. Hence the mechanical model contains each chain link and pulley as a separate body. While a chain link is part of a strand, only joint forces act on the chain links, whereas, in the contact situation, additional impulsive and frictional forces in the contact zones, between the chain links rocker pins and the tapered surfaces of the pulleys, have to be taken into account. In order to calculate the contact forces, Coulombs friction law as well as a time saving continuous approximation are applied. Due to the fact that the deformation of the pulleys has a decisive influence on the dynamics of the complete chain drive, the pulleys are described as an elastic multibody system. Simulation results show the influence of the cone pulleys deformation on the power transmission. A comparison of calculated and measured results verifies the model presented.

167. A model based approach for the optimisation of gearshifting in automatic transmissions

Pfeiffer In this paper, an approach for the optimisation of gear shift operations in automatic transmissions based on a verified mechanical model for a powertrain is presented. The control data for the clutch pressure in the gearbox and the engine load-reduction are used as optimisation parameters. The objective of the optimisation is to determine an optimal set of control data, so that the performance of the automatic transmission with respect to passengers comfort and gear shift duration is improved. Two sets of optimisation criteria are developed. The first one consists of three different cost functions for the description of the passengers comfort, whereas the second one contains two cost functions for the evaluation for the evaluation of the operating life of the shift elements. The nonlinear optimisation problem is solved with a sequential quadratic programming algorithm. The results show a significant improvement in terms of the optimisation criteria for different load cases.

169. Fiber characteristics of qiviut and guard hair from wild muskoxen (Ovibos moschatus)

Abstract

In response to increasing commercial interest and the high market value of qiviut (the downy underwool of the muskox), we have employed standards and measurements used in the wool and cashmere industries to describe qiviut fiber characteristics. Fleece samples (qiviut with guard hair) were shaved from the midshoulder of 299 wild muskox hides of known sex and age (1, 2, 3, and 4+ yr) during the Banks Island, Canada, muskox harvest in November 1997. Samples were analyzed for fiber diameter distribution of raw fiber and qiviut, scoured and qiviut yields, and lengths of guard hair and qiviut fiber. We found a sex x age interaction for average fiber diameter (AFD) in raw fiber (P = 0.002) and qiviut (P [is less than] 0.001) only. Adult males had significantly coarser AFD than females (21.5 [micro]m, males vs 20.1 [micro]m, females and 18.2 [micro]m, males vs 17.5 [micro]m, females) for raw fiber and qiviut, respectively. Qiviut AFD from yearlings was 1.7 [micro]m finer than the AFD of adult qiviut. Fiber diameter distribution (SD) decreased with age in the raw sample (P [is less than] 0.003) and qiviut (P [is less than] 0.001) and qiviut SD was greater (P [is less than] 0.001) in males than in females. Qiviut theoretical yield (% mass of fibers [is less than or equal to] 30 [micro]m) increased (P [is less than] 0.001) with age, and females had higher theoretical yields than males (P [is less than] 0.001). Scoured yield did not vary between sexes in any age class and averaged 93.3%. Qiviut staple length did not differ with either age or sex. In summary, differences between the sexes were small up to the 3rd yr, and these differences were not likely to be of commercial importance. However, considering that AFD is a primary commercial criterion of value, AFD changes from 16.5 [micro]m in yearlings to 18.2 [micro]m in adults and from 17.5 [micro]m in adult females to 18.2 [micro]m in adult males would be expected to result in significant differences in commercial value. The first practical application of X-rays, more than 100 years ago, was for biomedical imaging. Although not central to the initial development of 3 rd generation synchrotron light sources, imaging techniques have today established themselves as a core element of the research program at all major light sources, including the ESRF, where X-ray imaging has been identified as one of five main themes for its Upgrade Programme [1].

Abstract

We report advances and complementary results concerning a recently developed method for high-sensitivity grating-based hard x-ray phase tomography. We demonstrate how the soft tissue sensitivity of the technique can be used to obtain in-vitro tomographic images of a tumor bearing rat brain specimen, without use of contrast agents. In particular, we demonstrate that brain tumors and the white and gray brain matter structure in a rat's cerebellum can be resolved by this approach. The findings are potentially interesting from a clinical point of view, since a similar approach using three transmission gratings can be implemented with more readily available x-ray sources, such as standard x-ray tubes. Moreover, open the results the way to in-vivo experiments in the near future.

Abstract

We report numerical and experimental results demonstrating accurate region-of-interest computed tomography (CT) reconstruction based on differential phase-contrast projection (DPC) images. The approach removes the constraint of covering the entire sample within the field of view of the image detector. Particularly for biomedical applications, the presented DPC-CT region-of-interest approach will allow for the visualization of previously inaccessible details deep inside an entire animal or organ. We envisage that this development will also be of interest for potential future clinical applications, because grating-based DPC-CT can be implemented with standard x-ray tube sources.

Abstract

Nondestructive microscale investigation of objects is an invaluable tool in life and materials sciences. Currently, such investigation is mainly performed with X-ray laboratory systems, which are based on absorption-contrast imaging and cannot access the information carried by the phase of the X-ray waves. The phase signal is, nevertheless, of great value in X-ray imaging as it is complementary to the absorption information and in general more sensitive to visualize features with small density differences. Synchrotron facilities, which deliver a beam of high brilliance and high coherence, provide the ideal condition to develop such advanced phase-sensitive methods, but their access is limited. Here we show how a small modification of a laboratory setup yields simultaneously quantitative and 3D absorption and phase images of the object. This single-shot method is based on correlation of X-ray near-field speckles and represents a significant broadening of the capabilities of laboratory- based X-ray tomography., QC 20220215

Abstract

At the time of its first publication, halomucin from Haloquadratum walsbyi strain HBSQ001 was the largest archaeal protein known (9159 aa). It has a predicted signal sequence, making it likely to be an extracellular or secreted protein. Best BLAST matches were found to be mammalian mucins that protect tissues to dehydration and chemical stress. It was hypothesized that halomucin participates in protection against desiccation by retaining water in a hull around the halophilic organisms that live at the limits of water activity. We visualized Haloquadratum cells by staining their intracellular polyhydroxybutyrate granules using Nile Blue. Halomucin was stained by immunofluorescence with antibodies generated against synthetic peptides derived from the halomucin amino acid sequence. Polyhydroxybutyrate stained cells were reconstructed in 3D which highlights not only the highly regular square shape but also the extreme flatness of Haloquadratum. Double-staining proves halomucin to be extracellular but to be only loosely associated to cells in agreement with its hypothesized function.

Abstract

The Protein Information Resource (PIR; http://www-nbrf.georgetown. edu/pir/) supports research on molecular evolution, functional genomics, and computational biology by maintaining a comprehensive, non-redundant, well-organized and freely available protein sequence database. Since 1988 the database has been maintained collaboratively by PIR-International, an international association of data collection centers cooperating to develop this resource during a period of explosive growth in new sequence data and new computer technologies. The PIR Protein Sequence Database entries are classified into superfamilies, families and homology domains, for which sequence alignments are available. Full-scale family classification supports comparative genomics research, aids sequence annotation, assists database organization and improves database integrity. The PIR WWW server supports direct on-line sequence similarity searches, information retrieval, and knowledge discovery by providing the Protein Sequence Database and other supplementary databases. Sequence entries are extensively cross-referenced and hypertext-linked to major nucleic acid, literature, genome, structure, sequence alignment and family databases. The weekly release of the Protein Sequence Database can be accessed through the PIR Web site. The quarterly release of the database is freely available from our anonymous FTP server and is also available on CD-ROM with the accompanying ATLAS database search program.

197. Metformine bij patiënten met diabetes mellitus met hartfalen of chronische nierinsufficiëntie

Abstract

Metformine is een van de meest gebruikte orale antidiabetische middelen. Naast zijn antidiabetische werking heeft het als voordelen dat het in tegenstelling tot de andere orale producten gewichtsneutraal is, een verbetering geeft van de cardiovasculaire mortaliteit en een verwaarloosbaar risico inhoudt op het ontwikkelen van hypoglykemie. Het gebruik van metformine wordt echter overschaduwd door een zeer zeldzame, maar levensgevaarlijke bijwerking: het ontwikkelen van lactaatacidose. Dit risico heeft ertoe geleid dat men alle situaties die voorbeschikken tot het ontwikkelen van lactaatacidose beschouwt als een tegenaanwijzing voor metformine. Hiertoe behoren hartfalen, leverfalen en chronisch obstructief longlijden (COPD), maar ook chronische nierinsufficiëntie aangezien metformine uitsluitend renaal geklaard wordt. Wanneer al deze tegenaanwijzingen strikt worden nageleefd, zouden heel veel patiënten met diabetes mellitus (DM) niet meer in aanmerking komen voor het gebruik van metformine en de vele voordelen niet meer kunnen genieten. Meer en meer worden sommige van deze tegenaanwijzingen voor metformine in vraag gesteld. De laatste jaren werd er dan ook veel onderzoek verricht naar het gebruik van metformine bij patiënten met de klassieke tegenaanwijzingen, zoals hartfalen en chronische nierinsufficiëntie. Dit artikel geeft een overzicht van deze recente inzichten en beschrijft specifiek het gebruik van metformine bij patiënten met DM met hartfalen en chronische nierinsufficiëntie.
