161. Fractionnement et caractérisation des lixiviats de centres d'enfouissement technique de déchets ménagers: intérêt de la chromatographie liquide haute performance sur le gel d'exclusion stérique

Abstract

L'ultrafiltration et la chromatographie d'exclusion stérique haute performance sont utilisées pour la séparation et la caractérisation des composés organiques présents dans les lixiviats de centres d'enfouissement technique de déchets ménagers. Le fractionnement de la matière organique est obtenu sur des colonnes type TSK PW, en élution eau pH 4 et eau-méthanol. La spectroscopie en UV-visible et en fluorescence, un détecteur évaporatif à diffusion de lumière sont utilisés pour la caractérisation des fractions. Cette méthode rapide de séparation associée à une multidétection permet une mise en évidence, dans les fractions issues de l'ultrafiltration, de composés organiques caractéristiques. Dans la fraction de poids moléculaires inférieurs à 1000 Daltons, trois familles sont détectées. Les substances humiques et les protéines sont les principaux groupes présents dans la fraction de poids moléculaires supérieurs à 10000 Daltons., Landfill leachates represent a source of organic pollution characterized by an important organic load, with high chemical oxygen demand in recent sanitary landfills and some organic compounds refractory to biodegradation. Several researchers have examined the organic matter in these landfill leachates. In addition to measuring parameters such as chemical and biological oxygen demands (COD and BOD) and UV-absorbance, different analytical techniques were applied: gas chromatography with flame ionization or mass detection; high performance liquid chromatography; infrared spectroscopy; nuclear magnetic resonance spectrometry; and elementary analysis. Raw leachates or samples after fractionation on Sephadex gel were characterized by ultrafiltration or adsorption on XAD resins.Conclusions from these earlier studies were as follows: - physico-chemical properties of leachates revealed not only a high organic pollution but also diversity and variability according to the age of the sanitary landfill and the climatic conditions; - gel permeation chromatography and ultrafiltration revealed two main fractions in the leachates: one with molecular weights below 1000 Daltons (Da), and another with molecular weights above 5000-10000 Da; - infrared spectroscopy and nuclear magnetic resonance spectrometry showed functional groups present in the humic and fulvic acid fractions of natural organic matter; - a varying number of peaks detected by gas chromatography with flame ionization or mass detection proved the complexity of the matrix. Few compounds were identified and quantified, with the exception of fatty acids. Moreover this technique was only applicable to molecules with low molecular weight.The purpose of the present work was to develop a new method of fractionation of organic matter in landfill leachates and to study their characterization and treatment biodegradation. 183. Structural and Dynamic Approach of Early Hydration Steps in Erodable Polymers by ATR−FTIR and Fluorescence Spectroscopies

Abstract

The understanding of mechanisms involved in the controlled release of active molecules from the polymer matrix requires the study of hydration phenomenon. Fluorescence and attenuated total reflection Fourier transform infrared (ATR−FTIR) spectroscopies were used to analyze the earlier steps of hydration in acrylic copolymers. The effect of hydration on the anisotropy of the PRODAN fluorescent probe showed a change in the dynamics of the macromolecular chains. By using ATR−FTIR spectroscopy, the structure of dissolved water in polymers was studied. The evolution of this structure was performed as hydration proceeds by band decomposition of the νOH vibration on the basis of the four-state model which allows discriminating bound from free molecules:  in the “early” stages of hydration, the water network appears drastically perturbed as compared to that of bulk pure water, this perturbation vanishing in the “later” stages of hydration. Besides, this study reveals that the polymer carbonyl groups constitutes the major water binding sites as it was deduced from their progressive engagement in H bonds in the time course of hydration. The rate and extent of this process was observed to be correlated with the hydrophobicity of polymers. The present findings, obtained on similar time scales from noninvasive techniques, indicate that both dynamic and structural consequences of hydration are correlated in most of the polymers here studied.

186. Small bowel obstruction: role and contribution of sonography

Abstract

The purpose of this study was to determine whether sonography provides additional clinical information in patients suspected of small bowel (SB) obstruction. During a period of 30 months, in a prospective setting, we evaluated with sonography 123 patients suspected of SB obstruction. Sonographic examinations of the entire abdomen were performed with state-of-the-art, real-time, grey-scale equipment. Fourteen patients were labelled ’gassy' and no added information was provided following abdominal ultrasound. Sonography confirmed the SB obstruction in 82 cases with 5 false positives, resulting in a specificity of 82.1 %. Sonographic examinations were negative in 27 cases with 4 false negatives and a sensitivity of 95 %. The accuracy was 91.7 % when the ’gassy' patients were excluded and 81.3 % overall. The aetiology of the ileus was detected by sonography in 13 cases of paralytic ileus (54.1 %) and in 57 cases of mechanical ileus (71.4 %). It is concluded that ultrasound, which is a non-invasive, portable and even bedside imaging procedure, appears accurate in confirming a SB obstruction and in determining the aetiology of SB obstruction.

Abstract

Background: A neurocognitive phenotype of post-COVID-19 infection has recently been described that is characterized by a lack of awareness of memory impairment (i.e., anosognosia), altered functional connectivity in the brain's default mode and limbic networks, and an elevated monocyte count. However, the relationship between these cognitive and brain functional connectivity alterations in the chronic phase with the level of cytokines during the acute phase has yet to be identified.Aim: Determine whether acute cytokine type and levels is associated with anosognosia and functional patterns of brain connectivity 6-9 months after infection.Methods: We analyzed the predictive value of the concentration of acute cytokines (IL-1RA, IL-1 beta, IL-6, IL-8, IFN beta, G-CSF, GM-CSF) (cytokine panel by multiplex immunoassay) in the plasma of 39 patients (mean age 59 yrs, 38-78) in relation to their anosognosia scores for memory deficits via stepwise linear regression. Then, associations between the different cytokines and brain functional connectivity patterns were analyzed by MRI and multivariate partial least squares correlations for the whole group. Results: Stepwise regression modeling allowed us to show that acute TNFa levels predicted (R-2 = 0.145; beta =-0.38; p = .017) and were associated (r =-0.587; p < .001) with scores of anosognosia for memory deficits observed 6-9 months post-infection. Finally, high TNFa levels were associated with hippocampal, temporal pole, accumbens nucleus, amygdala, and cerebellum connectivity. Conclusion: Increased plasma TNFa levels in the acute phase of COVID-19 predict the presence of long-term anosognosia scores and changes in limbic system functional connectivity.
