Abstract

Technology utilizing human induced pluripotent stem cells (iPS cells) has enormous potential to provide improved cellular models of human disease. However, variable genetic and phenotypic characterization of many existing iPS cell lines limits their potential use for research and therapy. Here we describe the systematic generation, genotyping and phenotyping of 711 iPS cell lines derived from 301 healthy individuals by the Human Induced Pluripotent Stem Cells Initiative. Our study outlines the major sources of genetic and phenotypic variation in iPS cells and establishes their suitability as models of complex human traits and cancer. Through genome-wide profiling we find that 546% of the variation in different iPS cell phenotypes, including differentiation capacity and cellular morphology, arises from differences between individuals. Additionally, we assess the phenotypic consequences of genomic copy-number alterations that are repeatedly observed in iPS cells. Abstract

Mitigation of SARS-CoV-2 transmission from international travel is a priority. We evaluated the effectiveness of travellers being required to quarantine for 14-days on return to England in Summer 2020. We identified 4,207 travel-related SARS-CoV-2 cases and their contacts, and identified 827 associated SARS-CoV-2 genomes. Overall, quarantine was associated with a lower rate of contacts, and the impact of quarantine was greatest in the 16-20 age-group. 186 SARS-CoV-2 genomes were sufficiently unique to identify travel-related clusters. Fewer genomically-linked cases were observed for index cases who returned from countries with quarantine requirement compared to countries with no quarantine requirement. This difference was explained by fewer importation events per identified genome for these cases, as opposed to fewer onward contacts per case. Abstract

We examined preferences around spiritual support services delivered by chaplains among U.S. adults in outpatient and inpatient healthcare settings using a cross-sectional national survey of U.S. adults (n = 1,020). For outpatient settings, 5% reported interest in chaplain services, whereas 16% reported interest in inpatient settings. In both settings, a higher perceived value of chaplain services (p < 01) and previous experience with a chaplain (p <.01) were associated with a greater interest in a chaplain consult when adjusting for demographic factors. In both settings, the most desired services were to explore what was most important in the event of a serious illness or injury (46–47%), values related to treatment decisions (43–46%), and connecting to resources for personal strength and resilience (36–39%), whereas exploring more religious concerns was less desirable. Abstract

Introduction Tobacco 21 (T21) policies, which prohibit tobacco sales to individuals under 21, aim to reduce youth tobacco use by limiting youth access to these products. Little, however, is known about the longitudinal effect of T21 policies on youth tobacco use behaviors at the national level. Methods Participants aged 15–21 years from a longitudinal study conducted between May 2014 and May 2019 (n  = 13,990) were matched to geocoded T21 policies. Generalized linear mixed models examined the association of direct and bordering T21 policy exposure and cigarette and e-cigarette use and intention to use, accounting for individual characteristics. Results Controlling for sociodemographic and psychosocial covariates, there were statistically significant positive associations between T21 exposure and e-cigarette use (OR = 1.45 [1.03,2.06], p  < .003) and intention to use e-cigarettes (OR = 1.54 [1.05,2.26], p  < .027). We found no association between T21 policy exposure and cigarette use or intention to use cigarettes. Furthermore, exposure to T21 policies did not significantly modify the relationship between age and either cigarette outcome. Conclusions This is the first longitudinal study to evaluate state and local T21 policies at the national level. Our analyses demonstrate that existing T21 policies are not sufficient to reduce youth tobacco use and intentions to use, and suggest that T21 policies need to be a part of a comprehensive tobacco policy landscape. Our findings suggest further research is warranted on state and local T21 policy enforcement and implementation, including how T21 may differentially impact cigarette and e-cigarette use, and may have implications for the federal T21 policy. Implications This research evaluates state and local T21 policies in the United States longitudinally, finding that T21 policies are not sufficient to stem e-cigarette use among adolescents and young adults. These findings support further policy action and suggest that local and state T21 policies are not sufficient to reduce tobacco use and rather, need to be part of a broader, more comprehensive set of tobacco control policies. 194. Retroactive Adjustment of Perceived Time

Abstract

Accurately timing acoustic events in dynamic scenes is fundamental to scene analysis. To detect events in busy scenes, listeners must often identify a change in the "pattern" of ongoing fluctuation, resulting in many ubiquitous events being detected later than when they occurred. This raises the question of how delayed detection time affects the manner in which such events are perceived relative to other events in the environment. To model these situations, we use sequences of tone-pips with a time-frequency pattern that changes from regular to random ("REG-RAND") or vice versa ("RAND-REG"). REG-RAND transitions are detected rapidly, but the emergence of regularity cannot be established immediately, and thus RAND-REG transitions take significantly longer to detect. Using a temporal order judgment task, and a light-flash as a temporal marker, we demonstrate that listeners do not perceive the onset of RAND-REG transitions at the point of detection ([approximately]530 ms post transition), but automatically re-adjust their estimate [approximately]300 ms closer to the nominal transition. These results demonstrate that the auditory system possesses mechanisms that survey the proximal history of an ongoing stimulus and automatically adjust perception to compensate for prolonged detection time, allowing listeners to build meaningful representations of the environment.
