Abstract

AIM:To examine the effect of intravitreal adenoviral vector-mediated tristetraprolin (Ad-TTP) on VEGF mRNA expression in a rat model of laser-induced choroidal neovascularization.METHODS:Ad-TTP was prepared using a commercial kit. Retinal laser-induced photocoagulation (10 spots per eye) was performed on rats in this experimental choroidal neovascularization (CNV) model. Rats were divided into four groups:control (single intravitreal injection of balanced salt solution, n=10), laser-induced CNV (photocoagulation only, n=20), laser-induced CNV plus Ad-TTP injection (photocoagulation plus a single intravitreal Ad-TTP injection, n=20) and Ad-TTP injection only (n=10). Changes in choroidal morphology were evaluated in ten rats in the laser only and the laser plus Ad-TTP groups. Two weeks after laser injury, the size of CNV was calculated by perfusion with high-molecular-weight fluorescein isothiocyanate (FITC)-dextran. VEGF mRNA expression in retina-choroid tissue from ten rats in each group was measured by reverse transcription polymerase chain reaction (RT-PCR).RESULTS:Two weeks after treatment, the area of laser-induced CNV was reduced by approximately 60% in the rats given the Ad-TTP injection compared with that in the laser-only group. There was a tendency toward decreased VEGF mRNA expression in the Ad-TTP injection groups.CONCLUSION:A single intravitreal injection of Ad-TTP significantly suppressed CNV size in this experimental laser-induced CNV model. Ad-TTP injection also decreased VEGF mRNA expression compared with that in the laser-induced CNV group. The present study is meaningful as the first study to investigate the effect of tristetraprolin delivered via intravitreal injection.

Abstract

Deacylated lipooligosaccharides (dLOSs) were developed as human vaccine adjuvants. The dLOSs have significant immune activities and markedly lower toxicities than their precursors, lipopolysaccharides (LPSs). In this study, we developed a novel liquid chromatography mass spectrometry (LC–MS) method for characterization of the dLOSs. The structural identification and characterization of the dLOSs were carried out using electrospray ionization tandem mass spectrometric experiments. Finally, we propose the most likely structure of the dLOSs as being composed of a diacyl form of lipid A with seven sugars, 3 HEP + 2 KDO + 1 GlcN + 1 Glc. [ABSTRACT FROM AUTHOR]

198. A Quantitative Proteomic Analysis to Reveal Effects of N-acetylcysteine on H2O2-induced Cytotoxicity

Abstract

Aims: This study aimed to perform a quantitative analysis of the proteomic changes in neural cells under H2O2 exposure and N-acetylcysteine (NAC) treatment. Background: NAC is a potent antioxidant and an effective free radical scavenger that has been used as a potential treatment of several neurological disorders. Although the molecular mechanisms of neuroprotective action by NAC have been studied, more efforts are still needed for the elucidation of the mechanisms through a quantitative proteomic analysis. Objective: This study aimed to identify differentially expressed proteins among control, H2O2-treated group, and NAC+H2O2-treated group as well as reveal proteins involved in the protection of neural cells from H2O2-induced toxicity. Methods: SK-N-MC cells were untreated (control), treated with H2O2 (disease group), pretreated with NAC, and then treated with H2O2 (NAC group). Proteins were digested to peptides and analyzed using liquid chromatography-tandem mass spectrometry with the data-independent acquisition. Skyline was used to quantify peptides and proteins. MSstats was used for statistical analysis. Gene ontology and protein-protein interactions were performed using the Differentially Expressed Proteins (DEPs). Results: Cytoprotective effects of NAC on the cell against H2O2-induced toxicity were first proven using a cell viability study and lactate dehydrogenase assay. The proteomic analysis found 93 DEPs in three comparisons. Among them, 37 proteins were differentially expressed under H2O2 exposure. Only 10 DEPs were rescued in the case of NAC pretreatment. Aspartate aminotransferase and L-lactate dehydrogenase B chain were two DEPs involved in the cysteine and methionine metabolism pathway, which might relate to the mechanisms of NAC protective effects. Conclusion: The findings of cell studies and proteomic analysis were in agreement with previous results, confirming the cytoprotective effects of NAC on neural cells against oxidative stress.
