Abstract

Synthetic biology aspires to construct natural and non-natural pathways to useful compounds. However, pathways that rely on multiple promiscuous enzymes may branch, which might preclude selective production of the target compound. Here, we describe the assembly of a six-enzyme pathway in Escherichia coli for the synthesis of C[subscript 50]-astaxanthin, a non-natural purple carotenoid. We show that by judicious matching of engineered size-selectivity variants of the first two enzymes in the pathway, farnesyl diphosphate synthase (FDS) and carotenoid synthase (CrtM), branching and the production of non-target compounds can be suppressed, enriching the proportion of C[subscript 50] backbones produced. We then further extend the C[subscript 50] pathway using evolved or wild-type downstream enzymes. Despite not containing any substrate- or product-specific enzymes, the resulting pathway detectably produces only C[subscript 50] carotenoids, including ~90% C[subscript 50]-astaxanthin. 158. Development of a regulatable low-temperature protein expression system using the psychrotrophic bacterium, Shewanella livingstonensisAc10, as the host

Abstract

ABSTRACTA low-temperature protein expression system is useful for the production of thermolabile proteins. We previously developed a system that enables constitutive protein production at low temperatures, using the psychrotrophic bacterium Shewanella livingstonensisAc10 as the host. To increase the utility of this system, in the present study, we introduced a repressible promoter of the trpoperon of this bacterium into the system. When ß-lactamase was produced under the control of this promoter at 18°C and 4°C, the yields were 75 and 33 mg/L-culture, respectively, in the absence of L-Trp, and the yields were decreased by 72% and 77%, respectively, in the presence of L-Trp. We also found that 3-indoleacrylic acid, a competitive inhibitor of the Escherichia coli trprepressor, increased the expression of the reporter gene. This repressible gene expression system would be useful for regulatable recombinant protein production at low temperatures.

Abstract

平成21年度宇宙輸送シンポジウム(2010年1月14日-15日, 宇宙航空研究開発機構宇宙科学研究本部), Space Transportation Symposium (January 14-15, 2010. Institute of Space and Astronautical Science, Japan Aerospace Exploration Agency(JAXA)), Sagamihara, Kanagawa Japan, As an observation method to estimate the density of neutral particles in the upper atmosphere, a remote sensing system using an artificial ion source is developed and characterized. An ion beam originating from an ion thruster causes charge exchange collisions (CEX) with neighboring neutral particles and results in Energetic Neutral Atoms (ENAs). We can estimate the density of the neutral particles in the upper atmosphere by analyzing these ENAs. To investigate this new ENA particle detection method, we started fundamental experiments of how to detect ENAs by using an Avalanche Photodiode (APD). In these experiments we investigated the characteristics of APDs as ENA detectors. We obtain results that confirm APDs to be good ENA detectors in terms of resolution compared to a quartz crystal measurement. Also, we estimated the amount of ENAs which can be detected in a demonstration experiment using a sounding rocket and showed that APDs are feasible detectors for ENA detection. Additionally, we investigated the dependence of the APD's gain with respect to the energy and the species of the ENAs. 182. Identification of SAMT family proteins as substrates of MARCH11 in mouse spermatids

Abstract

MARCH11, a RING-finger transmembrane ubiquitin ligase, is predominantly expressed in spermatids and localized to the trans-Golgi network (TGN) and multivesicular bodies (MVBs). Because ubiquitination acts as a sorting signal of cargo proteins, MARCH11 has been postulated to mediate selective protein sorting via the TGN-MVB pathway. However, the physiological substrate of MARCH11 has not been identified. In this study, we have identified and characterized SAMT1, a member of a novel 4-transmembrane protein family, which consists of 4 members. Samt1 mRNA and its expression product were found to be specific to the testis and were first detected in germ cells 25 days after birth in mice. Immunohistochemical analysis further revealed that SAMT1 was specifically expressed in haploid spermatids during the cap and acrosome phases. Confocal microscopic analysis showed that SAMT1 co-localized with MARCH11 as well as with fucose-containing glycoproteins, another TGN/MVB marker, and LAPM2, a late endosome/lysosome marker. Furthermore, we found that MARCH11 could increase the ubiquitination of SAMT1 and enhance its lysosomal delivery and degradation in an E3 ligase activity-dependent manner. In addition, the C-terminal region of SAMT1 was indispensable for its ubiquitination and proper localization. The other member proteins of the SAMT family also showed similar expression profile, intracellular localization, and biochemical properties, including ubiquitination by MARCH11. These results suggest that SAMT family proteins are physiological substrates of MARCH11 and are delivered to lysosomes through the TGN-MVB pathway by a ubiquitin-dependent sorting system in mouse spermatids.
