Abstract

The opportunities of the hot wall aerosol synthesis, i.e. conventional spray pyrolysis (CSP) method are demonstrated for the generation of highly spherical three-dimensional (3D) nanostructured phosphor particles with uniformly distributed components, phases and nano-clustered inner structure. With the presumption that certain particle morphology is formed during the evaporation/drying stage, the aerosol transport properties and powder generation are correlated with the particles structural and morphological features. With the help of various analyzing techniques like Field Emission Scanning Electron Microscopy (FE-SEM), Transmission Electron Microscopy (TEM) coupled with energy dispersive X-ray Analysis and STEM mode (TEM/EDS), X-ray Powder Diffraction (XRPD) and fluorescence measurements the feasible processing of up-conversion rare-earth Y2O3:Er, Yb phosphors powders are discussed. 156. Structural, morphological and luminescence properties of nanocrystalline up-converting Y1.89Yb0.1Er0.01O3 phosphor particles synthesized through aerosol route

Abstract

Nanocrystalline up-converting Y2O3:Yb3+, Er3+ phosphor particles were processed in a dispersed system-aerosol, generated ultrasonically at 1.3 MHz from common nitrate precursor solution having fixed ytterbium-to-erbium concentration ratio. The appropriate process parameters: residence time 21 s, carrier gas (air) flow rate 1.6 dm(3)/min, synthesis temperature 900 degrees C, led to the formation of un-agglomerated spherical nanostructured secondary particles, having mean particle size of approx 450 nm, composed of primary nanoscaled (20 nm) subunits. In order to reach targeting phase crystallinity, the as-prepared particles were additionally annealed at 1100 degrees C in air for 12, 24 and 48 h, respectively. Particle structure, morphology and purity were analyzed by X-ray powder diffraction (XRPD), scanning electron microscopy (FESEM/SEM), analytical and high resolution transmission electron microscopy (TEM/HRTEM) in combination with energy dispersive X-ray analysis and Fourier Transform Infrared Spectroscopy (FTIR). All samples crystallized in a cubic bixbyte-structure, space group Ia-3. The crystallite size changed with annealing time from 30 nm in as-prepared sample to 135 nm in sample annealed for 48 h, respectively. Emission spectra were assigned to the following trivalent erbium f-f electronic transitions: H-2(9/2) - GT I-4(15/2) (blue: 407-420 nm), (H-2(11/2), S-4(3/2)) - GT I-4(15/2) (green: 510-590 nm), and F-4(9/2) - GT I-4(15/2) (red: 640-720 nm). The significant improvement of the emission decay times were observed after thermal treatment and this effect is correlated further with the structural and morphological particles characteristics. For the annealing time of 12 h a quite high emission decay times were achieved (blue: 0.14 ms, green: 0.32 ms and red: 0.39 ms).

Abstract

Helicobacter pylori (HP) infection, a cause of multifocal atrophic gastritis, is considered an important factor related to the evolution of the human gastric mucosa from normal to intestinal-type adenocarcinoma. We examined cell proliferation and both double and single strand DNA damage in situ in 35 patients undergoing gastrectomy for adenocarcinoma with HP-infected gastric mucosa by immunolocalization of Ki-67, terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling, and in situ nick translation. We also studied the distribution of intraepithelial neutrophils by elastase immunolocalization. HP infection was confirmed in all cases by serum anti-HP antibodies, ureas testing, and histopathological examination. HP-infected gastric mucosa was classified according to the degree of inflammation and intestinal metaplasia. Ki-67, terminal deoxynucleotidyl transferase-mediated labeling, in situ nick translation, and intraepithelial neutrophil indices all increased with the progression of gastritis and were highest in glands with incomplete intestinal metaplasia. All indices were lowest in gastric glands with complete intestinal metaplasia. Significant positive correlations were observed among these markers. Increased proliferative activity in HP-associated chronic gastritis in response to cell damage or injury was clearly demonstrated, suggesting that both HP-associated toxins and intraepithelial neutrophils are important in HP-related gastric epithelial injury. Increased cell turnover associated with incomplete intestinal metaplasia may result in DNA instability and subsequent development of intestinal-type gastric adenocarcinoma in HP-infected mucosa.

Abstract

Topoisomerase II (topo II) separates chromosomes at the end of mitosis and is also the target for various chemotherapeutic agents. Expression of this enzyme has been demonstrated to increase rapidly at the end of the S to G2/M phase and decrease after the completion of mitosis. We immunolocalized topo II in specimens of both normal and neoplastic human gastric mucosas to evaluate expression of this enzyme. Three different antibodies were used for the immunostaining of topo II (anti-topo II alpha isoform, anti-topo II beta isoform and anti-topo II alpha and -beta isoforms). There were no significant differences in topo II labeling index (LI) between frozen and paraffin-embedded tissue sections obtained from the same cases. Topo II LI was significantly correlated with Ki67 LI in all of the specimens examined. The area of cells positive for Topo II was much narrower than that of Ki67 in the normal gastric glands, and the pattern of Topo II immunolocalization in both adenomas and adenocarcinomas was also essentially the same as that of Ki67. The topo II LI values (positive cells/1000 cells) for normal gastric gland, adenoma, intestinal-type adenocarcinoma, and diffuse-type adenocarcinoma were 114.7 +/- 2.2, 266.7 +/- 18.8, 277.6 +/- 19.2, and 324.5 +/- 5.3, respectively. Significant differences in topo II LI and topo II/Ki67 index were observed between normal and neoplastic mucosas (P < 0.0001) and between adenomas or intestinal-type adenocarcinoma and diffuse-type adenocarcinoma (P < 0.001 and P < 0.01, respectively). Simultaneous measurement of topo II alpha and nuclear DNA content by two-parameter flow cytometry revealed that the Jurkat cell line established from acute lymphocytic leukemia cells expressed the enzyme in cells at other than S and G2/M phases of the cell cycle whereas topo-II alpha-positive cells were predominantly observed in S and G2/M phases of the cell cycle in the cells from normal lymph nodes. These findings suggest that dys-regulation or qualitative changes of topo II alpha expression are associated with malignancy. Topo II immunostaining can thus detect proliferating cells in routinely processed tissue sections and can indicate the altered topo II alpha expression in human cancers, which may be related to the sensitivity to topo-II-targeted chemotherapeutic agents.

179. Nature of structural changes in LSM-YSZ nanocomposite material during thermal treatments

Abstract

Nanocomposite powders in the system (La1-xSx)(a)MnO3-YSZ((1-y)) [LSM-YSZ], as an attractive cathode material for high operating solid oxide fuel cell (SOFC), were obtained through ultrasonic spray pyrolysis (1.7 MHz) method. The principle of the synthesis is based on a single-step high temperature aerosol decomposition of atomized precursor solutions composed from yttria stabilized ZrO2 sol (average particle size similar to 60 nm) and corresponding aqueous nitrate and hydrogen peroxide solutions of La, Sr and Mn. Two starting systems were examined: (a) (La0.76Sr0.24)(0.93)MnO3 with 0% YSZ, and (b) (La0.77Sr0.23)(0.92)MnO3-YSZ(0.81) with corresponding molar ratio LSM/YSZ = 48:52. X-ray diffraction analysis of as-prepared powders showed the presence of a lanthanum magnesium oxide and yttrium zirconium oxide. Compositional homogeneity and particle morphology were determined by SEM analysis. LSM-YSZ obtained particles are spherical in shape, submicron in size and composed of primary subgrains. TEM analysis of ultra-cross section revealed dense-packed nanosized grains, several tens of nanometers in diameter. After powders calcination TEM observations confirmed dense particle structure composed of primary grains, with a mean diameter of around 100 nm. The structural changes expressed through the crystallite growth were analyzed by nonisothermal differential scanning calorimetry (DSC).
