152. Epidemiology of Injuries Among National Basketball Association Players: 2013-2014 Through 2018-2019

Abstract

Background: Understanding the epidemiology of injuries to athletes is essential to informing injury prevention efforts.Hypothesis: The incidence and impact of basketball-related injuries among National Basketball Association (NBA) players from 2013-2014 through 2018-2019 is relatively stable over time.Study Design: Descriptive epidemiology study.Level of Evidence: Level 3.Methods: Injuries from 2013-2014 through 2018-2019 were analyzed using the NBA Injury and Illness Database from an electronic medical record system. Descriptive statistics were calculated for injuries by season, game-loss, and onset. Incidence rates were estimated using Poisson models and linear trend tests.Results: Between 552 and 606 players participated in ≥1 game per season during the study. Annual injury incidence ranged from 1550 to 1892, with 33.6% to 38.5% resulting in a missed NBA game. Game-loss injury rates ranged from 5.6 to 7.0 injuries per 10,000 player-minutes from 2014-2015 through 2018-2019 (P= 0.19); the rate was lower in 2013-2014 (5.0 injuries per 10,000 player-minutes), partly due to increased preseason injury rates and transition of reporting processes. The 6-year game-loss injury rate in preseason and regular season games was 6.9 (95% CI 6.0, 8.0) and 6.2 (95% CI 6.0, 6.5) injuries per 10,000 player-minutes; the rate in playoff games was lower (P< 0.01) at 2.8 (95% CI 2.2, 3.6). Most (73%) game-loss injuries had acute onset; 44.4% to 52.5% of these involved contact with another player.Conclusion: From 2013-2014 through 2018-2019, over one-third of injuries resulted in missed NBA games, with highest rates of game-loss injuries in preseason games and lowest rates in playoff games. Most game-loss injuries had acute onset, and half of those involved contact with another player.Clinical Relevance: These findings - through reliable data reporting by team medical staff in an audited system - can guide evidence-based injury reduction strategies and inform player health priorities.

Abstract

Synthetic aperture radar (SAR) uses relative motion to produce ne resolution images from microwave frequencies and is a useful tool for regular monitoring and mapping applications. Unfortunately, if target distance is estimated poorly, then phase errors are incurred in the data, producing a blurry reconstruction of the image. In this thesis, we introduce a new multistatic methodology for determining these phase errors from interferometry-inspired combinations of signals. To motivate this, we rst consider a more general problem called phase retrieval, in which a signal is reconstructed from linear measurements whose phases are either unreliable or unavailable. We make signi cant theoretical progress on the phase retrieval problem, to include characterizing injectivity in the complex case, devising the theory of almost injectivity, and performing a stability analysis. We then apply certain ideas from phase retrieval to resolve phase errors in SAR. Speci cally, we use bistatic techniques to measure relative phases, and then we apply a graph-theoretic phase retrieval algorithm to recover the phase errors. We conclude by devising an image reconstruction procedure based on this algorithm, and we provide simulations that demonstrate stability to noise., The original document contains color images.

155. Phase retrieval from very few measurements

Abstract

In many applications, signals are measured according to a linear process, but the phases of these measurements are often unreliable or not available. To reconstruct the signal, one must perform a process known as phase retrieval. This paper focuses on completely determining signals with as few intensity measurements as possible, and on efficient phase retrieval algorithms from such measurements. For the case of complex M-dimensional signals, we construct a measurement ensemble of size 4M-4 which yields injective intensity measurements; this is conjectured to be the smallest such ensemble. For the case of real signals, we devise a theory of "almost" injective intensity measurements, and we characterize such ensembles. Later, we show that phase retrieval from M+1 almost injective intensity measurements is NP-hard, indicating that computationally efficient phase retrieval must come at the price of measurement redundancy.

163. Local responses of the respiratory mucosa to bacterial colonization

Abstract

The respiratory mucosa of mammals possesses sophisticated systems to regulate microbial activities and populations in the adjacent lumen. This tissue is a highly coordinated unit with epithelial, glandular and structural components that can respond to diverse microbial signals. While many mucosal interactions have been characterized, this report describes a number of novel microbial stimuli and mucosal responses that occur in the respiratory tract. It is shown that colonization of mouse nasal mucosa induces MIP2 secretion, an important chemotactic factor, that recruits neutrophils into the mucosal lumen. These neutrophils interact intimately with bacteria, but are effective in clearing the microbes only under certain conditions. Colonization also stimulates submucosal glands to produce and secrete host defenses such as siderocalin and mucus. Respiratory epithelial cell culture models of these in vivo processes demonstrate that bacterial toxins and siderophores induce pro-inflammatory responses via novel pattern recognition systems. This work suggests that there is a great deal left to learn about the impact of microbial ecology and physiology on host tissue responses, and, conversely, also a great deal to learn about the impact of the host innate immune response on microbial physiology, lifestyle and ecology. It is clear that colonizing microbes and their hosts are co-evolving partners, responding to each others' physiology and biochemistry, frequently achieving stable interactions in the long run. Further, one can speculate, based on these novel systems, that the calculus by which the respiratory mucosa determines an appropriate response involves integration of a broad array of signals.

Abstract

A novel chemistry reduction scheme incorporating neural networks and a pre-existing reduction algorithm called Global Pathway Selection (GPS) is proposed and validated. GPS is a state-of-the-art chemistry reduction tool that can provide accurate predictions for any given detailed mechanism. However, this accuracy becomes limited by the overfitting of some reactions for the entire combustion process. We propose a new adaptive GPS approach that conducts GPS reductions and creates multiple reduced mechanisms. The adaptive GPS method is then used to train a supervised neural network to enable the identification of important species for the given state vector. This network is then used to dynamically update the simulator's reduced mechanism according to any given condition. This method is referred to as supervised learning-aided GPS (SL-GPS) in this study. Because the mechanism is continuously modified to better represent the simulation's state, and each such modification requires only a call of a pre-trained network, this method significantly improves the accuracy of predictions using the reduced mechanisms with reduced computational cost. The adaptive GPS method is tested in 0D simulations of methane combustion using the GRI 3.0 detailed mechanism. The results obtained using the adaptive GPS and SL-GPS are compared to the predictions of the detailed mechanism and classical GPS. The SL-GPS method is shown to perform at greater accuracies than the classic GPS while using fewer species and reactions, and at a similar accuracy to the adaptive GPS with which the neural network was trained. The capabilities of adaptive and SL-GPS methods are even more profound for mechanisms with a greater number of species and reactions, i.e., PCRL-Mech1 mechanism for ethanol combustion which has been used for validation in this study. Abstract: Prion formation involves the conversion of soluble proteins into an infectious amyloid form. This process is highly specific, with prion aggregates templating the conversion of identical proteins. However, in some cases non-identical prion proteins can interact to promote or inhibit prion formation or propagation. These interactions affect both the efficiency with which prion diseases are transmitted across species and the normal physiology of yeast prion formation and propagation. Here we examine two types of heterologous prion interactions: interactions between related proteins from different species (the species barrier) and interactions between unrelated prion proteins within a single species. Interestingly, although very subtle changes in protein sequence can significantly reduce or eliminate cross-species prion transmission, in Saccharomyces cerevisiae completely unrelated prion proteins can interact to affect prion formation and propagation. Abstract

Background aims. Dendritic cells (DC) are increasingly being used as cellular vaccines to treat cancer and infectious diseases. While there have been some promising results in early clinical trials using DC-based vaccines, the inability to visualize non-invasively the location, migration and fate of cells once adoptively transferred into patients is often cited as a limiting factor in the advancement of these therapies. A novel perflouropolyether (PFPE) tracer agent was used to label human DC ex vivo for the purpose of tracking the cells in vivo by 19F magnetic resonance imaging (MRI). We provide an assessment of this technology and examine its impact on the health and function of the DC. Methods. Monocyte-derived DC were labeled with PFPE and then assessed. Cell viability was determined by examining cell membrane integrity and mitochondrial lipid content. Immunostaining and flow cytometry were used to measure surface antigen expression of DC maturation markers. Functional tests included bioassays for interleukin (IL)-12p70 production, T-cell stimulatory function and chemotaxis. MRI efficacy was demonstrated by inoculation of PFPE-labeled human DC into NOD-SCID mice. Results. DC were effectively labeled with PFPE without significant impact on cell viability, phenotype or function. The PFPE-labeled DC were clearly detected in vivo by 19F MRI, with mature DC being shown to migrate selectively towards draining lymph node regions within 18 h. Conclusions. This study is the first application of PFPE cell labeling and MRI cell tracking using human immunotherapeutic cells.
