From October 1993 to August 1994 four distal stomach resections (three Billroth-II reconstructions and one Billroth-I reconstruction) were performed laparoscopically. The first patient, who was 83 years old, presented with a stenosing bleeding, pre-pyloric, malignant non-Hodgkin-lymphoma. The second patient, who was 84 years old, presented with an ulcerated, non-malignant leiomyoblastoma of the antrum. Due to the histological type of the tumours a radical lymphadenectomy was not performed. The other two patients presented with a persisting pre-pyloric ulcer combined with an almost complete stenosis of the pylorus. All operations followed the principles of conventional open surgery. Anastomoses were stapled intracorporally. The specimen was removed in a lap sac through a 3.5cm infra-umbilical incision. Intra-abdominal complications did not occur. However, the first patient (with non-Hodgkin-lymphoma) died on the 21st post-operative day from cardio-pulmonary failure. The other three patients were discharged from hospital after an uneventful recovery.

The supraspinal control of colonic motility and anorectal motility is poorly documented. We had the opportunity to study colonic function, esophageal function, and urinary bladder function in three patients who presented with vascular lesions limited to the anterior (case 1) or the posterior area (cases 2 and 3) of the pons. Esophageal manometry, urodynamic examination, whole and segmental transit time measurements (using radiopaque markers) and anorectal motility were systematically performed. The results were the following: (1) in the first case esophageal motility was not altered, whereas abnormal micturition, right colonic inertia, and absence of rectoanal inhibitor reflex were observed; (2) in cases 2 and 3, there was a poor esophageal coordination, the micturition and rectoanal inhibitor reflex were normal, and the transit time of the left colon was increased. Our observations are consistent with the previously described localization of neurological areas controlling swallowing and micturition; they also favor the pons as the possible level of supraspinal control of colonic and anorectal motility.

The growth rate of rodent embryonic neuroblasts and human neuroblastoma cell lines is regulated in part by autocrine or paracrine actions of neuropeptides of the family that includes vasoactive intestinal peptide (VIP), peptide histidine isoleucine (PHI), and pituitary adenylate cyclase-activating peptide (PACAP). These peptides act via seven transmembrane G-protein-linked receptors coupled to cAMP elevation, phospholipase C activation, intracellular Ca2+ release, and/or of mitogen-activated protein (MAP) kinase activation. Here we investigated the action of these peptides on the mouse neuroblastoma cell line Neuro2a. PHI and VIP inhibited proliferation at concentrations as low as 10(-13) M and 10(-10) M, respectively. In contrast, PACAP action was biphasic, with stimulation occurring at subnanomolar doses and inhibition at higher doses. Peptide actions were studied further by measuring cAMP and ERK1/2 MAP kinase activity and by assessing 3H-thymidine incorporation in conjunction with a panel of signal transduction pathways inhibitors. The data obtained indicated that the PHI-inhibitory and PACAP-stimulatory activities were mediated by corresponding changes in activity of the MAP kinase pathway and independent of protein kinase A (PKA) or protein kinase C (PKC). In contrast, the inhibitory actions of VIP and PACAP were specifically blocked by antagonists of PKA. Northern blot analysis revealed gene expression for only the PACAP-preferring (PAC1) receptor. However, binding experiments using 125I-labeled PACAP27, PHI, and VIP, demonstrated the presence of PACAP-preferring sites, bivalent VIP/PACAP sites, and PHI-binding sites that did not interact with VIP. The studies demonstrate potent regulatory actions of PACAP, PHI, and VIP on neuroblastoma cell proliferation which appear to be mediated by multiple subsets of receptors which differentially couple to MAP kinase and PKA signaling pathways.

The comparison of the CT-guided percutaneous wire localization and CT-guided percutaneous Indigocarmine Blue localization in preoperative detection of pulmonary nodules shows a great advantage of the former. Wire localization allows the detection and thoracoscopic resection even of deeply localized nodules. In the following article we present our experience with both methods and discuss critically the results.

Three phenotypically distinct subclones (SH-SY-5Y, SH-EP, SH-IN) of the human neuroblastoma cell line SK-N-SH were found to possess vasoactive intestinal polypeptide (VIP) precursor mRNA, release immunoreactive VIP, and express high-affinity VIP receptors coupled to adenylate cyclase. The apparent molecular mass for the receptor polypeptide, as determined by covalent cross-linking of 125I-VIP, was 49 kDa.

We have previously shown that the mono [125I]iodinated vasoactive intestinal peptide (125I-VIP) could be covalently cross-linked on intact colonic adenocarcinoma cells (HT29). A major Mr 67,000 and a minor Mr 120,000 cross-linked polypeptides have been characterized [Muller, Luis, Fantini, Abadie, Giannellini, Marvaldi & Pichon (1985) Eur. J. Biochem. 151, 411-417]. The glycoprotein nature of these species was investigated using endo-beta-acetylglucosaminidase F (Endo F) treatment, enzymic and chemical desialylation and wheat germ agglutinin (WGA)-Sepharose affinity chromatography. Affinity-labelled VIP-binding proteins solubilized by Nonidet P-40 bound to WGA-Sepharose and could be eluted specifically with N-acetyl-D-glucosamine. Treatment with Endo F resulted in an increased electrophoretic mobility of both polypeptides. The major and the minor VIP-binding proteins were converted respectively into Mr 47,000 and 100,000 species, indicating removal of 20 kDa of N-linked oligosaccharides. Deglycosylation with trifluoromethanesulphonic acid also led to a 20 kDa loss in mass of the Mr 67,000 component, indicating the absence of additional O-linked sugars on this polypeptide. The presence of sialic acid on the major VIP-binding protein was demonstrated after treatment of intact cells with neuraminidase or by chemical desialylation with hydrochloric acid. We conclude from this study that the VIP receptor from intact HT29-D4 cells is a glycoprotein with N-linked oligosaccharide side chains containing sialic acid.

Summary We show that the greedy algorithm introduced in [1] and [5] to perform the parallel QR decomposition of a dense rectangular matrix of sizem×n is optimal. Then we assume thatm/n2 tends to zero asm andn go to infinity, and prove that the complexity of such a decomposition is asymptotically2n, when an unlimited number of processors is available.

HT29 cells originating from a human colon adenocarcinoma, spread very rapidly after seeding on their own extracellular matrix (ECM). Preincubation of cells with the inhibitor of protein glycosylation tunicamycin (TM) or with the ionophore monensin substantially suppressed cell spreading in serum-free medium without affecting cell adhesion to ECM. Addition of the drugs after cell attachment and spreading inhibited cell growth. TM-treated cells remained viable after 6 days of exposure to 2μgml−1 ′ of TM and resumed their normal growth rate and shape after removing the drug from the medium. On the contrary, monensin inhibition of cell growth was not reversible: after 3 days, cells detached from the ECM and were unable to exclude Trypan Blue. At the ultrastructural level, a swollen Golgi apparatus with numerous vacuoles was observed after treatment for 2h in either TM or monensin-preincubated cells. These results suggest that TM and monensin interfere with the insertion and, or, function of one or more cell surface glycoproteins, possibly interacting with cytoskeleton and involved in cell spreading and growth.

Abstract The authors present a study of the intrinsic anatomy of the gluteus medius m. and of its innervation through the caudal branch of the superior gluteal n. The existence of an intramuscular tendon in the thickness of the gluteus medius was constantly prooved in 40 muscles. The relations of the intrinsic fibrous structure of the muscle and its innervation were studied. The authors deduce from that the topography of a gluteus medius incision, with respect to a safety area towards its innervation, which leads to an exposure of the acetabulum that is satisfying and gives opportunities of a sound repair after the surgery of the hip joint through the transgluteal approach. They propose the “anterior hemimyotomy of the gluteus medius m” designation.
