153. A genome-wide association study of early gamma-band response in a schizophrenia case-control sample.

Abstract

Objectives: Disturbances in the gamma-frequency band of electroencephalography (EEG) measures are among the most consistently observed intermediate phenotypes in schizophrenia. We assessed whether genetic variations are associated with gamma-band activity. Methods: We performed a genome-wide association analysis of the early auditory evoked gamma-band response in schizophrenia affected subjects and healthy control individuals (in total N = 315). Results: No marker surpassed the threshold for genome-wide significant association. Several of the markers that were closest to significance mapped to genes involved in neuronal development and the Neuregulin-ErbB signalling network, such as NRG2 and KALRN. Using a gene-set enrichment analysis, we found suggestive evidence for association with genes involved in EEG abnormality (P = .048). Conclusions: We identified no marker genome-wide significantly associating with gamma response; independent replication of the gene-set analysis result and larger sample sizes will be required to provide leads to cellular pathways involved in gamma-band activity. Abstract

A major goal of functional mri measurements is the localization of the neural correlates of sensory, motor, and cognitive functions and to compare brain activation responses between treatment groups. This includes the proper comparison of response amplitudes in brain areas with respect to various experimental conditions. In order to reach solid conclusions about the localization and functional properties of specialized brain areas, a number of analysis steps need to be performed. These analysis steps include preprocessing operations to reduce imaging artifacts and noise components, alignment of functional and anatomical data sets, and inferential statistics. In order to efficiently generalize statistical analyses to the population level, corresponding brain regions need to be identified across individuals. Abstract

Since its invention 20 years ago, functional magnetic resonance imaging (fmri) has become one of the most widely used and probably the publicly most visible noninvasive technique to measure brain activation. Fmri has played a central role in the development of cognitive neuroscience, and several new fields, including social neuroscience, neuroeconomics, and genetic imaging, may not have developed had it not been for the unique opportunities afforded by fmri. The particular strengths of this technique are in its spatial resolution and fidelity, ability to reach deep subcortical structures, and whole-brain coverage, enabling the mapping of functionally connected networks and the extraction of information from activation patterns that are distributed across different brain areas. In the psychiatric domain, fmri has made major contributions to the understanding of psychopathology and the effects of risk genes on cognitive and affective networks (linden 2012a, b), and in neurology fmri has become a central technique for mapping neuroplasticity, for example, in recovery from stroke (seitz 2010), and for presurgical mapping. Abstract

Objectives: Aberrant salience mainly is attributed to excessive dopaminergic processing in the ventral striatum. Increased gamma power during sensory processing of physical salience has been shown to be associated with positive trait schizotypy. In the present study, this is assessed in patients with schizophrenia. Methods: The early evoked visual gamma-band response (GBR) at 40 Hz was assessed for a schizophrenia patient group (N = 22) and a matched healthy control group (N = 22) applying EEG time-frequency analysis. The GBR was assessed for two conditions within a visual detection paradigm: a target with or without a physically salient distracter and evaluated in relation to the PANSS. Results: A 2 × 2 ANOVA revealed a significant main effect of condition and a trend interaction of group and condition for the GBR, with highest power for schizophrenia patients in the physically salient distracter condition. Moreover, evoked GBR power in this condition was correlated with positive (r = 0.664; P = 0.001**) and disorganised (r = 0.618; P = 0.002**) schizophrenia symptoms. Conclusions: Evoked GBR power during processing of physical salience in schizophrenia was associated with positive symptoms. We suggest that abnormal processing of physically salient stimuli might be involved in the pathophysiological genesis of positive symptoms. [ABSTRACT FROM AUTHOR]

Abstract

The main objective of functional neuroimaging is to detect and characterise (in space and time) relevant changes in brain states and their relation to neuronal activity. Functional mri (fmri), electroencephalography (eeg) and magnetoencephalography (meg) are the most widespread noninvasive techniques that are available to experimental and clinical neuroscientists to achieve this objective starting from in vivo measures of brain electrical activity. Both fmri and eeg assume that a given brain state can be decoded from the precise anatomical localisation and the detailed temporal evolution of neuroelectrical brain activation signals, respectively. Starting from these common assumptions, fmri neuroscientists have developed many different approaches for mapping brain states at a spatial resolution of a few millimetres and testing many different neurophysiological and neuropathological hypotheses in normal and clinical populations, despite the limited temporal resolution of the available signals (see previous chapters). On the other hand, eeg neuroscientists have posed analogous questions and addressed similar problems by developing different approaches for the detailed temporal analysis of eeg recordings, despite the limited spatial detail in their findings. The previous chapter illustrated how fmri can be used by eeg neuroscientists to improve the quality of eeg results and to help with the problem of source localisation. The purpose of this chapter is to illustrate how the fmri neuroscientist can integrate detailed temporal information by incorporating simultaneously recorded eeg signals into standard as well as sophisticated fmri spatiotemporal modelling. We discuss how this can be achieved in such a way that new effects become detectable in the fmri domain even when the original event or state change causing possible fmri effects can only be characterised at very rapid temporal scales (e.g. Milliseconds) or frequency bands (above 1 hz). 168. The Time Course of Activity within the Dorsal and Rostral-Ventral Anterior Cingulate Cortex in the Emotional Stroop Task.

Abstract

Growing evidence from neuroimaging studies suggest that emotional and cognitive processes are interrelated. Anatomical key structures in this context are the dorsal and rostral-ventral anterior cingulate cortex (dACC and rvACC). However, up to now, the time course of activations within these regions during emotion-cognition interactions has not been disentangled. In the present study, we used event-related potentials (ERP) and standardized low-resolution electromagnetic tomography (sLORETA) region of interest (ROI) source localization analyses to explore the time course of neural activations within the dACC and rvACC using a modified emotional Stroop paradigm. ERP components related to Stroop conflict (N200, N450 and late negativity) were analyzed. The time course of brain activations in the dACC and rvACC was strikingly different with more pronounced initial responses in the rvACC followed by increased dACC activity mainly at the late negativity window. Moreover, emotional valence modulated the earlier N450 stage within the rvACC region with higher neural activations in the positive compared to the negative and neutral conditions. Emotional arousal modulated the late negativity stage; firstly in the significant arousal × congruence ERP effect and then the significant higher current density in the low arousal condition within the dACC. Using sLORETA source localization, substantial differences in the activation time courses in the dACC and rvACC could be found during the emotional Stroop task. We suggest that during late negativity, within the dACC, emotional arousal modulated the processing of response conflict, reflected in the correlation between the ex-Gaussian µ and the current density in the dACC.
