The ultimobranchial (UB) glands of the common toad Bufo bufo consist of several cellular masses containing two quite different cell types which line a central lumen filled with amorphous material. The morphologically defined Type I cell is akin to a typical calcitonin secretory cell as observed for all vertebrates, with small dense-core secretory granules. On the contrary the Type II cell displays large apical dense bodies which may be related to the secretion and/or absorption of the amorphous material. Cells morphologically related to Type II cells have been described in the UB glands of Sauropsidea and in the UB follicles of mammalian thyroid gland. An immunocytochemical stain using an antiserum raised against synthetic salmon calcitonin demonstrated the specific localization of an immunoreactive product in both Type I and Type II cell granules, suggesting that both cell types could be involved in calcitonin metabolism. Moreover, the presence of immunoreactive calcitonin in the Type II cells of the Bufo UB gland raises the question of the function of the morphologically equivalent cells of other species.

Plasma levels of calcitonin (CT), parathyroid hormone (PTH), and estradiol (E2) were measured in the rat during the estrous cycle. Changes in the circulating levels of CT were observed, in particular a significant fall during estrus. No significant changes in PTH levels were recorded using a N terminal assay. E2 levels were not correlated with CT or PTH levels.

Plasma levels of immunoreactive calcitonin were measured in patients with age-related osteoporosis and with postmenopausal osteoporosis and compared to the levels in control subjects. In all studies immunoreactive calcitonin was lower in the osteoporotic patients. Parathyroid hormone levels were measured in women with postmenopausal osteoporosis and in control women; no difference was observed between the two groups. The results are consistent with the hypothesis that calcitonin but not parathyroid hormone plays a role in postmenopausal and age-related osteoporosis.

Forty-nine women received a combination of cis-platinum and hexamethylmelamine (36 also received doxorubicin) for advanced ovarian cancer progressing after therapy that included an alkylating agent or extended field radiation. Twenty-six (53%) had an objective remission that lasted a median of 6 months from start of treatment. Response rate was independent of age, extent of prior therapy, and performance status. A long interval from initial diagnosis to entry, response to therapy, and ambulatory performance predicted improved survival from entry. No patient is surviving free of disease. Myelosuppression and vomiting were moderately severe but tolerable. Azotemia and peripheral neuropathy were infrequent and milk. These drugs have major activity in this poor-risk group and should be studied as part of initial therapy when enhanced efficacy and reduced toxicity are to be expected.
