Bacteria have evolved many different signal transduction systems that sense signals and generate a variety of responses. Generally, most abundant are transcriptional regulators, sensor histidine kinases and chemoreceptors. Typically, these systems recognize their signal molecules with dedicated ligand-binding domains (LBDs), which, in turn, generate a molecular stimulus that modulates the activity of the output module. There are an enormous number of different LBDs that recognize a similarly diverse set of signals. To give a global perspective of the signals that interact with transcriptional regulators, sensor kinases and chemoreceptors, we manually retrieved information on the protein-ligand interaction from about 1,200 publications and 3D structures. The resulting 811 proteins were classified according to the Pfam family into 127 groups. These data permit a delineation of the signal profiles of individual LBD families as well as distinguishing between families that recognize signals in a promiscuous manner and those that possess a well-defined ligand range. A major bottleneck in the field is the fact that the signal input of many signaling systems is unknown. The signal repertoire reported here will help the scientific community design experimental strategies to identify the signaling molecules for uncharacterised sensor proteins. [ABSTRACT FROM AUTHOR]

Bacteria possess different systems to sense and respond to environmental signals (Galperin, 2018). Most abundant are one-component systems (Ulrich et al., 2005), two-component systems (Stock & Zhulin, 2017) and chemosensory pathways (Porter et al., 2011). While one- and two-component systems mainly control gene expression (Stock & Zhulin, 2017, Ulrich et al., 2005), chemosensory pathways mediate chemotaxis or are associated with alternative functions (Porter et al., 2011, Wadhams & Armitage, 2004). The analysis of the increasing number of bacterial genomes shows that about half of bacteria have genes encoding chemosensory pathways (Ortega et al., 2017). The central element of a chemosensory pathway is the ternary complex formed by chemoreceptors, the CheA autokinase and the CheW coupling proteins (Wadhams & Armitage, 2004). Ligand mediated stimulation of the chemoreceptor causes an alteration of CheA activity that in turn modulates transphosphorylation to CheY that in its phosphorylated form binds to the flagellar motor causing ultimately chemotaxis (Wadhams & Armitage, 2004). The chemoreceptor typically is composed by a periplasmic ligand binding domain, a transmembrane module and a cytoplasmic signaling domain (Parkinson et al., 2015). The function and signals recognized of most chemoreceptors are unknown. The results presented in this thesis increase the knowledge about the chemotactic chemosensory pathways, reveal the signals sensed and cast light into the sensing mechanisms, which is important information to understand important physiological processes such as bacterial migration, colonization, infection and virulence. Future work will determine the potential application of this knowledge, for instance the determination of the role of these chemoreceptors in the promotion of plant root colonization or infection. The demonstration that chemoreceptors respond to chemoeffectors and structurally related antagonists may be a potential strategy to interfere specifically with chemotaxis and virulence., Tesis Univ. This paper seeks to show the impact generated by the Argentine economy in SMEs, because it represents about 98% of companies in this country. This research will help SMEs to make decisions in times of crisis as well as to transform their weaknesses into strengths. On the other hand, it should be taken into account that Colombia and Argentina have large differences in several aspects, however, the percentage of SMEs that exist in each economy is high, for this reason it was considered pertinent to carry out this work from the two points of view. The methodology of this research requires an in-depth visualization due to the fact that strategies will be provided, that is why it was decided to consult people immersed in the business sector, all this thanks to the knowledge acquired during the international journey Administrador de empresas http://unidadinvestigacion.usta.edu.co Pregrado

- Abstract
Chemotaxis and energy taxis permit directed bacterial movements in gradients of environmental cues. Nitrate is a final electron acceptor for anaerobic respiration and can also serve as a nitrogen source for aerobic growth. Previous studies indicated that bacterial nitrate taxis is mediated by energy taxis mechanisms, which are based on the cytosolic detection of consequences of nitrate metabolism. Here we show that Pseudomonas aeruginosa PAO1 mediates nitrate chemotaxis on the basis of specific nitrate sensing by the periplasmic PilJ domain of the PA2788/McpN chemoreceptor. The presence of nitrate reduced mcpN transcript levels, and McpN-mediated taxis occurred only under nitrate starvation conditions. In contrast to the NarX and NarQ sensor kinases, McpN bound nitrate specifically and showed no affinity for other ligands such as nitrite. We report the three-dimensional structure of the McpN ligand binding domain (LBD) at 1.3-Å resolution in complex with nitrate. Although structurally similar to 4-helix bundle domains, the ligand binding mode differs since a single nitrate molecule is bound to a site on the dimer symmetry axis. As for 4-helix bundle domains, ligand binding stabilized the McpN-LBD dimer. McpN homologues showed a wide phylogenetic distribution, indicating that nitrate chemotaxis is a widespread phenotype. These homologues were particularly abundant in bacteria that couple sulfide/sulfur oxidation with nitrate reduction. This work expands the range of known chemotaxis effectors and forms the basis for the exploration of nitrate chemotaxis in other bacteria and for the study of its physiological role.IMPORTANCE Nitrate is of central importance in bacterial physiology. Previous studies indicated that movements toward nitrate are due to energy taxis, which is based on the cytosolic sensing of consequences of nitrate metabolism. Here we present the first report on nitrate chemotaxis. This process is initiated by specific nitrate binding to the periplasmic ligand binding domain (LBD) of McpN. Nitrate chemotaxis is highly regulated and occurred only under nitrate starvation conditions, which is helpful information to explore nitrate chemotaxis in other bacteria. We present the three-dimensional structure of the McpN-LBD in complex with nitrate, which is the first structure of a chemoreceptor PilJ-type domain. This structure reveals striking similarities to that of the abundant 4-helix bundle domain but employs a different sensing mechanism. Since McpN homologues show a wide phylogenetic distribution, nitrate chemotaxis is likely a widespread phenomenon with importance for the life cycle of ecologically diverse bacteria., We acknowledge the European Synchrotron Radiation Facility (ID23-1 and ID30A-1,3) and thank the beam line staff members for their invaluable support. - Abstract

- Abstract
Reticulitermes flavipes, one of the most harmful subterranean termite pests, is reported for the first time from Tenerife (Canary Islands, Spain). Cytochrome oxidase II was sequenced from five specimens in order to confirm the identification. To date, this invasive species has been detected in a limited area in the northeast of the island, affecting buildings, crops and native plant species. Another colony with the identical haplotype found in the southwest, 60 km away from the main population, indicates that this invasive insect may be more widespread over the island.

Dissertação de Mestrado Integrado em Arquitetura apresentada à Faculdade de Ciências e Tecnologia The thesis here presented intends to conceive a theoretical approach to the urban, political and social dimensions of Buenos Aires, assumed as a planned space, through an historical and alarming situation that has been increasing worldwide: the villas and the informal human settlements, therefore assumed as improvised urban spaces. The role of the governmental authorities, as well as the society’s, since the slum’s origins around 1930 and until 1983 – when the country reestablishes the democracy – has, with limited exceptions, guided its policies towards the ideal of excluding the social groups from the society along with its precarious means of living, for decades, seen as excesses of the civil society. The Federal City was mainly appreciated as the designated place for the social elites, for whom the “drawbacks” threads of poverty represented a resistance to the materialization of the perfect city that had always been designed, to that end promoting the redistribution or the evacuation of these social groups to the extensive areas of the metropolitan periphery. Historically, the different policies – mainly urban and social – have been implemented with different articulation levels, creating a series of conflicts between the political and legal entities and the organized groups known as movimientos villeros. In the new millennium, after decades of conflicts and demands for an equal and stable practice of the urban space, these social groups finally gained some legitimacy before the Government and the society, initially guaranteed by the official recognition of the right to a decent dwelling by the National Argentine Constitution, by extension meaning the legal validation of the right to the city – or le droit à la ville –, and furthermore allowing the active participation of the informal residents in the urban and public decisions, promoting the urbanization of their neighborhoods – or the humanization of their life condition. In Latin America, where an extensive and growing social segment improvises its own housing solutions by the means of informality, self-construction and self-management, it becomes essential to understand the influence of the governmental solutions over these groups along the agitated 20th century. Wavering between tolerance and intolerance policies, and with more or less physical and psychological brutality and political success, learning about the continuous attempts to solve this complex matter also allows to understand to what extent is the right to the city being accomplished or not in Buenos Aires nowadays.

- Abstract
Bacteria possess a large number of signal transduction systems that sense and respond to different environmental cues. Most frequently these are transcriptional regulators, two-component systems and chemosensory pathways. A major bottleneck in the field of signal transduction is the lack of information on signal molecules that modulate the activity of the large majority of these systems. We review here the progress made in the functional annotation of sensor proteins using high-throughput ligand screening approaches of purified sensor proteins or individual ligand binding domains. In these assays, the alteration in protein thermal stability following ligand binding is monitored using Differential Scanning Fluorimetry. We illustrate on several examples how the identification of the sensor protein ligand has facilitated the elucidation of the molecular mechanism of the regulatory process. We will also discuss the use of virtual ligand screening approaches to identify sensor protein ligands. Both approaches have been successfully applied to functionally annotate a significant number of bacterial sensor proteins but can also be used to study proteins from other kingdoms. The major challenge consists in the study of sensor proteins that do not recognize signal molecules directly, but that are activated by signal molecule-loaded binding proteins.

- Abstract
Many bacteria can move chemotactically to a variety of compounds and the recognition of chemoeffectors by the chemoreceptor ligand binding domain (LBD) defines the specificity of response. Many chemoreceptors were found to recognize different amino and organic acids, but the McpU chemoreceptor from Pseudomonas putida was identified as the first chemoreceptor that bound specifically polyamines. We report here the three-dimensional structure of McpU-LBD in complex with putrescine at a resolution of 2.4 Å which fitted well a solution structure generated by small-angle X-ray scattering. Putrescine bound to a negatively charged pocket in the membrane distal module of McpU-LBD. Similarities exist in the binding of putrescine to McpU-LBD and taurine to the LBD of the Mlp37 chemoreceptor of Vibrio cholerae. In both structures, the primary amino group of the respective ligand is recognized by hydrogen bonds established by two aspartate and a tyrosine side chain. This feature may be used to predict the ligands of chemoreceptors with unknown function. Analytical ultracentrifugation revealed that McpU-LBD is monomeric in solution and that ligand binding does not alter this oligomeric state. This sensing mode thus differs from that of the well-characterised four-helix bundle domains where ligands bind to two sites at the LBD dimer interface. Although there appear to be different sensing modes, results are discussed in the context of data, indicating that chemoreceptors employ the same mechanism of transmembrane signaling. This work enhances our understanding of CACHE domains, which are the most abundant sensor domains in bacterial chemoreceptors and sensor kinases.

- Abstract
Introduction: Rocky Mountain spotted fever is a highly lethal infectious disease, particularly if specific treatment with doxycycline is given belatedly. Objective: To describe the clinical profile of fatal Rocky Mountain spotted fever cases in hospitalized patients in the state of Sonora, México.Materials and methods: We conducted a cross-sectional study on a series of 47 deaths caused by Rickettsia rickettsii from 2013 to 2016. The diagnosis of Rocky Mountain spotted fever was confirmed in a single blood sample by polymerase chain reaction (PCR) or by a four-fold increase in immunoglobulin G measured in paired samples analyzed by indirect immunofluorescence. Clinical and laboratory characteristics were compared stratifying subjects into two groups: pediatric and adult. Results: There were no differences in clinical characteristics between groups; petechial rash was the most frequent sign (96%), followed by headache (70%) and myalgia (67%). Although that doxycycline was administered before the fifth day from the onset of symptoms, death occurred in 55% of patients. In clinical laboratory, thrombocytopenia, and biomarkers of liver acute failure and acute kidney failure were the most frequent. Conclusion: Rocky Mountain spotted fever remains as one of the most lethal infectious diseases, which may be related not only to the lack of diagnostic suspicion and delayed administration of doxycycline, but to genotypic characteristics of Rickettsia rickettsii that may play a role in the variability of the fatality rate that has been reported in other geographical regions where the disease is endemic., Introducción.

- Abstract
Histamine is a key biological signaling molecule. It acts as a neurotransmitter in the central and peripheral nervous systems and coordinates local inflammatory responses by modulating the activity of different immune cells. During inflammatory processes, including bacterial infections, neutrophils stimulate the production and release of histamine. Here, we report that the opportunistic human pathogen Pseudomonas aeruginosa exhibits chemotaxis toward histamine. This chemotactic response is mediated by the concerted action of the TlpQ, PctA, and PctC chemoreceptors, which display differing sensitivities to histamine. Low concentrations of histamine were sufficient to activate TlpQ, which binds histamine with an affinity of 639 nM. To explore this binding, we resolved the high-resolution structure of the TlpQ ligand binding domain in complex with histamine. It has an unusually large dCACHE domain and binds histamine through a highly negatively charged pocket at its membrane distal module. Chemotaxis to histamine may play a role in the virulence of P. aeruginosa by recruiting cells at the infection site and consequently modulating the expression of quorum-sensing-dependent virulence genes. TlpQ is the first bacterial histamine receptor to be described and greatly differs from human histamine receptors, indicating that eukaryotes and bacteria have pursued different strategies for histamine recognition

- Abstract
A virtual element discretisation for the numerical approximation of the three-field formulation of linear poroelasticity introduced in R. Oyarzúa and R. Ruiz-Baier, (SIAM J. Numer. Anal.54 2951–2973, 2016) is proposed. The treatment is extended to include also the transient case. Appropriate poroelasticity projector operators are introduced and they assist in deriving energy bounds for the time-dependent discrete problem. Under standard assumptions on the computational domain, optimal a priori error estimates are established. These estimates are valid independently of the values assumed by the dilation modulus and the specific storage coefficient, implying that the formulation is locking-free. Furthermore, the accuracy of the method is verified numerically through a set of computational tests. [ABSTRACT FROM AUTHOR]

The Amblyomma maculatum Koch group of ixodid ticks consists of three species: A. maculatum, A. triste, and A. tigrinum. However, since Koch described this group in 1844, the systematics of its members has been the subject of ongoing debate. This is especially true of A. maculatum and A. triste; recent molecular analyses reveal insufficient genetic divergence to separate these as distinct species. Further confounding this issue is the discovery in 2014 of A. maculatum group ticks in southern Arizona (AZ), USA, that share morphological characteristics with both A. triste and A. maculatum. To biologically evaluate the identity of A. maculatum group ticks from southern Arizona, we analyzed the reproductive compatibility between specimens of A. maculatum group ticks collected from Georgia (GA), USA, and southern Arizona. Female ticks from both Arizona and Georgia were mated with males from both the Georgia and Arizona Amblyomma populations, creating two homologous and two heterologous F1 cohorts of ticks: GA ♀/GA ♂, AZ ♀/AZ ♂, GA ♀/AZ ♂, and AZ ♀/GA ♂. Each cohort was maintained separately into the F2 generation with F1 females mating only with F1 males from their same cohort. Survival and fecundity parameters were measured for all developmental stages. The observed survival parameters for heterologous cohorts were comparable to those of the homologous cohorts through the F1 generation. However, the F1 heterologous females produced F2 egg clutches that did not hatch, thus indicating that the Arizona and Georgia populations of A. maculatum group ticks tested here represent different biological species. Demographic aging has led to an increase in the prevalence of different diseases, including the so-called geriatric syndromes (GS), like depression, disability and frailty phenotype (FP). Spirituality and religious practices (RP) have been associated with positive health outcomes and could be a protective factor against the development of some GS. Study aims was to determine the frequency of spirituality and RP, and their associations between some GS in adults 60 years of age or older, who attended a geriatric clinic in a university hospital in Mexico. Cross-sectional study in adults aged ≥ 60 years, recruited in 2018, was performed. Participants underwent a comprehensive geriatric assessment, with which spirituality and religious practices model and the diagnosis of GS were obtained. Linear regression analyses were determined to establish the association between RP and GS. We included 128 subjects; mean age was 79 years (SD ± 8). Lowers scores for mass and communion assiduity were associated with worst disability and FP scores (b = − 0.35, P = 0.02; b = − 0.37, P = 0.03). The RP model explained 8% (R
2 = 0.083) of the total variance of the depressive symptoms score (P = 0.03). This study showed that the prevalence of RP is higher in Mexican older adults. The RP model can explain 8 and 9% of the variation in dependent variables (depressive symptoms and disability). These results suggest the importance of monitoring RP, as they seem to have a positive impact on health status of the elderly. [ABSTRACT FROM AUTHOR]- Published
