Bone morphogenic proteins (BMPs) and the Notch pathway regulate quiescence and self-renewal of stem cells of the subventricular zone (SVZ), an adult neurogenic niche. Here we analyze the role at the intersection of these pathways of Tis21 (Btg2/PC3), a gene regulating proliferation and differentiation of adult SVZ stem and progenitor cells. In Tis21-null SVZ and cultured neurospheres, we observed a strong decrease in the expression of BMP4 and its effectors Smad1/8, while the Notch anti-neural mediators Hes1/5 and the basic helix-loop-helix (bHLH) inhibitors Id1-3 increased. Consistently, expression of the proneural bHLH gene NeuroD1 decreased. Moreover, cyclins D1/2, A2, and E were strongly up-regulated. Thus, in the SVZ Tis21 activates the BMP pathway and inhibits the Notch pathway and the cell cycle. Correspondingly, the Tis21-null SVZ stem cells greatly increased; nonetheless, the proliferating neuroblasts diminished, whereas the post-mitotic neuroblasts paradoxically accumulated in SVZ, failing to migrate along the rostral migratory stream to the olfactory bulb. The ability, however, of neuroblasts to migrate from SVZ explants was not affected, suggesting that Tis21-null neuroblasts do not migrate to the olfactory bulb because of a defect in terminal differentiation. Notably, BMP4 addition or Id3 silencing rescued the defective differentiation observed in Tis21-null neurospheres, indicating that they mediate the Tis21 pro-differentiative action. The reduced number of granule neurons in the Tis21-null olfactory bulb led to a defect in olfactory detection threshold, without effect on olfactory memory, also suggesting that within olfactory circuits new granule neurons play a primary role in odor sensitivity rather than in memory.

Exposure to exercise increases the generation of new neurons in the adult neurogenesis. However, only few studies have investigated the beneficial effect of physical exercise in impaired neurogenesis paradigms. Here we use a mouse model in which the ablation of Btg1 produces a post?natal transient increase of hippocampal neurogenesis, followed by severe depletion of stem and progenitor cells during the adulthood. In this study, we examine whether running could reverse the severe phenotype observed in the adult Btg1 knockout mouse. To this aim, for the first time we evaluate how running influences the cell cycle kinetics of distinct stem and precursor subpopulations of wild type and Btg1 knockout mice, by adopting a recently developed method to measure the cell cycle length. Our data shown that running fully restores the proliferation in the Btg1 knockout mice, by shortening the S?phase length and consequently the overall cell cycle duration of stem (Sox2) and progenitor (Neurod1) cells. Conversely, in the wild?type mice exercise affects exclusively the cell cycle lengthening of NeuroD1 positive progenitor cells. These events are sufficient and necessary to reactivate the hyperproliferation observed in the Btg1 null early postnatal mice and to completely replenish the loss of adult neurogenesis. The sustained increase of cell proliferation in Btg1 null mice after running provides a large pool of activated neural stem cells that results in a long?lasting increment of differentiation, production and survival of newborn neurons. This study shows for the first time that running changes cell cycle kinetics of newly?generated neurons in adult hippocampus.

27th European Photovoltaic Solar Energy Conference and Exhibition; 266-270, Multijunction cells are composed of multiple layers of semiconductors, with different bandgaps. Among them, GaInP/GaAs/Ge cells are the most popular ones, due to their high performances [1]. These cells have higher efficiencies that silicon ones and should be really convenient in a concentrating photovoltaic plants. Photons are absorbed by a layer if their energy is equal or higher to the semiconductor bandgap. Otherwise they are transmitted to the lower semiconductor. All the photons with less energy than the lowest bandgap are converted into waste heat. They represent about the 5% of the total solar spectrum [2]. Moreover, the Germanium layer usually produces a higher current that the other subcells and even the excess current is converted into heat. Cooling is an important task in designing CPV: the higher the concentration the higher the temperature. Thus, a passive cooling effort can be obtained reducing the infrared radiation absorbed by the cell. An infrared reflecting layer placed on the top of the cell encapsulation should achieve this goal. This solution should reduce the quote of low-energy photons absorbed by the cell, decreasing both the sunlight directly converted into heat and the sunlight converted into excess current by the Germanium layer. Infrared reflecting coverglasses have been successfully tested on space applications. This research focuses on PMMA application for IR reflecting purposes in a terrestrial high CPV system.

Natural allyl sulfur compounds show antiproliferative effects on tumor cells, but the biochemical mechanisms underlying the antitumorigenic properties of the organ sulfur compounds are not yet fully understood. Sodium 2-propenyl-thiosulfate is a garlic water-soluble organo-sulfane sulfur compound able to promote apoptosis in cancer cells, affecting the 'managing' of the redox state in the cell. Our studies show that sodium 2-propenyl-thiosulfate reacts spontaneously with reduced glutathione at physiological pH, leading to the formation of S-allyl-mercapto-glutathione, radicals and peroxyl species, which are able to induce inhibition of enzymes with cysteine in the catalytic site, such as sulfurtransferases. S-Allyl-mercapto-glutathione was purified and characterized by NMR and MS, and its cytotoxic effect at 500 μm on HuT 78 cells was analyzed, showing activation of the p38-MAPK pathway. Many allyl sulfur compounds are also able to promote chemoprevention by induction of xenobiotic-metabolizing enzymes, inducing down-activation or detoxification of the carcinogens. Thus, the effects of the S-allyl-mercapto-glutathione on proteins involved in the cellular detoxification system, such as glutathione S-transferase, have been evaluated both in vitro and in HuT 78 cells. Although the antitumor properties of water-soluble sulfur compounds may arise from several mechanisms and it is likely that more cellular events occur simultaneously, a relevant role is played by the formation of both reduced glutathione conjugates and radical species that affect the activity of the thiol-proteins involved in fundamental cellular processes.

Adult neurogenesis in the dentate gyrus plays a critical role in hippocampus-dependent spatial learning. It remains unknown, however, how new neurons become functionally integrated into spatial circuits and contribute to hippocampus-mediated forms of learning and memory. To investigate these issues, we used a mouse model in which the differentiation of adult-generated dentate gyrus neurons can be anticipated by conditionally expressing the pro-differentiative gene PC3 (Tis21/BTG2) in nestin-positive progenitor cells. In contrast to previous studies that affected the number of newly generated neurons, this strategy selectively changes their timing of differentiation. New, adult-generated dentate gyrus progenitors, in which the PC3 transgene was expressed, showed accelerated differentiation and significantly reduced dendritic arborization and spine density. Functionally, this genetic manipulation specifically affected different hippocampus-dependent learning and memory tasks, including contextual fear conditioning, and selectively reduced synaptic plasticity in the dentate gyrus. Morphological and functional analyses of hippocampal neurons at different stages of differentiation, following transgene activation within defined time-windows, revealed that the new, adult-generated neurons up to 3–4 weeks of age are required not only to acquire new spatial information but also to use previously consolidated memories. Thus, the correct unwinding of these key memory functions, which can be an expression of the ability of adult-generated neurons to link subsequent events in memory circuits, is critically dependent on the correct timing of the initial stages of neuron maturation and connection to existing circuits., Author Summary Previous studies have implicated adult-born hippocampal neurons in the formation of spatial and contextual memories by using mouse models where newly generated neurons are either eliminated or increased in number. Nonetheless, how new neurons are integrated in the existing circuits and contribute to memory formation still awaits clarification. Toward this end, we have developed a different approach, using a mouse model that accelerates the differentiation of the newly generated neurons without altering their number, and offers the possibility to induce the process at any chosen moment. We show that the new neurons pass through their early stages of maturation faster and, though establishing connections with the existing neuronal circuits, fail to function properly. In fact, mice are not only unable to learn new spatial information, but they are also unable to use previously acquired memories. These results demonstrate that the appropriate timing of maturation of new neurons is important for their adult performance in memory circuits, i.e., to integrate new memory traces and recall previous events., The early stages of hippocampal neuron maturation are critical for establishing the connectivity to existing circuits that is required for normal spatial learning and memory.

Histone deacetylase 4 (HDAC4) negatively regulates skeletal myogenesis by associating with the myocyte enhancer factor 2 (MEF2) transcription factors. Our data indicate that the gene PC4 (interferon-related developmental regulator 1 [IFRD1], Tis7), which we have previously shown to be required for myoblast differentiation, is both induced by MyoD and potentiates the transcriptional activity of MyoD, thus revealing a positive regulatory loop between these molecules. Enhancement by PC4 of MyoD-dependent activation of muscle gene promoters occurs selectively through MEF2 binding sites. Furthermore, PC4 localizes in the nucleus of differentiating myoblasts, associates with MEF2C, and is able to counteract the HDAC4-mediated inhibition of MEF2C. This latter action can be explained by the observed ability of PC4 to dose dependently displace HDAC4 from MEF2C. Consistently, we have observed that (i) the region of PC4 that binds MEF2C is sufficient to counteract the inhibition by HDAC4; (ii) PC4, although able to bind HDAC4, does not inhibit the enzymatic activity of HDAC4; and (iii) PC4 overcomes the inhibition mediated by the amino-terminal domain of HDAC4, which associates with MEF2C but not with PC4. Together, our findings strongly suggest that PC4 acts as a coactivator of MyoD and MEF2C by removing the inhibitory effect of HDAC4, thus exerting a pivotal function during myogenesis.
