153. Management and outcome of neonates with a prenatal diagnosis of esophageal atresia type A: A population-based study.

Abstract

Objective: Evaluate the neonatal management and outcomes of neonates with prenatal diagnosis of esophageal atresia (EA) type A.Methods: This population-based study was conducted using data from the French National Register for infants with EA born from 2008 to 2014, including all cases of EA type A. We compared prenatal and neonatal characteristics and outcomes in children with prenatal diagnosis of EA type A with those with a postnatal diagnosis until the age of 1.Results: A total of 1118 live births with EA were recorded among which 88 (7.9%) were EA type A. Prenatal diagnoses were performed in 75 cases (85.2%), and counselling with a prenatal specialist was conducted in 84.8% of the prenatal group. Still within that group, the gestational age at delivery was significantly higher than in the postnatal group (36 [35-38] versus 34 [32-36] weeks; P = .048). Inborn births were more frequent in the prenatal group (86.1% vs 7.7%, P < .0001), and mortality and outcome were similar in both groups.Conclusion: Prenatal diagnosis is high in EA type A, which enables to offer an antenatal parental counseling and which avoids postnatal transfers. Prognosis of EA types A does not appear to be influenced by the prenatal diagnosis. [ABSTRACT FROM AUTHOR]

156. Characterization of the volatile profile of Antarctic bacteria by using solid-phase microextraction-gas chromatography-mass spectrometry

Abstract

Bacteria belonging to the Burkholderia cepacia complex (Bcc) are significant pathogens in Cystic Fibrosis (CF) patients and are resistant to a plethora of antibiotics. In this context, microorganisms from Antarctica are interesting because they produce antimicrobial compounds inhibiting the growth of other bacteria. This is particularly true for bacteria isolated from Antarctic sponges. The aim of this work was to characterize a set of Antarctic bacteria for their ability to produce new natural drugs that could be exploited in the control of infections in CF patients by Bcc bacteria. Hence, 11 bacterial strains allocated to different genera (e.g., Pseudoalteromonas, Arthrobacter and Psychrobacter) were tested for their ability to inhibit the growth of 21 Bcc strains and some other human pathogens. All these bacteria completely inhibited the growth of most, if not all, Bcc strains, suggesting a highly specific activity toward Bcc strains. Experimental evidences showed that the antimicrobial compounds are small volatile organic compounds, and are constitutively produced via an unknown pathway. The microbial volatile profile was obtained by SPME-GC-MS within the m/z interval of 40-450. Solid phase micro extraction technique affords the possibility to extract the volatile compounds in head space with a minimal sample perturbation. Principal component analysis and successive cluster discriminant analysis was applied to evaluate the relationships among the volatile organic compounds with the aim of classifying the microorganisms by their volatile profile. These data highlight the potentiality of Antarctic bacteria as novel sources of antibacterial substances to face Bcc infections in CF patients.

Abstract

The bacterial diversity in three Antarctic lakes located at Crater Cirque (CC), Inexpressible Island (INI) and Luther Peak (LH) in the Victoria Land was investigated by a combination of culture-dependent and culture-independent techniques. The in situ abundance of different bacterial groups was determined by fluorescent in situ hybridization (FISH), while bacterial diversity among the cultivable microflora was investigated by culturing and genetic fingerprinting. Differences in the composition of bacterial assemblages were observed among the lakes investigated, when using the two approaches reported above. Overall, the detection rate by FISH of DAPI-stained cells varied from 48.4% to 68.9% with the general bacterial probe EUB338. Bacteria hybridizing with the group-specific probe CF319a were found to be abundant in the three lakes. A total of 478 strains were isolated from R2A agar plates and grouped by restriction analysis technique; sequencing of representative 16S rDNAs was performed to elucidate the taxonomic positions of isolates. Overall, isolates were placed within five different taxa: ?-Proteobacteria (39%), Bacteroidetes (35%), ?-Proteobacteria (8%), Actinobacteria (7%) and ?-Proteobacteria (5%). Finally, the 5.8% of total isolates shared the highest degree of sequence identity with unclassified bacteria. Members of the ?-Proteobacteria predominated at INI, whereas isolates from CC and LH mainly belonged to the Bacteroidetes. Among the ?-Proteobacteria, the genus Pseudomonas was predominant, whereas Flavobacterium spp. were very common among the Bacteroidetes. Finally, several Actinobacteria were closely related to unknown Antarctic bacteria.
