155. Dietary fiber and bacterial SCFA enhance oral tolerance and protect against food allergy through diverse cellular pathways

Abstract

The incidence of food allergies in western countries has increased dramatically in recent decades. Tolerance to food antigens relies on mucosal CD103(+) dendritic cells (DCs), which promote differentiation of regulatory T (Treg) cells. We show that high-fiber feeding in mice improved oral tolerance and protected from food allergy. High-fiber feeding reshaped gut microbial ecology and increased the release of short-chain fatty acids (SCFAs), particularly acetate and butyrate. High-fiber feeding enhanced oral tolerance and protected against food allergy by enhancing retinal dehydrogenase activity in CD103(+) DC. This protection depended on vitamin A in the diet. This feeding regimen also boosted IgA production and enhanced T follicular helper and mucosal germinal center responses. Mice lacking GPR43 or GPR109A, receptors for SCFAs, showed exacerbated food allergy and fewer CD103(+) DCs. Dietary elements, including fiber and vitamin A, therefore regulate numerous protective pathways in the gastrointestinal tract, necessary for immune non-responsiveness to food antigens.

Abstract

For a variety of reasons, including the common use of deception in psychology experiments, participants often disbelieve experimenters' assertions about important task parameters. This can lead researchers to conclude incorrectly that participants are behaving non-normatively. The problem can be overcome by deriving and testing normative models that do not assume full belief in key task parameters. A real experimental example is discussed.

Abstract

Since 2008, Chiron has been continuously running toxicology and new psychoactive substance (NPS) reference standard-focused synthesis research projects. This work has been carried out in collaboration with the German rapid test provider nal-von-minden and a variety of Nordic institutions: St. Olavs Hospital, The Norwegian Public Health Institute (now Oslo University Hospital), and, more recently, with Linköping University, Sweden. A considerable knowledge-base on systematic NPS drug trend analysis, synthesis of emerging and prophetic NPS reference standards and NPS metabolite reference standards has been accumulated through the EUROSTARS-funded projects QUANTUM SPICE (2013–2016), PSYCHOMICS (2016–2019), and the current NPS REFORM (2019–2022). The aim of this presentation is to provide an overview of what has been learnt and how the NPS market is expected to develop in future. The outcomes of the current and previous EUROSTARS projects will be presented and the future expected developments in the field will be discussed.

Abstract

There is evidence that betting on longshots increases in the last race of a day of horse racing. Previous accounts have assumed that the phenomenon is driven by bettors who have lost money and are trying to recoup their losses. To test this assumption of 'reference dependence,' three laboratory experiments simulated a day at the races: In each of several rounds, participants chose either (i) a gamble with a small probability of a large gain and a large probability of a small loss (the 'longshot') or (ii) a gamble with a moderate chance of a small gain or a small loss (the 'favorite'). The first two experiments employed a game played for points, while a third experiment included monetary incentives and stimuli drawn from a real day of racing. These experiments provide a clear demonstration of the last race effect in a laboratory setting. However, the results indicate that the effect is largely reference independent: Participants were more likely to choose the longshot in the last round regardless of whether, and how much, they had won or lost in previous rounds. Abstract

Contracting Parties to the OSPAR Convention for the Protection of the Maine Environment of the North-East Atlantic are required to undertake monitoring and assessment of both inorganic and organic contaminants. There is a requirement to assess contaminants across different trophic levels on an ecosystem-specific basis. However, this is currently constrained by the availability of relevant samples to cover the full range of trophic levels. This study investigates the variability (inter- and intra-species variation) of the concentrations and distributions of thirty-two polychlorinated biphenyl (PCB) congeners and nine polybrominated diphenyl ether (PBDE) congeners in twenty-six species covering four trophic levels from different geographic locations around Scotland. Trophic magnification factors (TMFs) were calculated using a traditional method and a balanced method for both the ICES-7 PCBs and BDE47, to refine and improve the application of TMFs to assess and predict biomagnification risk to biota in the marine environment. There were clear differences in congener percentage distribution between sample categories and species, with differences influenced by physiological processes and eco-biological parameters. Trophic magnification was found to occur for the ICES-7 PCBs and BDE47 using the traditional method, with the highest degree of trophic magnification reported for CB52. An unbalanced dataset was found to influence the calculated TMF and in some cases, the overall conclusion of the trophic transfer of PCB and PBDE congeners. The balanced method is highly recommended for calculating TMFs to ensure that the TMF is a true indication of the biomagnification potential, particularly when conducting regional comparisons for which sampling requirements are difficult to achieve.
