Abstract

Central serous chorioretinopathy (CSC) is a common disease affecting younger people and may lead to vision loss. CSC shares phenotypic overlap with age-related macular degeneration (AMD). As recent studies have revealed a characteristic increase of choroidal thickness in CSC, we conducted a genome-wide association study on choroidal thickness in 3,418 individuals followed by TaqMan assays in 2,692 subjects, and we identified two susceptibility loci: CFH rs800292, an established AMD susceptibility polymorphism, and VIPR2 rs3793217 (P = 2.05 × 10-10 and 6.75 × 10-8, respectively). Case-control studies using patients with CSC confirmed associations between both polymorphisms and CSC (P = 5.27 × 10-5 and 5.14 × 10-5, respectively). The CFH rs800292 G allele is reportedly a risk allele for AMD, whereas the A allele conferred risk for thicker choroid and CSC development. This study not only shows that susceptibility genes for CSC could be discovered using choroidal thickness as a defining variable but also, deepens the understanding of differences between CSC and AMD pathophysiology.

Abstract

The Simons Observatory (SO) is a set of cosmic microwave background instruments that will be deployed in the Atacama Desert in Chile. The key science goals include setting new constraints on cosmic inflation, measuring large scale structure with gravitational lensing, and constraining neutrino masses. Meeting these science goals with SO requires high sensitivity and improved calibration techniques. In this paper, we highlight a few of the most important instrument calibrations, including spectral response, gain stability, and polarization angle calibrations. We present their requirements for SO and experimental techniques that can be employed to reach those requirements

Abstract

Atmosphere is one of the most important noise sources for ground-based cosmic microwave background (CMB) experiments. By increasing optical loading on the detectors, it amplifies their effective noise, while its fluctuations introduce spatial and temporal correlations between detected signals. We present a physically motivated 3D-model of the atmosphere total intensity emission in the millimeter and sub-millimeter wavelengths. We derive a new analytical estimate for the correlation between detectors time-ordered data as a function of the instrument and survey design, as well as several atmospheric parameters such as wind, relative humidity, temperature and turbulence characteristics. Using an original numerical computation, we examine the effect of each physical parameter on the correlations in the time series of a given experiment. We then use a parametric-likelihood approach to validate the modeling and estimate atmosphere parameters from the polarbear-i project first season data set. We derive a new 1.0% upper limit on the linear polarization fraction of atmospheric emission. We also compare our results to previous studies and weather station measurements. The proposed model can be used for realistic simulations of future ground-based CMB observations.

Abstract

We constrain anisotropic cosmic birefringence using four-point correlations of even-parity E-mode and odd-parity B-mode polarization in the cosmic microwave background measurements made by the POLARization of the Background Radiation (POLARBEAR) experiment in its first season of observations. We find that the anisotropic cosmic birefringence signal from any parity-violating processes is consistent with zero. The Faraday rotation from anisotropic cosmic birefringence can be compared with the equivalent quantity generated by primordial magnetic fields if they existed. The POLARBEAR nondetection translates into a 95% confidence level (C.L.) upper limit of 93 nanogauss (nG) on the amplitude of an equivalent primordial magnetic field inclusive of systematic uncertainties. This four-point correlation constraint on Faraday rotation is about 15 times tighter than the upper limit of 1380 nG inferred from constraining the contribution of Faraday rotation to two-point correlations of B-modes measured by Planck in 2015. Metric perturbations sourced by primordial magnetic fields would also contribute to the B-mode power spectrum. Using the POLARBEAR measurements of the B-mode power spectrum (two-point correlation), we set a 95% C.L. upper limit of 3.9 nG on primordial magnetic fields assuming a flat prior on the field amplitude. This limit is comparable to what was found in the Planck 2015 two-point correlation analysis with both temperature and polarization. We perform a set of systematic error tests and find no evidence for contamination. This work marks the first time that anisotropic cosmic birefringence or primordial magnetic fields have been constrained from the ground at subdegree scales.

Abstract

We report an improved measurement of the cosmic microwave background (CMB) $B$-mode polarization power spectrum with the POLARBEAR experiment at 150 GHz. By adding new data collected during the second season of observations (2013-2014) to re-analyzed data from the first season (2012-2013), we have reduced twofold the band-power uncertainties. The band powers are reported over angular multipoles $500 \leq \ell \leq 2100$, where the dominant $B$-mode signal is expected to be due to the gravitational lensing of $E$-modes. We reject the null hypothesis of no $B$-mode polarization at a confidence of 3.1$\sigma$ including both statistical and systematic uncertainties. We test the consistency of the measured $B$-modes with the $\Lambda$ Cold Dark Matter ($\Lambda$CDM) framework by fitting for a single lensing amplitude parameter $A_L$ relative to the Planck best-fit model prediction. We obtain $A_L = 0.60 ^{+0.26} _{-0.24} ({\rm stat}) ^{+0.00} _{-0.04}({\rm inst}) \pm 0.14 ({\rm foreground}) \pm 0.04 ({\rm multi})$, where $A_{L}=1$ is the fiducial $\Lambda$CDM value, and the details of the reported uncertainties are explained later in the manuscript., Comment: 16 pages, 10 figures. Abstract

Almost all experimental investigations on concepts of time (T), distance (D) and velocity (V) since 1946 have been based on the Piagetian theory. However, there are several controversial points in Piaget's investigations. In the experiment described in this paper, developmental changes of concepts concerning relation of T, D, and V were examined using 148 children from 4- to 10-years-old and 20 undergardaute students, with the aim of addressing the controversy in Piaget's investigations. Results of these investigations include: (1) almost all children over 5 years of age could understand the proportional relation between D and T, and V and D through concrete operation, though they could not verbalize the reasons for their understanding; (2) it was difficult for young children to understand the inversely proportional relation between V and T even through concrete operations, but, the verbalization of reasons developed rather smoothly and rapidly after age 8; (3) in young children under 6-and-a-half-years-old, all relations between two variables seemed to be proportional and they seemed to pay little attention to the third constant variable; (4) over 8-years-old, most children correctly understood two proportional relations and one inversely proportional relation; and (5) it is not clear if children over 9 had the intercorrelational concept among three variables. Abstract

Bilateral neovascular age-related macular degeneration (AMD) causes much more handicaps for patients than unilateral neovascular AMD. Although several AMD-susceptibility genes have been evaluated for their associations to bilaterality, genome-wide association study (GWAS) on bilaterality has been rarely reported. In the present study, we performed GWAS using neovascular AMD cases in East Asian. The discovery stage compared 581,252 single nucleotide polymorphisms (SNPs) between 803 unilateral and 321 bilateral Japanese cases but no SNP showed genome-wide significance, while SNPs at six regions showed P-value < 1.0 × 10-5, STON1-GTF2A1L/LHCGR/FSHR, PLXNA1, CTNNA3, ARMS2/HTRA1, LHFP, and FLJ38725. The first replication study for these six regions comparing 36 bilateral and 132 unilateral Japanese cases confirmed significant associations of rs4482537 (STON1-GTF2A1L/LHCGR/FSHR), rs2284665 (ARMS2/HTRA1), and rs8002574 (LHFP) to bilaterality. In the second replication study comparing 24 bilateral and 78 unilateral cases from Singapore, rs4482537 (STON1-GTF2A1L/LHCGR/FSHR) only showed significant association. Meta-analysis of discovery and replication studies confirmed genome-wide level significant association (P = 2.61 × 10-9) of rs4482537 (STON1-GTF2A1L/LHCGR/FSHR) and strong associations (P = 5.76 × 10-7 and 9.73 × 10-7, respectively) of rs2284665 (ARMS2/HTRA1) and rs8002574 (LHFP). Our GWAS for neovascular AMD bilaterality found new genetic loci STON1-GTF2A1L/LHCGR/FSHR and confirmed the previously reported association of ARMS2/HTRA1.

Abstract

Angiotensin-I converting enzyme (ACE) occupies a pivotal role in cardiovascular homeostasis. Major loci for plasma ACE have been identified at ACE on Chromosome 17 and at ABO on Chromosome 9. We sought to characterise the genetic architecture of plasma ACE at finer resolution in two populations. We carried out a GWAS in 1810 individuals of Japanese ethnicity; this identified signals at ACE and ABO that together accounted for nearly half of the population variability of the trait. We conducted measured haplotype analysis at the ABO locus in 1425 members of 248 British families using haplotypes of three SNPs, which together tagged the alleles responsible for the principal blood group antigens A1, A2, B and O. Type O alleles were associated with intermediate plasma ACE activity compared to Type A1 alleles (in whom plasma ACE activity was ̃36% lower) and Type B alleles (in whom plasma ACE activity was ̃36% higher). We demonstrated heterogeneity among A alleles: A2 alleles were associated with plasma ACE activity that was very similar to the O alleles. Variation at ACE accounted for 35% of the trait variance, and variation at ABO accounted for 15%. A further 10% could be ascribed to polygenic effects. © 2013 John Wiley and Sons Ltd.

Abstract

We carried out a genome-wide association study of lung cancer (3,259 cases and 4,159 controls), followed by replication in 2,899 cases and 5,573 controls. Two uncorrelated disease markers at 5p15.33, rs402710 and rs2736100 were detected by the genome-wide data (P = 2 × 10-7 and P = 4 × 10-6) and replicated by the independent study series (P = 7 × 10-5 and P = 0.016). The susceptibility region contains two genes, TERT and CLPTM1L, suggesting that one or both may have a role in lung cancer etiology. © 2008 Nature Publishing Group.

Abstract

We describe the design of a cryogenic rotation stage (CRS) for use with the cryogenic half-wave plate (CHWP) polarization modulator on the POLARBEAR-2b and POLARBEAR-2c (PB2b/c) cosmic microwave background (CMB) experiments, the second and third installments of the Simons Array. Rapid modulation of the CMB polarization signal using a CHWP suppresses 1/f contamination due to atmospheric turbulence and allows a single polarimeter to measure both polarization states, mitigating systematic effects that arise when differencing orthogonal detectors. To modulate the full detector array while avoiding excess photon loading due to thermal emission, the CHWP must have a clear-aperture diameter of > 450 mm and be cooled to < 100 K. We have designed a 454 mm clear-aperture, < 65 K CRS using a superconducting magnetic bearing driven by a synchronous magnetic motor. We present the specifications for the CRS, its interfacing to the PB2b/c receiver cryostat, its performance in a stand-alone test, and plans for future work. [ABSTRACT FROM AUTHOR]

Abstract

POLARBEAR-2 (PB-2) is a cosmic microwave background (CMB) polarization experiment that will be located in the Atacama highland in Chile at an altitude of 5200 m. Its science goals are to measure the CMB polarization signals originating from both primordial gravitational waves and weak lensing. PB-2 is designed to measure the tensor to scalar ratio, r, with precision {\sigma}(r) < 0.01, and the sum of neutrino masses, {\Sigma}m{\nu}, with {\sigma}({\Sigma}m{\nu}) < 90 meV. To achieve these goals, PB-2 will employ 7588 transition-edge sensor bolometers at 95 GHz and 150 GHz, which will be operated at the base temperature of 250 mK. Abstract

The polarbear cosmic microwave background (CMB) polarization experiment has been observing since early 2012 from its 5,200 m site in the Atacama Desert in Northern Chile. polarbear's measurements will characterize the expected CMB polarization due to gravitational lensing by large scale structure, and search for the possible B-mode polarization signature of inflationary gravitational waves. polarbear's 250 mK focal plane detector array consists of 1,274 polarization-sensitive antenna-coupled bolometers, each with an associated lithographed band-defining filter and contacting dielectric lenslet, an architecture unique in current CMB experiments. The status of the polarbear instrument, its focal plane, and the analysis of its measurements are presented. © 2014 Springer Science+Business Media New York.

Abstract

Gravitational lensing due to the large-scale distribution of matter in the cosmos distorts the primordial Cosmic Microwave Background (CMB) and thereby induces new, small-scale $B$-mode polarization. This signal carries detailed information about the distribution of all the gravitating matter between the observer and CMB last scattering surface. We report the first direct evidence for polarization lensing based on purely CMB information, from using the four-point correlations of even- and odd-parity $E$- and $B$-mode polarization mapped over $\sim30$ square degrees of the sky measured by the POLARBEAR experiment. These data were analyzed using a blind analysis framework and checked for spurious systematic contamination using null tests and simulations. Evidence for the signal of polarization lensing and lensing $B$-modes is found at 4.2$\sigma$ (stat.+sys.) significance. The amplitude of matter fluctuations is measured with a precision of $27\%$, and is found to be consistent with the Lambda Cold Dark Matter ($\Lambda$CDM) cosmological model. This measurement demonstrates a new technique, capable of mapping all gravitating matter in the Universe, sensitive to the sum of neutrino masses, and essential for cleaning the lensing $B$-mode signal in searches for primordial gravitational waves., Comment: 7 pages, 3 figures. Abstract

POLARBEAR-2 is a next-generation receiver for precision measurements of the polarization of the cosmic microwave background (Cosmic Microwave Background (CMB)). Scheduled to deploy in early 2015, it will observe alongside the existing POLARBEAR-1 receiver, on a new telescope in the Simons Array on Cerro Toco in the Atacama desert of Chile. For increased sensitivity, it will feature a larger area focal plane, with a total of 7,588 polarization sensitive antenna-coupled Transition Edge Sensor (TES) bolometers, with a design sensitivity of 4.1 uKrt(s). The focal plane will be cooled to 250 milliKelvin, and the bolometers will be read-out with 40x frequency domain multiplexing, with 36 optical bolometers on a single SQUID amplifier, along with 2 dark bolometers and 2 calibration resistors. To increase the multiplexing factor from 8x for POLARBEAR-1 to 40x for POLARBEAR-2 requires additional bandwidth for SQUID readout and well-defined frequency channel spacing. Extending to these higher frequencies requires new components and design for the LC filters which define channel spacing. The LC filters are cold resonant circuits with an inductor and capacitor in series with each bolometer, and stray inductance in the wiring and equivalent series resistance from the capacitors can affect bolometer operation. We present results from characterizing these new readout components. Integration of the readout system is being done first on a small scale, to ensure that the readout system does not affect bolometer sensitivity or stability, and to validate the overall system before expansion into the full receiver. Abstract

The companion paper describes a measurement of polarization lensing in cross-correlation; Primary fluctuations in both temperature and polarization of the Cosmic Microwave Background (CMB) reflect the properties of the Universe from the Big Bang until the photons decoupled from matter 380,000 years later. These primary fluctuations are then lensed by large-scale structures (such as clusters of galaxies and filaments of dark matter), with the result that the distribution and properties of dark matter, including the masses of neutrinos, can be determined more accurately by extracting the lensing information than through studying the primary fluctuations alone. Polarization lensing can give cleaner, higher resolution results than temperature lensing. The correlation of lensed CMB polarization with large-scale structure, traced through the Cosmic Infrared Background, was recently detected; however, this correlation does not trace all structure and depends on the relationship between the infrared flux from the galaxies and the underlying mass distribution. Here we report the detection of gravitational lensing directly from CMB polarization measurements. With these data, we have made a census of essentially all structure integrated along the line of sight through the full depth of the observable Universe on 30 square degrees of the sky, and we find good agreement with expectations from the standard Lambda cold-dark matter cosmology.

Abstract

We constrain anisotropic cosmic birefringence using four-point correlations of even-parity E-mode and odd-parity B-mode polarization in the cosmic microwave background measurements made by the POLARization of the Background Radiation (POLARBEAR) experiment in its first season of observations. We find that the anisotropic cosmic birefringence signal from any parity-violating processes is consistent with zero. The Faraday rotation from anisotropic cosmic birefringence can be compared with the equivalent quantity generated by primordial magnetic fields if they existed. The POLARBEAR nondetection translates into a 95% confidence level (C.L.) upper limit of 93 nanogauss (nG) on the amplitude of an equivalent primordial magnetic field inclusive of systematic uncertainties. This four-point correlation constraint on Faraday rotation is about 15 times tighter than the upper limit of 1380 nG inferred from constraining the contribution of Faraday rotation to two-point correlations of B-modes measured by Planck in 2015. Metric perturbations sourced by primordial magnetic fields would also contribute to the B-mode power spectrum. Using the POLARBEAR measurements of the B-mode power spectrum (two-point correlation), we set a 95% C.L. upper limit of 3.9 nG on primordial magnetic fields assuming a flat prior on the field amplitude. This limit is comparable to what was found in the Planck 2015 two-point correlation analysis with both temperature and polarization. We perform a set of systematic error tests and find no evidence for contamination. This work marks the first time that anisotropic cosmic birefringence or primordial magnetic fields have been constrained from the ground at subdegree scales.

Abstract

We present an overview of the design and status of the \Pb-2 and the Simons Array experiments. \Pb-2 is a Cosmic Microwave Background polarimetry experiment which aims to characterize the arc-minute angular scale B-mode signal from weak gravitational lensing and search for the degree angular scale B-mode signal from inflationary gravitational waves. The receiver has a 365~mm diameter focal plane cooled to 270~milli-Kelvin. The focal plane is filled with 7,588 dichroic lenslet-antenna coupled polarization sensitive Transition Edge Sensor (TES) bolometric pixels that are sensitive to 95~GHz and 150~GHz bands simultaneously. The TES bolometers are read-out by SQUIDs with 40 channel frequency domain multiplexing. Refractive optical elements are made with high purity alumina to achieve high optical throughput. The receiver is designed to achieve noise equivalent temperature of 5.8~$\mu$K$_{CMB}\sqrt{s}$ in each frequency band. \Pb-2 will deploy in 2016 in the Atacama desert in Chile. The Simons Array is a project to further increase sensitivity by deploying three \Pb-2 type receivers. The Simons Array will cover 95~GHz, 150~GHz and 220~GHz frequency bands for foreground control. Abstract

Atmosphere is one of the most important noise sources for ground-based cosmic microwave background (CMB) experiments. By increasing optical loading on the detectors, it amplifies their effective noise, while its fluctuations introduce spatial and temporal correlations between detected signals. We present a physically motivated 3d-model of the atmosphere total intensity emission in the millimeter and sub-millimeter wavelengths. We derive a new analytical estimate for the correlation between detectors time-ordered data as a function of the instrument and survey design, as well as several atmospheric parameters such as wind, relative humidity, temperature and turbulence characteristics. Using an original numerical computation, we examine the effect of each physical parameter on the correlations in the time series of a given experiment. We then use a parametric-likelihood approach to validate the modeling and estimate atmosphere parameters from the POLARBEAR-I project first season data set. We derive a new 1.0% upper limit on the linear polarization fraction of atmospheric emission. We also compare our results to previous studies and weather station measurements. Abstract

Age-related macular degeneration (AMD) is a major cause of blindness, but presents differently in Europeans and Asians. Here, we perform a genome-wide and exome-wide association study on 2,119 patients with exudative AMD and 5,691 controls, with independent replication in 4,226 patients and 10,289 controls, all of East Asian descent, as part of The Genetics of AMD in Asians (GAMA) Consortium. We find a strong association between CETP Asp442Gly (rs2303790), an East Asian-specific mutation, and increased risk of AMD (odds ratio (OR)=1.70, P=5.60 × 10(-22)). The AMD risk allele (442Gly), known to protect from coronary heart disease, increases HDL cholesterol levels by 0.17 mmol l(-1) (P=5.82 × 10(-21)) in East Asians (n=7,102). We also identify three novel AMD loci: C6orf223 Ala231Ala (OR=0.78, P=6.19 × 10(-18)), SLC44A4 Asp47Val (OR=1.27, P=1.08 × 10(-11)) and FGD6 Gln257Arg (OR=0.87, P=2.85 × 10(-8)). Our findings suggest that some of the genetic loci conferring AMD susceptibility in East Asians are shared with Europeans, yet AMD in East Asians may also have a distinct genetic signature.

Abstract

Anti-thyrotropin (TSH) receptor autoantibodies (TRAbs) have been known to be involved in Graves' disease. To understand the molecular mechanism for pathogenesis of TSAbs in Graves' disease, we isolated and reconstituted the Ig genes of EBV-transformed B cell clones producing monoclonal thyroid stimulating Ab (TSAb) obtained from patients with Graves' disease. The V region genes of Ig heavy (H) and light (L) chains of two TSAb clones, IgG clone B6B7 and IgM clone 101-2, were isolated by the PCR. Nucleotide sequencing analysis revealed that germ-line VH and VK segments widely used for autoantibodies including the previously isolated TRAbs were utilized in the two clones. A significant number of somatic mutations were found in V regions of both clones, indicating the involvement of somatic mutations for the TSAb specificity. Reconstituted Ig H and L chain genes of the two clones were stably introduced into myeloma cells for IgG1 production. IgGs purified from cultured supernatants of both transfectants exhibited significant TSAb activities, while they did not inhibit TSH binding to the receptor. The successful expression of recombinant TSAbs in eukaryotic cells will provide opportunities to apply them to various pathophysiologic, diagnostic and therapeutic investigations in autoimmune thyroid diseases.

Abstract

We present the design and characterization of the POLARBEAR experiment. POLARBEAR will measure the polarization of the cosmic microwave background (CMB) on angular scales ranging from the experiment's 3.5 arcminute beam size to several degrees. The experiment utilizes a unique focal plane of 1,274 antenna-coupled, polarization sensitive TES bolometers cooled to 250 milliKelvin. Employing this focal plane along with stringent control over systematic errors, POLARBEAR has the sensitivity to detect the expected small scale B-mode signal due to gravitational lensing and search for the large scale B-mode signal from inflationary gravitational waves. POLARBEAR was assembled for an engineering run in the Inyo Mountains of California in 2010 and was deployed in late 2011 to the Atacama Desert in Chile.
