Abstract

Nanoadjuvants that combine immunostimulatory properties and delivery systems reportedly bestow major improvements on the efficacy of recombinant, protein-based vaccines. Among these, self-assembled micellar formulations named ISCOMs (immune stimulating complexes) show a great ability to trigger powerful immunological responses against infectious pathogens. Here, a nanoadjuvant preparation, based on saponins from Quillaja brasiliensis, was evaluated together with an experimental Zika virus (ZIKV) vaccine (IQB80-zEDIII) and compared to an equivalent vaccine with alum as the standard adjuvant. The preparations were administered to mice in two doses (on days zero and 14) and immune responses were evaluated on day 28 post-priming. Serum levels of anti-Zika virus IgG, IgG1, IgG2b, IgG2c, IgG3 were significantly increased by the nanoadjuvant vaccine, compared to the mice that received the alum-adjuvanted vaccine or the unadjuvanted vaccine. In addition, a robust production of neutralizing antibodies and in vitro splenocyte proliferative responses were observed in mice immunized with IQB80-zEDIII nanoformulated vaccine. Therefore, the IQB80-zEDIII recombinant preparation seems to be a suitable candidate vaccine for ZIKV. Overall, this study identified saponin-based delivery systems as an adequate adjuvant for recombinant ZIKV vaccines and has important implications for recombinant protein-based vaccine formulations against other flaviviruses and possibly enveloped viruses.

Abstract

Here diffusion and Ware pinch are analyzed as opposed effects for plasma confinement, when instabilities are not considered. In this work it is studied the equilibrium inductive electric field where both effects annul each other in the sense that the average normal velocity is zero, that is, marginal velocity confinement is reached. The critical electric field defined in that way is studied for different values of elliptic elongation, Shafranov shift and triangularity. A short complementary analysis is also performed of the variation of the poloidal magnetic field along a magnetic line. Magnetohydrodynamic transport theory in the collisional regime is used as in recent publications. Abstract

In this paper we introduce an algebraic form of the dispersion relation for a non ideal inhomogeneous dusty plasma in order to improve drastically the calculation of the drift instability growth rate. This method makes use of the multipole approximation of the Z dispersion function, previously published, and valid for the entire range. A careful analysis of the solutions spectra of this kind of polynomial equation permits us to calculate easily the growth rate of the drift instability for the ion-dust and dust acoustic mode. The value of the parallel to magnetic field wavelength for which the instability reaches the maximal value is carefully localized and discussed. AbstractThe first occurrence of Leptusmites parasitizing different species of stingless bees is reported. The samples were collected in the Argentinian province of Misiones on flowers and managed stingless bee colonies. Although the frequency registered was very low, the current report is relevant as it will drive research efforts on the understanding of parasite-host dynamics, consequences at the community level, and disease management in wild and productive contexts.

198. Training Characteristics and Competitive Demands in Women Road Cyclists: A Systematic Review

Abstract

Purpose: To identify the main training characteristics and competitive demands in women’s road cycling. Methods: A systematic search was conducted on 5 databases according to PRISMA (Preferred Reporting Items for Systematic Review and MetaAnalysis) guidelines. The articles had to be primary studies, written after 1990 with a sample of competitive women between the ages of 15 and 50. The Quality Assessment Tool for Quantitative Studies and the Oxford Levels of Evidence scales were used. Results: The search yielded 1713 articles, of which 20 were included. Studies on training and competitive demands (n = 5) found that both external and internal loads are higher in women than in men. Studies on strength and endurance training (n = 5) showed that both velocity-based and heavy-load strength training programs performed at least 2 days per week and including 3 to 4 lowerbody exercises improved performance. Altitude-training studies (n = 3) found that “Live High–Train Low” was effective to increase performance during the first 9 days after the training camp. The 7 remaining studies focused on a range of topics. The methodological quality was strong for 12 studies and moderate for 8. In contrast, the level of evidence was high in 7 and low in the other 13. Conclusions: Endurance training and competitive demands in women’s road cycling are higher than those of men. Strength training is effective in women when the frequency, intensity, and number of exercises are appropriate, while altitude training should be completed a few days before competing. Abstract

The research aims to give new insights on the effect of administering selected bacterial strains, isolated from honey bee gut, and/or a commercial plant extract blend ( HiveAlive®) on Nosema ceranae. Analyses were first performed under laboratory conditions such as different infective doses of N. ceranae, the effect of single strains and their mixture and the influence of pollen administration. Daily survival and feed consumption rate were recorded and pathogen development was analysed using qPCR and microscope counts. Biomarkers of immunity and physiological status were also evaluated for the different treatments tested using one bacterial strain, a mixture of all the bacteria and/or a plant extract blend as treatments. The results showed an increase of abaecin transcript levels in the midgut of the honey bees treated with the bacterial mixture and an increased expression of the protein vitellogenin in the haemolymph of honey bees treated with two separate bacterial strains (Bifidobacterium coryneforme and Apilactobacillus kunkeei). A significant effectiveness in reducing N. ceranae was shown by the bacterial mixture and the plant extract blend regardless of the composition of the diet. This bioactivity was seasonally linked. Quantitative PCR and microscope counts showed the reduction of N. ceranae under different experimental conditions. The antiparasitic efficacy of the treatments at field conditions was studied using a semi-field approach which was adapted from research on insecticides for the first time, to analyse antiparasitic activity against N. ceranae. The approach proved to be reliable and effective in validating data obtained in the laboratory.
