BACKGROUND:Plasmodium falciparum parasitization of erythrocytes causes a substantial increase in the levels of intracellular fatty acids, notably oleic acid. How parasites acquire this monounsaturated fatty acid has remained enigmatic. Here, we report on the biochemical and enzymatic characterization of stearoyl-CoA desaturase (SCD) in P. falciparum. METHODOLOGY/PRINCIPAL FINDINGS:Metabolic labeling experiments allowed us to demonstrate the production of oleic acid from stearic acid both in lysates of parasites incubated with [(14)C]-stearoyl-CoA and in parasite-infected erythrocytes labeled with [(14)C]-stearic acid. Optimal SCD activity was detected in schizonts, the stage of maximal membrane synthesis. This activity correlated with a late trophozoite stage-specific induction of PFE0555w transcripts. PFE0555w harbors a typical SCD signature. Similar to mammalian SCDs, this protein was found to be associated with the endoplasmic reticulum, as determined with PFE0555w-GFP tagged transgenic P. falciparum. Importantly, these parasites exhibited increased rates of stearic to oleic acid conversion, providing additional evidence that PFE0555w encodes the plasmodial SCD (PfSCD). These findings prompted us to assess the activity of sterculic acid analogues, known to be specific Delta9-desaturase inhibitors. Methyl sterculate inhibited the synthesis of oleic acid both with parasite lysates and infected erythrocytes, most likely by targeting PfSCD. This compound exhibited significant, rapid and irreversible antimalarial activity against asexual blood stages. This parasiticidal effect was antagonized by oleic acid. CONCLUSION/SIGNIFICANCE:Our study provides evidence that parasite-mediated fatty acid modification is important for blood-stage survival and provides a new strategy to develop a novel antimalarial therapeutic based on the inhibition of PfSCD.

Diazocyclopentadiene (1) was photolyzed in N(2), CO, and other low-temperature matrices. The resulting carbene, cyclopentadienylidene (2), was characterized by its UV and IR spectra, and its thermal dimerization and reaction with CO were observed. Photolysis of 1 with plane-polarized light gave matrices exhibiting linear dichroism. Comparison of dichroic IR and UV spectra revealed that the photolysis proceeds via an excited A(l) state of the diazo compound. Plane-polarized irradiation of the corresponding ketene (4) in CO matrices resulted in photoreorientation of the molecules of 4 without significant loss.

MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) elicits motor deficits similar to those observed in Parkinson's disease. Before exerting its neurotoxic action, MPTP must be activated by brain monoamine oxidase (MAO) to the neurotoxic metabolite MPP+ (1-methyl-4-phenylpyridinium). MPTP derivatives differ in their reactivity as MAO substrates and in their neurotoxicity. A structure-reactivity relationship study based on literature data was undertaken in order to determine the key features in the structure of MPTP and analogs that are responsible for the reactivity towards MAO. Thirty-three MPTP derivatives (including MPTP itself) were included in the study. To explain the reactivity towards MAO of the 33 MPTP analogs, different statistical methods (principal component analysis, multiple linear regression analysis) as well as the CoMFA (Comparative Molecular Field Analysis) approach, a new tool in structure-activity correlations, were used. Linear regression analysis failed to yield any predictive model, but suggested some trends. In contrast, the CoMFA approach was successful in correlating structural features and MAO reactivity. Coefficient contour maps showed where differences in the steric field (van der Waals' interactions) are most highly associated with differences in MAO reactivity. Several positive (in the ortho- and meta-position of the phenyl group) and negative (in the para-position of the phenyl group; beyond the N-methyl group) interaction regions were identified. Some structural features of the MAO active site could be postulated. First, the N-methyl group has the ideal size and elicits ideal interactions within the MAO active pocket, while smaller or larger groups are less favorable; second, para-substituent on the phenyl ring produce steric hindrances and are unfavorable to reactivity; third, ortho- and meta-substituents may have stabilizing interactions within the active pocket and are favorable to the reactivity. Moreover the model derived by CoMFA allowed us to make successful predictions of reactivity towards MAO for several additional tetrahydropyridines.

Reaction of a number of 2-(cycloprop-1-en-1-yl)ethanol derivatives with bromine, acid or silver ion leads to ring expansion, either to 5,6-dihydro 2H-pyrans or to 3-methylenetetrahydrofurans.

Mycolic acids (MAs) are a major component of the cell walls of Mycobacterium tuberculosis and related organisms. These alpha-alkyl beta-hydroxy long fatty acids have been the subject of numerous studies for their immunological properties. We previously reported that an interaction between cholesterol and mycolic acids could be responsible for the low accuracy in the serodiagnosis of TB when using free mycolic acid in an ELISA assay. The aim of this work was to investigate if this interaction could be due to a similarity in the structural properties between mycolic acids and cholesterol. The investigation revealed that patient sera cross-reacted with mycolic acids and cholesterol in an ELISA experiment suggesting that both molecules may present related functionality in a similar structural orientation. This relation was further supported by the interaction of mycolic acids with Amphotericin B (AmB), a known binding agent to ergosterol and cholesterol. Using a resonant mirror biosensor, we observed that AmB recognised both cholesterol and mycolic acids. In addition, a specific attraction was observed between mycolic acid and cholesterol by the accumulation of cholesterol from liposomes in suspension onto immobilized mycolic acids containing liposomes, detected with a biosensor technique. Combined, these results suggest that mycolic acids can assume a three-dimensional conformation similar to a sterol. This requires that mycolic acid exposes its hydroxyl group and assumes rigidity in its chain structure to generate a hydrophobic surface topology matching that of cholesterol. A particular folded conformation would be required for this, of which a few different types have already been proven to exist in monolayers of mycolic acids.

In this study, we investigated the physicochemical properties of the cellulosic preparations obtained from both untreated perennial ryegrass leaves and de-juiced leaves. It was found that treatment at 22 degrees C with 18% NaOH and 18% KOH for 2h, and 10% NaOH and 10% KOH for 16 h yielded 28.2%, 28.8%, 22.7%, 23.4%, respectively, of 'cellulose' residue from untreated ryegrass leaves and 35.7%, 36.8%, 32.8% and 34.6%, respectively, from the de-juiced leaves. For each cellulosic fraction, the glucose content was 71.6%, 69.6%, 67.8%, 66.7%, 69.7%, 68.6%, 63.9% and 61.7%, respectively. The structure of the cellulose samples was examined using FTIR and CP/MAS (13)C NMR spectroscopy and X-ray diffraction. The cellulosic preparations were free of bound lignin except for noticeable amounts of residual hemicelluloses (28.4-38.3%), and had intrinsic viscosities between 275.1 and 361.0 mL/g, along with molecular weights from 144,130 to 194,930 g/mol. This study found that the cellulose samples isolated from both de-juiced ryegrass leaves and the untreated leaves had a much lower percent crystallinity (33.0-38.6%) than that from wood-based fibres (60-70%) and had much shorter fibres (0.35-0.49 mm) than those of either cereal straws, bagasse or wood. In addition, a partial disruption of the hydrogen bonds and microfibrils may occur during the de-juicing process by mechanical activity, which results in a decreased cellulose crystallinity and fibre length. These findings are significant in relation to hydrolysing ryegrass cellulose for bio-ethanol production.

Sequential three-stage treatments with 80% EtOH containing 0.2% NaOH, 2.5% H 2 O 2 –0.2% EDTA containing 1.5% NaOH and 2.5% H 2 O 2 –0.2% TAED containing 1.0% NaOH at 75 °C for 3 h released 8.0% and 10.4%, 79.1% and 77.0% and 12.9% and 12.5% of the original hemicelluloses from perennial grass and cocksfoot grass, respectively. It was found that the four alkaline peroxide-soluble hemicellulosic fractions contained higher amounts of xylose (33.4–38.2%), uronic acids (9.3–15.3%) and rhamnose (3.0–3.9%), but were lower in glucose (25.1–28.3%), galactose (13.3–15.3%) and mannose (0.4–1.5%) than those of the two alkaline EtOH-soluble hemicellulosic fractions in which glucose (32.9–36.0%), xylose (20.1–22.6%), arabinose (14.1–21.4%), galactose (16.6–19.9%), mannose (4.1–9.9%) and uronic acids (3.4–7.4%) were the major sugar components. 13 C NMR spectroscopy confirmed that all the six hemicellulosic fractions were composed of galactoarabinoxylans, 4- O -methylglucuronoarabinoxylans and β-glucan. In addition, the studies showed that the four alkaline peroxide-soluble hemicellulosic fractions were more linear and acidic and had larger molecular weights ( M w , 28,400–38,650 g mol −1 ) than those of the two alkaline EtOH-soluble hemicellulosic fractions ( M w , 16,460–17,420 g mol −1 ).

1-Trimethylsilyl-3-phenylcyclopropene undergoes a highly stereocontrolled ene-reaction to give a dimer and further reaction leads to one or more trimers derived through two ene-reactions.

Tuberculostearic acid (10R-methyloctadecanoic acid) and its 10S-enantiomer were synthesised by a chiral pool strategy, in four steps from citronellyl bromide.

Selective methods for the preparation of 3-phenylcyclopropene and its 1-substituted derivatives are provided. The parent cyclo- propene is readily trapped in (3+2)- and (4+2)-cycloadditions that lead toexo-3-phenyl-1,2-disubstituted cyclopropanes.Ab initiocalculations suggest that the lowest energy conformation has the plane of the benzene ring perpendicular to the cyclopropeneπ-bond but with a low rotation barrier.

Seven residual hemicellulosic preparations (19.6–45.0% of the original hemicelluloses) were extracted from wheat straw pre-treated with various organic solvents using 1.8% H2O2–0.18% cyanamide at 50 °C and pH 10.0 for 4 h. Their chemical compositions and physicochemical properties were determined using GC, HPLC, GPC, FT-IR and 13NMR spectroscopy. The results indicated that all the residual hemicellulosic preparations were heteropolysaccharides containing xylose, glucose, arabinose, galactose, mannose, rhamnose and 4-O-methyl-α- d -glucopyranosyluronic acid. The predominant monosaccharide was xylose, ranging between 67.7% and 81.9% of the total neutral sugars, composed mainly of l -arabino-(4-O-methyl- d -glucurono)- d -xylan. The content of contaminant lignin in the isolated residual hemicelluloses was 2.89–5.31%. The Mw values of the two residual hemicellulosic preparations H6 and H7 (42,710 and 44,080 g mol−1, respectively) obtained from the aqueous-alcohol pre-treated straw were much higher than those of H1–H5 (12,980–15,950 g mol−1) extracted from the organic acid pre-treated straw.

A 23-26-carbon chain length range of omega-19 (1'R,2'S) cyclopropane fatty acids, related to mycobacterial mycolic acids, has been prepared. The key cyclopropyl intermediate, (1'R,2'S)-(Z)-1-formyl-2-octadecylcyclopropane, underwent Wittig chemistry with various reagents to provide vinylic precursors, which were selectively reduced to the corresponding saturated omega-19 cyclopropane fatty acids or esters. The 24-carbon omega-19 cyclopropane ester was made by chain elongation of the 23-carbon ester. Saturated and unsaturated chiral cyclopropane acids and esters were assayed, using wall extracts of Mycobacterium smegmatis; the incorporation of 14C-acetate was used to measure inhibition or stimulation of mycolic acid synthesis. Minor inhibition (2-3%) was shown by the 23- and 24-carbon saturated esters; all the other compounds were stimulants. The most effective (38-55%) stimulators of mycolate synthesis were the unsaturated esters with 23- and 26-carbons and the saturated and unsaturated 25-carbon acids.

This review surveys both data obtained by the authors and published data on the partial or full hydrodehalogenation of di- and polyhalocyclopropanes (chlorides and bromides) with Grignard reagents catalyzed by titanium or zirconium compounds. The factors affecting the efficiency and selectivity of the hydrodebromination of bromocyclopropanes are considered: the nature of Grignard reagents (including isotopically labeled reagents), their transformations and effects in catalyzed and uncatalyzed reactions, the participation of solvents, catalytic and stoichiometric amounts of the catalyst, etc. A scheme is proposed in which the key steps of the mechanism of hydrodebromination of bromocyclopropanes includes three blocks of reactions: (a) the generation of a catalytically active Ti(II) species; (b) the hydrodehalogenation of bromocyclopropanes involving electron transfer from a low-valent catalyst species, formation of the cyclopropyl radical, and stabilization of this radical as a result of hydrogen atom transfer from the solvent molecule; and (c) transformations of previously formed radical species, such as dimerization and disproportionation (for example, of radical species generated from Grignard reagents or ether molecules) or the linking of alkyl radicals to radical species produced from solvent molecules.

The experimental Raman and IR vibrational spectra of 3,3-dimethyl-1-(trimethylsilyl)cyclopropene in the liquid phase were recorded. Total geometry optimisation was carried out at the HF/6-31G* level and the HF/6-31G*//HF/6-31G* force field was computed. This force field was corrected by scale factors determined previously (using Pulay's method) for correction of the HF/6-31G*//HF/6-31G* force fields of 3,3-dimethylbutene-1, 1-methyl-, 1,2-dimethyl-, and 3,3-dimethylcyclopropene. The theoretical vibrational frequencies calculated from the scaled quantum mechanical force field and the theoretical intensities obtained from the quantum mechanical calculation were used to construct predicted spectra and to perform the vibrational analysis of the experimental spectra.

We report a synthesis of a single enantiomer of a dicyclopropane containing mycolic acid from Mycobacterium tuberculosis; this method can be simply varied to modify the chain lengths or the absolute stereochemistry of either cyclopropane.

Five different assays, Gibbs, Prussian Blue, Folin-Ciocalteau, fluorescence quenching of added phenol and precipitation of phenolics with bovine serum albumin (BSA) were investigated for their suitability in measuring the phenolic content of freshwaters. Phenol and a hydrolysable tannic acid were used as standards for monophenolics and polyphenolics, respectively. The individual and simultaneous application of both standards in doubly distilled water and filtered freshwater samples showed no matrix interference for the Gibbs, the Prussian Blue and the Folin-Ciocalteau assays. The quenching of phenol fluorescence and incomplete precipitation of added tannic acid in the freshwater samples were thought to originate from complexation. The Gibbs assay was specific for monophenolics, monohydroxybenzenes, with a Criterion of Detection (CoD) of 0.027 mg l(-1). Evaluating the assay using twenty-two monophenolics of lignin origin showed, apart from phenol itself, the phenolic acids vanillic, isovanillic, ferulic and syringic to have a linear response between 0 and 10 microM. The other monophenolics were not responsive in the Gibbs assay. The oxidation-based assays Prussian Blue and Folin-Ciocalteau had a CoD of 0.169 and 0.025 mg l(-1), respectively. The ratio of response of both assays for each sample was taken as an indication of the degree of polymerisation of the phenolic content. The Folin-Ciocalteau assay was used directly on the samples, on samples spiked with tannic acid at 2 and 4 mg l(-1), and after precipitation of phenolics with BSA. The difference in tannic acid equivalents before and after treatment, assayed the amount of protein precipitated phenolics. The results of all assays allowed differentiation between monophenolics (Gibbs), polyphenolics (Prussian Blue), total phenolics (Folin-Ciocalteau), complexation of added phenol and protein-precipitated phenolics. The reaction mechanisms underlying the assays were matched onto those occurring during humification. The assays were applied to six filtered freshwater samples and two humic and two fulvic acids. The results showed a different pattern for each site and illustrated varying reactivity of the 'phenolic content' of freshwater.

The genes from the oxygenase cluster nagAaGHAbAcAd of naphthalene-degrading Ralstonia sp. strain U2 were cloned and overexpressed. Salicylate 5-hydroxylase (S5H) activity, converting salicylate to gentisate, was present in vitro only in the single extract of cells with overexpressed nagAaGHAb or in a mixture of three cell extracts containing, respectively, NagGH (the oxygenase components), NagAa (ferredoxin reductase), and NagAb (ferredoxin). Each of the three extracts required for S5H activity was rate limiting in the presence of excess of the others but, when in excess, did not affect the rate of catalysis. S5H catalyzed the 5-hydroxylation of the aromatic rings of 3- and 4-substituted salicylates. However, the methyl group of 5-methylsalicylate was hydroxylated to produce the 5-hydroxymethyl derivative and the 6-position on the ring of 5-chlorosalicylate was hydroxylated, producing 5-chloro-2,6-dihydroxybenzoate. In an assay for the nag naphthalene dioxygenase (NDO) based on the indole-linked oxidation of NADH, three extracts were essential for activity (NagAcAd, NagAa, and NagAb). NDO and S5H were assayed in the presence of all possible combinations of the nag proteins and the corresponding nah NDO proteins from the “classical” naphthalene degrader P. putida NCIMB9816. All three oxygenase components functioned with mixed combinations of the electron transport proteins from either strain. The S5H from strain U2 is a unique monooxygenase which shares sequence similarity with dioxygenases such as NDO but is also sufficiently similar in structure to interact with the same electron transport chain and probably does so in vivo during naphthalene catabolism in strain U2.

A new method for the stereoselective synthesis of the (R,R)-β-hydroxy-α-alkyl fatty acid fragment of mycolic acids, via an asymmetric anti-aldol reaction is reported. The ‘mycolic acid motif’ fragment was prepared in three steps and >98% ee.
