Major depressive disorder (MDD) is associated with abnormal functional interactions among large-scale brain networks. Here we report findings from a randomized, active-controlled trial examining whether mindfulness-based therapy can regulate specific patterns of resting-state functional connectivity in clinically depressed patients. We show that a brief, clinically effective mindfulness intervention functionally decouples top-down control regions from brain areas implicated in sensory, affective, and attentional processing. While previous work has demonstrated the clinical impact of mindfulness training, the present findings shed light on the precise neural targets, providing new insight into the specificity of this therapeutic approach. more...

- Abstract
We present a publicly available dataset of 227 healthy participants comprising a young (N=153, 25.1±3.1 years, range 20-35 years, 45 female) and an elderly group (N=74, 67.6±4.7 years, range 59-77 years, 37 female) acquired cross-sectionally in Leipzig, Germany, between 2013 and 2015 to study mind-body-emotion interactions. During a two-day assessment, participants completed MRI at 3 Tesla (resting-state fMRI, quantitative T1 (MP2RAGE), T2-weighted, FLAIR, SWI/QSM, DWI) and a 62-channel EEG experiment at rest. During task-free resting-state fMRI, cardiovascular measures (blood pressure, heart rate, pulse, respiration) were continuously acquired. Anthropometrics, blood samples, and urine drug tests were obtained. Psychiatric symptoms were identified with Standardized Clinical Interview for DSM IV (SCID-I), Hamilton Depression Scale, and Borderline Symptoms List. Psychological assessment comprised 6 cognitive tests as well as 21 questionnaires related to emotional behavior, personality traits and tendencies, eating behavior, and addictive behavior. - Abstract
We organized 10Kin1day, a pop-up scientific event with the goal to bring together neuroimaging groups from around the world to jointly analyze 10,000+ existing MRI connectivity datasets during a 3-day workshop. In this report, we describe the motivation and principles of 10Kin1day, together with a public release of 8,000+ MRI connectome maps of the human brain. Ongoing grand-scale projects like the European Human Brain Project (1), the US Brain Initiative (2), the Human Connectome Project (3), the Chinese Brainnetome (4) and exciting world-wide neuroimaging collaborations such as ENIGMA (5) herald the new era of big neuroscience. In conjunction with these major undertakings, there is an emerging trend for bottom-up initiatives, starting with small-scale projects built upon existing collaborations and infrastructures. As described by Mainen et al. (6), these initiatives are centralized around self-organized groups of researchers working on the same challenges and sharing interests and specialized expertise. These projects could scale and open up to a larger audience and other disciplines over time, eventually lining up and merging their findings with other programs to make the bigger picture. more...

Modern scientific discovery depends on collecting large heterogeneous datasets with many sources of variability, and applying domain-specific pipelines from which one can draw insight or clinical utility. For example, macroscale connectomics studies require complex pipelines to process raw functional or diffusion data and estimate connectomes. Individual studies tend to customize pipelines to their needs, raising concerns about their reproducibility, which add to a longer list of factors that may differ across studies and result in failures to replicate (including sampling, experimental design, and data acquisition protocols). Mitigating these issues requires multi-study datasets and the development of pipelines that can be applied across them. We developed NeuroData’s MRI to Graphs (NDMG) pipeline using several functional and diffusion studies, including the Consortium for Reliability and Reproducability, to estimate connectomes. Without any manual intervention or parameter tuning, NDMG ran on 25 different studies (≈6,000 scans) from 19 sites, with each scan resulting in a biologically plausible connectome (as assessed by multiple quality assurance metrics at each processing stage). For each study, the connectomes from NDMG are more similar within than across individuals, indicating that NDMG is preserving biological variability. Moreover, the connectomes exhibit near perfect consistency for certain connectional properties across every scan, individual, study, site and modality; these include stronger ipsilateral than contralateral connections and stronger homotopic than heterotopic connections. Yet, the magnitude of the differences varied across individuals and studies—much more so when pooling data across sites, even after controlling for study, site, and basic demographic variables (i.e., age, sex, and ethnicity). This indicates that other experimental variables (possibly those not measured or reported) are contributing to this variability, which if not accounted for can limit the value of aggregate datasets, as well as expectations regarding the accuracy of findings and likelihood of replication. We therefore provide a set of principles to guide the development of pipelines capable of pooling data across studies while maintaining biological variability and minimizing measurement error. Features of ongoing experience are common across individuals and cultures. However, certain people express specific patterns of thought to a greater extent than others. Contemporary psychological theory assumes that individual differences in thought patterns occur because different types of experience depend on the expression of different neurocognitive processes. Consequently, individual variation in the underlying neurocognitive architecture is hypothesised to determine the ease with which certain thought patterns are generated or maintained. Our study (N = 178) tested this hypothesis using multivariate pattern analysis to infer shared variance among measures of cognitive function and neural organisation and examined whether these latent variables explained reports of the patterns of on-going thoughts people experienced in the lab. We found that relatively better performance on tasks relying primarily on semantic knowledge, rather than executive control, was linked to a neural functional organisation associated, via meta-analysis, with task labels related to semantic associations (sentence processing, reading and verbal semantics). Variability of this functional mode predicted significant individual variation in the types of thoughts that individuals experienced in the laboratory: neurocognitive patterns linked to better performance at tasks that required guidance from semantic representation, rather than those dependent on executive control, were associated with patterns of thought characterised by greater subjective detail and a focus on time periods other than the here and now. These relationships were consistent across different days and did not vary with level of task demands, indicating they are relatively stable features of an individual's cognitive profile. Together these data confirm that individual variation in aspects of ongoing experience can be inferred from hidden neurocognitive architecture and demonstrate that performance trade-offs between executive control and long-term semantic knowledge are linked to a person's tendency to imagine situations that transcend the here and now.
