In spite of the application of different cleaning procedures, the marble used for the portrait bust of Queen Margherita di Savoia continued to show permanent discoloration, consisting of an unevenly distributed grayish alteration, mainly on the front part. In this work, a multi-analytical, non-invasive approach was proposed using spectrocolorimetry, reflectance spectroscopy and multispectral imaging. The initial assumption, suggesting the presence of altered protective materials based on organic products (such as waxes or oils,) applied in the past according to traditional practices, was excluded, revealing instead the presence of deposits of particulate matter, which penetrated inside the crystalline structure of the marble, leading to a variation in its shade. Cleaning tests were also carried out to define the best product, using sustainable chemicals such as Polar Varnish Rescue®, alkoxyde surfactant, disodium EDTA, GLDA and Politect® Base in order to identify the best methodology and materials for sustainable cleaning, respecting the integrity of the original matter. Politect® Base demonstrated better action in comparison to the other products tested, and similar results were obtained with GLDA, which could be applied in areas where the Politect® is less efficient (e.g., lace).

Sarcopenia (SP) is a syndrome characterized by age-associated loss of skeletal muscle mass and function. SP worsens both acute and late radiation-induced toxicity, prognosis, and quality of life. Myosteatosis is a pathological infiltration of muscle tissue by adipose tissue which often precedes SP and has a proven correlation with prognosis in cancer patients. Sarcopenic obesity is considered a “hidden form” of SP (due to large fat mass) and is independently related to higher mortality and worse complications after surgery and systemic treatments with worse prognostic impact compared to SP alone. The evaluation of SP is commonly based on CT images at the level of the middle of the third lumbar vertebra. On this scan, all muscle structures are contoured and then the outlined surface area is calculated. Several studies reported a negative impact of SP on overall survival in patients undergoing RT for tumors of the head and neck, esophagus, rectum, pancreas, cervix, and lung. Furthermore, several appetite-reducing side effects of RT, along with more complex radiation-induced mechanisms, can lead to SP through, but not limited to, reduced nutrition. In particular, in pediatric patients, total body irradiation was associated with the onset of SP and other changes in body composition leading to an increased risk of cardiometabolic morbidity in surviving adults. Finally, some preliminary studies showed the possibility of effectively treating SP and preventing the worsening of SP during RT. Future studies should be able to provide information on how to prevent and manage SP before, during, or after RT, in both adult and pediatric patients.

Background: The safe use of radiotherapy (RT) requires compliance with dose/volume constraints (DVCs) for organs at risk (OaRs). However, the available recommendations are sometimes conflicting and scattered across a number of different documents. Therefore, the aim of this work is to provide, in a single document, practical indications on DVCs for OaRs in external beam RT available in the literature. Material and Methods: A multidisciplinary team collected bibliographic information on the anatomical definition of OaRs, on the imaging methods needed for their definition, and on DVCs in general and in specific settings (curative RT of Hodgkin’s lymphomas, postoperative RT of breast tumors, curative RT of pediatric cancers, stereotactic ablative RT of ventricular arrythmia). The information provided in terms of DVCs was graded based on levels of evidence. Results: Over 650 papers/documents/websites were examined. The search results, together with the levels of evidence, are presented in tabular form. Conclusions: A working tool, based on collected guidelines on DVCs in different settings, is provided to help in daily clinical practice of RT departments. This could be a first step for further optimizations.

Eosinophils are bone marrow-derived granulocytes that, under homeostatic conditions, account for as much as 1-3% of peripheral blood leukocytes. During inflammation, eosinophils can rapidly expand and infiltrate inflamed tissues, guided by cytokines and alarmins (such as IL-33), adhesion molecules and chemokines. Eosinophils play a prominent role in allergic asthma and parasitic infections. Nonetheless, they participate in the immune response against respiratory viruses such as respiratory syncytial virus and influenza. Notably, respiratory viruses are associated with asthma exacerbation. Eosinophils release several molecules endowed with antiviral activity, including cationic proteins, RNases and reactive oxygen and nitrogen species. On the other hand, eosinophils release several cytokines involved in homeostasis maintenance and Th2-related inflammation. In the context of SARS-CoV-2 infection, emerging evidence indicates that eosinophils can represent possible blood-based biomarkers for diagnosis, prognosis, and severity prediction of disease. In particular, eosinopenia seems to be an indicator of severity among patients with COVID-19, whereas an increased eosinophil count is associated with a better prognosis, including a lower incidence of complications and mortality. In the present review, we provide an overview of the role and plasticity of eosinophils focusing on various respiratory viral infections and in the context of viral and allergic disease comorbidities. We will discuss the potential utility of eosinophils as prognostic/predictive immune biomarkers in emerging respiratory viral diseases, particularly COVID-19. Finally, we will revisit some of the relevant methods and tools that have contributed to the advances in the dissection of various eosinophil subsets in different pathological settings for future biomarker definition.

Abstract The review discloses the historical and technological evolution of enzyme‐based field‐effect transistors (EnFETs) underlying the importance of gate electrode modification toward the implementation of novel FETs configurations such as extended‐gate FET (EG‐FETs) or EG organic FETs (EG‐OFETs). The working principle of the EnFETs as postulated by Bergveld in 1970, who defined the EnFET as an ion‐selective FET (ISFET) modified with enzyme‐membrane, is also discussed considering the analytical equations related to the EnFET output response. For each category, namely EnFETs, EG‐FETs, and EG‐OFETs, we reviewed the key devices’ configurations that addressed the research in this field in the last 40 years with particular attention to the analytical figures of merit.

Flavobacterium psychrophilum is the causative agent of Bacterial Cold Water Disease (BCWD)/ Rainbow Trout Fry Syndrome (RTFS) in rainbow trout Oncorhynchus mykiss. The disease is associated with significant mortality in trout farms and thereby responsible for severe economical losses in this part of the aquaculture industry. Vaccination of the very young life cycle stages is not successful due to the immature development of the adaptive immunity in yolksac larvae and early fry. This explains the extensive usage of antibiotics for control of BCWD/RTFS. Selective breeding of RTFS resistant fish may be a solution, and identification of markers associated with natural susceptibility/resistance to the disease may elevate breeding efforts towards more robust strains. We suggest a QTL (SNP Affx-88941461) for partial disease resistance on chromosome 25 (Omy25) based on our experimental F. psychrophilum challenges of outbred fish and subsequent GWAS analyses. The favourable SNP allele was designated Q, whereas q indicated the unfavourable allele. We validated this QTL in two subsequent challenge experiments by F. psychrophilum exposure of QQ, Qq and qq trout. In the first trial we produced trout carrying at least one favourable allele (QQ and Qq) associated with partial resistance by fertilizing trout eggs from non-selected (outbred) females with sperm from homozygous QQ males. In the second trial we also produced homozygous offspring by fertilizing trout eggs from QQ females with sperm from QQ males. The resistance profiles of these groups were then determined in the laboratory by triplicate or duplicate challenge experiments (common garden water bath exposure to F. psychrophilum), which showed a significantly higher survival in trout carrying the favourable allele. Field observations supplemented the laboratory studies. Under farm conditions QQ and Qq trout showed a higher survival rate compared to qq trout. In the present study chromosome 25 (Omy25) QTL was found associated with a partial resistance to F. psychrophilum. However, we recommend to include additional SNPs in future breeding studies because several QTL are associated with resistance.

This study investigates the potential of fatty acid methyl esters (FAME) as environmentally sustainable alternatives to traditional solvents for the removal of low-polarity materials commonly found in cultural heritage artefacts. Recognizing the environmental and health concerns associated with conventional solvents, this research focuses on FAME to remove low/non-polarity or non-polar substances used in cultural heritage preservation. Laboratory samples coated with low molecular synthetic resins (LMW) such as Regalrez 1094 and microcrystalline wax were treated with FAME applied in gel to prevent solvent–substrate interactions. Photographic documentation under UV-vis light, optical microscope observations, and spectrocolorimetric analysis were used for assessing surface “cleanness”, while FTIR-ATR spectroscopy was used to detect possible residue from treatment. Moreover, SEM analysis was used for a better understanding of the results. The best results were obtained using FAME base on methyl stearate according to Hansen-RED.

Here we have developed an advanced surface-enhanced Raman scattering (SERS) platform that enables the ultrasensitive, rapid and highly specific identification of tumour biomarkers in liquid biopsies. Our focus is on the detection of Thyroglobulin (Tg), the most important tumour biomarker for the diagnosis and prognosis of thyroid cancer. Specifically, SERS-active substrates fabricated by nanosphere lithography on chip or on tips of optical fiber (OF) were functionalised with Tg Capture antibodies. Gold nanoparticles were functionalized with Detection antibodies and conjugated with a Raman reporter. The sandwich assay platform was validated in the planar configuration and a detection limit of only 7 pg/ml was successfully achieved. A careful morphological characterisation of the SERS substrates allowed us to strictly correlate the coverage area with the Tg concentration. The same approach was successfully demonstrated in the washout of fine needle aspiration biopsies from cancer patients. The strategy was transferred to the Lab On Fiber (LOF) SERS platform and successfully used to detect Tg concentration. The proposed SERS-enhanced immunoassay platform has proven to be highly versatile and can be used with both microfluidic chip POC devices and SERS-OF-based optrodes to perform sensitive, specific and rapid ex vivo assays for Tg detection in liquid intraoperative biopsies.

With political and economic scenarios changing at an ever faster pace, it is necessary to understand the potential effects on asset prices. Today, the topic of rising inflation in the US as well as in the Eurozone, although still considered temporary by central banks, confronts us with the "unexpected risk" of a deviation from the baseline scenario. This implies the risk of having an aggressive monetary policy in the US, in a restrictive direction, therefore harmful to the financial markets. In this context, the question arises: is it possible to contemplate these events beforehand and act in good time? The answer is Yes and good risk management practices are important, using stress testing / scenario analysis techniques to accompany risk measures such as VaR and Expected Shortfall. Implementing this concept, through the implementation of stress test / scenario analysis - Bloomberg Economics Forecast Models® and Bloomberg Factor Models® - the present work seeks to consider plausible adverse scenarios that may arise and to assess the related impacts in terms of portfolio. The final aim is to improve the information set for the investor, allowing him to avoid potential market falls, as far as possible, that could prevent him from achieving his investment objectives.

One of the challenges in studying desert dust aerosol along with its numerous interactions and impacts is the paucity of direct in situ measurements, particularly in the areas most affected by dust storms. Satellites typically provide column-integrated aerosol measurements, but observationally constrained continuous 3D dust fields are needed to assess dust variability, climate effects and impacts upon a variety of socio-economic sectors. Here, we present a high-resolution regional reanalysis data set of desert dust aerosols that covers Northern Africa, the Middle East and Europe along with the Mediterranean Sea and parts of central Asia and the Atlantic and Indian oceans between 2007 and 2016. The horizontal resolution is 0.1∘ latitude × 0.1∘ longitude in a rotated grid, and the temporal resolution is 3 h. The reanalysis was produced using local ensemble transform Kalman filter (LETKF) data assimilation in the Multiscale Online Nonhydrostatic AtmospheRe CHemistry model (MONARCH) developed at the Barcelona Supercomputing Center (BSC). The assimilated data are coarse-mode dust optical depth retrieved from the Moderate Resolution Imaging Spectroradiometer (MODIS) Deep Blue Level 2 products. The reanalysis data set consists of upper-air variables (dust mass concentrations and the extinction coefficient), surface variables (dust deposition and solar irradiance fields among them) and total column variables (e.g. dust optical depth and load). Some dust variables, such as concentrations and wet and dry deposition, are expressed for a binned size distribution that ranges from 0.2 to 20 µm in particle diameter. Both analysis and first-guess (analysis-initialized simulation) fields are available for the variables that are diagnosed from the state vector. A set of ensemble statistics is archived for each output variable, namely the ensemble mean, standard deviation, maximum and median. The spatial and temporal distribution of the dust fields follows well-known dust cycle features controlled by seasonal changes in meteorology and vegetation cover. The analysis is statistically closer to the assimilated retrievals than the first guess, which proves the consistency of the data assimilation method. Independent evaluation using Aerosol Robotic Network (AERONET) dust-filtered optical depth retrievals indicates that the reanalysis data set is highly accurate (mean bias = −0.05, RMSE = 0.12 and r = 0.81 when compared to retrievals from the spectral de-convolution algorithm on a 3-hourly basis). Verification statistics are broadly homogeneous in space and time with regional differences that can be partly attributed to model limitations (e.g. poor representation of small-scale emission processes), the presence of aerosols other than dust in the observations used in the evaluation and differences in the number of observations among seasons. Such a reliable high-resolution historical record of atmospheric desert dust will allow a better quantification of dust impacts upon key sectors of society and economy, including health, solar energy production and transportation. IntroductionDespite the recent approval of several therapies in the adjuvant setting of melanoma, tumor relapse still occurs in a significant number of completely resected stage III-IV patients. In this context, the use of cancer vaccines is still relevant and may increase the response to immune checkpoint inhibitors. We previously demonstrated safety, immunogenicity and preliminary evidence of clinical efficacy in stage III/IV resected melanoma patients subjected to a combination therapy based on peptide vaccination together with intermittent low-dose interferon-α2b, with or without dacarbazine preconditioning (https://www.clinicaltrialsregister.eu/ctr-search/search, identifier: 2008-008211-26). In this setting, we then focused on pre-treatment patient immune status to highlight possible factors associated with clinical outcome.MethodsMultiparametric flow cytometry was used to identify baseline immune profiles in patients’ peripheral blood mononuclear cells and correlation with the patient clinical outcome. Receiver operating characteristic curve, Kaplan-Meier survival and principal component analyses were used to evaluate the predictive power of the identified markers.ResultsWe identified 12 different circulating T and NK cell subsets with significant (p ≤ 0.05) differential baseline levels in patients who later relapsed with respect to patients who remained free of disease. All 12 parameters showed a good prognostic accuracy (AUC>0.7, p ≤ 0.05) and 11 of them significantly predicted the relapse-free survival. Remarkably, 3 classifiers also predicted the overall survival. Focusing on immune cell subsets that can be analyzed through simple surface staining, three subsets were identified, namely regulatory T cells, CD56dimCD16- NK cells and central memory γδ T cells. Each subset showed an AUC>0.8 and principal component analysis significantly grouped relapsing and non-relapsing patients (p=0.034). These three subsets were used to calculate a combination score that was able to perfectly distinguish relapsing and non-relapsing patients (AUC=1; p=0). Noticeably, patients with a combined score ≥2 demonstrated a strong advantage in both relapse-free (p=0.002) and overall (p=0.011) survival as compared to patients with a score

This article presents a comprehensive review of the key documents and laws governing early childhood education in the 0-6 age range, which have contributed to the development of an integrated and unified system in Italy. The analysis of these materials reveals how international policies implemented since the 2000s have progressively shaped a multifaceted concept of inclusion. Moving beyond the notion of eliminating barriers and addressing injustice faced by children in difficulty, there has been a paradigm shift towards a holistic understanding of the child, emphasizing the harmonization of physical, social, and cognitive aspects. The legislative guidelines increasingly incorporate important “constants” such as the significance of early childhood education and care (ECER) in early identification of difficulties, the connection to family, community, and peer environments, and the need for high-quality initial and ongoing teacher training. These recommendations offer valuable insights for all stakeholders involved. While reviewing the milestones of Italian legislation, this study acknowledges the progress made and the challenges addressed, yet it also recognizes the importance of further implementation of certain recommendations.
