151. Wheat grain Avenin-like protein dynamics in relation to genotypes and environments

Abstract

The recently discovered non-gluten prolamins, avenin-like proteins (ALPs) in wheat can improve flour baking qualities. In our study, 15 TaALP genes were identified and mapped to chromosomes 7A, 4A and 7D. Phylogenetic analysis showed that TaALP genes formed three major clades, types a, b, and c. The allelic variation of ALP genes in a wild emmer wheat (Triticum turgidum ssp. dicoccoides) populations from Israel were investigated to study the evolution of TdALP genes under different micro environments. In total, 49 alleles were identified at 4 TdALP loci. Correlations between the sites in which wild emmer wheat accessions were collected in Israel and the diversity of their ALP allelles suggested that at least some alleles were selected for by environmental factors. In this project, we found that TaALP genes are pathogen-inducible. Bioinformatics predicted the presence of pathogenesis-related nucleotide motifs in the promoter regions of TaALP genes. Expression levels of TaALP genes and some PR genes were analysed by quantitative RT-PCR in developing caryopses at 7, 13 and 42 days after pollination. Differential expression patterns of TaALP genes were identified in plants infected by Fusarium graminearum. Recombinant TaALP-encoded proteins significantly inhibited the fungal growth in vitro. mRNA in situ hybridization confirmed that TaALP transcripts were upregulated in aleurone, sub-aleurone, and embryos after infection. Genome-wide Fusarium head blight (FHB) index association analysis indicated that certain TaALP alleles were significantly correlated with FHB resistance. The ALPs may act as pathogen resistance proteins mediated by systemic acquired resistance (SAR). Our research indicated that TaALP genes, characterized by typical gliadin domains, are broad-spectrum, partial-resistance genes that contribute to sustainable control of wheat pathogen disease and possibly other fungus-induced disease in wheat. 165. Genetic architecture of wheat (Triticum aestivum L.) phenology to maximize yield in water limited environments

Abstract

Water deficiency during the critical growth stages is one major concern affecting successful wheat production in the rainfed agriculture systems used in Australia and other semi-arid regions of the world. Bypassing the sensitive growth phases from a water stress period can result in higher grain yields in water limited environments. Therefore, this study was concentrated on the adjustment of phenology genes to synchronize the critical growth phases with the rainfall pattern or water availability of target environments. Vernalization (Vrn), photoperiod (Ppd) and earliness per se are the three major components of flowering pathway in wheat. A series of experiments were conducted to determine the role of allelic variations of vernalization and photoperiod genes in heading and subsequent grain production. The first experiment with double haploid (DH) lines differing for winter/spring alleles of Vrn-1 loci (vrn-A1, vrn-B1 and vrn-D1) revealed that combination of all three spring alleles confers the earliest flowering while the combination of all three winter alleles results in the latest flowering. On the other hand a combination of any one winter allele with two spring alleles results in intermediate heading but performed better in terms of a higher number of grain per spike, thousands kernels weight and test weight. Inclusion of a photoperiod insensitive allele with any combination of Vrn-1 loci reduces the heading time further by at least 20 days except for the combination with all winter alleles. The results also revealed that allelic variation in the Vrn and Ppd genes also alters the water requirement rate and total water consumption by modifying the duration of the growth phases. Investigation of the role of photoperiod sensitive/insensitive alleles of Ppd-D1 on heading and spike development revealed the relationship of photoperiod insensitive alleles Ppd-D1a with the gibberellin regulated pathway in addition to the effects of day length on flowering. 166. Sulphur and nitrogen fertilization strategy for wheat grain quality improvement and the underlying mechanism

Abstract

Australia is the fourth largest wheat (Triticum aestivum) exporter around the world,exporting about18 million tonnes annually. To achieve a desirable profit, the focus of the Australian wheat industry is to concurrently increase grain yield (GY) and grain protein content(GPC). However, due to the negative correlation between wheat GY and GPC,this is a challenging task. Apart from complex genetic approaches, modifyingnitrogen and sulphur fertilizer regime is a strategic method to achieve this goal. It is known that without adequate sulphur application, wheat cannot reach its full yield potential and make efficient use of nitrogen for protein biosynthesis. During grain filling, changes in nitrogen availability mainly affect GPC, while variable sulphur concentration exerts major impact on grain protein compositions. In this study, the impacts of a range of nitrogen and sulphur treatments on a series of nitrogen use efficiency (NUE) related agronomic traits, morphological traits and protein parameters were investigated using multi-year & site field trials and glasshouse experiments, and the accumulation patterns of individual storage protein composition. Six varying quality Australian bread wheat varieties,including Spitfire, Mace, Wyalkatchem, Westonia, Bonnie Rock,and Livingston,were selected for this study. Grain protein yield is used as a parameter to represent protein efficiency which is obtained by GY multiplying GPC. The results indicate that nitrogen application increases grain protein yield and optimizes grain protein parameters, but the NUE in relation to GY and grain protein yield (NUE-GY & NUE-PY) are both decreased. The correlation of neck diameter with GY and NUE-GY under different nitrogen treatments indicates that the improvement of neck diameter can be achieved through optimizing nitrogen treatments, which ultimately increases GY and NUE-GY. 168. Contribution to Breadmaking Performance of Two Different HMW Glutenin 1AyAlleles Expressed in Hexaploid Wheat

Abstract

Two expressed alleles of the 1Ay high-molecular-weight glutenin subunit (HMW-GS), 1Ay21*and 1AyT1, previously introduced in durum and bread wheat, were separately introgressed into the Australian bread wheat (Triticum aestivumL.) cv. Livingston. The developed lines had different allelic compositions compared to that of the parental cultivar (1Ax1), having either 1Ax21+1Ay21*or 1Ax1+1AyT1at the Glu-A1locus. Since 1Ax21and 1Ax1are known to have the same effects on quality, differences observed between the two sets of the developed lines are attributed to the two introgressed Aygenes. Yield and agronomic performance of the lines were evaluated in the field, and the protein, dough, and baking quality attributes were evaluated by large-scale quality testing. Results demonstrated that the subunit 1Ay21* increased unextractable polymeric protein by up to 14.3% and improved bread loaf volume by up to 9.2%. On the other hand, subunit 1AyT1increased total grain protein by up to 9% along with dough elasticity. Furthermore, milling extraction was higher, and flour ash was lower in the 1Ay21*lines compared to the lines integrating 1AyT1. Both sets of the 1Ay introgression lines reduced dough-mixing time compared to the recurrent parent Livingston. The results also showed that 1Ay21*had a higher potential to improve the baking quality than 1AyT1under the Livingston genetic background. Both alleles showed the potential to be utilized in breeding programs to improve the breadmaking quality.

Abstract

BACKGROUND: High‐molecular‐weight glutenin subunits (HMW‐GSs) have important effects on bread‐making quality. Allelic variations of HMW‐GS in bread wheat varieties contribute in different ways to dough properties and bread volume. However, no systematic analysis has been done on the effects of allelic variation on bread‐crumb structure, an important parameter when evaluating bread‐making quality. In this study, seven Glu‐1 deletion lines and one intact line harboring different encoding loci and derived from a cross between two spring wheat cultivars were used to investigate the contribution of a single Glu‐1 locus, or combination of Glu‐1 loci, to the crumb structure. RESULTS: Deletion of HMW‐GS locus combinations resulted in a decline in slice size, brightness, and fineness of the bread crumb. A desirable crumb structure correlated well with preferred subunit combinations: high levels of GMPs, superior dough properties, and loaf volume. The effects of the HMW‐GS combinations were ranked as Dx5 + Dy10 > Bx17 + By18 > Ax1 + Null. The Ax1 + Null allele affected the crumb structure by interacting with the Bx17 + By18 or Dx5 + Dy10. 173. MoS2confined within CMK-3 as multifunctional nanoreactor towards high-performance Li–S battery

Abstract

Lithium-sulfur (Li–S) battery has been considered a promising energy-storage devices due to its high energy density and low cost. However, the low utilization of sulfur and rapid capacity degradation resulted from low conductivity of sulfur, shuttle effect of sulfides intermediates and slow reaction kinetics restrain its practical application. In this work, a sulfur host composite constructed by CMK-3 confining MoS2nanoparticles was developed through a simple hydrothermal and post-annealing method. In this composite, most nano-size MoS2nanoparticles are confined within the pores and channels of CMK-3 (MoS2@CMK-3), acting as a “restricted nanoreactor” shows multiple effect for lithium polysulfides (LiPSs). On the one hand, the highly conductive CMK-3 not only accelerate the electrons transfer, but also can physically capture soluble LiPSs through its abundant mesopores. More significantly, the active MoS2within the nanoreactor exhibits a synergistic effect of strong chemical adsorption and subsequent catalytic conversion effect for LiPSs, which efficiently inhibits the shuttle effect. As a result, the as-prepared cathode based-MoS2-50@CMK-3 delivers a high specific capacity of 950.3 mA h g−1at 2 C and still maintain a discharge specific capacity of 440.3 mA h g−1after 1600 cycles with a capacity decay rate of 0.033 % per cycle. This work would guide the rational design of advanced sulfur cathodes for Li–S batteries.

180. Co/N co-doped graphene-like nanocarbon for highly efficient oxygen reduction electrocatalyst

Abstract

The development of efficient and inexpensive graphene-based electrocatalysts is of great significance to promote the commercial application of fuel cell and metal-air batteries. In this paper, a new type of Co and N co-doped graphene-like nanocarbon (Co/N-GLC) material was prepared by nano-silicon protection and high temperature pyrolysis. The obtained Co/N-GLC catalyst not only has a similar morphology of graphene, but also possesses a high specific surface area (809 m2g−1) with hierarchical porous structure (micropores/mesopores), and relative high active dopants content. These properties endow it with a good oxygen reduction activity in alkaline media, which can be comparable to commercial Pt/C catalyst. Moreover, the assembled zinc-air batteries using Co/N-GLC catalyst as the air electrode display a better discharge performance and higher stability compared to that of Pt/C electrode. This work demonstrates that the prepared graphene-like carbon catalyst has a good prospect, which can replace noble metal catalyst at the cathode in metalair batteries. 本文通过纳米硅作为保护层结合高温热解得到一种新型钴/氮共掺杂多孔类石墨烯纳米碳材料氧还原催化剂(Co/N-GLC). 结果显 示, Co/N-GLC具有类似于石墨烯的碳纳米薄层结构, 并展现出分级多孔(微孔/介孔)特性, 其比表面积高达809 m2g−1; 此外, Co/N-GLC还 拥有较高的吡啶氮和石墨氮含量. 这些优异的特性使得Co/N-GLC在碱性介质中具有出色的氧还原活性, 接近于商业Pt/C催化剂. 同时, 在 锌空气电池测试中, Co/N-GLC具有接近Pt/C电极的放电性能和优异的稳定性, 表明该催化剂有望替代贵金属催化剂, 具有很好的应用前 景.

Abstract

The ideal theranostic nanoplatform for tumors is a single nanoparticle that has a single semiconductor or metal component and contains all multimodel imaging and therapy abilities. The design and preparation of such a nanoparticle remains a serious challenge. Here, with FeS2 as a model of a semiconductor, the tuning of vacancy concentrations for obtaining 'all-in-one' type FeS2 nanoparticles is reported. FeS2 nanoparticles with size of ≈30 nm have decreased photoabsorption intensity from the visible to near-infrared (NIR) region, due to a low S vacancy concentration. By tuning their shape/size and then enhancing the S vacancy concentration, the photoabsorption intensity of FeS2 nanoparticles with size of ≈350 nm (FeS2-350) goes up with the increase of the wavelength from 550 to 950 nm, conferring the high NIR photothermal effect for thermal imaging. Furthermore, this nanoparticle has excellent magnetic properties for T2-weighted magnetic resonance imaging (MRI). Subsequently, FeS2-350 phosphate buffer saline (PBS) dispersion is injected into the tumor-bearing mice. Under the irradiation of 915-nm laser, the tumor can be ablated and the metastasis lesions in liver suffer significant inhibition. Therefore, FeS2-350 has great potential to be used as novel 'all-in-one' multifunctional theranostic nanoagents for MRI and NIR dual-modal imaging guided NIR-photothermal ablation therapy (PAT) of tumors. Abstract

As an abundant ROS, hydrogen peroxide (H2O2) plays pivotal roles in plant growth and development. In this work, we conducted for the first time an iTRAQ-based quantitative proteomic analysis of wheat seedling growth under different exogenous H2O2 treatments. The growth of seedlings and roots was significantly restrained by increased H2O2 concentration stress. Malondialdehyde, soluble sugar, and proline contents as well as peroxidase activity increased with increasing H2O2 levels. A total of 3425 proteins were identified by iTRAQ, of which 157 showed differential expression and 44 were newly identified H2O2-responsive proteins. H2O2-responsive proteins were mainly involved in stress/defense/detoxification, signal transduction, and carbohydrate metabolism. It is clear that up-regulated expression of signal transduction and stress/defence/detoxification-related proteins under H2O2 stress, such as plasma membrane intrinsic protein 1, fasciclin-like arabinogalactan protein, and superoxide dismutase, could contribute to H2O2 tolerance of wheat seedlings. Increased gluconeogenesis (phosphoenol-pyruvate carboxykinase) and decreased pyruvate kinase proteins are potentially related to the higher H2O2 tolerance of wheat seedlings. A metabolic pathway of wheat seedling growth under H2O2 stress is presented.

191. Developing Wheat for Improved Yield and Adaptation Under a Changing Climate: Optimization of a Few Key Genes

Abstract

Wheat grown under rain-fed conditions is often affected by drought worldwide. Future projections from a climate simulation model predict that the combined effects of increasing temperature and changing rainfall patterns will aggravate this drought scenario and may significantly reduce wheat yields unless appropriate varieties are adopted. Wheat is adapted to a wide range of environments due to the diversity in its phenology genes. Wheat phenology offers the opportunity to fight against drought by modifying crop developmental phases according to water availability in target environments. This review summarizes recent advances in wheat phenology research, including vernalization (Vrn), photoperiod (Ppd), and also dwarfing (Rht) genes. The alleles, haplotypes, and copy number variation identified for Vrnand Ppdgenes respond differently in different climatic conditions, and thus could alter not only the development phases but also the yield. Compared with the model plant Arabidopsis, more phenology genes have not yet been identified in wheat; quantifying their effects in target environments would benefit the breeding of wheat for improved drought tolerance. Hence, there is scope to maximize yields in water-limited environments by deploying appropriate phenology gene combinations along with Rhtgenes and other important physiological traits that are associated with drought resistance.

Abstract

The wheat NAM-B1 and NAM-A1 genes are positively associated with grain protein content (GPC) in wheat. We conducted molecular characterisation of the NAM-1 genes in 51 Australian wheat varieties (Triticum aestivum L.), with the aim of improving GPC and nitrogen-usage efficiency in Australian wheat. In summary, the wild type NAM-B1 gene, which originated from Israel, was identified in two Australian wheat varieties. Five varieties contained a deletion allele, whereas the majority (43) harboured a non-functional NAM-B1 allele and one variety contained both functional and non-functional alleles. Twenty-six Australian wheat varieties contained the NAM-A1a haplotype, which was similar to its well-characterised homoeolog NAM-B1 wild type and associated with high GPC. The NAM-D1 gene in the 51 wheat varieties was also characterised, and no gene variation in the exon regions was noted; only two single-nucleotide polymorphisms in introns 1 and 2 were found among the 51 varieties.
