Cell proliferation was evaluated by cell counting kit-8 and 5-ethynyl-20-deoxyuridine (EdU) staining assay. Cell invasion and migration were detected by transwell assay. Cell apoptosis was evaluated by flow cytometry. The relationship between RAET1K and miR-503-5p, as well as miR-503-5p and INPP4B, was determined by luciferase reporter assay and RNA immunoprecipitation (RIP) assay. In addition, the tumorigenesis of leukemia cells was evaluated by using a xenograft mouse model in vivo.Results: RAET1K was significantly upregulated and miR-503-5p was markedly downregulated in bone marrow tissues and cell lines (HL-60 and THP-1). Silencing of RAET1K (si-RAET1K) and overexpression of miR-503-5p inhibited cell proliferation, migration, and invasion but promoted apoptosis of HL-60 and THP-1 cells. 160. Function of the Polyketide Synthase Domains of Schizochytriumsp. on Fatty Acid Synthesis in Yarrowia lipolytica

Abstract

It is well known that polyunsaturated fatty acids (PUFAs) in Schizochytriumsp. are mainly synthesized via the polyketide synthase (PKS) pathway. However, the specific mechanism of PKS in fatty acid synthesis is still unclear. In this work, the functions of ORFA, ORFB, ORFC, and their individual functional domain genes on fatty acid synthesis were investigated through heterologous expression in Yarrowia lipolytica. The results showed that the expression of ORFA, ORFB, ORFC, and their individual functional domains all led to the increase of the very long-chain PUFA content (mainly eicosapentaenoic acid). Furthermore, the transcriptomic analysis showed that except for the 3-ketoacyl-ACP synthase (KS) domain of ORFB, the expression of an individual functional domain, including malonyl-CoA: ACP acyltransferase, 3-hydroxyacyl-ACP dehydratase (DH), 3-ketoacyl-ACP reductase, and KS domains of ORFA, acyltransferase domains of ORFB, and two DH domains of ORFC resulted in upregulation of the tricarboxylic acid cycle and pentose phosphate pathway, downregulation of the triacylglycerol biosynthesis, fatty acid synthesis pathway, and β-oxidation in Yarrowia lipolytica. These results provide a theoretical basis for revealing the function of PKS in fatty acid synthesis in Y. lipolyticaand elucidate the possible mechanism for PUFA biosynthesis.

164. The effects of PM2.5 on lung cancer-related mortality in different regions and races: A systematic review and meta-analysis of cohort studies.

Abstract

Given the increasing environmental pollution around the world, air pollutants such as particulate matters (PM) have been becoming typical carcinogenic factors to cause lung cancer. The PM with aerodynamic diameter less than or equal to 2.5 μm (PM2.5) plays an important role in lung cancer mortality. However, effects of PM2.5 on the mortality of lung cancer patients have not been investigated well. The purpose of this study is to assess the associations between PM2.5 exposure and lung cancer-related mortality, by regions and races. This systematic review was developed and conducted according to PRISMA protocol. A comprehensive literature search for peer-reviewed studies published up to October 2021 was conducted using the electronic databases PubMed, EMBASE, and Cochrane Library. The identified records were carefully evaluated following the PICOS criteria. Relative risks (RRs) and 95% confidence interval (CI) of lung cancer mortality associated with PM2.5 were extracted and meta-analyzed. The Chi-squared test, Begg's test, and Eggerr analysis were performed to assess the heterogeneity and publication bias. Sensitivity analysis and subgroup analysis were also conducted to make sure the authenticity and stability of the present study. In total, 16 studies met the inclusion criteria. For the overall effects of PM2.5 on lung cancer-related mortality, there was heterogeneity among the included studies (I2 = 62.30%, P < 0.01); thus, the random effects model was chosen to conduct related analysis. The pooled adjusted RR of lung cancer mortality associated with PM2.5 was 1.12 (95% CI: 1.09–1.15) for per 10 µg/m3 increase in PM2.5 concentrations. For different continents, the RRs of subgroup analysis in North-America, Asia, and Europe were 1.16 (1.09–1.22), 1.08 (CI: 1.07–1.09), and 1.14 (CI: 1.04–1.23), respectively. Subgroup analysis by race showed that for Mongoloid RR = 1.08 (1.07–1.09), and for Caucasoid RR = 1.13 (CI: 1.11–1.15). With per 10 µg/m3 increase in PM2.5, the mortality rate of Caucasoid lung cancer patients increased by 1.13 times, greater than that of the Mongoloid. Increase in PM2.5 concentrations is associated positively with the mortality of lung cancer patients. For different regions and races, the adverse effects of PM2.5 are discrepant. Abstract

Summary: Climate extremes pose enormous threats to natural ecosystems. Arbuscular mycorrhizal (AM) fungi are key plant symbionts that can affect plant community dynamics and ecosystem stability. However, knowledge about how AM fungal communities respond to climate extremes in natural ecosystems remains elusive.Based on a grassland extreme drought experiment in Inner Mongolia, we investigated the response of AM fungal communities to extreme drought in association with plant communities. The experiment simulated two types of extreme drought (chronic/intense) of once‐in‐20‐year occurrence.AM fungal richness and community composition exhibited high sensitivity to extreme drought and were more sensitive to intense drought than chronic drought. This community sensitivity (i.e. decline in richness and shifts in community composition) of AM fungi can be jointly explained by soil moisture, plant richness, and aboveground productivity. Notably, the robustness of the plant–AM fungal community co‐response increased with drought intensity.Our results indicate that AM fungal communities are sensitive to climate extremes, and we propose that the plant community mediates AM fungal community responses. Abstract

Extreme drought decreases aboveground net primary production (ANPP) in most grasslands, but the magnitude of ANPP reductions varies especially in C3‐dominated grasslands. Because the mechanisms underlying such differential ecosystem responses to drought are not well resolved, we experimentally imposed an extreme 4‐yr drought (2015–2018) in two C3 grasslands that differed in aridity. These sites had similar annual precipitation and dominant grass species (Leymus chinensis) but different annual temperatures and thus water availability. Drought treatments differentially affected these two semiarid grasslands, with ANPP of the drier site reduced more than at the wetter site. Structural equation modeling revealed that community‐weighted means for some traits modified relationships between soil moisture and ANPP, often due to intraspecific variation. Specifically, drought reduced community mean plant height at both sites, resulting in a reduction in ANPP beyond that attributable to reduced soil moisture alone. Higher community mean leaf carbon content enhanced the negative effects of drought on ANPP at the drier site, and ANPP–soil‐moisture relationships were influenced by soil C:N ratio at the wetter site. Importantly, neither species richness nor functional dispersion were significantly correlated with ANPP at either site. Overall, as expected, soil moisture was a dominant, direct driver of ANPP response to drought, but differential sensitivity to drought in these two grasslands was also related to soil fertility and plant traits. Abstract

To study the effects of an axial magnetic field on the discharge characteristics of Ar inductively coupled plasma, a set of discharge plasma generators was designed. The plasma parameters such as electron temperature and electron density were diagnosed with a Langmuir probe. The research showed that as the air pressure was 10 Pa, with the increase in axial magnetic field intensity, the electron temperature and electron density reduced continuously in the central discharge region, while the threshold power of discharge mode transition increased constantly. The analysis suggested that due to the circumnutation of charged particles acted upon by Lorentz force, the introduction of the axial magnetic field had a constraint effect on the particle movement and energy transfer and decreased the collision between the high-energy electron in the discharge sheath and the electron in the central region, thereby reducing the electron density and inductive coupling efficiency. From further analysis of the electron energy probability function, it could be found that in the E mode, the constraint effect of the axial magnetic field on electron motion was more obvious. The proportion of the high-energy electron (>27 eV) was apparently higher than that in the H mode, and the electron energy distribution was more even. This was caused by less electron collision.
