Abstract

In the developing neural tube, precursors at defined positions along the dorsal-ventral axis respond to Sonic hedgehog (Shh) secreted from the notocord and floorplate and elaborate distinct neurons identified by homeodomain protein expression. Oligodendrocyte lineage genes (Olig1/2) that encode a novel class of bHLH proteins are expressed in the oligodendrocyte lineage in the CNS. Moreover, their early expression is regulated by Shh in a dosage-dependent manner, suggesting that Olig genes may play roles in pattern formation and neuronal and/or glial cell fate determination. To study possible Olig functions during development, we analyzed endogenous Olig1/2 expression and found that Olig genes were initially expressed in the mouse spinal cord as early as 8.5 dpc and that expression at 11.5 dpc occurred in a restricted domain. Olig expression overlaps Nkx2.2 expression, where V3 interneurons (INs) are generated. We next generated transgenic mice that ectopically express Olig1 in the ventricular zone under control of the Hoxb4 enhancer. Global disruption of pattern was not observed by analysis of markers for neural precursors (NPs), motor neurons, and V2 INs. In contrast, misexpression Oligl evidently is detrimental to production of V3 INs, as Sim1-expressing cells were absent in these transgenic mice. Moreover, increased levels of cell death were observed. These data suggest that Olig genes may antagonize the development of certain neuronal subtypes and may refine pattern formation in NPs of the ventral neural tube.

182. Design and feasibility study of calcitonin and cathepsin K inhibitor for colon delivery

Abstract

The degradation of human calcitonin (hCT) by enzymes and mucosa from different gastrointestinal (GI) compartments was studied. The data from these studies are essential for the development of a hydrogel based colon-specific hCT delivery system. Lulminal and brush border membrane (BBM) enzymes from the colon and small intestine (SI) or the rabbit were isolated, and their enzymatic activity toward hCT in vitro was evaluated. Human fecalase was used to mimic the luminal enzymatic activity in the human colon, and its degradation ability was assessed. Excised intact rabbit intestinal tissues from both the colon and the SI were used to study the degradation patterns of hCT by intact mucosa. Detection of intact hCT was performed using reverse phase high performance liquid chromatography (HPLC). In vitro study showed that isolated luminal enzymes and BBM enzymes from the SI were more potent id degrading intact hCT. Moreover, BBM enzymes were for more abundant in the SI than in the colon. Compared with rabbit colonic enzymes, the degradation potency of human fecalase was further abated. Reduced proteolytic activity suggests that the colon is an advantageous site of peptide delivery. The primary structures of the peptide fragments were identified by Matrix Assisted desorption/Ionization Time-of-Flight (MALDI) Mass Spectrometry analysis. A synthetic copolymer, which has the same structure as the primary chain in hydrogels degradation in the colon, was synthesized by free radical precipitation copolymerization of N, N-dimethylacrylamide, acrylic acid, N-tert-butylacrylamide, and N-methacryloylglycylglycine p-nitrohenyl ester, followed by aminolysis to convert the nitrophenyl ester group into hydroxypropionamide. Fibrillation study using UV/Vis and fluorescence spectroscopy methods indicates incorporation of polymer into the buffer solution can significantly decrease the aggregation of hCT thus improving the physical stability of hCT in aqueous solution. These results suggest the hydrogel system will have the synergetic effect on physical stabilization of hCT as a delivery system. A polymetric cathepsin K inhibitor was designed and synthesized to inhibit the cathepsin K activity in osteoclasts thus deterring the collagen degradation. The inhibitor was synthesized by conjugating a special designed short peptide to a functionalized semitelechelic poly[N-(2-hydroxypropyl)methacrylamide. A competitive inhibition of the surrogate enzyme papain indicates this polymeric inhibitor has the potential to be a potent antiosteoclastic drug in the treatment of osteoclast by co administrating with hCT. Furthermore, the colon specific biodegradable hydrogel will protect the cathepsin K inhibitor in the same manner as hCT through the GI tract, and consequently enhances its clinical efficacy.

Descriptive textual information deposited with gene expression data in the Gene Expression Omnibus (GEO) is an underutilized resource because the textual information is unstructured and difficult to query. Rendering this information in a structured format utilizing standard medical terms would facilitate better searching and data reuse. Such a procedure would significantly increase the clinical utility of biomedical data repositories. Methods: The thesis is divided into two sections. The first section compares how well four medical terminologies were able to represent textual information deposited in GEO. The second section implements free-text search and faceted search and evaluates how well they are able to answer clinical queries with varying levels of complexity. Part I: 120 samples were randomly extracted from samples deposited in the GEO database from six clinical domains-breast cancer, colon cancer, rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), type I diabetes mellitus (IDDM), and asthma. These samples were previously annotated manually and structured textual information was obtained in a tag:value format. Data was mapped to four different controlled terminologies: NCI Thesaurus, MeSH, SNOMED-CT, and ICD- 10. The samples were assigned a score on a three-point scale that was based on how well the terminology was able to represent descriptive textual information. Part II: Faceted and free-text search tools were implemented, with 300 GEO samples included for querying. Eight natural language search questions were selected randomly from scientific journals. Abstract

The San Francisco Bay Area has one of the most congested metropolitan corridors in both California and nationwide, with very high demand for both passenger and air-freight transport. It is also a main entrance to the United States for the huge Asia market, and thus critical for the United States to play a leading role in the global economy. On one hand, traffic congestion in the main corridors through the Bay Area is severe and is becoming worse with the rapid increase of population and the development of the local economy, in which a substantial impact is created by truck-related activities such as the ever increasing air freight business (performed by companies such as Federal Express, UPS, DHL, and CNF). On the other hand, the San Francisco Bay Area Rapid Transit District (BART) operates a regional environmentally-green transit system has excess capacity during non-commute periods and during the commute on some lines in some reverse-commute direction. If the BART system were to be used by the air-freight delivery service providers, BART could probably provide reliable service to integrated air freight carriers to meet their limited-time window delivery service needs. This would lead to additional revenue generation for BART. For the traveling public as well as local, regional, and state government it would reduce truck activity, and its corresponding negative impacts on traffic, environment, safety, land use and the economy. Using BART for air freight movement as a model for combined goods and passenger movement can be generalized to other critical corridors nationwide to effectively relieve corridor congestion problem. Improving movement through these critical metropolitan corridors could yield significant benefits in terms of reduced travel time and delays and increased reliability and predictability of both passenger and freight movement.

199. Building 'Smaller, Cheaper, Faster' Satellites within the Constraints of an Academic Environment

Abstract

Student driven satellite projects are working under the constraints of extremely limited budgets, short development times due to student turnover, limited technical expertise, and other academic time commitment pressures. These are issues prevalent throughout the university system, but many programs do not directly address these realities. So how can educational programs promote a "smaller, cheaper, faster" philosophy and at the same time effectively realize its potential? This question was asked of the students in Stanford University's Satellite Systems Development Laboratory (SSDL). In response, the Satellite QUIck Research Testbed (SQUIRT) program was developed to give students the opportunity to participate in the entire lifecycle of a satellite development. This paper will summarize the technical solutions they have achieved, given the environmental constraints the program is under. In particular, the issues addressed are those relevant to the subsystems which the authors manage: Communications, Thermal Control, and Attitude Determination & Control. Each subsystem section will describe its goals, constraints, and technical approaches, as well as the integral role of industry. It is hoped that this research and approach is transportable to other universities that wish to build their own small satellites. The successful matriculation of such "smaller, cheaper, faster" programs are not only fantastic educational opportunities for young engineers and future program managers, but eventually will prove beneficial to industry as this paradigm becomes the competitive norm.
