Abstract

We report genome-wide ancient DNA from 49 individuals forming four parallel time transects in Belize, Brazil, the Central Andes, and the Southern Cone, each dating to at least ∼9,000 years ago. The common ancestral population radiated rapidly from just one of the two early branches that contributed to Native Americans today. We document two previously unappreciated streams of gene flow between North and South America. One affected the Central Andes by ∼4,200 years ago, while the other explains an affinity between the oldest North American genome associated with the Clovis culture and the oldest Central and South Americans from Chile, Brazil, and Belize. However, this was not the primary source for later South Americans, as the other ancient individuals derive from lineages without specific affinity to the Clovis-associated genome, suggesting a population replacement that began at least 9,000 years ago and was followed by substantial population continuity in multiple regions.

Abstract

Hypoxia inducible factor-1 (HIF-1) is a transcriptional factor that induces genes involved in glucose metabolism. HIF-1 is formed by a regulatory α-subunit (HIF-1α) and a constitutive β-subunit (HIF-1β). The white spot syndrome virus (WSSV) induces a shift in glucose metabolism and oxidative stress. HIF-1α is associated with the induction of metabolic changes in tissues of WSSV-infected shrimp. However, the contributions of HIF-1 to viral load and antioxidant responses in WSSV-infected shrimp have been not examined. In this study, the effect of HIF-1 silencing on viral load and the expression and activity of antioxidant enzymes (superoxide dismutase-SOD, glutathione S-transferase-GST, and catalase) along with oxidative damage (lipid peroxidation and protein carbonyl) in tissues of white shrimp infected with the WSSV were studied. The viral load increased in hepatopancreas and muscle after WSSV infection, and the accumulative mortality was of 100% at 72 h post-infection. The expression and activity of SOD, catalase, and GST decreased in each tissue evaluated after WSSV infection. Protein carbonyl concentrations increased in each tissue after WSSV infection, while lipid peroxidation increased in hepatopancreas, but not in muscle. Silencing of HIF-1α decreased the WSSV viral load in hepatopancreas and muscle of infected shrimp along with shrimp mortality. Silencing of HIF-1α ameliorated the antioxidant response in a tissue-specific manner, which translated to a decrease in oxidative damage. These results suggest that HIF-1 is essential for restoring the antioxidant response, which counters the oxidative injury associated with WSSV infection.

Abstract

Addiction treatment can improve its impact by providing evidence-based care for the variety of problems that accompany substance use disorders. We conducted a retrospective evaluation of a new treatment program in California that aimed at providing multifaceted services through affiliated licensed and certified outpatient providers. The process evaluation used a logic model, focusing on program inputs, activities, and outputs, to understand the services received by the initial 18 clients who entered treatment. Outcomes for these patients were not assessed. Results indicated that clients received a variety of services: On average clients contracted for 118 treatment sessions and received 143 sessions. Among the many types of services provided, the most frequently received were integrative healthcare (averaging 42 sessions), group therapy (32 sessions), and individual therapy (32 sessions). This logic-model process evaluation indicated that a range of services were provided. The comprehensive approach may have promise for extending addiction treatment beyond its usual boundaries.

Abstract

Some recent academic and popular literature implies that the problem of the colonization of the Americas has been largely resolved in favor of one specific model: a Pacific coastal migration, dependent on high marine productivity, from the Bering Strait to South America, thousands of years before Clovis, the earliest widespread cultural manifestation south of the glacial ice. Speculations on maritime adaptations and typological links (stemmed points) across thousands of kilometers have also been advanced. A review of the current genetic, archeological, and paleoecological evidence indicates that ancestral Native American population expansion occurred after 16,000 years ago, consistent with the archeological record, particularly with the earliest securely dated sites after ~15,000 years ago. These data are largely consistent with either an inland (ice-free corridor) or Pacific coastal routes (or both), but neither can be rejected at present. Systematic archeological and paleoecological investigations, informed by geomorphology, are required to test each hypothesis.

Abstract

Inferior vena cava agenesis (IVCA) is a rare anomaly predisposing affected people to lower-limb venous thrombosis with low frequency of pulmonary embolism. Antenatal thrombosis and inherited thrombophilia have been suggested as causes of IVCA. However, there is little evidence on the clinical course and management of this condition. We designed a patient registry to assess the thrombotic risk and features of IVCA.

164. ICTs, ESPs and ZPD through Microlessons in Teacher Education

Abstract

This paper presents the initial results of the use of dialogic interaction enhanced by the use of technology in teaching English in different settings and subjects of teacher education. Technology is used in three different ways: as a support (video) for analysis through teacher-instructor interaction, as a means of social interaction and use of language for education between teacher and students (use of the computer for instruction), and as the creation of own designed materials for language training (through the use of technology). Data is obtained from video-recordings related to teaching and learning English as a foreign language by three different teachers in three subjects from Primary and Infant Education at Universidad de Alcalá. Results indicate that microteaching is not only valid as a training method but also to introduce new content and concepts that have not been previously introduced in the classes. The interaction in the Zone of Proximal Development (ZPD) between the teacher-instructor and the teacher-students also proves to have a powerful effect in motivation, teaching improvement and language for education skills development through self-reflection. Abstract

The thesis describes two desymmetrization approaches to two different natural products. The first chapter contains background on the salvileucalins and the Buchner reaction along with the work done towards the synthesis of salvileucalin B. This includes the synthesis of the diazo substrates for the Buchner reaction and the results of experimentation on a key Buchner reaction step in the sequence. The second chapter contains background on emetine and the work done towards its synthesis. This includes the synthesis of the dihydropyridine intermediate, the results of experimentation on the key step of acidic hydrolysis of the dihydropyridine to facilitate a double Pictet–Spengler reaction, exploration of an oxidative cleavage strategy, and the results on a model system to find conditions for a successful Pictet–Spengler reaction. Future directions of the project are also discussed, which include a Mukaiyama hydration strategy and modulation of the groups on the dihydropyridine nitrogen.

172. “Analysis of Macular Pigment Optical Density in Childhood: A Systematic Review”

Abstract

ABSTRACTPurposeThis systematic review studies the relationship between Macular Pigment Optical Density (MPOD) values and cognitive and visual function in childhood.MethodsIt included cross-sectional, observational studies or controlled clinical trials in humans between 0 and 18 years of age, analyzing MPOD values in 3 main databases: PubMed, Scopus and Web of Science. The study was performed according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement recommendations.ResultsThirteen studies were included in this systematic review. The relationship of cognitive function, visual function and diverse variables with MPOD was analyzed in 4, 4 and 5 studies, respectively. The age of the participants ranged between premature infants to 12 years. Most of the studies used Heterochromatic Flicker Photometry (HFP) with macular densitometer to obtain MPOD values. MPOD values ranged between 0 (undetectable) to 0.66 ± 0.03 d.u. Only 4 articles studied the relationship between MPOD values and dietary intake of lutein and zeaxanthin using questionnaires about diet.ConclusionsLutein and zeaxanthin accumulation plays an important role during the maturational stage and childhood development. Although cognitive function is more strongly correlated with MPOD values, the relationship with visual function remains unclear, and further studies are required to support this relationship.

Abstract

European Spallation Source instruments like the macromolecular diffractometer, NMX, require an excellent neutron detection efficiency, high-rate capabilities, time resolution, and an unprecedented spatial resolution in the order of a few hundred micrometers over a wide angular range of the incoming neutrons. For these instruments solid converters in combination with Micro Pattern Gaseous Detectors (MPGDs) are a promising option. A GEM detector with gadolinium converter was tested on a cold neutron beam at the IFE research reactor in Norway. The {\mu}TPC analysis, proven to improve the spatial resolution in the case of $^{10}$B converters, is extended to gadolinium based detectors. For the first time, a Gd-GEM was successfully operated to detect neutrons with a measured efficiency of 11.8% at a wavelength of 2 {\AA} and a position resolution better than 250 {\mu}m., Comment: 18 pages, 10 figures, 2 tables

Abstract

We generated genome-wide data from 69 Europeans who lived between 8,000-3,000 years ago by enriching ancient DNA libraries for a target set of almost four hundred thousand polymorphisms. Enrichment of these positions decreases the sequencing required for genome-wide ancient DNA analysis by a median of around 250-fold, allowing us to study an order of magnitude more individuals than previous studies and to obtain new insights about the past. We show that the populations of western and far eastern Europe followed opposite trajectories between 8,000-5,000 years ago. At the beginning of the Neolithic period in Europe, ~8,000-7,000 years ago, closely related groups of early farmers appeared in Germany, Hungary, and Spain, different from indigenous hunter-gatherers, whereas Russia was inhabited by a distinctive population of hunter-gatherers with high affinity to a ~24,000 year old Siberian6 . By ~6,000-5,000 years ago, a resurgence of hunter-gatherer ancestry had occurred throughout much of Europe, but in Russia, the Yamnaya steppe herders of this time were descended not only from the preceding eastern European hunter-gatherers, but from a population of Near Eastern ancestry. Western and Eastern Europe came into contact ~4,500 years ago, as the Late Neolithic Corded Ware people from Germany traced ~3/4 of their ancestry to the Yamnaya, documenting a massive migration into the heartland of Europe from its eastern periphery. This steppe ancestry persisted in all sampled central Europeans until at least ~3,000 years ago, and is ubiquitous in present-day Europeans. These results provide support for the theory of a steppe origin of at least some of the Indo-European languages of Europe.

188. "The L test under single and dual-task conditions in women with fibromyalgia: A psychometric study".

Abstract

This study aimed to investigate the test-retest reliability of the L test under single and dual-task conditions in women with fibromyalgia. To analyze the concurrent validity of the L test and Timed Up and Go test (TUG) and the relationship between the L test and the impact of the disease. A cross-sectional study with 22 women with fibromyalgia. The Intraclass Correlation Coefficient (ICC) 3,1 evaluated the reliability. The concurrent validity was established between the L test and the TUG. The Pearson product-moment correlation assessed the correlation between the L test and the impact of the disease. Results revealed an ICC of 0.95 (0.876–0.979) in the single condition with the stopwatch and 0.95 (0.872–0.978) with the Chronopic. In the dual-task condition 0.93 (0.832–0.971) with the stopwatch and 0.94 (0.854–0.975) with the Chronopic. The L test and the TUG showed significant relationships with a stopwatch in the single (test-r = 0.839; retest-r = 0.897) and the dual-task conditions (test-r = 0.793; retest-r = 0.882). These findings were also found with the Chronopic in the single (test-r = 0.876; retest-r = 0.901) and the dual-task conditions (test-r = 0.872; retest-r = 0.897). Significant relationships were found between the L test and the impact of the disease with a stopwatch in the single (test-r = 0.502; retest-r = 0.647) and the dual-task conditions (test-r = 0.428; retest-r = 0.571). And with the Chronopic in the single (test-r = 0.528; retest-r = 0.663) and the dual-task conditions (test-r = 0.482; retest-r = 0.608). L test is a reliable tool under single and dual-task conditions in women with fibromyalgia. This test is a valid method when compared with the TUG, and it is related to the impact of the disease. Abstract

BACKGROUND: Prone positioning (PP) has demonstrated its potential for improving outcomes in patients with ARDS who require invasive mechanical ventilation. However, the ability of prolonged proning to reduce mortality in patients with COVID-19 specifically, sessions lasting > 24 h remains uncertain. METHODS: In this retrospective cohort study, we examined 158 subjects with COVID-19 pneumonia who required mechanical ventilation due to moderate-to-severe ARDS. Seventy-six subjects were placed in standard PP and 82 in extended PP, defined as prone sessions lasting at least 32 h. Our primary aim was to evaluate the effect of EPP on 90-d survival in subjects with COVID-19 with acute severe respiratory failure. To ensure the reliability of our findings and to minimize bias, we applied 3 adjustment approaches: cardinality matching (CM), matching weighting (MW), and inverse probability of treatment weighting with stabilized and trimmed weights (SW). We used Kaplan-Meier curves and Cox proportional hazard models to analyze the effects of EPP on 90-d mortality and sensitivity analysis by calculating E-values. RESULTS: The overall crude 90-d mortality rate was 31.7%. The unadjusted 90-d mortality rates were 19.5% in the EPP group and 44.7% in the SPP group (hazard ratio [HR] 0.35 [95% CI 0.19- 0.63], P < .001). After adjustment for confounding factors using CM, MW, and SW, baseline covariates were balanced between the 2 groups. Subjects in the EPP group exhibited lower 90-d mortality rates after adjustment using CM (HR 0.42 [95% CI 0.23-0.79], P = .007), MW (HR 0.45 [95% CI 0.21-0.95], P = .036), or SW (HR 0.29 [95% CI 0.15-0.56], P < .001). CONCLUSIONS: Extended PP was associated with improved 90-d survival in subjects with COVID-19 undergoing mechanical ventilation for severe ARDS. These findings suggest the potential benefit of EPP in the management of COVID-19-related respiratory failure. Abstract

The objective is to determine the effects that are found in the siblings of intellectually disabled children. Previous studies, have concluded that there are certain effects that intellectually disabled children have on their siblings. The effects that are considered in the paper are stress; anxiety levels, internalizing and externalizing behaviors; how they deal with problems, and lastly self-perception; whether they are mentally stable and confident. Negative effects are going to be the focus of the study, however, there are certain positive effects that are exposed. Levels of warmth and affection are higher when a child has a sibling with a disability. There were various forms of instruments used through the different articles: Personal Information Questionnaire , the Rosenberg Self- Esteem Scale and the Trait Anxiety Scales; Wechsler Intelligence Scale for Children-4th Edition, the Vineland Adaptive Behavior Scales, the Sibling Relationship Questionnaire, the Social Skills Rating System-Secondary Level, and the Achenbach Child Behavior Checklist; Semantic Differentials. An interview was also conducted on a single person, who proved the hypothesis of all the articles.

Abstract

The modulation of AMPA receptor (AMPAR) content at synapses is thought to be an underlying molecular mechanism of memory and learning. AMPAR content at synapses is highly plastic and is regulated by numerous AMPAR accessory transmembrane proteins such as TARPs, cornichons, and CKAMPs. SynDIG (synapse differentiation-induced gene) defines a family of four genes (SynDIG1-4) expressed in distinct and overlapping patterns in the brain. SynDIG1 was previously identified as a novel transmembrane AMPAR-associated protein that regulates synaptic strength. The related protein SynDIG4 [also known as Prrt1 (proline-rich transmembrane protein 1)] has recently been identified as a component of AMPAR complexes. In this study, we show that SynDIG1 and SynDIG4 have distinct yet overlapping patterns of expression in the central nervous system, with SynDIG4 having especially prominent expression in the hippocampus and particularly within CA1. In contrast to SynDIG1 and other traditional AMPAR auxiliary subunits, SynDIG4 is de-enriched at the postsynaptic density and colocalizes with extrasynaptic GluA1 puncta in primary dissociated neuron culture. These results indicate that, although SynDIG4 shares sequence similarity with SynDIG1, it might act through a unique mechanism as an auxiliary factor for extrasynaptic GluA1-containing AMPARs. J. Abstract

The exact timing, route, and process of the initial peopling of the Americas remains uncertain despite much research. Archaeological evidence indicates the presence of humans as far as southern Chile by 14.6 thousand years ago (ka), shortly after the Pleistocene ice sheets blocking access from eastern Beringia began to retreat. Genetic estimates of the timing and route of entry have been constrained by the lack of suitable calibration points and low genetic diversity of Native Americans. We sequenced 92 whole mitochondrial genomes from pre-Columbian South American skeletons dating from 8.6 to 0.5 ka, allowing a detailed, temporally calibrated reconstruction of the peopling of the Americas in a Bayesian coalescent analysis. The data suggest that a small population entered the Americas via a coastal route around 16.0 ka, following previous isolation in eastern Beringia for ~2.4 to 9 thousand years after separation from eastern Siberian populations. Following a rapid movement throughout the Americas, limited gene flow in South America resulted in a marked phylogeographic structure of populations, which persisted through time. All of the ancient mitochondrial lineages detected in this study were absent from modern data sets, suggesting a high extinction rate. To investigate this further, we applied a novel principal components multiple logistic regression test to Bayesian serial coalescent simulations. The analysis supported a scenario in which European colonization caused a substantial loss of pre-Columbian lineages.
