Abstract

Residual presence of pesticides in a variety of fruits, vegetables and agricultural crops nowadays can often exceed maximum residue limits. Exposure to organophosphorus pesticides through direct contact, ingestion or inhalation can have a deleterious effect on human health ; additional residue in food also contributes to the risk of exposure. The neurological system is particularly susceptible to adverse effects from either direct pesticide exposure or through food. Selected pesticides such as methamidophos, fenamiphos, ethoprophos, phosalone, and acetamiprid are pesticides with possible potential to inhibit acetylcholinesterase (AChE), the essential enzyme for neurotransmission. Disruption of AChE biosynthesis process by certain pesticides has also been evidenced, and was considered as one of the mechanisms for the long-term effects of these compounds. In order to fully understand the action of these pesticides, we investigated their effects and mechanisms on enzymatic activity of both AChE and its analog enzyme butyrylcholinesterase (BChE). The objective of this study was to reveal alterations caused by selected pesticides on the enzymatic activity and expression of AChE in selected cell lines. Moreover, a possible detoxication mechanism was tested, taking into account BChE as endogenous bioscavenger of xenobiotics.

Abstract

Due to their role in the protection of herbs, pesticides are globally widespread and can have a deleterious effect on human health. The neurological system is particularly susceptible to adverse effects from pesticide exposure. Selected pesticides such as metamiphos, phenamidophos, ethoprophos, phosalone, and acetamiprid are neuroactive insecticides with neurotransmitter cascades as their main targets. Inhibition of acetylcholinesterase (AChE) by organophosphate (OP) compounds is a well-known mechanism of intoxication. The disruption of nicotine acetylcholine receptors (nAChRs) through agonists such as acetamiprid can lead to toxic effects by inhibiting AChE in the synaptic cleft. To gain better insight for the purposes of intoxication monitoring, alterations in cholinesterase activity were observed together with interactions of pesticides with active site residues of cholinesterases, revealing the possible orientation of the inhibitors. The focus of this study was to achieve a better comprehension of the action mechanisms and effects that selected pesticides have on cholinergic neurotransmission and afterwards attain the best intervention solution for pesticide intoxication. Abstract

Acetylcholinesterase (AChE, EC 3.1.1.7) plays important roles in cholinergic neurotransmission and has been widely recognized as a biomarker for monitoring pollution by organophosphate (OP) and carbamate pesticides. Dioxin is an emerging environmental AChE disruptor and is a typical persistent organic pollutant with multiple toxic effects on the nervous system. Growing evidence has shown that there is a significant link between dioxin exposure and neurodegenerative diseases and neurodevelopmental disorders, most of which involve AChE and cholinergic dysfunctions. Therefore, an in-depth understanding of the effects of dioxin on AChE and the related mechanisms of action might help to shed light on the molecular bases of dioxin impacts on the nervous system. In the past decade, the effects of dioxins on AChE have been revealed in cultured cells of different origins and in rodent animal models. Unlike OP and carbamate pesticides, dioxin-induced AChE disturbance is not due to direct inhibition of enzymatic activity; instead, dioxin causes alterations of AChE expression in certain models. As a widely accepted mechanism for most dioxin effects, the aryl hydrocarbon receptor (AhR)-dependent pathway has become a research focus in studies on the mechanism of action of dioxin-induced AChE dysregulation. In this mini-review, the effects of dioxin on AChE and the diverse roles of the AhR pathway in AChE regulation are summarized. Additionally, the involvement of AhR in AChE regulation during different neurodevelopmental processes is discussed. These AhR-related findings might also provide new insight into AChE regulation triggered by diverse xenobiotics capable of interacting with AhR. Abstract

In this study, two types of soil columns were constructed to investigate the effect of the Enhanced permeability layers (EPL) on permeability and efficiency. A series of pre-tests of experimental conditons also were performaned to obtain the optimal Pb removal conditions, preventing the influence of the conditions on permeability. The results showed that disodium Ethylenediaminetetraacetic acid (EDTA) had optimal removal ability, followed by Oxalic acid (OA), citric acid, Diethylenetriaminepentaacetic acid (DTPA), and acetic acid (HAc). After the 45 days of column leaching (experimental conditions: 0.20 mol/L of EDTA, pH = 4 ± 0.02, at environmental temperature), the respective removal ratios of column 2#, 15(cm) and 35(cm) were 80.60% and 88.40%, while those of column 1#, 15(cm) and 35(cm) were 69.46% and 74.67%. BET analyzed that the surface area of column 1# soil was 7.07 m2/g, smaller than the 12.87 m2/g in column 2#, verifying a change in soil pores and specific surface area. SEM and EDS analysis of soil particles showed that the leaching altered the sub-micron structures of soil, and the particle size distribution showed that the particles became smaller after leaching. The permeability coefficient (K) fit analysis result showed that the K of column 2# soil was about 9.78 × 10−3 mm/min, twice that in column 1#, which was 4.27 × 10−3 mm/min. These results demonstrated that the EPL increases permeability of soil heap stackand the leaching mass transfer.

Abstract

Intoxication monitoring and intervention solution for pesticides intoxication are essential issues for public health surveillance and control world-wildly. The well-known action mechanism for OP pesticides intoxication is through the inhibition of acetylcholinesterase (AChE), an enzyme critical for cholinergic neurotransmission. Disruption of AChE biosynthesis process by certain pesticides has been evidenced, and was considered as one of the mechanisms for the long-term effects of these compounds. Thus, in order to fully understand the action of these pesticides, their effects and mechanisms on both AChE expression and enzymatic activity merit in- depth investigations. The objectives was to reveal the alterations in the enzymatic activity and expression of AChE caused by selected pesticides (Metamiphos, Phenamidophos, Ethoprophos, Phosalone, and Acetamiprid), to investigate the action mechanisms by in silico methods, and to elucidate cell signaling mechanisms for the AChE dysregulation.

Abstract

Electronic metal–support interactions (EMSI) describe the electron flow between metal sites and a metal oxide support. It is generally used to follow the mechanism of redox reactions. In this study of CuO‐CeO2 redox, an additional flow of electrons from metallic Cu to surface carbon species is observed via a combination of operando X‐ray absorption spectroscopy, synchrotron X‐ray powder diffraction, near ambient pressure near edge X‐ray absorption fine structure spectroscopy, and diffuse reflectance infrared Fourier transform spectroscopy. An electronic metal–support–carbon interaction (EMSCI) is proposed to explain the reaction pathway of CO oxidation. The EMSCI provides a complete picture of the mass and electron flow, which will help predict and improve the catalytic performance in the selective activation of CO2, carbonate, or carbonyl species in C1 chemistry. Abstract

The toxicity of 2,3,7,8-Tetrachlorodibenzo- p -dioxin (TCDD) is generally believed to be mediated by aryl hydrocarbon receptor (AhR), but some evidence suggests that the effects of TCDD can also be produced through AhR-independent mechanisms. In previous experiments, we found that mainly AhR-dependent mechanism was involved in the migration inhibition of glioblastoma U87 cells by TCDD. Due to the heterogeneity of glioblastomas, not all tumor cells have significant AhR expression. The effects and mechanisms of TCDD on the migration of glioblastomas with low AhR expression are still unclear. We employed a glioblastoma cell line A172 with low AhR expression as a model, using wound healing and Transwell® assay to detect the effect of TCDD on cell migration. We found that TCDD can inhibit the migration of A172 cells without activating AhR signaling pathway. Further, after being pre-treated with AhR antagonist CH223191, the inhibition of TCDD on A172 cells migration was not changed, indicating that the effect of TCDD on A172 cells is not dependent on AhR activation. By transcriptome sequencing analysis, we propose dysregulation of the expression of certain migration-related genes, such as IL6, IL1B, CXCL8, FOS, SYK, and PTGS2 involved in cytokines, MAPK, NF-κB, and IL-17 signaling pathways, as potential AhR-independent mechanisms that mediate the inhibition of TCDD migration in A172 cells.
