Abstract

Microfluidic chip electrophoresis has been widely employed for separation of various biochemical species owing to its advantages of low sample consumption, low cost, fast analysis, high throughput, and integration capability. In this article, we reviewed the development of four different modes of microfluidics‐based electrophoresis technologies including capillary electrophoresis, gel electrophoresis, dielectrophoresis, and field (electric) flow fractionation. Coupling detection schemes on microfluidic electrophoresis platform were also reviewed such as optical, electrochemical, and mass spectrometry method. We further discussed the innovative applications of microfluidic electrophoresis for biomacromolecules (nucleic acids and proteins), biochemical small molecules (amino acids, metabolites, ions, etc.), and bioparticles (cells and pathogens) analysis. Abstract

The emerging consensus on organic matter (OM) cycling in soil and sediment proposes that a continuum of biological and geochemical processes in the micro-environment controls the fate of OM. However, spatio-temporal observation of the biogeochemical nature and behaviour of OM at the soil–water interfaces (SWIs) is impeded by the heterogonous and opaque nature of their microenvironment. Herein, we used a novel SoilChip method (soil microarrays incubated with a predefined solution) to continuously mimic and trace the OM biogeochemistry at SWIs for 21 days. Combining X-ray photoelectron spectroscopy and ion sputtering on SoilChips, we provided the first direct evidence that a nanoscale organic film with a distinct composition and thickness gradually formed at the SWI within 21 days of cultivation. Although the OM coating on the SWI quickly reached equilibrium within 4 days, the formation of thicker mineral–organic association (MOA, 20–130 nm) and microbial biomass (>130 nm) continued, partially at the cost of the thin MOA (<20 nm). Consistent with the thickening SWI, the bioavailability of nutrients (dissolved organic C and ammonium) decreased gradually over 21 days, which restrained the microbial activities. Collectively, thickening SWIs act as a biogeochemical gate to regulate the bioavailability of specific organic compounds and determine their preservation or microbial mineralization. Further, thickening SWIs in the z-axis direction provide direct structural insight to increase carbon sequestration in soil and sediment.

187. Chemically Edited Exosomes with Dual Ligand Purified by Microfluidic Device for Active Targeted Drug Delivery to Tumor Cells

Abstract

Exosomes, which are lipid membrane-bound nanovesicles (50–150 nm in diameter), have aroused extensive attention for their potential applications in invasive molecular and stand for a new therapeutic delivery system. However, they are limited by poor targeting ability and a lack of efficient isolation techniques. Here, we present a three-dimensional nanostructured microfluidic chip, in which arrays of micropillars were functionalized with crisscrossed multiwall carbon nanotubes by chemical deposition, to capture exosomes with high efficiency through a combination of a specific recognition molecule (CD63) and the unique topography of the nanomaterials. As is proven, this nanostructured interface substantially made the immuno capturing of exosomes more efficient. A high percentage of intact vesicles <150 nm were readily purified. As a further application, we added functionality to the exosomes by a chemical editing approach for targeted drug delivery. Donor cells were labeled chemically with dual ligands (biotin and avidin) in the phospholipid membrane and encapsulated drugs in the cytosol. Though the engineered donor cells secreted exosomes, the dual ligands, together with the drugs, were inherited by the exosomes, which were then isolated with the microfluidic chip. Then, the isolated exosomes were used as drug delivery vehicles and showed strong targeting abilities to tumor cells and highly efficient receptor-mediated cellular uptake when exposed to recipient cells. Thus, the anticancer effect of chemotherapeutic drugs was improved significantly. It suggested that this platform could provide a useful tool for isolating intact exosomes with high efficiency and exploiting their natural carrier function to deliver chemotherapeutic drugs to tumor cells with increased efficacy and targeting capacity.

188. Advances in coupling microfluidic chips to mass spectrometry

Abstract

Microfluidic technology has shown advantages of low sample consumption, reduced analysis time, high throughput, and potential for integration and automation. Coupling microfluidic chips to mass spectrometry (Chip-MS) can greatly improve the overall analytical performance of MS-based approaches and expand their potential applications. In this article, we review the advances of Chip-MS in the past decade, covering innovations in microchip fabrication, microchips coupled to electrospray ionization (ESI)-MS and matrix-assisted laser desorption/ionization (MALDI)-MS. Development of integrated microfluidic systems for automated MS analysis will be further documented, as well as recent applications of Chip-MS in proteomics, metabolomics, cell analysis, and clinical diagnosis.

199. Single-cell Analysis with Microfluidic Devices

Abstract

A microfluidic device as a pivotal research tool in chemistry and life science is now widely recognized. Indeed, microfluidic techniques have made significant advancements in fundamental research, such as the inherent heterogeneity of single-cells studies in cell populations, which would be helpful in understanding cellular molecular mechanisms and clinical diagnosis of major diseases. Single-cell analyses on microdevices have shown great potential for precise fluid control, cell manipulation, and signal output with rapid and high throughput. Moreover, miniaturized devices also have open functions such as integrating with traditional detection methods, for example, optical, electrochemical or mass spectrometry for single-cell analysis. In this review, we summarized recent advances of single-cell analysis based on various microfluidic approaches from different dimensions, such as in vitro, ex vivo, and in vivoanalysis of single cells.
