152. A Performance Evaluation Method for Long and Steep Uphill Sections of Heavy-Haul Railway Lines

Abstract

Any system for evaluating the safety service performance of heavy-haul railway lines must effectively reflect the real-time service status of the line. The working conditions of heavy-load lines are complex and diverse, particularly on uphill sections. Existing evaluation systems struggle to accurately reflect the service conditions of long and steep uphill sections bearing heavy loads, posing a significant threat to the safe operation of these lines. To address this problem, we propose a new method for evaluating the safety service performance of long and steep uphill sections of heavy-haul railway lines by establishing a scoring system based on the Analytic Hierarchy Process (AHP). First, damage indicators for heavy-haul lines are categorized into three groups: track geometry status indicators, track structure status indicators, and track traffic status indicators. Using data from existing heavy-haul lines and maintenance experiences, we determine a score deduction standard, classifying lines into four levels based on their safety service quality. Next, we establish a coefficient table for the service performance of long and steep uphill sections after the corresponding scores are deducted. Using data for the length and elevation grade of the actual uphill section, we adjust the deducted scores of the track structure status indicators, enhancing the evaluation system’s accuracy in describing the working conditions. Finally, we verify the stability of the entire system by conducting a sensitivity analysis of the indicator evaluation results using the One-At-a-Time (OAT) method. This method fills a critical gap in the safe operation and maintenance of heavy-haul railways and provides a safety guarantee for the operation of long uphill sections of heavy-haul railways.

Abstract

The breast is a dynamic organ whose response to physiological and pathophysiological conditions alters its disease susceptibility, yet the specific effects of these clinical variables on cell state remain poorly annotated. We present a unified, high-resolution breast atlas by integrating single-cell RNA-seq, mass cytometry, and cyclic immunofluorescence, encompassing a myriad of states. We define cell subtypes within the alveolar, hormone-sensing, and basal epithelial lineages, delineating associations of several subtypes with cancer risk factors, including age, parity, and BRCA2 germline mutation. Of particular interest is a subset of alveolar cells termed basal-luminal (BL) cells, which exhibit poor transcriptional lineage fidelity, accumulate with age, and carry a gene signature associated with basal-like breast cancer. We further utilize a medium-depletion approach to identify molecular factors regulating cell-subtype proportion in organoids. Together, these data are a rich resource to elucidate diverse mammary cell states.

159. ACTFL Chinese Reading Proficiency Guidelines: Verifying the Difficulty Hierarchy

Abstract

The "ACTFL Proficiency Guidelines" serve as a roadmap for foreign/second language (L2) instruction, assessment, curriculum development, and textbook writing. However, very little research has investigated the validity claims of the "ACTFL Guidelines." This study empirically verifies the difficulty hierarchy suggested by descriptors in the "ACTFL Chinese Proficiency Guidelines--Reading" by scaling teacher-perceived descriptor difficulty. Seventeen ACTFL descriptors that represent essential components of L2 Chinese reading ability were selected and compiled as a Likert-scale questionnaire. Ninety-five experienced Chinese language teachers evaluated L2 Chinese learners' reading proficiency using this questionnaire. Data collected were subject to Rasch analysis and analysis of variance test. Data analysis results revealed that the mean difficulties of descriptors on three major ACTFL proficiency levels (Intermediate, Advanced, and Superior) were significantly different. The difficulty of individual descriptors ascended from Intermediate, Advanced, to Superior level, with few exceptions. These findings confirmed the difficulty hierarchy suggested by the "ACTFL Chinese Proficiency Guidelines--Reading."

Abstract

Inspection of tissues using a light microscope is the primary method of diagnosing many diseases, notably cancer. Highly multiplexed tissue imaging builds on this foundation, enabling the collection of up to 60 channels of molecular information plus cell and tissue morphology using antibody staining. This provides unique insight into disease biology and promises to help with the design of patient-specific therapies. However, a substantial gap remains with respect to visualizing the resulting multivariate image data and effectively supporting pathology workflows in digital environments on screen. We, therefore, developed Scope2Screen, a scalable software system for focus+context exploration and annotation of whole-slide, high-plex, tissue images. Our approach scales to analyzing 100GB images of 10^9 or more pixels per channel, containing millions of cells. A multidisciplinary team of visualization experts, microscopists, and pathologists identified key image exploration and annotation tasks involving finding, magnifying, quantifying, and organizing ROIs in an intuitive and cohesive manner. Building on a scope2screen metaphor, we present interactive lensing techniques that operate at single-cell and tissue levels. Lenses are equipped with task-specific functionality and descriptive statistics, making it possible to analyze image features, cell types, and spatial arrangements (neighborhoods) across image channels and scales. A fast sliding-window search guides users to regions similar to those under the lens; these regions can be analyzed and considered either separately or as part of a larger image collection. A novel snapshot method enables linked lens configurations and image statistics to be saved, restored, and shared. We validate our designs with domain experts and apply Scope2Screen in two case studies involving lung and colorectal cancers to discover cancer-relevant image features.

Abstract

We perform a study of the $D_s^+ \to a_0(980)(f_0(980)) e^+ \nu_e$ reactions investigating the different sources of isospin violation which make the production of the $a_0(980)$ possible. We find that loops involving kaons in the production mechanism provide a source of isospin violation since they do not cancel due to the different mass of charged and neutral kaons, but we also find that the main source comes from the breaking of isospin in the meson-meson transition $T$ matrices, which contain information on the nature of the low lying scalar mesons. The reaction is thus very sensitive to the nature of the $a_0(980)$ and $f_0(980)$ resonances. Our results are consistent with the present upper bound for $a_0(980)$ production and only a factor three smaller, indicating that future runs with more statistics should find actual numbers for this reaction from where we can learn more about the origin of the scalar resonances and their nature., Comment: 17 pages, 4 figures, 3 tables; v2: discussion added, references added, version to appear in Eur.Phys.J.C

Abstract

The imminent release of tissue atlases combining multi-channel microscopy with single cell sequencing and other omics data from normal and diseased specimens creates an urgent need for data and metadata standards that guide data deposition, curation and release. We describe a Minimum Information about highly multiplexed Tissue Imaging (MITI) standard that applies best practices developed for genomics and other microscopy data to highly multiplexed tissue images and traditional histology.
