However, the pathogenesis of AM is not clearly understood. We aimed to investigate the pathophysiological changes and molecular mechanism in AM.Methods: Single-cell RNA sequencing (scRNA-seq) was employed to construct a transcriptomic atlas of various cell subsets from the ectopic endometrium (EC) and eutopic endometrium (EM) of one AM patient and evaluate differential expression. The Cell Ranger software pipeline (version 4.0.0) was applied to conduct sample demultiplexing, barcode processing and mapping reads to the reference genome (human GRCh38). Different cell types were classified with markers with the “FindAllMarkers” function, and differential gene expression analysis was performed with Seurat software in R. The findings were confirmed by Reverse Transcription Real-Time PCR using samples from three AM patients.Results: We identified nine cell types: endothelial cells, epithelial cells, myoepithelial cells, smooth muscle cells, fibroblasts, lymphocytes, mast cells, macrophages and unknown cells. A number of differentially expressed genes, including CLO4A1, MMP1, TPM2 and CXCL8, were identified from all cell types. Functional enrichment showed that aberrant gene expression in fibroblasts and immune cells was related to fibrosis-associated terms, such as extracellular matrix dysregulation, focal adhesion and the PI3K-Akt signaling pathway. We also identified fibroblast subtypes and determined a potential developmental trajectory related to AM. In addition, we identified increased cell–cell communication patterns in EC, highlighting the imbalanced microenvironment in AM progression.Conclusion: Our results support the theory of endometrial–myometrial interface disruption for AM, and repeated tissue injury and repair could lead to increased fibrosis in the endometrium. Therefore, the present study reveals the association between fibrosis, the microenvironment, and AM pathogenesis. Abstract

Polylactic acid (PLA) is a kind of biodegradable polymer that has applications in a wide range of biomedical fields and food-contact packaging. However, the mechanical disadvantage limits the application of PLA-based material. The present study modified the pure PLA by orientation and investigated its mechanical and antibacterial properties. The crystallinity and the tensile strength were both drawing ratio dependent. They increased along with the draw ratio to a maximum value of 600% and then decreased. The antibacterial effect of PLA against Staphylococcus aureus and Vibrio paraemolyicus both first increased and then decreased along with the drawing ratio, reaching their highest effect at the drawing ratio of 600%. The inhibition effect of PLA reduced generally with the aging days. 175. The study on the influence factors of intention to continue using MOOCs: integrating UTAUT model and social presence.

Abstract

Massive Open Online Courses (MOOCs) are the product of “internet + education”, offering open educational resources to global students. This study analyzed the factors influencing students’ intention to continue using MOOCs based on their satisfaction. To achieve research objectives this study integrates the unified theory of acceptance and use of technology (UTAUT) and social presence. A total of 481 valid samples were used to verify the hypotheses proposed using structural equation modeling (SEM) and PROCESS. The results showed that the UTAUT model (performance expectancy, effort expectancy, social influence, and facilitating conditions) had significant positive effects on student satisfaction with MOOCs. That is, it had a positive impact on the intention to continue using MOOCs. More importantly, social presence had a significant moderating effect on the relationship between UTAUT scores and satisfaction with MOOCs. Social presence weakened the impact of the UTAUT on satisfaction. Based on the research findings, implications, and limitations of this study are discussed. [ABSTRACT FROM AUTHOR]

Abstract

Mediator is a multiprotein complex integral to the transcription machinery, mediated by RNA polymerase II. Some Mediator subunits have been found to have critical functions in plants' responses to abiotic stresses. However, the role of plant Mediator subunits in drought responses remains largely enigmatic. Here, we identified a Mediator subunit, RhMED15a, in roses (Rosa hybrida). Its expression was greatly and swiftly induced by dehydration treatment in the root. The promoter sequence of RhMED15a contains cis-acting elements that respond to abscisic acid (ABA) and methyl jasmonate (MeJA). In addition, the expression of RhMED15a was significantly up-regulated with ABA treatment and inversely down-regulated with MeJA treatment. Silencing RhMED15a using virus-induced gene silencing (VIGS) in roses significantly reduced drought tolerance in rose plants. This resulted in a significant increase in the malondialdehyde (MDA) level and a decreased survival rate in comparison to TRV controls. Moreover, we found that the expression of five drought-related genes, including dehydration responsive element binding factor 1B (DREB1B), responsive to desiccation stress 29A (RD29A), responsive to desiccation stress 29B (RD29B), early response to dehydration 14 (ERD14), and 9-cis-epoxycarotenoid dioxygenase 1 (NCED1), was considerably suppressed in RhMED15a-silenced plants during drought stress. Taken together, our results present that the Mediator tail module subunit RhMED15a serves as an enhancer of drought tolerance in rose, probably through the modulation of the expression of some drought-related genes. 197. Reliability optimisation of the supply chain system based on seruproduction

Abstract

This paper focuses on the manufacturers implementing seruproduction, and constructs an integer programming model aiming at maximising the reliability of supply chain system based on considering the supplier's on-time supply rate of parts, worker assignment, and production plan comprehensively. In view of the complexity of proposed problem, a hybrid search strategy is designed by combining genetic algorithm (GA) and variable neighbourhood search (VNS), i.e., a hybrid genetic algorithm with variable neighbourhood search (HGAVNS) algorithm based on the multi-level encoding method. To verify the advance of proposed HGAVNS algorithm, the comparison with GA and VNS is presented, and experimental results show that HGAVNS performs better in solving proposed problem.
