Abstract Acoustic emission (AE) characterization is an effective technique to indirectly capture the failure process of quasi brittle rock. In previous studies, both experiments and numerical simulations were adopted to investigate the AE characteristics of rocks. However, as the most popular numerical model, the moment tensor model (MTM) cannot be constrained by the experimental result because there is a gap between MTM and experiments in principle, signal processing and energy analysis. In this paper, we developed a particle-velocity-based model (PVBM) that enabled direct monitoring and analysis of the particle velocity in the numerical model and had good robustness. The PVBM imitated the actual experiment and could fill in gaps between the experiment and MTM. AE experiments of marine shale under uniaxial compression were carried out, and the results were simulated by MTM. In general, the variation trend of the experimental result could be presented by MTM. Nevertheless, the magnitudes of AE parameters by MTM presented notable differences of more than several orders of magnitude compared with those by the experiment. We sequentially used PVBM as a proxy to analyse these discrepancies and systematically evaluate the AE characterization of rocks from the experiment to numerical simulation, considering the influence of wave reflection, energy geometrical diffusion, viscous attenuation, particle size and progressive deterioration of rock material. The combination of MTM and PVBM could reasonably and accurately acquire AE characteristics of the actual AE experiment of rocks by making full use of their respective advantages.

Abstract IL6 is one of the most elevated cytokines during chimeric antigen receptor (CAR) T cell cytokine release syndrome (CRS), and IL6R blockade by Tocilizumab has successfully relieved the most life-threatening aspects of CRS in patients. In addition, latest studies demonstrated the essential role of IL1 in driving CART induced neurotoxicity in mouse models. Here we present a clinical investigation (ChiCTR2000032124; ChiCTR2000031868) of anti-CD19 and anti-BCMA CART (41BBζ) secreting an anti-IL6 scFv and IL1 receptor antagonist (IL1RA) in treating patients with hematologic malignancy. Our results revealed that IL6 and IL1B were maintained at low levels without significant elevation during CRS, rendering Tocilizumab dispensable. Moreover, treated patients did not show neurotoxicity during CRS and exhibited mild to moderate CRS. Notably, we observed high rate of complete response (CR) and significant CART expansion during treatment. In sum, we conclude that CART-secreting anti-IL6 scFv and IL1RA could self-neutralize IL6 storm and maintain low levels of IL1B during CART therapy to minimize IL6- and IL1-associated cytokine toxicity and neurotoxicity without impairing therapeutic efficacy.

Abstract Cancer is one of the most fatal diseases that threaten human health, whereas more than 90% mortality of cancer patients is caused by tumor metastasis, rather than the growth of primary tumors. Thus, how to effectively control or even reverse the migration of tumor cells is of great significance for cancer therapy. CtBP, a transcriptional cofactor displaying high expression in a variety of human cancers, has become one of the main targets for cancer prediction, diagnosis, and treatment. The roles of CtBP in promoting tumorigenesis have been well studied in vitro, mostly based on gain-of-function, while its physiological functions in tumor invasion and the underlying mechanism remain largely elusive. Snail (Sna) is a well-known transcription factor involved in epithelial-to-mesenchymal transition (EMT) and tumor invasion, yet the mechanism that regulates Sna activity has not been fully understood. Using Drosophila as a model organism, we found that depletion of CtBP or snail (sna) suppressed RasV12/lgl -/- -triggered tumor growth and invasion, and disrupted cell polarity-induced invasive cell migration. In addition, loss of CtBP inhibits RasV12/Sna-induced tumor invasion and Sna-mediated invasive cell migration. Furthermore, both CtBP and Sna are physiologically required for developmental cell migration during thorax closure. Finally, Sna activates the JNK signaling and promotes JNK-dependent cell invasion. Given that CtBP physically interacts with Sna, our data suggest that CtBP and Sna may form a transcriptional complex that regulates JNK-dependent tumor invasion and cell migration in vivo.

The prediction of stock prices holds significant implications for researchers and investors evaluating stock value and risk. In recent years, researchers have increasingly replaced traditional machine learning methods with deep learning approaches in this domain. However, the application of deep learning in forecasting stock prices is confronted with the challenge of overfitting. To address the issue of overfitting and enhance predictive accuracy, this study proposes a stock prediction model based on a gated recurrent unit (GRU) with reconstructed datasets. This model integrates data from other stocks within the same industry, thereby enriching the extracted features and mitigating the risk of overfitting. Additionally, an auxiliary module is employed to augment the volume of data through dataset reconstruction, thereby enhancing the model’s training comprehensiveness and generalization capabilities. Experimental results demonstrate a substantial improvement in prediction accuracy across various industries.

Yaw misalignment can make a wake steer, which is an effective method to increase the power of wind farms but it also increases the fatigue load of the turbines. In this paper, the combination of yaw offset and pitch control (CYMP) is studied to analyse the potential mitigation of wake, focusing on the wind velocity and turbulence of the wake distribution, power increment, and fatigue load reduction. The simulation case study shows that the method of CYMP can reduce the fatigue load by 10.29% and increase the total power by 1.7% compared with only wake steering in FAST.Fram. The Collaborative MPC (CoMPC) method based on CYMP is proposed to the real-time wake control in this research, which can increase power by more than 2% and reduce thrust by more than 4% than greedy control under 10 m/s turbulent wind.

Thin and muscular have been characterised as body shape ideals for women and men, respectively, yet each sex misperceives what the other sex desires; women exaggerate the thinness that men like and men exaggerate the muscularity that women like. Body shape ideals align with stereotypic perceptions of femininity in women and masculinity in men. The present study investigates whether misperception of opposite-sex desires extends to femininity/masculinity in facial morphology. We used interactive 3D head models to represent faces varying in sexual dimorphism. White European heterosexual men and women were asked to choose their own and ideal face shape, the ideal shape of a short- and a long-term partner, and the face shape they thought the opposite sex would most like for a short- and a long-term partner. Women overestimated the facial femininity that men prefer in a partner and men overestimated the facial masculinity that women prefer in a partner. The discrepancy between own and ideal sexual dimorphism (an index of appearance dissatisfaction) covaried with by the misperception of what the opposite-sex desires. These results indicate misperception of opposite-sex facial preferences and that mistaken perceptions may contribute to dissatisfaction with own appearance. [ABSTRACT FROM AUTHOR]

Phytoplankton play an irreplaceable role as producers in maintaining lake ecosystems. Nevertheless, scant attention has been given to investigating the dispersion of phytoplankton communities and the factors influencing them across expansive areas. In this study, we present the results of a survey on the distribution of phytoplankton community and the effects of different driving factors in 11 lakes along Inner Mongolia in July–August 2020. Non-metric multidimensional scaling analysis and variance decomposition (VPA) were used to elucidate the distribution of phytoplankton communities and the response of drivers. A total of 169 species of phytoplankton from 8 phyla were detected. Both the abundance and diversity of phytoplankton in the Inner Mongolia lakes showed a trend of high in the east and low in the west (with Daihai Lake as the boundary). The Margalef index of phytoplankton significantly negatively correlated with salinity (r  = −0.707, P  < 0.05) and total dissolved solids (r  = −0.720, P  < 0.05), and both density and biomass highly significantly positively correlated with the suspended solids, Chlorophyll a and trophic level index. The VPA explained 38.9% of the changes in the phytoplankton community with the highest rate of explanation of land use. Therefore, preventing anthropogenic impacts, as well as reducing nutrient loads, can effectively ensure the ecological diversity of lake phytoplankton in lake populations with large geographical spans and varying levels of nutrients. Currently, accelerating the construction of a new development pattern with dual economic circulations is strategically significant for enhancing the autonomy and sustainability of China's economic development. First of all, this paper, from the perspective of green innovation in marine science and technology, measures its efficiency using the SBM super-efficiency model. Then, a coupling coordination model calculates the coordination degree of dual economic circulations after measuring the domestic and international economic circulations based on the TOPSIS method. Secondly, a two-way fixed effects model is employed to examine the impact of green innovation in marine science and technology on domestic economic circulation, international economic circulation, and dual economic circulations. The study finds that green innovation in marine science and technology effectively promotes domestic economic circulation, international economic circulation, and the dual economic circulations. Additionally, green innovation in marine science and technology mitigates the impact of inter-regional resource misalignment on domestic economic circulation and influences international economic circulation by promoting the high-quality development of the marine economy. Finally, it is discovered that green innovation in marine science and technology exhibits a single-threshold heterogeneous effect on domestic economic circulation and international economic circulation. The development of bioactive carrier with controlled delivery of Bone morphogenetic protein (BMP-2) is an important part in bone regeneration. Our study fabricated novel modified nanomaterials based on poly-l-lysine functionalized graphene oxide (GO-PLL) and explore its potential applications for BMP-2 delivery. GO-PLL was used to modify BMP-2 encapsulated bovine serum albumin (BSA) nanoparticles (GO-PLL/BMP-2) by self-assembling technology. The physiochemical and biological properties of GO-PLL modified BSA nanoparticles were investigated in vitro. Additionally, rabbit calvarial bone defects were created to investigate bone formation ability in the presence of GO-PLL/BMP-2 loaded alginate/gelatin scaffolds. The results suggested BSA particles were spatially interacted with GO-PLL complexes by strong electrostatic attraction. When compared with BMP-2 encapsulated BSA nanoparticles, a pronounced decrease in BMP-2 gentle release was observed in the presence of GO-PLL/BMP-2 nanoparticles, and extended to over 14-day release periods. More importantly, GO-PLL/BMP-2 nanoparticles significantly enhanced alkaline phosphatase activity (ALP) and matrix mineralization, and exhibited favorable bone formation ability when loaded into alginate/gelatin scaffolds. These results demonstrated that PLL functionalized GO complexes are considered as appropriate delivery system for BSA modification, and have great potential for BMP-2 delivery in bone regeneration.

Throughout their lifespan, monopile foundations supporting offshore wind turbines inevitably experience horizontal loads from waves, winds, and currents, resulting in cumulative deformation. It has been believed that deformation caused by horizontal loading weakens the interaction between the pile and the soil, leading to a reduction in the ultimate uplift bearing capacity of the pile foundation. However, there is a scarcity of literature investigating this issue, particularly regarding monopiles used in offshore wind turbine installations. Therefore, this study aims to explore the impact of horizontal cyclic loads on the ultimate uplift bearing capacity of monopile, focusing on the pile–soil interaction. To achieve this, a series of 1 g model tests were conducted on a rigid model pile embedded in silt with varying relative compaction. The test results indicate that the ultimate uplift bearing capacity of the pile is significantly diminished after experiencing horizontal cyclic loading, and the extent of reduction is closely linked to the amplitude of the horizontal deformation. A semi-empirical model is developed to predict the ultimate uplift bearing capacity of the pile foundation following horizontal cyclic loading. The key findings of this study are as follows: (1) The earth pressure in the active zone gradually decreases with an increasing number of cycles, while the earth pressure in the passive zone experiences a slight increase under horizontal cyclic loading. (2) The position of the pile rotation center under horizontal cyclic loading is approximately 0.84 times the depth at which the pile is buried, and this relationship appears to be independent of soil density and cyclic load ratio. (3) The variation of earth pressure corresponding to the horizontal deformation of the pile in the active zone can be divided into three phases: a rapid decline phase, a slow decline phase, and a stable phase. (4) The reduction in the ultimate uplift capacity is influenced by the cyclic ratio and number of cycles but does not appear to have a significant relationship with soil density.
