Large dip‐slip earthquakes have a major contribution to mountain building while earthquake‐induced landslides lower mountains simultaneously. The amount of the coseismic uplift and landslides may dominate long‐term mountain evolution. However, how earthquakes contribute to mountain evolution through coseismic uplift and landslides is less constrained in real cases. We present the regional coseismic uplift of the 1999 Mw7.6 Chi‐Chi earthquake by using synthetic aperture radar (SAR) images and GPS. The coseismic uplift pattern is consistent with field observations showing increasing movement to the north with ~8 m of uplift toward the northern end. We estimated uplifted rock volume of 2.60 ± 1.09 km3, which is five times greater than the coseismic landslide volume. Intense erosion of the Taiwan orogen may erode elevated rocks rapidly, but the uplift and landslide distributions do not match and correlate more inversely, suggesting the frontal orogenic topography should be increased rather than annulled over earthquake cycles. Plain Language Summary: Earthquakes can raise mountains but they can cause landslides to lower the mountains at the same time. For large earthquakes, mountains could reduce the size due to more landslides than uplift. It is important to analyze earthquakes occurred within mountain ranges to examine how much volume of rocks is elevated and how much volume of rocks is trampled down. Because complete observations of uplift and landslides for large earthquakes are rare, we used geodetic methods to estimate the volume of the elevated rocks for one of the largest earthquakes in mountain ranges in the world, the Chi‐Chi, Taiwan, earthquake in 1999. The volume of elevated rocks is much larger than the landslide volume, and the uplift area is close to the frontal mountains and the landslide areas are distributed in the deeper mountains. If this finding holds for other mountain ranges in similar settings, this means that large earthquakes should raise mountains, especially in the frontal part of the mountains. We report the fabrication, characterization, and field emission properties of (silicon oxide) SiO
x coated thin-multiwalled carbon nanotubes (t-MWNTs). The coated t-MWNTs show improved field emission behavior. Initially, raw t-MWNTs (diameter ∼5–8 nm) were functionalized by acid treatment. Using spin on glass as a Si precursor, SiOx was coated on the nanotubes by routine chemical methods. The coated samples were characterized by high resolution transmission electron microscopy (HRTEM), Fourier transform infrared (FTIR) spectroscopy, and thermogravimetric analysis (TGA) techniques. The HRTEM results show that the local thickness of the coating varies from ∼1 to 2 nm. The FTIR investigations show the formation of nanophases, such as Si–C, Si–O–C, and intercalated Si–O, at the coating/nanotube interface. The TGA reveals that the coating prevents the high-temperature oxidation and degradation of the nanotubes. The field emission characteristics of the coated, functionalized, and raw nanotubes show that the turn-on fields and current density are improved for the coated nanotubes. This improvement was attributed to the lowering of the work function and dielectric constant of the C/SiOx interface layer and the localization of the density of states close to the Fermi energy for the coated nanotubes. The analysis of the emission stability spectra shows that the coated nanotubes have a more favorable lifetime. The observed enhancement was attributed to the protection of the nanotubes from the reactive sputter etching during the field emission process. The details of the analysis are presented. While many transition metal (TM) nitrides—including TiN, ZrN, and TaN—have been widely studied and are currently used as hard wear-resistant coatings, diffusion barriers, and optical coatings, little is known about a related TM nitride, HfN. Here, we report the results of a systematic investigation of the growth and physical properties of HfNx layers, with 0.80<=x<=1.50, deposited on MgO(001) by ultrahigh vacuum reactive magnetron sputtering at 650 °C in mixed N2/Ar discharges. HfNx layers with 0.80<=x<=1.20 crystallize in the B1–NaCl structure with a cube-on-cube epitaxial relationship to the MgO(001) substrate, while films with 1.24<=x<=1.50 contain a N-rich second phase. The relaxed bulk lattice parameter of HfNx(001) decreases only slightly with increasing N/Hf ratio, ranging from 0.4543 nm with x=0.80 to 0.4517 nm with x=1.20. The room-temperature resistivity ρ of stoichiometric HfN(001) is 14.2 μΩ cm and ρ(x) increases with both increasing and decreasing x to 140 μΩ cm with x=0.80 and 26.4 μΩ cm with x=1.20. The hardness H and elastic modulus E of HfN(001) are 25.2 and 450 GPa, respectively. H(x) initially increases for both over- and understoichiometric layers due to defect-induced hardening, while E(x) remains essentially constant. Single-phase HfNx(001) is metallic with a positive temperature coefficient of resistivity (TCR) between 50 and 300 K and a temperature-independent carrier density. It is also superconducting with the highest critical temperature, 9.18 K, obtained for layers with x=1.00. In the two phase regime, ρ ranges from 59.8 μΩ cm with x=1.24 to 2710 μΩ cm with x=1.50. TCR becomes positive with x>=1.38, no superconducting transition is observed, and both H and E decrease. [ABSTRACT FROM AUTHOR]

Stoichiometric HfN layers, with N/Hf=1.0±0.03, were grown on MgO(001) substrates at 650 °C by ultrahigh-vacuum magnetically unbalanced magnetron sputter deposition in mixed N2/Ar discharges at 20 mTorr (2.67 Pa). High-resolution x-ray diffraction ω-2θ and azimuthal [lowercase_phi_synonym] scans combined with cross-sectional transmission electron microscopy establish that HfN grows epitaxially with a cube-on-cube orientational relationship to the substrate: (001)HfN∥(001)MgO and [100]HfN∥[100]MgO. The layers are fully relaxed at the growth temperature and have a room-temperature bulk lattice constant of 0.4524 nm. Electronic transport measurements show that HfN is metallic with a room-temperature resistivity of 14.2 μΩ cm, an n-type carrier concentration of 4.8×1021 cm-3, and an electron mobility of 86 cm2 V-1 s-1. The resistivity ρ remains constant at 3.5 μΩ cm, limited by defect scattering, between 10 and 50 K, while at higher temperatures ρ increases linearly and is limited primarily by phonon scattering. HfN(001) is also superconducting with a critical temperature of 9.18 K. We have grown single-crystal NaCl-structure δ-TiN[sub x] layers with x ranging from 0.67 to 1.00 on MgO(001) at 700 °C by ultra-high-vacuum reactive magnetron sputtering of Ti in mixed Ar/N[sub 2] discharges in order to investigate microstructural evolution and the physical properties of TiN[sub x] as a function of the N vacancy concentration. High-resolution x-ray diffraction and transmission electron microscopy results show that all layers grow with a cube-on-cube epitaxial relationship to the substrate, (001)[sub TiN]∥(001)[sub MgO] and [100][sub TiN]∥[100][sub MgO]. The relaxed lattice parameter a[sub o](x) decreases linearly from 4.240 Å with x=1.00 to 4.226 Å with x=0.67. Stoichiometric TiN(001) layers are fully relaxed at the growth temperature while layers with 0.67<=x<=0.92 are fully coherent with their substrates. Surface morphologies vary dramatically with x. TiN[sub x](001) layers with x=0.67–0.82 have very flat surfaces arising from large cation surface diffusion lengths approaching values corresponding to step flow. However, the surfaces of the TiN[sub 0.92](001) and TiN[sub 1.00](001) layers, which were grown at higher N[sub 2] partial pressures, consist of a periodic two-domain ripple structure along the <110> directions due to kinetic roughening associated with lower cation surface mobilities resulting from higher steady state N coverages. TiN[sub 1.0](001) layers grown in pure N[sub 2] exhibit growth mounds that are predominantly square with edges aligned along the <110> directions. The room-temperature resistivity, 13 μΩ cm with x=1.00, increases from 52 μΩ cm for TiN[sub x](001) layers with x=0.92 to 192 μΩ cm with x=0.67, due primarily to increased carrier scattering from N vacancies. © 2004 American Institute of Physics. While NaCl-structure transition-metal nitrides have been widely studied over the past two decades, little is known about the corresponding NaCl-structure rare-earth nitrides. Polycrystalline CeN, for example, has been reported by different groups to be both a wide band-gap semiconductor and a metal. To address this controversy, we have grown epitaxial CeN layers on MgO(001) and measured their physical properties. The films were grown at 700°C by ultrahigh vacuum reactive magnetron sputter deposition in mixed Ar/N[SUB2] discharges maintained at 4 mTorr)(0.53 Pa). X-ray diffraction and transmission electron microscopy results establish the film/substrate epitaxial relationship as cube-on-cube, (001)[SUBCeN]‖(001)[SUBMgO] with [100][SUBCeN]‖[100][SUBMgO], while Rutherford backscattering spectroscopy shows that the layers are stoichiometric with N/Ce=0.99Plusmn;0.02. CeN is metallic with a positive temperature coefficient of resistivity and a temperature-independent carrier concentration, as determined by Hall effect measurements, of 2.8±0.2×10[SUP22]cm[SUP-3] with a room temperature mobility of 0.31 cm[SUP2]V[SUP-1]s[SUP-1]. At temperatures between 2 and 50 K, the resistivity is limited by defect scattering and remains constant at 29 μΩ cm, while at higher temperatures it increases linearly, limited primarily by phonon scattering, to reach a room-temperature value of 68.5 μΩ cm. The hardness and elastic modulus of CeN(001) were determined from nanoindentation measurements to be 15.0±0.9 and 330±16 GPa. [ABSTRACT FROM AUTHOR]

Leptospirosis remains one of the most widespread zoonotic diseases caused by spirochetes of the genus Leptospira, which accounts for high morbidity and mortality globally. Leptospiral infections are often found in tropical and subtropical regions, with people exposed to contaminated environments or animal reservoirs are at high risk of getting the infection. Leptospirosis has a wide range of clinical manifestations with non-specific signs and symptoms and often misdiagnosed with other acute febrile illnesses at early stage of infection. Despite being one of the leading causes of zoonotic morbidity worldwide, there is still a gap between pathogenesis and human immune responses during leptospiral infection. It still remains obscure whether the severity of the infection is caused by the pathogenic properties of the Leptospira itself, or it is a consequence of imbalance host immune factors. Hence, in this review, we seek to summarize the past and present milestone findings on the biomarkers of host immune response aspects during human leptospiral infection, including cytokine and other immune mediators. A profound understanding of the interlink between virulence factors and host immune responses during human leptospirosis is imperative to identify potential biomarkers for diagnostic and prognostic applications as well as designing novel immunotherapeutic strategies in future.

The electromigration damage in flip chip solder bumps of eutectic SnPb and SnAg[sub 3.8]Cu[sub 0.7] was studied after current stressing at 120 °C with 1.5 A. The diameter of the bumps was about 200 μm. The under-bump metallization (UBM) on the chip side was electroless Ni and on the board side was electroplated Cu. Surface marker motion was used to measure the atomic flux driven by electromigration and to calculate the effective charge number, Z[sup *], of the solder. For eutectic SnPb, Z[sup *] is about 36–100 after 39.5 h of electromigration, but for SnAg[sub 3.8]Cu[sub 0.7] the marker movement was too small to measure Z[sup *] even after 200 h of current stressing. While the effect of electromigration in SnAg[sub 3.8]Cu[sub 0.7] is much smaller than that in eutectic SnPb, hillocks of intermetallic compounds at the anode have been observed in the former. An extensive growth of Ni–Cu–Sn intermetallic compounds in the matrix of both solder bumps was found. A localized dissolution of electroless Ni UBM was observed under current stressing, yet no dissolution of the Cu UBM by electromigration was found. © 2001 American Institute of Physics. The reaction kinetics of eutectic SnPb solder on Cu were studied and compared in the liquid state at 200 to 240 °C and in the solid state aged at 125-170 °C. The ternary phase diagrams of SnPbCu, the morphology of intermetallic compound (IMC), and the kinetics of growth of the intermetallics were used in the comparison. The temperature difference between these two reactions is only 30 °C, but the kinetics of reaction, as well as the morphology of IMC formation, are very different. The kinetics in the wetting reaction is four orders of magnitude faster than that in solid state aging. The Cu[sub 6]Sn[sub 5] intermetallic morphology in solid state aging is a layer type, but it has a scallop-type morphology in the wetting reaction. The morphology strongly affects the kinetics. While the kinetic difference can be attributed to the difference in atomic diffusivity between the liquid state and the solid state, it is the morphology that determines the kinetic path in these reactions. We conclude that a fast rate of reaction, which leads to a high rate of Gibbs free energy change, controls the reaction, not the Gibbs free energy change itself. © 2001 American Institute of Physics. The electromigration of eutectic SnPb solder interconnects between a Si chip and a FR4 substrate was studied at 120 °C for up to 324 h with current stressing of 10[sup 4] amp/cm[sup 2]. Hillocks were observed at the anode and voids at the cathode. The dominant diffusing species was found to be Pb, confirmed by its accumulation at the anode. Diffusion markers were used to measure the electromigration flux and calculate the effective charge of atomic diffusion in the solder. The morphology of interfacial reaction products between four lead-free solder alloys on electroless Ni-P was characterized. The four Pb-free solders were 99.3Sn0.7Cu, 95.5Sn3.8Ag0.7Cu, 96.5Sn3.5Ag, and 96Sn2Ag2Bi (in wt%) alloys. After reflow, the interfacial intermetallics in the first two solders that contain Cu (99.3Sn0.7Cu and 95.5Sn3.8Ag0.7Cu) had good adhesion with electroless Ni-P. However, the 96.5Sn3.5Ag and 96Sn2Ag2Bi alloys formed interfacial intermetallics with a needle shaped morphology that spalled off the surface of electroless Ni-P. This difference is attributed to the role of Cu in the solders (which modified the chemical potential of the interfacial intermetallics), the volume change that occurs during intermetallic formation, and the interfacial properties of the compound. In solid state aging experiments, the consumption of electroless Ni-P by intermetallic growth was not significant (approximately 1 μm) and all the intermetallics had good adhesion to the electroless Ni-P. The electroless Ni-P exhibited some damage at the outer edge of the bond pad due to stress imposed during solid state aging. Large Ag[sub 3]Sn and Cu[sub 6]Sn[sub 5] intermetallics were observed in the bulk of the solders (except for SnAgBi solder), and these intermetallics are discussed in terms of soldering reaction at the interface and phase equilibrium. © 2000 American Institute of Physics. Background: Clinical and molecular factors are essential to define the prognosis in patients with glioblastoma (GBM). O6-methylguanine-DNA methyltransferase (MGMT) methylation status, age, Karnofsky Performance Status (KPS), and extent of surgical resection are the most relevant prognostic factors. Our investigation of the role of gender in predicting prognosis shows a slight survival advantage for female patients. Methods: We performed a prospective evaluation of the Project of Emilia Romagna on Neuro-Oncology (PERNO) registry to identify prognostic factors in patients with GBM who received standard treatment. Results: A total of 169 patients (99 males [58.6%] and 70 females [41.4%]) were evaluated prospectively. MGMT methylation was evaluable in 140 patients. Among the male patients, 36 were MGMT methylated (25.7%) and 47 were unmethylated (33.6%); among the female patients, 32 were methylated (22.9%) and 25 were unmethylated (17.9%). Survival was longer in the methylated females compared with the methylated males (P = 0.028) but was not significantly different between the unmethylated females and the unmethylated males (P = 0.395). In multivariate analysis, gender and MGMT methylation status considered together (methylated females vs. methylated males; hazard ratio [HR], 0.459; 95% confidence interval [CI], 0.242-0.827; P = 0.017), age (HR, 1.025; 95% CI, 1.002-1.049; P = 0.032), and KPS (HR, 0.965; 95% CI, 0.948-0.982; P < 0.001) were significantly correlated with survival. Conclusions: Survival was consistently longer among MGMT methylated females compared with males. Gender can be considered as a further prognostic factor.

Objectives:To estimate a regional progression pattern of amyloid deposition from cross-sectional amyloid-sensitive PET data and evaluate its potential for in vivo staging of an individual's amyloid pathology.Methods:Multiregional analysis of florbetapir (18F-AV45)–PET data was used to determine individual amyloid distribution profiles in a sample of 667 participants from the Alzheimer's Disease Neuroimaging Initiative cohort, including cognitively normal older individuals (CN) as well as patients with mild cognitive impairment and Alzheimer disease (AD) dementia. The frequency of regional amyloid positivity across CN individuals was used to construct a 4-stage model of progressing amyloid pathology, and individual distribution profiles were used to evaluate the consistency of this hierarchical stage model across the full cohort.Results:According to a 4-stage model, amyloid deposition begins in temporobasal and frontomedial areas, and successively affects the remaining associative neocortex, primary sensory-motor areas and the medial temporal lobe, and finally the striatum. Amyloid deposition in these brain regions showed a highly consistent hierarchical nesting across participants, where only 2% exhibited distribution profiles that deviated from the staging scheme. The earliest in vivo amyloid stages were mostly missed by conventional dichotomous classification approaches based on global florbetapir-PET signal, but were associated with significantly reduced CSF Aβ42 levels. Advanced in vivo amyloid stages were most frequent in patients with AD and correlated with cognitive impairment in individuals without dementia.Conclusions:The highly consistent regional hierarchy of PET-evidenced amyloid deposition across participants resembles neuropathologic observations and suggests a predictable regional sequence that may be used to stage an individual's progress of amyloid pathology in vivo.

Although many advances in understanding the cellular and molecular mechanisms for Alzheimer's disease have been made in the past decades, drug development has failed to significantly improve drug treatment of the disease. This is partly because failed clinical trials have focused on efficacy in terms of cognitive improvement rather than the rigorous evaluation of putative disease mechanisms and the effect of the tested drugs on these mechanisms. The use of imaging biomarkers of neurodegenerative disease, especially those from magnetic resonance imaging (MRI) and positron emission tomography (PET), could substantially accelerate the development of novel therapeutics by providing quantitative metrics of disease progression that relate directly to the underlying pathology. Herein, new MRI and PET imaging techniques relevant to neurodegeneration are briefly introduced.

Objective:To comprehensively assess neurobiological effects of the protective APOE2 allele in the aged brain using a cross-sectional multimodal neuroimaging approach.Methods:Multimodal neuroimaging data were obtained from a total of 572 older individuals without dementia (cognitively normal and mild cognitive impairment) enrolled in the Alzheimer's Disease Neuroimaging Initiative and included assessments of regional amyloid load with AV45-PET, glucose metabolism with fluorodeoxyglucose-PET, and gray matter volume with structural MRI. Imaging indexes of APOE2 carriers were contrasted to risk-neutral APOE3 homozygotes, and analyses were controlled for age, sex, education, and clinical diagnosis. Additional models examined genotype-specific effects of age on the imaging markers.Results:In region-of-interest–based analyses, APOE2 carriers had significantly less precuneal amyloid pathology and did not show the typical age-related increase in amyloid load, although the age × genotype interaction was only trend-level significant. In contrast, parietal metabolism and hippocampal volume did not differ between APOE2 and APOE3 genotypes, and both groups showed comparable negative effects of age on these markers. The amyloid specificity of APOE2-related brain changes was corroborated in 2 complementary analyses: spatially unbiased voxel-wise analyses showing widespread reductions in amyloid deposition but no differences in gray matter volume or metabolism and an analysis of CSF-based biomarkers showing a significant effect on amyloid but not on tau pathology.Conclusions:Regarding the range of Alzheimer disease biomarkers considered in the present study, the APOE2 allele appears to have a relatively selective effect on reduced accumulation of amyloid pathology in the aged brain.

Weight changes are common in aging and Alzheimer's disease (AD) and postmortem findings suggest a relation between lower body mass index (BMI) and increased AD brain pathology. In the current multicenter study, we tested whether lower BMI is associated with higher core AD brain pathology as assessed by cerebrospinal fluid (CSF)-based biological markers of AD in 751 living subjects: 308 patients with AD, 296 subjects with amnestic mild cognitive impairment (MCI), and 147 elderly healthy controls (HC). Based upon a priori cutoff values on CSF concentration of total tau and beta-amyloid (Aβ(1-42)), subjects were binarized into a group with abnormal CSF biomarker signature (CSF+) and those without (CSF-). Results showed that BMI was significantly lower in the CSF+ when compared with the CSF- group (F = 27.7, df = 746, p0.001). There was no interaction between CSF signature and diagnosis or apolipoprotein E (ApoE) genotype. In conclusion, lower BMI is indicative of AD pathology as assessed with CSF-based biomarkers in demented and nondemented elderly subjects.

Contains fulltext : 108202.pdf (Publisher’s version ) (Closed access) Identifying genetic variants influencing human brain structures may reveal new biological mechanisms underlying cognition and neuropsychiatric illness. The volume of the hippocampus is a biomarker of incipient Alzheimer's disease and is reduced in schizophrenia, major depression and mesial temporal lobe epilepsy. Whereas many brain imaging phenotypes are highly heritable, identifying and replicating genetic influences has been difficult, as small effects and the high costs of magnetic resonance imaging (MRI) have led to underpowered studies. Here we report genome-wide association meta-analyses and replication for mean bilateral hippocampal, total brain and intracranial volumes from a large multinational consortium. The intergenic variant rs7294919 was associated with hippocampal volume (12q24.22; N = 21,151; P = 6.70 x 10(-16)) and the expression levels of the positional candidate gene TESC in brain tissue. Additionally, rs10784502, located within HMGA2, was associated with intracranial volume (12q14.3; N = 15,782; P = 1.12 x 10(-12)). We also identified a suggestive association with total brain volume at rs10494373 within DDR2 (1q23.3; N = 6,500; P = 5.81 x 10(-7)). 01 mei 2012

Loss-of-function mutations in the genes associated with primary microcephaly (MCPH) reduce human brain size by about two-thirds, without producing gross abnormalities in brain organization or physiology and leaving other organs largely unaffected [Woods CG, et al. (2005) Am J Hum Genet 76:717–728]. There is also evidence suggesting that MCPH genes have evolved rapidly in primates and humans and have been subjected to selection in recent human evolution [Vallender EJ, et al. (2008) Trends Neurosci 31:637–644]. Here, we show that common variants of MCPH genes account for some of the common variation in brain structure in humans, independently of disease status. We investigated the correlations of SNPs from four MCPH genes with brain morphometry phenotypes obtained with MRI. We found significant, sex-specific associations between common, nonexonic, SNPs of the genes CDK5RAP2 , MCPH1 , and ASPM , with brain volume or cortical surface area in an ethnically homogenous Norwegian discovery sample ( n = 287), including patients with mental illness. The most strongly associated SNP findings were replicated in an independent North American sample ( n = 656), which included patients with dementia. These results are consistent with the view that common variation in brain structure is associated with genetic variants located in nonexonic, presumably regulatory, regions.

Posterior cingulate cortex (PCC) hypometabolism as measured by FDG PET is an indicator of Alzheimer’s disease (AD) in prodromal stages, such as in mild cognitive impairment (MCI), and has been found to be closely associated with hippocampus atrophy in AD dementia. We studied the effects of local and remote atrophy and of local amyloid load on the PCC metabolic signal in patients with different preclinical and clinical stages of AD. We determined the volume of the hippocampus and PCC grey matter based on volumetric MRI scans, PCC amyloid load based on AV45 PET, and PCC metabolism based on FDG PET in 667 subjects participating in the Alzheimer’s Disease Neuroimaging Initiative spanning the range from cognitively normal ageing through prodromal AD to AD dementia. In cognitively normal individuals and those with early MCI, PCC hypometabolism was exclusively associated with hippocampus atrophy, whereas in subjects with late MCI it was associated with both local and remote effects of atrophy as well as local amyloid load. In subjects with AD dementia, PCC hypometabolism was exclusively related to local atrophy. Our findings suggest that the effects of remote pathology on PCC hypometabolism decrease and the effects of local pathology increase from preclinical to clinical stages of AD, consistent with a progressive disconnection of the PCC from downstream cortical and subcortical brain regions.

Single-crystal NaCl-structure delta-TiNx layers with x raging from 0.67 to 1.00 are grown at 700 degree C by ultra-high-vacuum magnetron sputtering of Ti in mixed Ar/N2 discharges to investigate microstructural evolution and the physical properties of TiNx as a function of the N vacancy concentration. Findings show that room-temperature resistivity, 13 Mu Omega cm with x=1.00, increases from 53 Mu Omega cm for TiNx(001) layers with x=0.92 to 192 Mu Omega cm with x=0.67, due primarily to increased carrier scattering from N vacancies.

Subcortical brain regions form circuits with cortical areas to coordinate movement, learning, memory and motivation, and altered circuits can lead to abnormal behaviour and disease. To investigate how common genetic variants affect the structure of these brain regions, here we conduct genome-wide association studies of the volumes of seven subcortical regions and the intracranial volume derived from magnetic resonance images of 30,717 individuals from 50 cohorts. We identify five novel genetic variants influencing the volumes of the putamen and caudate nucleus. We also find stronger evidence for three loci with previously established influences on hippocampal volume and intracranial volume. These variants show specific volumetric effects on brain structures rather than global effects across structures. The strongest effects were found for the putamen, where a novel intergenic locus with replicable influence on volume (rs945270; P = 1.08 x 10(-33); 0.52% variance explained) showed evidence of altering the expression of the KTN1 gene in both brain and blood tissue. Variants influencing putamen volume clustered near developmental genes that regulate apoptosis, axon guidance and vesicle transport. Identification of these genetic variants provides insight into the causes of variability in human brain development, and may help to determine mechanisms of neuropsychiatric dysfunction. 6 p.

- Abstract
Often multiple instances of an object occur in the same scene, for example in a warehouse. Unsupervised multi-instance object discovery algorithms are able to detect and identify such objects. We use such an algorithm to provide object proposals to a convolutional neural network (CNN) based classifier. This results in fewer regions to evaluate, compared to traditional region proposal algorithms. Additionally, it enables using the joint probability of multiple instances of an object, resulting in improved classification accuracy. The proposed technique can also split a single class into multiple sub-classes corresponding to the different object types, enabling hierarchical classification., QC 20171130

In a large multicentre sample of cognitively normal subjects, as a function of age, gender and APOE genotype, we studied the frequency of abnormal cerebrospinal fluid levels of Alzheimers disease biomarkers including: total tau, phosphorylated tau and amyloid-β1-42. Fifteen cohorts from 12 different centres with either enzyme-linked immunosorbent assays or Luminex® measurements were selected for this study. Each centre sent nine new cerebrospinal fluid aliquots that were used to measure total tau, phosphorylated tau and amyloid-β1-42 in the Gothenburg laboratory. Seven centres showed a high correlation with the new Gothenburg measurements; therefore, 10 cohorts from these centres are included in the analyses here (1233 healthy control subjects, 4084 years old). Amyloid-β amyloid status (negative or positive) and neurodegeneration status (negative or positive) was established based on the pathological cerebrospinal fluid Alzheimers disease cut-off values for cerebrospinal fluid amyloid-β1-42 and total tau, respectively. While gender did not affect these biomarker values, APOE genotype modified the age-associated changes in cerebrospinal fluid biomarkers such that APOE ε4 carriers showed stronger age-related changes in cerebrospinal fluid phosphorylated tau, total tau and amyloid-β1-42 values and APOE ε2 carriers showed the opposite effect. At 40 years of age, 76% of the subjects were classified as amyloid negative, neurodegeneration negative and their frequency decreased to 32% at 85 years. The amyloid-positive neurodegeneration-negative group remained stable. The amyloid-negative neurodegeneration-positive group frequency increased slowly from 1% at 44 years to 16% at 85 years, but its frequency was not affected by APOE genotype. The amyloid-positive neurodegeneration-positive frequency increased from 1% at 53 years to 28% at 85 years. Abnormally low cerebrospinal fluid amyloid-β1-42 levels were already frequent in midlife and APOE genotype strongly affects the levels of cerebrospinal fluid amyloid-β1-42, phosphorylated tau and total tau across the lifespan without influencing the frequency of subjects with suspected non-amyloid pathology.
