Abstract

BACKGROUND—: Atherosclerosis is driven by synergistic interactions between pathological biomechanical, inflammatory and lipid metabolic factors. Our previous studies demonstrated that absence of caveolin-1 (Cav1)/caveolae in hyperlipidemic mice strongly inhibits atherosclerosis, which was attributed to activation of endothelial nitric oxide synthase (eNOS) and increased production of nitric oxide (NO), reduced inflammation and low-density lipoprotein (LDL) trafficking. However, the contribution of eNOS activation and NO production in the athero-protection of Cav1 and the exact mechanisms by which Cav1/caveolae controls the pathogenesis of diet-induced atherosclerosis are still not clear. METHODS—: Triple knockout mouse lacking expression of eNOS, Cav1 and Ldlr were generated to explore the role of NO production in Cav1-dependent atheo-protective function. The effects of Cav1 on lipid trafficking, extracellular matrix (ECM) remodeling and vascular inflammation were studied both in vitro and in vivo using a mouse model of diet-induced atherosclerosis. The expression of Cav1 and distribution of caveolae regulated by flow were analyzed by immunofluorescence staining and transmission electron microscopy. RESULTS—: We found that absence of Cav1 significantly suppressed atherogenesis in Ldlr(−/−)eNOS(−/−) mice, demonstrating that athero-suppression is independent of increased NO production. Instead, we find that absence of Cav1/caveolae inhibited LDL transport across the endothelium, pro-atherogenic fibronectin deposition, and disturbed flow-mediated endothelial cell (EC) inflammation. Consistent with the idea that Cav1/caveolae may play a role in early flow-dependent inflammatory priming, distinct patterns of Cav1 expression and caveolae distribution were observed in athero-prone and athero-resistant areas of the aortic arch even in wild-type mice. CONCLUSIONS—: The above findings support a role for Cav1/caveolae as a central regulator of atherosclerosis that links biomechanical, metabolic and inflammatory pathways independent of endothelial eNOS activation and NO production.

Abstract

Attempts to optimize pharmacokinetic properties in a promising series of pyrrolopyrimidinone MARK inhibitors for the treatment of Alzheimer's disease are described. A focus on physical properties and ligand efficiency while prosecuting this series afforded key tool compounds that revealed a large discrepancy in the rat in vitro-in vivo DMPK (Drug Metabolism/Pharmacokinetics) correlation. These differences prompted an in vivo rat disposition study employing a radiolabeled representative of the series, and the results from this experiment justified the termination of any further optimization efforts.

180. Investigation of Pneumatic Inlet and Diffuser Blowing on a Ducted Fan Propulsor in Static Thrust Operation

Abstract

Tilting ducted fans present a solution for the lifting and forward flight propulsion requirements of VTOL aircraft. However, the geometry of the duct enshrouding the propeller has great a effect on the efficiency of the fan in various flight modes. Shroud geometry controls the velocity and pressure at the face of the fan, while maintaining a finite loading out at the tips of the fan blades. A duct tailored for most efficient generation of static lifting thrust will generally suffer from performance deficiencies in forward flight. The converse is true as well, leaving the designer with a difficult trade affecting the overall performance and sizing of the aircraft. Ideally, the shroud of a vertical lifting fan features a generous bell mouth inlet promoting acceleration of flow into the face of the fan, and terminating in a converging nozzle at the exit. Flow entering the inlet is accelerated into the fan by the circulation about the shroud, resulting in an overall increase in thrust compared to an open propeller operating under the same conditions . The accelerating shroud design is often employed in lifting ducted fans to benefit from the thrust augmentation; however, such shroud designs produce significant drag penalties in axial flight, thus are unsuitable for efficient forward flight applications. Decelerating, or diffusing, duct designs are employed for higher speed forward flight configurations. The lower circulation on the shroud tends to decelerate the flow into the face of the fan, which is detrimental to static thrust development; however, net thrust is developed on the shroud while the benefits of finite blade loading are retained. With judicious shroud design for intended flight speeds, a net increase in efficiency can be obtained over an open propeller. In this experiment, conducted under contract to NASA LaRC (contract NAG-1-02093) circulation control is being applied to a mildly diffusing shroud design, intended for improved forward flight performance, to generate circulation in the sense of an accelerating duct design. The intent is to improve static thrust performance of a ducted fan tailored for high speed axial flight, while at the same time significantly reduce the pressure signature on the ground plane. Circulation control on the fan shroud is achieved by the Coanda effect.

Abstract

The ocean biogeochemistry components of the Canadian Earth System Model v. 5 are presented and compared to observations and other models. CanESM5 employs the same biogeochemistry module as CanESM2 whereas CanESM5-CanOE ("Canadian Ocean Ecosystem model") is a new, more complex biogeochemistry module developed for Sixth Coupled Model Intercomparison Project (CMIP6), with multiple food chains, flexible phytoplankton elemental ratios, and a prognostic iron cycle. This new model is described in detail and the outputs compared to CanESM5 and CanESM2, as well as to observations and other CMIP6 models. Both CanESM5 models show gains in skill relative to CanESM2, which are attributed primarily to improvements in ocean circulation. CanESM5-CanOE shows improved skill relative to CanESM5 in some areas. CanESM5-CanOE includes a prognostic iron cycle, and maintains high nutrient / low chlorophyll conditions in the expected regions (in CanESM2 and CanESM5, iron limitation is specified as a temporally static 'mask'). Surface nitrate concentrations are biased low in the subarctic Pacific and equatorial Pacific, and high in the Southern Ocean. Export production in CanESM5-CanOE is among the lowest for CMIP6 models; in CanESM5 it is among the highest, but shows the most rapid decline after about 1980. CanESM5-CanOE has relatively low concentrations of zooplankton and detritus relative to phytoplankton, and a high and relatively constant living phytoplankton fraction of total particulate organic matter. In most regions, large and small phytoplankton show decoupled seasonal cycles with greater abundance of large phytoplankton in the productive seasons. Cumulative ocean uptake of anthropogenic carbon dioxide through 2014 is lower in both CanESM5 models than in observation-based estimates or the model ensemble mean, and is lower in CanESM5-CanOE (122PgC) than in CanESM5 (132PgC). 195. Redesigning Community as an Ecovillage

Abstract

This chapter explores, analyses and celebrates the potential for regenerative human habitat and sustainable solutions offered by ecovillages. Broadly speaking, ecovillages are a form of residential community where inhabitants are motivated by eco- and social-centric values to grow place-based human ecosystems; these renewed eco-cultural lifeways provide experimental alternatives to the trajectory of ecocide and social crisis un-reflexively pursued by most modern societies. They participate in regeneration and adaptation of biodiversity; they practice various forms of democratic decision-making and inclusive social practices; and, they innovate forms of economic exchange whose polyvalent metrics measure success differently than do neoliberal economists. The chapter begins by providing background information and context for the emergence of ecovillages and key identifying practices and goals of ecovillages, and then proceeds to provide a case study example of an off-the grid ecovillage located in the southern Appalachian mountains in North Carolina, U.S.: Earthaven Ecovillage. The mission of Earthaven is ‘to create a village which is a living laboratory and educational seed bank for a sustainable human future’. The chapter includes insights from an Earthaven resident who offers insights into how ecovillagers at Earthaven work on village building, land-based livelihoods, small-scale agro-ecological carbon farming, place-based economics, wisdom traditions, permaculture, renewable energy, and radical responsibility. They share how the goal of this work is to help Earthaven residents and visitors attempt to develop lifeways that indigenise humans and gather a cultural seed bank for living more sustainably on a hot and full Earth.
