Background: Myelin oligodendrocyte glycoprotein (MOG) may be implicated in the immunopathogenesis of multiple sclerosis (MS) inducing demyelination in the animal model of MS. In adults reported anti-MOG antibody frequencies have been variable across a number of studies and can also be detected in controls.Objective: To measure antibodies against MOG in paediatric patients with demyelinating disorders of the central nervous system and in controls.Methods: Serum antibodies against MOG and myelin basic protein were measured by ELISA, flow cytometry (FACS) and in the liquid phase in 11 children with acute disseminated encephalomyelitis (ADEM), 22 children with MS, seven children with acute viral encephalitis and 13 healthy controls. The serostatus of Epstein—Barr virus (EBV) infections were assessed.Results: Anti-MOG antibodies, measured either by ELISA or FACS were exclusively detected in children with demyelination. In ADEM these antibodies were highly reactive. Anti-MBP reactivity was detectable equally in all groups. The presence of either autoantibodies did not associate with EBV serostatus, age, gender or disease course.Conclusions: This study independently corroborates recently published results of seroprevalence and specificity of the assay. Due to their low sensitivity anti-MOG antibodies will not serve as disease-specific biomarkers, but could help to support the diagnosis of ADEM in difficult cases. [ABSTRACT FROM PUBLISHER]

The aim of this paper is to investigate the interface between the formal and informal support provided to very old people against a background of increasing need for care and a decreasing number of potential informal caregivers. We used a sample of 323 community-dwelling octogenarians participating in the Swiss Interdisciplinary Longitudinal Study on the Oldest Old (SWILSOO) (n=1441 interviews). Descriptive analyses and a multilevel modelwere used to testwhether formal and informal services complemented or substituted one another. The study revealed that the amount of informal services increased significantly as the frequency of formal aid increased, indicating that the two networks were complementary in the majority of the cases. In 21.2% of the cases, the formal network partly substituted the informal network (as an adjustment) and only in 6.4% of the cases did the informal support end after the formal support had increased (radical substitution). The concern that the introduction of formal services may curb the readiness of relatives and friends to provide care is thus unfounded. [ABSTRACT FROM AUTHOR]

Abstract: Conformity is a type of social learning that has received considerable attention among social psychologists and human evolutionary ecologists, but existing empirical research does not identify conformity cleanly. Conformity is more than just a tendency to follow the majority; it involves an exaggerated tendency to follow the majority. The “exaggerated” part of this definition ensures that conformists do not show just any bias toward the majority, but a bias sufficiently strong to increase the size of the majority through time. This definition of conformity is compelling because it is the only form of frequency-dependent social influence that produces behaviorally homogeneous social groups. We conducted an experiment to see if players were conformists by separating individual and social learners. Players chose between two technologies repeatedly. Payoffs were random, but one technology had a higher expected payoff. Individual learners knew their realized payoffs after each choice, while social learners only knew the distribution of choices among individual learners. A subset of social learners behaved according to a classic model of conformity. The remaining social learners did not respond to frequency information. They were neither conformists nor nonconformists, but mavericks. Given this heterogeneity in learning strategies, a tendency to conform increased earnings dramatically. [Copyright &y& Elsevier]

Background: Galactocerebroside, the major glycolipid of central nervous system myelin, is a known target for pathogenic demyelinating antibody responses in experimental allergic encephalomyelitis (EAE), the animal model of multiple sclerosis (MS). Objective: To address the importance of anti-galactocerebroside (α-GalC) antibodies in MS and to evaluate them as biomarkers of disease. Methods: α-GalC IgGs were quantified from sera of patients with MS and in marmoset EAE by a new immunosorbent assay. Results: We report a significant difference in serum α-GalC IgG titers between patients with relapsing-remitting (RR)–MS and healthy controls (HCs; P < .001). The frequencies of α-GalC antibody-positive subjects (α-GalC titers ≥ mean HC titers+3 SD) are also significantly elevated in RR-MS compared with HC (40% vs 0%; P =.0033). Immunoaffinity purified α-GalC IgGs from human serum bind to cultured human oligodendrocytes, indicating that the ELISA detects a biologically relevant epitope. Corroborating these findings, α-GalC antibody responses in marmoset EAE were similarly found to be specifically associated with the RR forms and not the peracute or progressive forms, in contrast with other anti-myelin antibodies (P =.0256). Conclusion: (1) α-GalC antibodies appear MS-specific and are not found in healthy subjects, unlike antibodies against myelin proteins; (2) when present, α-GalC antibodies identify mostly RR-MS and may be an indicator of ongoing disease activity. • Safety data on rabies vaccination in persons with multiple sclerosis (MS) are lacking. • It is unknown whether MS patients have an increased for relapse after RV. • A self-controlled retrospective cohort of 55 MS patients was followed in the year before and the year after prophylactic RV. • There were fewer relapses in the year after vaccination, including the three-month post-vaccination exposure period, than in the year before. • Rabies vaccination was not associated with MS relapse. No studies assessing rabies vaccine (RV) tolerability in persons with multiple sclerosis (MS) have been conducted. Given the lack of safety data, RV is recommended essentially only for post-exposure prophylaxis, which is difficult to administer effectively in many rabies-endemic countries. We sought to determine whether RV administration as pre-exposure prophylaxis was associated with MS relapse. This retrospective cohort study compared the clinical courses of MS patients in the year before and after rabies vaccination. The year before vaccination was defined as the pre-exposure risk period, the three months thereafter as the exposure-risk period, and the following nine months as the post-risk period. All adult MS patients immunized with RV between 2014 and 2018 and with available medical records in the two-year window were included. The primary outcome was the incidence of symptomatic MS relapse in the exposure-risk period versus the pre-exposure period. Fifty-five patients received at least one dose of RV. Most (38/55, 69%) were female; mean age was 38.5 years (SD ±9.2). While 21 (38%) patients experienced 24 relapses in the year before vaccination, only three (5%) experienced one relapse each in the post-vaccination exposure-risk period; three others (5%) experienced a total of four relapses in the subsequent post-risk period. The annualized relapse rates in the pre-exposure, exposure-risk, and post-risk periods were 0.44, 0.22, and 0.10, respectively (rate ratio for exposure-risk to pre-exposure periods, 0.509 [95% CI 0.098–1.677]). In this cohort, rabies vaccination was not associated with clinical MS relapse. Larger, prospective studies are needed to confirm these results. [ABSTRACT FROM AUTHOR]
