Abstract

Colorectal cancer (CRC) accounts for over 600 000 deaths annually worldwide. The current study aims to evaluate the value of proto‐oncogene PIM1 as a therapeutic target in CRC and investigate the anticancer activity of hispidulin, a naturally occurring phenolic flavonoid compound, against CRC. Immunohistochemistry analysis showed that PIM1 was upregulated in CRC tissue. The role of PIM1 as an oncogene was evidenced by the fact that PIM1 knockdown inhibits cell growth, induces apoptosis, and suppresses invasion. Our results showed that hispidulin exerts antitumor activity in CRC through inhibiting the expression of PIM1. Moreover, our findings revealed that hispidulin downregulated the expression of PIM1 by inhibiting JAK2/STAT3 signaling by generating reactive oxygen species. Furthermore, our in vivo studies showed that hispidulin can significantly inhibit tumor growth and metastasis in CRC. Collectively, our results provide an experimental basis for trialing hispidulin in CRC treatment. PIM1 can be considered a potential therapeutic target in CRC.

Abstract

We determined the spatial distribution pattern, source, risk level and influencing factor of heavy metal pollution in a typical high-tech industrial development zone of Changsha, China. A total of 123 topsoil samples were excavated from 11 different land-uses. Quite strong pollution (Nemerow) occurred in the whole zone. The Igeo pollution levels were from slight to strong for these elements. Four primary components were extracted from PCA with 79.36% of total variance explained. It could be deduced from the primary components that Cu, Co, Ni normally resulted from electronic industry; As and Cr were affected by both industrial and natural source; Pb and Zn were originated from traffic activities; and Cd was correlated with the activities of General equipment manufacturing industry and pharmaceutical manufacturing industry. The concentrations of Pb and Zn decreased from densely populated areas to suburbs, while the high concentrations of the other six elements were mainly found in the industrial lands. Heavy metal concentration was in weak relationship with green cover ratio (GCR) and built time, inferring that increase of GCR might not be contributed to improvement of the heavy metal pollution. Abstract

Context adaptive binary arithmetic coding (CABAC) is the entropy coding tool applied in the latest video coding standard, High Efficiency Video Coding (H.265/HEVC). CABAC achieves high coding efficiency but involves the bin-to-bin data dependency of Binary Arithmetic Encoder (BAE). This paper proposes a range updating structure with pre-bitwise-NOT (PBN) operation to shorten the critical paths in the range updating stage of the BAE, and also proposes a BAE architecture with optimized Lookahead rLPS. Synthesized results in SMIC 40nm show that the proposed architecture can operate at 436.7MHz and the throughput is 1777Mbin/s.

159. Perspectives on Cu–Ag Bimetallic Catalysts for Electrochemical CO2Reduction Reaction: A Mini-Review

Abstract

The atmospheric CO2concentration has increased significantly since the Industrial Revolution, leading to various environmental issues and energy crisis. The electrochemical reduction of CO2powered by renewable energy is attracting widespread attention, due to its capacity to transform CO2into value-added chemicals and fuels and contribute to carbon neutrality. Among Cu-based catalysts, Cu–Ag bimetallic catalysts exhibit superior selectivity for high value-added multicarbon compounds. This mini-review provides an overview of Cu–Ag bimetallic catalysts including alloy system and composite structure system, emphasizing the relationship between catalysts’ structure and the influencing factors for the enhanced selectivity toward C2+. At the end of this review, we outline the challenges and perspectives for the better development of electrochemical CO2reduction by Cu–Ag bimetallic catalysts.

Abstract

An atomically dispersed structure is attractive for electrochemically converting carbon dioxide (CO2) to fuels and feedstock due to its unique properties and activity. Most single-atom electrocatalysts are reported to reduce CO2to carbon monoxide (CO). Herein, we develop atomically dispersed indium (In) on a nitrogen-doped carbon skeleton (In–N–C) as an efficient catalyst to produce formic acid/formate in aqueous media, reaching a turnover frequency as high as 26771 h–1at −0.99 V relative to a reversible hydrogen electrode (RHE). Electrochemical measurements show that trace amounts of In loaded on the carbon matrix significantly improve the electrocatalytic behavior for the CO2reduction reaction, outperforming conventional metallic In catalysts. Further experiments and density functional theory (DFT) calculations reveal that the formation of intermediate *OCHO on isolated In sites plays a pivotal role in the efficiency of the CO2-to-formate process, which has a lower energy barrier than that on metallic In.

Abstract

A novel poly(glycine) (p-GLY)/graphene oxide (GO) composite based sensor (p-GLY/GO) was successfully prepared on glassy carbon electrode by simple electropolymerization. Electrochemical responses of analytes on p-GLY/GO modified electrode were studied by cyclic voltammetry and differential pulse voltammetry. The results demonstrated that p-GLY/GO modified electrode showed a favorable application for the simultaneous determination of dopamine (DA), uric acid (UA), guanine (GU) and adenine (AD). Owing to the synergistic effect of p-GLY and GO, the oxidation peaks of four analytes separated well from each other, and the potential separations of oxidation peaks of DA-UA, UA-GU and GU-AD were large up to 170, 350 and 300 mV, respectively. As-prepared p-GLY/GO modified electrode offered wide linear responses for DA, UA, GU and AD over the ranges of 0.20–62, 0.10–105, 0.15–48 and 0.090–103 μM with detection limits of 0.011, 0.061, 0.026 and 0.030 μM (S/N = 3), respectively. Moreover, p-GLY/GO modified electrode presented favorable selectivity, stability and reproducibility, which was a promising candidate as an electrochemical sensor for the simultaneous determination of DA, UA, GU and AD. 184. Transcriptional mechanism of IRF8 and PU.1 governs microglial activation in neurodegenerative condition

Abstract

Microglial activation occurs in divergent neuropathological conditions. Such microglial event has the key involvement in the progression of CNS diseases. However, the transcriptional mechanism governing microglial activation remains poorly understood. Here, we investigate the microglial response to traumatic injury-induced neurodegeneration by the 3D fluorescence imaging technique. We show that transcription factors IRF8 and PU.1 are both indispensible for microglial activation, as their specific post-developmental deletion in microglia abolishes the process. Mechanistically, we reveal that IRF8 and PU.1 directly target the gene transcription of each other in a positive feedback to sustain their highly enhanced expression during microglial activation. Moreover, IRF8 and PU.1 dictate the microglial response by cooperatively acting through the composite IRF-ETS motifs that are specifically enriched on microglial activation-related genes. This action of cooperative transcription can be further verified biochemically by the synergetic binding of IRF8 and PU.1 proteins to the composite-motif DNA. Our study has therefore elucidated the central transcriptional mechanism of microglial activation in response to neurodegenerative condition.

Abstract

The corrosion and passivation behaviors of zinc electrodes in 7.0 M KOH saturated with ZnO in the presence of dodecyltrimethylammonium bromide (DTAB) surfactant were evaluated by Tafel, linear sweep voltammetry, electrochemical impedance spectroscopy (EIS) and field emission scanning electron microscopy (FE-SEM) tests. It was found that DTAB had a considerable effect on inhibiting zinc corrosion by the adsorption of DTAB. The adsorption of DTAB obeyed the Freundlich isotherm. Tafel polarization curves revealed that DTAB worked as an anodic-type inhibitor. The highest corrosion inhibition efficiency of DTAB was up to 80.2% in the presence of 0.07 wt% DTAB. In addition, potentials of both passivation and anodic dissolution current peak positively shifted in the presence of DTAB, indicating that the passivation was delayed. Based on the EIS tests and FE-SEM analysis in the presence and absence of DTAB, it was shown that the adsorption and favorable moisture retention of DTAB were beneficial to the formation of uniform and fluffy protective layer on the surface of zinc electrode. Such a morphological modification of zinc surface greatly improved the diffusion rate of ions and the utilization of zinc electrode. [ABSTRACT FROM AUTHOR]

Abstract

For increasing scalability and reducing cost, transition metal dichalcogenides-based electrocatalysts presently have been proposed as substitutes for noble metals to generate hydrogen, but these alternatives usually suffer from inferior performance. Here, a Ravenala leaf-like W xC@WS2 heterostructure is grown via carbonizing WS2 nanotubes, whose outer walls being partially unzipped along with the W x C 'leaf-valves' attached to the inner tubes during the carbonization process. This heterostructure exhibits a catalytic activity for hydrogen evolution reaction with low overpotential of 146 mV at 10 mA cm−2 and Tafel slope of 61 mV per decade, outperforming the performance of WS2 nanotubes and W xC counterparts under the same condition. Density functional theory calculations are performed to unravel the underlying mechanism, revealing that the charge distribution between W xC and WS2 plays a key role for promoting H atom adsorption and desorption kinetics simultaneously.
