The main scientific goal of LHAASO-WCDA is to survey gamma-ray sources with energy from 100 GeV to 30 TeV. To observe high-energy shower events, especially to measure the energy spectrum of cosmic rays from 100 TeV to 10 PeV, a dynamic range extension system (WCDA++) is designed to use a 1.5-inch PMT with a dynamic range of four orders of magnitude for each cell in WCDA-1. The dynamic range is extended by using these PMTs to measure the effective charge density in the core region of air shower events, which is an important parameter for identifying the composition of primary particles. The system has been running for more than one year. In this paper, the details of the design and performance of WCDA++ are presented.

The influence of soil variability on three-dimensional (3D) probabilistic slope stability analysis has been investigated previously for soils that display isotropic spatial variability features or anisotropic horizontal fabric patterns. However, due to various soil deposition processes, weathering, filling or tectonic movements, the assumptions of isotropy or horizontal layering may not always be realistic. This study presents 3D analyses of slopes with spatially variable soils associated with rotated transverse anisotropy features. The results show that for cross-dip slopes where the strike direction of soil strata is perpendicular to the out-of-plane direction of the slope, the reliability depends on various factors including strata rotation angle and autocorrelation distances, and differs significantly from slopes with horizontally deposited soil fabric. The influence of strata orientation is also pronounced for dip slopes and reverse dip slopes, and these are presented in terms of reliability indices of the slopes and statistics of the length of sliding mass and elaborated by considering the failure mechanism under different scenarios. Through these analyses, this paper discusses the key features of slope reliability considering rotated transverse anisotropy in soil properties, and their major differences from situations involving horizontal soil layers or two-dimensional probabilistic assessments.

GWAC will have been built an integrated FOV of 5,000 degree2 and have already built 1,800 square degree2. The limit magnitude of a 10-second exposure image in the moonless night is 16R. In each observation night, GWAC produces about 0.7TB of raw data, and the data processing pipeline generates millions of single frame alerts. We describe the GWAC Data Processing and Management System (GPMS), including hardware architecture, database, detection-filtering-validation of transient candidates, data archiving, and user interfaces for the check of transient and the monitor of the system. GPMS combines general technology and software in astronomy and computer field, and use some advanced technologies such as deep learning. Practical results show that GPMS can fully meet the scientific data processing requirement of GWAC. It can online accomplish the detection, filtering and validation of millions of transient candidates, and feedback the final results to the astronomer in real-time. During the observation from October of 2018 to December of 2019, we have already found 102 transients.

Defunctioning stoma is an effective method to reduce symptomatic anastomotic leakage after rectal cancer surgery. It is of concern that about 1 in 5 defunctioning stomas will not be restored, that is, becoming permanent. And that is usually beyond expectation by physicians and patients, which deserves enough attention. The causes are complex, including anastomotic complications, tumor progression, perioperative death, poor anal function and patient willingness. Possible risk factors include symptomatic anastomotic leakage, age, tumor location, neoadjuvant therapy, anal function, TNM stage, ASA score, hospital factors, etc. Those factors may occur in various stages of patient referral such as before neoadjuvant therapy, prior to surgery, intra or post-operative period, and follow-up. Adequate physician-patient communication and shared decision-making, comprehensive tumor and patient function assessment, rational treatment strategy, careful manipulation during operation and good quality control, and meticulous perioperative management are important steps to reduce the permanent stoma. When shared decision-making, patients' needs should be fully considered while unnecessary expectations of anal preservation should be avoided. The risk of perpetuation of defunctioning stoma should be fully informed. Safe operation, especially anastomosis, is the key to avoid permanent stoma. Triplet regimens based on pomalidomide and dexamethasone have been applied to treat relapsed/refractory multiple myeloma, but the safety and efficacy are not yet very clear. This meta-analysis aimed at comparing the safety and efficacy of different triplet therapies and analyzing the best therapy regimen.A comprehensive literature search identified a total of 615 studies, and 22 studies assessing 1,889 subjects met the inclusion criteria of this meta: phase II/III trial, over 2 median lines of prior therapy, and detailed efficacy outcomes like overall response rate (ORR), overall survival, and progression-free survival (PFS). All statistical analyses were performed by Revman version 5.3, and the heterogeneity was tested by I2 (25% indicating low heterogeneity, 50% moderate, and 75% high). For those with less heterogeneity, fixed-effect model was used. With a significant high heterogeneity, a random-effect model was used.Pooled analysis showed ORR 66.2% across all triplet regimens based on pomalidomide and dexamethasone. Among all triplet regimens, therapy containing bortezomib showed the highest ORR (90.3%), and the one containing elotuzumab showed the lowest ORR (41.2%). The pooled ORRs for the remaining treatment regimens are as follows: cyclophosphamide (70.1%), isatuximab (66.3%), daratumumab (61.2%), clarithromycin (60.0%), pembrolizumab (47.3%). A total of 21 adverse events appeared in the included studies, with neutropenia being the highest incidence of hematologic adverse events (32.1%) and cough being the highest incidence of non-hematologic adverse events (43.3.%).Three-drug regimens based on pomalidomide and dexamethasone could yield excellent overall response rate to relapsed/refractory multiple myeloma, but there are still various adverse events; therefore, consequent studies should address these adverse events.

This study explored the usefulness of genomic copy number variation sequencing (CNV-Seq) in the prenatal diagnosis of pregnant women.Based on prenatal diagnostic indications, CNV-Seq analysis was done in the samples from the 579 pregnant women of the 7 subgroups that included advanced maternal age (group A), high risk noninvasive prenatal test (NIPT) (group B), high risk Down's (Group C), abnormal ultrasound findings (Group D), adverse pregnancy history (Group E), chromosome abnormalities in couples (Group F), and the mixed group (Group G).A total of 57 (9.84%) cases have abnormal CNV-Seq results. Among them, 21 cases were aneuploid chromosomal number abnormalities (3.63%, 21/579), and 36 cases were CNV abnormalities (6.22%, 36/579), including 7 cases of pathogenic copy number alteration (pCNA) (1.21%, 7/579) and 29 cases variants of uncertain significance (VUS) (5.01%, 29/579). The total detection rates of abnormal CNV-Seq in Group G and Group B were 20.27% (15/74) and 15.91% (14/88), which were significantly higher than those in other groups (p0.05). Among 36 cases of abnormal CNV-Seq, 7 cases were chromosome fragment deletion or duplication, which were pathogenic CNV, and some rare chromosomal diseases were detected.Patients with a high risk of NIPT or multiple indications of prenatal diagnosis are highly suspected of chromosomal diseases. CNV-Seq is a useful tool for detecting chromosome abnormalities for prenatal diagnosis of pregnant women more accurately and provides more comprehensive information for prenatal diagnosis to reduce birth defects.

Pedestrian dead reckoning (PDR) can solve the position information by its inertial measurement unit (IMU), which is a method to achieve indoor autonomous positioning without deploying positioning base stations in advance. However, affected by the accumulated error, the positioning accuracy will decrease with the increase of the moving distance. To improve the indoor positioning accuracy, the step length estimation of PDR is improved. A PDR method combining the information on walking frequency, acceleration variance, and fixed step is proposed. Firstly, the pedestrian's walking steps are detected and estimated by the threshold peak method; secondly, the step frequency and acceleration variance are calculated, and the step length estimation model is fitted with the fixed step length of the pedestrian to estimate the step length of each step; then, the fusion algorithm of magnetometer, accelerometer, and gyroscope are used to estimate the heading; finally, combined with the step number, step length, and heading angle, the position information of walking is calculated by dead reckoning. The experimental results show that in the practical distance of 42.6 m, the accuracy of dead reckoning distance based on the combination of step frequency, acceleration variance, and fixed step length can reach 98.47%, which is better than the fixed step length and linear step length version, 5.09% higher than the linear step length and 0.35% higher than the fixed step length version.

In Surface Relations Vivian L. Following inscrutability in literature, visual culture, and performance art since 1965, Huang articulates how Asian American artists take up the aesthetics of Asian inscrutability—such as invisibility, silence, unreliability, flatness, and withholding—to express Asian American life. Through analyses of diverse works by performance artists (Tehching Hsieh, Baseera Khan, Emma Sulkowicz, Tseng Kwong Chi), writers (Kim Fu, Kai Cheng Thom, Monique Truong), and video, multimedia, and conceptual artists (Laurel Nakadate, Yoko Ono, Mika Tajima), Huang challenges neoliberal narratives of assimilation that erase Asianness. Nitric oxide (NO) is a well-known gaseous mediator that maintains vascular homeostasis. Extensive evidence supports that a hallmark of endothelial dysfunction, which leads to cardiovascular diseases, is endothelial NO deficiency. Thus, restoring endothelial NO represents a promising approach to treating cardiovascular complications. Despite many therapeutic agents having been shown to augment NO bioavailability under various pathological conditions, success in resulting clinical trials has remained elusive. There is solid evidence of diverse beneficial effects of the treatment with low-power near-infrared (NIR) light, defined as photobiomodulation (PBM). Although the precise mechanisms of action of PBM are still elusive, recent studies consistently report that PBM improves endothelial dysfunction via increasing bioavailable NO in a dose-dependent manner and open a feasible path to the use of PBM for treating cardiovascular diseases via augmenting NO bioavailability. In particular, the use of NIR light in the NIR-II window (1000-1700 nm) for PBM, which has reduced scattering and minimal tissue absorption with the largest penetration depth, is emerging as a promising therapy. In this review, we update recent findings on PBM and NO.

Indonesia is one of the top three countries in the world for fisher fatalities due to a lack of stability monitoring, poor vessel design, and the fishers’ poor safety culture. Improved safety through technology, in particular stability awareness, has been identified as a key aspect to prevent capsizing and sinking. Despite the low technology uptake by Indonesian fishers, they are known to make extensive use of smart mobile phones as a primary form of communication. These phones are equipped with various sensors, including motion sensors that can be used by different mobile applications. This paper proposes the use of a mobile application to aid Indonesian fishers in assessing the stability of their vessel by using results from a roll decay test or realtime roll motion data monitored when operating at sea. For the pre-departure test, the mobile application uses the onboard gyroscope to record the roll angle time series which can then be used to quantify the roll period. An offline calculation is then carried out to approximate the vessel’s metacentric height. This is then visually presented using a traffic light system to provide the fishermen with a simple indication of whether the vessel is stable or not. Should the boat be unstable, additional guidance is provided to improve the vessel’s stability. The realtime monitoring system alerts the fishers when the thresholds are exceeded, to ensure realtime awareness of increased risk to the stability of the vessel.

Rehabilitation of disabled children with autism has become a challenge for current rehabilitation centres. This study conducted psychological investigations on disabled children and analysed the symptoms and characteristics of autism in these children to develop more reasonable rehabilitation treatment plans that would help the children receive psychological counselling and effective rehabilitation.This study investigated 107 disabled children from the Disabled Rehabilitation Research Centre of the South China Minority Autonomous Region. Using the PEP-3 scale as a research tool, a questionnaire was developed to investigate and collect data on the mental health of disabled children. The survey was conducted from 2017 to 2021, and 107 children's mental health data were collected in the form of questionnaires based on PEP-3 evaluation indicators. After cleaning the data, the questionnaire data were screened and processed. Descriptive statistical and correlation analysis tools were used for model analysis to understand the overall data distribution and the potential relationships among various data variables.The results of correlation analysis showed that cognition, language expression, language understanding, emotion, and social interaction in the subtest of developmental behaviour were the main indicators of the degree of autism in children. These indicators had a strong and significant correlation with the comprehensive score. Moreover, these indicators had a significant correlation with the individual self-care and adaptive behaviours reported by the children's caregivers. Small muscles, big muscles, and imitation (vision and movement) indicators had a significant correlation with problematic behaviours and physical fitness, and language and cognitive indicators also had a strong correlation with emotion and social interaction.Emphasis should be placed on the improvement of the language and cognitive abilities of disabled children with autism, and corresponding rehabilitation plans, and training can be formulated according to children with different degrees of illness to get a better rehabilitation outcome. Further, identification of key indicators of autism will be of help in aiding the development of rehabilitation treatment for disabled children with autism and formulation of long-term rehabilitation plans.

This is the second editorial commentary in a three-part series that addresses introductions, implications, and interestingness. The first, focusing on the introduction design, is about what the beginning of a paper should look like. The second, focusing on practice implications, is about what the end of a paper should look like. The third discusses how to develop and fit attention-grabbing ideas into the academic conversation in the literature. We hope this editorial series provides inspiration and ideas about publishing papers that people want to engage with.

The inaccuracy of anthropogenic emission inventories on a high-resolution scale due to insufficient basic data is one of the major reasons for the deviation between air quality model and observation results. A bottom-up approach, which is a typical emission inventory estimation method, requires a lot of human labor and material resources, whereas a top-down approach focuses on individual pollutants that can be measured directly as well as relying heavily on traditional numerical modeling. Lately, the deep neural network approach has achieved rapid development due to its high efficiency and nonlinear expression ability. In this study, we proposed a novel method to model the dual relationship between an emission inventory and pollution concentrations for emission inventory estimation. Specifically, we utilized a neural-network-based comprehensive chemical transport model (NN-CTM) to explore the complex correlation between emission and air pollution. We further updated the emission inventory based on back-propagating the gradient of the loss function measuring the deviation between NN-CTM and observations from surface monitors. We first mimicked the CTM model with neural networks (NNs) and achieved a relatively good representation of the CTM, with similarity reaching 95 %. To reduce the gap between the CTM and observations, the NN model suggests updated emissions of NOx, NH3, SO2, volatile organic compounds (VOCs) and primary PM2.5 changing, on average, by −1.34 %, −2.65 %, −11.66 %, −19.19 % and 3.51 %, respectively, in China for 2015. Such ratios of NOx and PM2.5 are even higher (∼ 10 %) in regions that suffer from large uncertainties in original emissions, such as Northwest China. The updated emission inventory can improve model performance and make it closer to observations. The mean absolute error for NO2, SO2, O3 and PM2.5 concentrations are reduced significantly (by about 10 %–20 %), indicating the high feasibility of NN-CTM in terms of significantly improving both the accuracy of the emission inventory and the performance of the air quality model.

The presence of clustered data is common in the sociobehavioral sciences. One approach that specifically deals with clustered data but has seen little use in education is the generalized estimating equations (GEEs) approach. We provide a background on GEEs, discuss why it is appropriate for the analysis of clustered data, and provide worked examples using both continuous and binary outcomes. Comparisons are made between GEEs, multilevel models, and ordinary least squares results to highlight similarities and differences between the approaches. Detailed walkthroughs are provided using both R and SPSS Version 26.

Quasi-periodic (QP) solutions of a damped nonlinear QP Mathieu equation with cubic nonlinearity are investigated by using the incremental harmonic balance (IHB) method with two time scales. The damped nonlinear QP Mathieu equation contains two incommensurate harmonic excitation frequencies, one is a small frequency while the other nearly equals twice the linear natural frequency. It is found that Fourier spectra of QP solutions of the equation consist of uniformly spaced sidebands due to cubic nonlinearity. The IHB method with two time scales, which relates to the two excitation frequencies, is adopted to trace solution curves of the equation in an automatical way and find all frequencies of solutions and their corresponding amplitudes. Effects of parametric excitation are studied in detail. Based on approximation of QP solutions by periodic solutions with a large period, Floquet theory is used to study the stability of QP solutions. Three types of QP solutions can be obtained from the IHB method, which agree very well with results from numerical integration. However, the perturbation method using the double-step method of multiple scales (MMS) obtains only one type of QP solutions since the ratio of the small frequency to the linear natural frequency of the first reduced-modulation equation is nearly 1 in the second perturbation procedure, while the other two types of QP solutions from the IHB method with two time scales do not need the ratio. Furthermore, the results from the double-step MMS are different from those numerical integration and the IHB method with two time scales.

Autophagy is a lysosomal mediated catabolic process that helps maintain cell balance and survival under extracellular or intracellular stress by degrading different cytoplasmic components. Autophagy is also a significant way for the body to defend pathogen invasion, which is swallowed by the phagosomes of host cells. When the phagosomes mature, they form autophagosomes, which are fused with lysosomes to form autophagolysosomes. Autophagolysosomes degrade due to the action of various hydrolases and remove pathogens at the same time. By interfering with the autophagy level of host cells, pulmonary atypical pathogens can escape from the host cells and be cleared by autophagy, and reproduce in host cells, finally reaching the goal of infecting the host. Human milk oligosaccharides (HMOs) are a family of unconjugated soluble glycans found in human breast milk that exhibit a myriad of biological activity. While recent studies have uncovered numerous biological functions for HMOs (antimicrobial, anti-inflammatoryprobiotic properties), the receptors and protein binding partners involved in these processes are not well characterized. This can be attributed largely in part to the low affinity and transient nature of soluble glycan-protein interactions, precluding the use of traditional characterization techniques to survey binding partners in live cells. Here, we present the use of synthetic photoactivatable HMO probes to capture, enrich and identify HMO protein targets in live cells using mass spectrometry-based chemoproteomics. Following initial validation studies using purified lectins, we profiled the targets of HMO probes in live mouse macrophages. Using this strategy, we mapped hundreds of HMO binding partners across multiple cellular compartments, including many known glycan-binding proteins as well as numerous proteins previously not known to bind glycans. We expect our findings to inform future investigations of the diverse roles of how HMOs may regulate protein function.

Here we present the first total and linearly polarized intensity images of this source obtained with the unprecedented 20 μas resolution of the EHT. J1924–2914 is a very compact flat-spectrum radio source with strong optical variability and polarization. In April 2017 the source was observed quasi-simultaneously with the EHT (April 5–11), the Global Millimeter VLBI Array (April 3), and the Very Long Baseline Array (April 28), giving a novel view of the source at four observing frequencies, 230, 86, 8.7, and 2.3 GHz. These observations probe jet properties from the subparsec to 100 pc scales. We combine the multifrequency images of J1924–2914 to study the source morphology. We find that the jet exhibits a characteristic bending, with a gradual clockwise rotation of the jet projected position angle of about 90° between 2.3 and 230 GHz. Linearly polarized intensity images of J1924–2914 with the extremely fine resolution of the EHT provide evidence for ordered toroidal magnetic fields in the blazar compact core. © 2022. There is solid evidence of the beneficial effect of photobiomodulation (PBM) with low-power near-infrared (NIR) light in the NIR-I window in increasing bioavailable nitric oxide (NO). However, it is not established whether this effect can be extended to NIR-II light, limiting broader applications of this therapeutic modality. Since we have demonstrated PBM with NIR laser in the NIR-II window, we determined the causal relationship between NIR-II irradiation and its specific biological effects on NO bioavailability. We analyzed the impact of NIR-II irradiation on NO release in cultured human endothelial cells using a NO-sensitive fluorescence probe and single-cell live imaging. SUMMARYPancreatic islets contain several endocrine cell types that coordinate to maintain blood glucose homeostasis. While β and α cells are thought to be the main drivers of glucose homeostasis through insulin and glucagon secretion respectively, the contribution of δ cells and somatostatin (SST) secretion to establishing the glycemic set point remains unresolved. Here we remove local SST signaling from δ cells within the pancreatic islet to investigate their contribution to the glycemic set point. Our data demonstrate that ablating δ cells or SST leads to a sustained decrease in the glycemic set point. This coincides with a decreased glucose threshold for insulin response from β cells, leading to increased insulin secretion to the same glucose challenge. In contrast, α cell ablation had no effect on glycemic set point. Collectively, these data establish the physiological role of δ cells in determining the glycemic set point through their interaction with β cells.
